  • Morphological differences were studied using the analysis of variance between various partial length and shell length of three populations of Saxidomus purpuratus on the Korean waters. The Relative growth equations, that is, SH-SL, SW-SL, TW-SL of S. purpuratus by sex were estimated. The analysis of variance of four morphological characters proved that each population has no sexual differences (p>0.01). But the three populations are significantly different in morphological characters (p<0.01).

  • Age and growth of purpulish Washington clam (Saxidomus purpuratus) was investigated from 480 samples monthly collected from January to December 2002 in Jinhae Bay, Korea. Examination of outer margins of the shell revealed that the translucent zone was formed once a year from March to April, it can be used as annulus. Ages were determined from ring radius of shell, and maximum age of the the clam was 9 years. The spawning period was from May to October, and the main spawning occurred in July. The relationship between shell length (SL) and shell height (SH) was $SH=0.8405{\times}SL^{-4.9709}\;(R^2=0.97)$ and that between the shell length (SL) and total weight (TW) was $TW=0.9580{\times}10-4{\times}SL^{3.220}(R^2=0.97).$ The von Bertalanffy growth equation were $SL_t=125.57(1-e^{-0.2523(t+0.5367)},\;TW_t=549.26(1-e^{-0.2523(t+0.5367)})^{3.220}.$

  • The reproductive biology of the Washington clam Saxidomus purpuratus, from Yeosu, Korea was investigated based on 699 individuals collected from January to December 2022. The sex ratio of this species was not significantly different from a 1:1 (male:female) sex ratio (P>0.05). Monthly changes in gonadosomatic index were analyzed to estimate the spawning season. The reproductive cycle of this species can be divided into the following stages: active (Jan.-Mar.), ripe (Mar.-Nov.), spent (Apr.-Dec.), and degenerative (Jun.-Dec.) stages. The spawning period ranged from March to December with the main periods being from April to June and from September to November. The shell length at 50% female group maturity (SL50) was estimated to be 60.2 mm.

  • Biochemical and biometric features of eggs of the Washington clam, Saxidomus purpuratus, are reported in this study. In addition, a polyclonal antibody against the purified clam egg was employed to quantitative assessment of eggs of the clams collected from Geoje Island, Korea in May 2002. Mean egg diameter was 70.81 $\pm$ 7.52 ${\mu}{\textrm}{m}$(histological preparation) or 88.56 $\pm$ 11.31 ${\mu}{\textrm}{m}$(purified oocytes), and individual egg weighed 95 ng in dry weigt. • The purpose of this study is to determine the appropriate substrate for larval settlement and spat growth in the purple clam, Saxidomus purpuratus in laboratory culture. Larvae were reared with 3 different types of sediments (mud, sand, and mixed) for 46 days in settlement experiment, and settled spats were further grown in 3 types of sediments for 36 weeks in growth experiment. The density of settled spats in muddy sediments was more than 2 times higher than those in mixed or sandy sediments. But, the average size of settled spats in muddy sediments was smaller than those in mixed or sandy sediments. After 36 weeks of growth period, growth rate decreased as shell length increased. When shell length was less than 2 mm, growth rate in mixed sediments was significantly higher than that in sandy sediments. When shell length was more than 2 mm, there was no significant difference in growth rate among different substrates. Sediment type affected growth rate only when the spats were relatively small (less than 2 mm). Muddy sediments seems better for larval settlement, while mixed sediments is best for spat growth. We suggest the laboratory procedure for enhancing seedling production of S. purpuratus.

  • An immunological method was developed in this study to quantify reproductive output of the female butter clam, Saxidomus purpuratus. A clam egg-specific polyclonal antibody was developed using the purified butter clam egg as an antigen. An indirect Enzyme-Linked Immunosorbent Assay (ELISA) was used in quantitative measurement of the eggs. Size of the butter clam eggs ranged from $70.81{\pm}7.52{\mu}m$ in histology or $88.56{\pm}11.31{\mu}m$ in intact eggs. The predominant egg constituent was protein (37.44%), followed by lipids (11.40%) and carbohydrates (9.68%). The SDS-PAGE showed that the egg proteins are composed of several peptides of molecular weights consisting of 247, 200, 99, 91, 54 and 47 kDa. ELISA indicated that the clams collected from Geoje Island in May 2002 produced 8.2 to 26.8% of their body weight as eggs or 9,307,309 to 31,156,333 with a mean of 16,931,893 eggs per individual clam. The results of this study thus suggest that indirect ELISA using rabbit anti-clam egg IgG as a primary antibody is a rapid, affordable and sensitive method to assess reproductive output of 5. purpuratus and possibly other bivalves using a small amount of eggs.

  • The purpose of this study is to compare the clearance rate (CR) and intake rate (IR) of juvenile purple clam, Saxidomus purpuratus when feeding on different unialgal diet of red tide dinoflagellates (RTDs), and to know what is the most important cell characteristic of RTDs to cause the differences in feeding parameters. Experiments were performed to measure the CR and IR of juvenile S. purpuratus as a function of algal concentration when food was either the standard food, Isochrysis galbana or one of 9 RTDs. Patterns of CR with increasing algal concentration were similar among different RTDs. The highest $C_{max}$ was observed when S. purpuratus was feeding on A. affine, while the lowest on C. polykrikoides. The patterns of IR with increasing algal concentration were also similar among different RTDs. However, there were great differences in the maximum value of IR ($I_{max}$) among different RTDs. The highest $I_{max}$ was observed when S. purpuratus was feeding on A. carterae, while the lowest on G. catenatum. Some RTDs similar in size showed different $C_{max}$. Other RTDs different in size showed similar $I_{max}$. Life form of each RTD affected significantly the $I_{max}$, which was higher for single-celled RTDs than chain-forming RTDs. There were no significant differences in feeding parameters between toxic and nontoxic RTDs. Moreover, a toxic dinoflagellate, A. carterae recorded the highest $I_{max}$ among RTDs. The most important characteristic of RTD as a factor affecting the feeding rate of S. purpuratus was life form, not size or toxicity of RTD species.
