Osteogenesis of Human Adipose Tissue Derived Mesenchymal Stem Cells (ATMSCs) Seeded in Bioceramic-Poly D,L-Lactic-co-Glycolic Acid (PLGA) Scaffold (Bioceramic-Poly D,L-Lactic-co-Glycolic Acid(PLGA) Scaffold에 접종한 인간지방조직-유래 중간엽 줄기세포의 골 형성)

  • The present experiment was performed to evaluate the osteogenic differentiation of human adipose tissue derived mesenchymal stem cells (ATMSCs) seeded in bioceramic-poly D,L-latic-co-glycolic acid (PLGA) scaffold. Osteogenic differentiation of ATMSCs were induced using the osteogenic induction (OI) medium. ATMSCs were cultured with OI medium during 28 days in well plate. The proliferation of ATMSCs in OI medium group was significantly increased for 14 days of plate culture but slowed after 21 days. On the other hand, proliferation in the control group showed constant increase for 28 days of culturing. The alkaline phosphatase (ALP) activity of ATMSCs in OI medium group increased during the 21 days of culture but decreased on 28 days. However, in control group ALP activity of ATMSCs was continuously decreased as time goes. Nodule was observed at 21 days of culture in OI medium group and confirmed accumulation of calcium in cell by alizarin red staining. ATMSCs were seeded in PLGA scaffold or in Bioceramic-PLGA scaffold, and cultured with OI medium. ALP activity of ATMSCs by osteoblast differentiation in each scaffold increased on 21 days of culture and decreased rapidly on 28 days. ALP activity of ATMSCs was increased highly in Bioceramic-PLGA scaffold compared to PLGA scaffold on 21 days of culturing. SEM-EDS analysis demonstrated that calcium and phosphate content and Ca/P ratio in Bioceramic-PLGA scaffold increased higher than in PLGA scaffold. Biodegradability of scaffold at 56 days after implantation showed that Bioceramic-PLGA scaffold was more biodegradable than PLGA scaffold. The results demonstrated that the differentiation of ATMSCs to osteoblast were more effective in scaffold culture than well plate culture. Bioceramic increased cell adhesion rate on scaffold and ALP activity by osteoblast differentiation. Also, bioceramic was considered to increase the calcium and phosphate in scaffold when ATMSCs was mineralized by osteogenic differentiation. Bioceramic-PLGA scaffold enhanced the osteogenesis of seeded ATMSCs compared to PLGA scaffold.

  • Scaffold is used to produce bio sensor. Scaffold is required high dimensional accuracy. 3D printer is used to manufacture scaffold. 3D printer can't detect defect during printing. Defect detection is very important in scaffold printing. Real-time defect detection is very necessary on industry. In this paper, we proposed the method for real-time scaffold defect detection. Real-time defect detection model is produced using CNN(Convolution Neural Network) algorithm. Performance of the proposed model has been verified through evaluation. Real-time defect detection system are manufactured on hardware. Experiments were conducted to detect scaffold defects in real-time. As result of verification, the defect detection system detected scaffold defect well in real-time.

A Study on Square Pore Shape Discrimination Model of Scaffold Using Machine Learning Based Multiple Linear Regression (다중 선형 회귀 기반 기계 학습을 이용한 인공지지체의 사각 기공 형태 진단 모델에 관한 연구)

  • In this paper, we found the solution using data based machine learning regression method to check the pore shape, to solve the problem of the experiment quantity occurring when producing scaffold with the 3d printer. Through experiments, we learned secured each print condition and pore shape. We have produced the scaffold from scaffold pore shape defect prediction model using multiple linear regression method. We predicted scaffold pore shapes of unsecured print condition using the manufactured scaffold pore shape defect prediction model. We randomly selected 20 print conditions from various predicted print conditions. We print scaffold five times under same print condition. We measured the pore shape of scaffold. We compared printed average pore shape with predicted pore shape. We have confirmed the prediction model precision is 99 %.

  • On the background of general idea and technique of bioscience, medicine and engineering, tissue engineering aim at maintenance, improvement and repair of human body function through manufacturing and transplantation of artificial tissue and organ exchangeable human body. Basic material used in the area is scaffold that aid tissue and organ formation. Making scaffold, solvent-casting and particulate leaching technique is widely used in manufacturing of porous polymer scaffold. There are many types of particle including salt and gelatin. Salt is a most commonly used particulate because it is easily available and very easy to handle and gelatin particle is another candidate for this method because it is known as a material, which enhances cell attachment and proliferation. But there is no comparative study of two kinds of materials. In this study we compared the biocompatibility of the two scaffolds made from salt(salt scaffold) and gelatin particle (gelatin scaffold). These results demonstrated that gelatin scaffold showed better attachment of cells at the initial stage and better proliferation of cells. The better performance of gelatin scaffold is contributed to the better connection of pores in the same porosity.

  • In this paper, We used the regression model of machine learning for improve the print quantity problem when which print scaffold with 400 ㎛ pore using FDM 3d printer. We have difficult to experiment with changing all factors in the field. So we reduced print quantity by selected two factors that most impact the pore size. We printed and measured scaffold 5 times under same conditions. We created regression model using scaffold pore size and print conditions. We predicted pore size of untested print condition using the regression model. After print scaffold with 400 ㎛ pore, we printed scaffold 5 times under same conditions. We compare the predicted scaffold pore size and the measured scaffold pore size. We confirmed that error is less than 1 % and we verified the results quantitatively.

A Comparative Study on Deep Learning Models for Scaffold Defect Detection (인공지지체 불량 검출을 위한 딥러닝 모델 성능 비교에 관한 연구)

  • When we inspect scaffold defect using sight, inspecting performance is decrease and inspecting time is increase. We need for automatically scaffold defect detection method to increase detection accuracy and reduce detection times. In this paper. We produced scaffold defect classification models using densenet, alexnet, vggnet algorithms based on CNN. We photographed scaffold using multi dimension camera. We learned scaffold defect classification model using photographed scaffold images. We evaluated the scaffold defect classification accuracy of each models. As result of evaluation, the defect classification performance using densenet algorithm was at 99.1%. The defect classification performance using VGGnet algorithm was at 98.3%. The defect classification performance using Alexnet algorithm was at 96.8%. We were able to quantitatively compare defect classification performance of three type algorithms based on CNN.

  • Warpage defect detecting of scaffold is very important in biosensor production. Because warpaged scaffold cause problem in cell culture. Currently, there is no detection equipment to warpaged scaffold. In this paper, we produced detection model for shape warpage detection using deep learning based CNN. We confirmed the shape of the scaffold that is widely used in cell culture. We produced scaffold specimens, which are widely used in biosensor fabrications. Then, the scaffold specimens were photographed to collect image data necessary for model manufacturing. We produced the detecting model of scaffold warpage defect using Densenet among CNN models. We evaluated the accuracy of the defect detection model with mAP, which evaluates the detection accuracy of deep learning. As a result of model evaluating, it was confirmed that the defect detection accuracy of the scaffold was more than 95%.

Chondrogenesis of Human Adipose Tissue Derived Mesenchymal Stem Cells (ATMSCs) Seeded in Gelatin-Chondroitin-Glucosamine Scaffold (Gelatin-Chondroitin-Glucosamine Scaffold에 접종한 인간지방조직-유래 중간엽 줄기세포의 연골형성)

  • The present experiment was performed to evaluate the chondrogenic differentiation potential of human adipose tissue-derived mesenchymal stem cells (ATMSCs) in the chondrogenic induction medium (CIM) with transforming growth factor-${\beta}1$ (TGF-${\beta}1$) and to evaluate the chondrogenic differentiation of ATMSCs seeded in gelatin-chondroitinglucosamine scaffold (GCG-scaffold). ATMSCs and mouse chondrocytes were cultured in the basic medium and CIM without TGF-${\beta}1$ (CIM1) or with TGF-${\beta}1$ (CIM2) for chondrogenic differentiation potential. The chondrogenic differentiation of ATMSCs was evaluated by glycosaminoglycan (GAG) synthesis and histochemical staining. In pellet culture, GAG synthesis of ATMSCs and chondrocyte was increased in culture on 14 days, but higher in CIM1 than basic medium, especially highest in CIM2. Cartilage matrix was observed in ATMSCs cultured in CIM2 on 14 days by Safranin O and trichrome staining. In well plate culture, proliferation of ATMSCs was continuously increased in culture on 10 days and higher in CIM than basic medium. The cell adhesion rate of ATMSCs seeded in flask or scaffolds was continuously increased during culture period, but higher in scaffold than flask. GAG synthesis of ATMSCs seeded in scaffolds showed no change in control group. In the CIM groups, GAG synthesis of ATMSCs was continuously increased than control group during culture period, especially very high in CIM2 and in the GCG-scaffold was slightly higher than the gelatin scaffold (G-scaffold). The present results demonstrated that ATMSCs showed an low chondrogenic differentiation potential, compared to mouse chondrocytes for 14 days of culture. TGF-${\beta}1$ is important factor in chondrogenic differentiation of ATMSCs. Gelatin scaffold was considered to increasing the effective chondrogenic differentiation environment. ATMSCs seeded in GCG-scaffold was more effective in chondrogenesis than in G-scaffold. Conclusively, the present results demonstrated that the treatment of chondroitin and glucosamine in the scaffold was more effective to promote the cartilage matrix formation.

Automatic design, planning and drawing of scaffolding system for constructions

  • Temporary scaffold systems in the construction site play an important role for ensuring the safety of the workers and for constructing the stable structures. To assemble the scaffold, the pipe scaffolding system, the wedge binding scaffolding system and the particular materials have been utilized. To design the material arrangement of a scaffold, firstly the configuration was determined considering the construction geometry. Then, the strength of the scaffold was confirmed and the quantity of the material was accounted. In this paper, the design method of the temporary scaffold was proposed for intending the semi-automatic procedure. In the proposed design method, the geometric design and the safety requirement were specified by the safety standard and the design flow was followed by the designer's knowledge. The size and the quantities of the materials were calculated by referring to the relation between the scaffold and the constructing structure. In the calculating procedure, three dimensional positions of each scaffold materials were calculated and recorded simultaneously. Then, three dimensional scaffold structural was drawn semi-automatically on the CAD software by using the obtained material sizes, positions and directions. The proposed design method provides us the precise quantities of scaffold materials and enables us to reduce the design effort and the cost estimation processes. In addition, the obtained results can be applied to BIM software after converting to IFC format.

Revision of Safety Standards through Analysis of Fatal Accidents Related to Mobile Scaffold in Construction Sites (건설현장 이동식 비계의 사망사고 분석을 통한 안전기준 개선 방안)

  • Mobile scaffold is used as a work platform in construction sites. According to industrial accident data, 10 fatalities occur annually on average due to falling from mobile scaffold in construction sites. To prevent such fatal accidents, safety standards must be improved by analyzing the causes of fatal accidents related to mobile scaffold. Accordingly, we analyzed domestic and international safety standards for mobile scaffold and analyzed the fatalities (117 persons) caused by mobile scaffold accidents in domestic construction sites during 2011-2020 and found that fall, overturn, and collapse accounted for 88% (103 persons), 10.3% (12 persons), and 1.7% (2 persons) of fatalities, respectively. The main causes of fatal accidents were found to be non-installation or insufficient installation of safety rails and outriggers and incorrect sequence of work. Based on the analysis results, we proposed strategies for improving the safety standards, such as the use of advanced guard rails and wheel-attached outriggers, to enhance the safety and field applicability of the mobile scaffold. Technology development and improvements in the safety standards for the use of mobile scaffold will aid in the prevention of fatal accidents caused by mobile scaffold.
