I investigated whether snake venom toxin(SVT) from Vipera lebetina turanica enhances the apoptosis ability of tumor necrosis factor(TNF)-related apoptosis-inducing ligand(TRAIL) in cancer cells. TRAIL inhibited HCT116 cell growth in a dose-dependent manner. Consistent with cell growth inhibition, the expression of TRAIL receptors; DR4 and DR5 was significantly increased as well as apoptosis related proteins such as cleaved caspase-3, 8, 9 and Bax. However, the expression of survival proteins(eg, cFLIP, survivin, XIAP and Bcl2) was suppressed by the combination treatment of SVT and TRAIL. Pretreatment with the reactive oxygen species(ROS) scavenger N-acetylcysteine reduced the SVT and TRAIL-induced upregulation of DR4 and DR5 expression and expression of the apoptosis related protein such as caspase-3 and-9 as well as cell growth inhibitory effects. The collective results suggest that SVT facilitates TRAIL-induced apoptosis in human colorectal cancer HCT116 cells through up-regulation of the TRAIL receptors; DR4 and DR5 via ROS pathway signals.

A new method is proposed for water resources using aquifers in coastal areas. These aquifers are generally filled with saline water due to seawater intrusion and consequently being left unutilized. Surface water can be injected into these aquifers and recovered for water quality enhancement and stored water. Injection and pumping wells are used. For this technique to be successful protection of pumping well from seawater intrusion is an essential issue. Salt water pumping can be used to prevent saline water upconing. Numerical analysis demonstrated that a properly designed and executed salt water pumping well can protect a freshwater pumping well from salt water intrusion.
This study was designed as a part of efforts to investigate the biochemical pollutant markers for diagnosis of marine pollutions by changes in oxygen radicals and their scavenger enzymes of the flounder (Paralichthys olivaceus) in the South Sea of Korea. Protein contents in brain and muscle of cultured flounder in the South Sea were remarkably lower ($19\~42\%$ and $25\~41\%$, respectively) than those of wild flounder in Pohang of the East Sea as a control group. Lipid Peroxide (LPO) levels in serum of cultured in the South Sea were considerably higher ($5\~33\%$) Elan those of wild flounder in Pohang. Hydroxyl radical formations in serum of cultured flounders in the South Sea were lower ($4\~25\%$) than those of wild flounders in Pohang. Superoxide dismutase (SOD) activities in serum of cultured flounders in South Sea were also significantly lower ($7\~31\%$) than those of wild flounders in Pohang. It is believed that significant decrease of protein contents in brain and muscle, and remarkable increases of LPO in serum of cultured flounders may be used as a biochemical pollutant markers for diagnosis of marine pollutions. These results suggest that near-coastal water as well as neritic water of the South Sea might be affected by pollutant input as well as the Yellow Sea of Korea.

Pyungwi-san(PWS) have been using as a basic prescription of digestive disorder in Korean traditional medicine. This study was performed to examine the in vitro antioxidant activity of the extract using different antioxidant tests including by 1,1-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging, superoxide anion radical scavenging, metal chelating hydrogen peroxide scavenging, lipid peroxydation protective effect and scavenging effect of nitric oxide and peroxynitrite. Herbal-acupuncture solution of PWS(PWS-HS) exhibited a concentration-dependent inhibition of DPPH radical adduct formation and it showed dose-dependent free radical scavenging activity onto superoxide anions. In addition, the result of metal chelating hydrogen peroxide scavenging and ammonium thiocyanate experiments showed that PWS-HS was an active scavenger of hydroxyl radicals. Furthermore, it was also found to be effective in scavenging nitric oxide and peroxynitrite, well-known cytotoxic species that can oxidize several cellular components such as proteins, lipids and DNA.
Batch tests were carried out to evaluate the thermal treatment of low volatile organic compounds in low-permeability soil. The chemical oxidation by sodium persulfate catalyzed by heat and Fe (II) was evaluated. Enhanced persulfate oxidation of n-decane (C-10), n-dodecane (C-12), n-tetradecane (C-14), n-hexadecane (C-16), and phenanthrene was observed with thermal catalyst, indicating increased sulfate radical production. Slight enhancement of the pollutants oxidation was observed when initial sodium persulfate concentration increased from 5 to 50 g/L. However, the removal efficiency greatly decreased as soil/water ratio increased. It indicates that mass transfer of the pollutants as well as the contact between the pollutants and sulfate radical were inhibited in the presence of solids. In addition, more pollutants can be adsorbed on soil particles and soil oxidant demand increased when soil/water ratio becomes higher. The oxidation of the pollutants was significantly improved when catalyzed by Fe(II). The sodium persulfate consumption increased at the same time because the residual Fe(II) acts as the sulfate radical scavenger.
An experimental study was presented for extinguishing characteristics of liquid fuel fire by water mist($Dv_{0.99}{\leq}200{\mu}m$) containing potassium acetate and sodium acetate trihydrate. To evaluate the extinguishing performance of water mist containing additives, the evaporation characteristics of a water droplet on a heated surface was examined. The evaporation process was recorded by a charge-coupled-device camera. Also, small-scale extinguishing tests were conducted for n-heptane pool fire in ventilated space. During the experiments, flame temperatures were measured, and concentrations of oxygen and carbon monoxide were analyzed by a combustion gas analyzer. The average evaporation rate of water droplet containing additives was lower than that of pure water at a given surface temperature and decreased with the concentration increase due to the precipitation of salt in the liquid-film and change of surface tension. In case of using additives, the fire extinguishing times was shorter than that of pure water at a given discharge pressure and it was because the momentum of a water droplet containing additives was increased. And also dissociated metal atoms, potassium or sodium, were reacted as a scavenger of the major radical species OH, H which were generated for combustion process. Moreover, at a high pressure of 4 MPa, the fire was extinguished through blowing effect as well as primary extinguishing mechanisms.
Exposure of isolated intact mitochondria to near UV to visible light resulted in not only loss of respiration, the most well-documented phenomenon regarding phototoxic effects in the respiring organelles, but also lipid peroxidation of membranes and mitochondrial swelling; these turned out to be O$_2$-dependent and thus prevented by anaerobiosis, enhanced by a partial deuteration of the suspension medium, and suppressed by the presence of a singlet oxygen ($^1O_2$) scavenger. Measurements of the spectral dependence of such detrimental effects of light on mitochondrial structure and function revealed that all the resulting spectra bear a significant resemblance to the action spectrum for photogeneration of $^1O_2$ from mitochondrial membranes, which in turn carries the spectral characteristics of light absorption by mitochondrial Fe-S centers. Futhermore, destructing the Fe-S centers by a mercurial treatment of mitochondria brought about a striking reduction of the light-induced membrane peroxidation and swelling of mitochondria. These results are consistent with the suggestion that the impairment of functional, structural integrity of mitochondria caused by strong irradiation is directly related to the production of $^1O_2$ in mitochondria, photosensitized by the Fe-S centers. This paper also presents kinetic data which indicate that, among various membrane-bound protein systems associated with mitochondrial energy metabolism, the respiratory chain is the primary target for photodamage.
The scavenging effects of two hyaroxycinnamic acids such as caffeic acid and chlorogenic acid on paraquat induced pulmonary toxicity were investigated. The results are summerized as follows: 1. In the 5-lipoxygenase assay, caffeic acid and chlorogenic acid inhibited the enzyme activities whose inhibition concentration (IC$\_$50/) were 4.1 and 9.6 ${\mu}$M respectively. 2. To evaluate the antiinflammatory effects on mediator related to the mechanism of inflammation, ADP-induced platelet aggregation assay and histamine degranulation assay were used. Caffeic acid and chlorogenic acid inhibited on ADP-induced platelet aggregation and histamine release at a concentration dependent manners. 3. Arachidonic acid-induced ear edema were inhibited by administration of caffeic acid and chlorogenic acid. 4. Cytologicad analysis of branchoalveolar lavage fluid (BALF) which was the useful tool for detection of an inflammatory response in the lungs of animals intoxicated with chemicals were used. Alveolar macrophages and neutrophils in BALF, as well as the protein content and the LDH activity in BALF supernatant increased by intoxication of paraquat, but decreased by administration of caffeic acid and chlorogenic acid. Therefore, two hydroxyeinnamic acids tested were the useful candidates for scavenger and antiinflammatory agents on paraquat induced pulmonary toxicity.

Nicotinamide (NAM) has been shown to suppress reactive oxygen species (ROS) production in primary human fibroblasts, thereby extending their replicative lifespan when added to the medium during long-term cultivation. Based on this finding, NAM is hypothesized to affect cellular senescence progression by keeping ROS accumulation low. In the current study, we asked whether NAM is indeed able to reduce ROS levels and senescence phenotypes in cells undergoing senescence progression and those already in senescence. We employed two different cellular models: MCF-7 cells undergoing senescence progression and human fibroblasts in a state of replicative senescence. In both models, NAM treatment substantially decreased ROS levels. In addition, NAM attenuated the expression of the assessed senescence phenotypes, excluding irreversible growth arrest. N-acetyl cysteine, a potent ROS scavenger, did not have comparable effects in the tested cell types. These data show that NAM has potent antioxidative as well as anti-senescent effects. Moreover, these findings suggest that NAM can reduce cellular deterioration caused by oxidative damage in postmitotic cells in vivo.
Although sanguinarine, a benzophenanthridine alkaloid, possesses anti-cancer properties against several cancer cell lines, the molecular mechanisms by which it inhibits cell growth and induces apoptosis have not been clearly understood. In order to further explore the critical events leading to apoptosis in sanguinarine-treated MCF-7 human breast carcinoma cells, the following effects of sanguinarine on components of the mitochondrial apoptotic pathway were examined: generation of reactive oxygen species (ROS), alteration of the mitochondrial membrane potential (MMP), and the expression changes of Bcl-2 family proteins. We show that sanguinarine-induced apoptosis is accompanied by the generation of intracellular ROS and disruption of MMP as well as an increase in pro-apoptotic Bax expression and a decrease of anti-apoptotic Bcl-2 and Bcl-xL expression. The quenching of ROS generation with N-acetyl-L-cysteine, the ROS scavenger, protected the sanguinarine-elicited ROS generation, mitochondrial dysfunction, modulation of Bcl-2 family proteins, and apoptosis. Based on these results, we propose that the cellular ROS generation plays a pivotal role in the initiation of sanguinarine-triggered apoptotic death.
