Cereal Scab Causing Mycotoxicoses in Korea and Present Status of Mycotoxin Researches (한국(韓國)에서의 진균중독증(眞菌中毒症)을 일으키는 맥류(麥類) 붉은 곰팡이병 및 진균독소(眞菌毒素) 연구(硏究)의 현황(現況))

  • In 1963, a severe epidemic of cereal scab caused by Gibberella zeae occurred in southern Korea and to a less extent in central and northern Korea. In some areas losses were $80{\sim}100$ percent. The epiphytotic was due to heavy rainfall during the heading and flowering season which provided a favorable environment for this severe epidemic. Yield losses resulted a great social problem because of the resultant food and feed grain shortage, lose of seeds for planting the following crops and mycotoxicoses to man and animals. In the same year, a nationwide research committee was organized including plant pathologists, microbiologists, agronomists and biochemists under the juristiction of the Ministry of Agriculture and Forestry. The committee initiated research on etiology, epidemiology, and control of the disease and on the toxic effect of infected grains to man and animals. The present paper will review some research carried out in Korea on cereal scab with special reference to epidemiology and mycotoxicoses to animals and man. In addition, the present status of research in Korea on aflatoxins in foods and toxic moldy rice will be briefly reviewed.

Genome Research on Peach and Pear

  • A lot of SSRs (simple sequence repeats) in peach and pear from enriched genomic libraries and in peach from a cDNA library were developed. These SSRs were applied to other related species, giving phenograms of 52 Prunus and 60 pear accessions. Apple SSRs could also be successfully used in Pyrus spp. Thirteen morphological traits were characterized on the basis of the linkage map obtained from an $F_2$ population of peach. This map was compiled with those morphological markers and 83 DNA markers, including SSR markers used as anchor loci, to compare different peach maps. Molecular markers tightly linked to new root-knot nematode resistance genes were also found. A linkage map including disease-related genes, pear scab resistance and black spot susceptibility, in the Japanese pear Kinchaku were constructed using 118 RAPD markers. Another linkage map, of the European pear Bartlett, was also constructed with 226 markers, including 49 SSRs from pear, apple, peach and chewy. Maps of other Japanese pear cultivars, i.e., Kousui and Housui, were also constructed. These maps were the first results of pear species.

Improved Method to Increase Conidia Production from Isolates of Different Pathotypes of Citrus Scab Pathogen Elsinoe spp.

  • Elsinoe fawcettii and E. australis are two currently recognized scab pathogens of citrus. E. fawcettii has at least six pathotypes while E. australis has at least two pathotypes. Colonies of E. fawcettii and E. australis do not sporulate in artificial media including potato dextrose agar (PDA). Whiteside's method has been widely used for preparing conidial inoculum in vitro. This study was carried out to develop efficient method for conidia production from artificial media. We developed a shaking method which included the following steps: 1) Colony grown on PDA was mashed with a steel spatula; 2) Mycelia fragments were cultured in 50 ml sterilized rain water in a rotary shaker-incubator (180 rpm) at $25^{\circ}C$ for 24 h: 3) The conidia suspension was filtered through two layers of cheesecloth. Average conidia production of all isolates tested using this shaking method was approximately 13.1 times higher than that from Whiteside's method in this study.

Induced systemic resistance and plant growth promotion of a phosphate-solubilizing bacterium, Enterobactor intermedium 60-2G (인산가용미생물, Enterobacterium intermedium 60-2G의 식물 생장 촉진 및 전신저항성 유도)

  • A phosphate-solubilizing bacterium, Enterobacter intermedium 60-2G, was examined for plant growth-promotion and induction of systemic resistance using a model system of cucumber and scab disease, caused by Cladosporium cucumerinum. Compared with a nonbacterized control, treatment of cucumber with E. intermedium significantly reduced the severity of scab disease after challenge-inoculation with C. cucumerinum. Treatment of cucumber with E. intermedium also enhanced cucumber growth. The 60-2G strain showed a strong antimicrobial activity against several plant pathogenic fungi including Fusarium soysporum and Magnaporthe grisea. These results suggest the E. intermedium 60-2G is a promising candidate as a biological control agent displaying multiple beneficial properties to promote plant health.

Characterization of Streptomyces Species Causing Potato Scab in Korea: Distribution, Taxonomy, and Pathogenicity

  • From 1996 to 1999, potato-growing areas in Korea were surveyed for identification and distribution of potato scab pathogens. Potato scab was widely distributed in the mass cultivation areas, especially in Jriu island, southern areas of Chonnam and Gyounggi provinces, and the alpine area of Gangwon province. Jeju island was the most affected area by this disease. A total of 55 Streptomyces strains were isolated from potato scab lesions, among which 40 strains were pathogenic on progeny tubers. Among the pathogenic strain, 21 strains were identified as previously described S. scabies, 7 Strains as S. turgidiscabies, and 5 Strains as S. acidiscabies, while 7 strains were observed as having distinct phenotypic properties. These strains were classified into six distinct clusters based on phenotypic characteristics and selected representative strains for each cluster. S. scabies (S33) had grey spores in a spiral chain. Mean-while, S. turgidiscabies (S27) had grey spores, S. acidiscabies (S71) had white spores, S. luridiscabiei (S63) had yellow-white spores, S. puniciscabiei (S77) had purple-red spores, and S. niveiscabiei (S78) had thin and compact white spores, all in a rectiflexuous chain. Pathogenicity was determined by the production of thaxtomin A and homologs of necl and ORFtnp genes. In TLC, representative strains S27, S71, S63, S77, and S78 produced a yellow band that co-migrated with the authentic thaxtomin A. However, thaxtomin A was not detected in chloroform extracts from oatmeal broth culture and Slice tuber tissue of S. luridiscabiei (S63) and S. puniciscabiei (S77) by HPLC analysis. In addition, no homologs of necl and ORFtnp genes in S. acidiscabies (S71), S. luridiscabiei (S63), S. puniciscabiei (S77), and S. niveiscabiei (S78) were detected by PCR and Southern hybridization analysis.

Review of Disease Incidence of Major Crops in 2003 (2003년 농작물 병해 발생개황)

  • The year of 2003 was characterized as a cool humid year. Low temperature and frequent rains were continued during March to July, resulting in 1.6 times higher rainfalls and 32% less sunshine period compared to the average yean Due to 2003's climatic condition, rice blast, and bacterial leaf blight occurred severely. Higher rainfalls caused severe epidemic of phytophthora disease and, in case of red-pepper, 55% of cultivation acreage was devastated by the disease over the country. Besides, crop diseases which become severe under cool-humid conditions, such as gray mold, sclerotinia rot, downy mildew, increased significantly compared to the previous year. In fruit trees, brown spot of apple, and pear scab occurred severely causing much yield loss.

  • Venturia nashicola is a fungal pathogen causing scab disease in Asian pears. It is particularly important in the Northeast Asia region where Asian pears are intensively grown. Venturia nashicola causes disease in Asian pear but not in European pear. Due to the highly restricted host range of Venturia nashicola, it is hypothesized that the small secreted proteins deployed by the pathogen are responsible for the host determination. Here we report the whole genome based phylogenetic analysis and predicted secretomes for V. nashicola isolates. We believe that our data will provide a valuable information for further validation and functional characterization of host determinants in V. nashicola.

Screening for Effective Organic Farming Materials for the Control of Cucumber Scab Caused by Cladosporium cucumerinum (오이 검은별무늬병 방제에 효과적인 유기농업자재 선발)

  • This study was conducted to evaluate the control efficacy of the organic farming materials on cucumber scab caused by Cladosporium cucumerinum PT1 (KACC 48094). The antifungal activities in vitro as well as the suppressive effect of 43 organic farming materials on the spore germination and germ tube growth by inoculating spore suspension on cucumber seedlings in vivo were investigated. Thirteen organic farming materials inhibited the mycelial growth of C. cucumerinum and nine of these were microbial agents. In the screening using cucumber seedlings, six organic farming materials were very effective with control efficacy value of 90%. Among them, Bacillus amyloliquefaciens M27 provided suppressive effect on both mycelial growth and spore germination against cucumber scab. Finally, nine organic farming materials were selected to test the protective and curative effects, and all chosen organic farming materials significantly suppressed disease incidence when applied in the preventive action, in comparison with the curative action. Especially, Bordeaux mixture I and III gave excellent protective control efficacy with control values of 96.7% and 73.3%, respectively, whereas its curative control effect was significant low. Among these, only Thymus quinquecostatus+Sophora extract showed curative activity, although the control value was as low as 50%. This study suggests that cucumber scab can be controlled by some organic farming materials in the farmhouses under comparatively cold and wet condition and protective treatment is more important and efficient.

Aspect of Incidence of the Major Citrus Diseases Recently (최근 주요 감귤 병의 발생 양상)

  • The percent of diseased fruit by Diaporthe citri was the most 16.8% in 2010 and the least 3.8% in 2005, and the average was 8.8% from 2003 to 2012. The degrees of disease incidence were relatively high, 17.0, 22.6, 19.2 and 18.9%, in 2003, 2004, 2007 and 2010, respectively. The percent of diseased fruit by Elsinoe fawcettii was the most 0.72% in 2004, and then gradually decreased to 0.08% in 2008, and the average was from 0.28% from 2004 to 2011. In case of canker by Xanthomonas citri subsp. citri, the percent of diseased fruit was the most 2.19% in 2004, and gradually decreased to 0.08 and 0.10% in 2009 and 2010, respectively. The average was 0.81% from 2004 to 2012. The melanose by D. citri was most in south-east area of Jeju Island, and east and west in case of canker. The melanose symptoms were initially appeared from mid June, mainly peaked late of July to late August. The canker symptoms were initially produced on leaves of natsudaidai and grapefruit cultivated in open field middle of June in 2010 and 2011 and late of May in 2012. The scab disease was firstly appeared approx. early or mid May and then rapidly increased late of May to middle of June. The inoculum was mainly produced from dead twigs late June to early of August.
