One of the most economically viable processes for the bioconversion of many lignocellulosic waste is represented by white rot fungi. Phanerochaete chrysosporium is one of the important commercially cultivated fungi which exhibit varying abilities to utilize different lignocellulosic as growth substrate. Examination of the lignocellulolytic enzyme profiles of the two organisms Phanerochaete chrysosporium and Rhizopus stolonifer show this diversity to be reflected in qualitative variation in the major enzymatic determinants (ie cellulase, xylanase, ligninase and etc) required for substrate bioconversion. For example P. chrysosporium which is cultivated on highly lignified substrates such as wood (or) sawdust, produces two extracellular enzymes which have associated with lignin deploymerization. (Mn peroxidase and lignin peroxidase). Conversely Rhizopus stolonifer which prefers high cellulose and low lignin containg substrates produce a family of cellulolytic enzymes including at least cellobiohydrolases and ${\beta}-glucosidases$, but very low level of recognized lignin degrading enzymes.
Ergosterol is an indicator chemical of fungi and involves in fungal cell growth as a major component in fungal cell membranes. Thus, this chemical can be used to estimate live fungal biomass within various solid substrates. Ergosterol content in liquid culture of Lentinula edodes had a linear relationship (r=0.98) with the hyphal mass of the fungus. This chemical content differed depending on the fungal strains, age of culture and water content levels of sawdust substrate. Ergosterol content was 0.13% in the 10 weeks old liquid culture while it was 0.10% in 20 weeks old one. The chemical content in the sawdust cultures of the fungus varied 0.015% to 0.042% depending upon strains and water content levels within sawdust substrate. Ergosterol content in the culture of a L. edodes strain, Sanrim 4, was higher by $20{\mu}g$ to $140{\mu}g$/g dry substrate than those of strains, Mok-H and Sanrim 6. And the chemical contents in the sawdust cultures with 125% or 175% water, 297 to 425 ${\mu}g$/g dry substrate, were higher than those with 75% or 225% water, 148 to $286{\mu}g$/g dry substrate. We conclude that ergosterol analysis can estimate the fungal biomass within solid substrate such as logs and sawdust.
The mycelia of Lepista nuda were isolated from the different basidiocarps, collected from the forests of Robinia pseudo-acaciae and mixed forests of R. pseudo-acaciae and Quercus serrata. On the different woods, the growths of mycelia collected were compared with those transferred from Applied Mycology Division, RDA. Also, the measurements of dry weight loss, as growth of mycelia, were correlated to the productions of carbon dioxide on the different wood substrates. The growth of mycelia was observed to be best on the woods of R. pseudo-acaciae; followed by the growth on the woods of Morus alba. The growth rate of mycelia appeared to be various, depending on the different kinds of wood substrates. The soild-cultures of the mycelia on the sawdust of the R. pseudo-acaciae woods and the grains of sorghum and corn were conducted, but the basidiocarps were not obtained from these. Therefore, this mycelium collected was believed to be important as new mushroom utilizing the different woods resources instead of Q. serrata.
The mating system of monokaryotic isolates in Pleurotus sajor-caju was controlled by two incompatibility factors A and B of tetrapola mating system. The mycelia of dikaryotic isolates grew faster than those of their component monokaryons, but no correlation between dikaryotic and their component monokaryotic isolates was found. The primodia formed well on the potato dextrose agar (PDA) media not only in the dikaryotic isolates but also in the monokaryons under irradiated conditions. The dikaryotic isolates produced normal sporophores; however, the monokaryotic isolates produced abnormal sporophores when they were cultivated with sawdust substrates. Some dikaryotic isolates derived by mating between monokaryotic isolates showed high yields of sporophores more than those of parental strains. Both the dikaryotic mycelial growth rate and primodia formation number on the PDA plate showed significant correlation with its sporophore products on sawdust substrates.

Some factors effecting on mycelial growth and cultural substrates using pine sawdust were investigated to develop an artificial cultivation technique. The optimal temperature and media pH for the mycelial growth were at $25^{\circ}C$ and 4.0, respectively. The mycelial growth was not different among three isolates tested. Among them ASI 19003 isolate showed the highest mycelial density and fruitbody yield. Among supplements added into pine sawdust, wheat bran was the best and its appropriate percentage was 20 for the mycelial growth. The mycelial densities were uprisen according to the amounts of the supplement. The highest yield, 107g/bottle of the sporophores of the mushroom was obtained from the substrates supplemented by 30% of wheat bran, but no fruitbody was formed on non supplemented one. Optimal moisture contents of the substrates was different for the mycelial growth, pinheading and fruitbody yields. Sixty five, 60 and 75% of moisture contents of the substrates were optimal for the mycelial growth, pinheading and fruitbody yields, respectively.

The composition of artificial soil in a containerized seedling production plays an important role in seedling quality as well as environmental issues. We investigated the effects of different types of biomaterials and mixed ratio with artificial soil on the growth of Fraxinus rhynchophylla seedlings. Soil medium was supplemented with 3 levels (0%, 10%, 20%) of pine bark, mushroom sawdust and rice husk. Root collar diameter (RCD), height growth, and biomass have significantly increased when rice husk was applied. Compared with the control, RCD and height growth showed highest in 20% rice husk treatment with an increase of 5.7% and 17.6%, respectively. In contrast, the treatments of pine bark and mushroom sawdust showed lower results in growth parameters (RCD, height growth, and total biomass) than control. Seedling quality index was also highest at the 20% rice husk treatment, but there was not statistically different among treatments. Our results suggested rice husk can be substituted up to 20% of substrates for containerized F. rhynchophylla seedling production system.

Oak mushroom is cultivated using logs and sawdust media as substrates. In this study, fungi were isolated during a monitoring of indoor air in the oak mushroom cultivation houses located in Cheongyang-gun of Chungnam, Geoje-gun of Gyeongnam, Gumi-si of Gyeongbuk, Jangheung-gun of Jeonnam and Yeoju-si of Gyeongggi-do. Identification of the fungi based on morphology and molecular analysis of the internal transcribed spacer region and 28S rDNA, translation elongation factor translation elongation factor 1 a gene, and ${\beta}-tubulin$ gene revealed that six fungi, Cenangium acuum, Neopestalotiopsis surinamensis, Metarhizium marquandii, Periconia macrospinosa, Trichoderma petersenii, and Trichoderma paratroviride that have not been recorded previously in Korea.

Sawdust bag cultivation of Shiitake mushroom (Lentinula edodes) is getting increase. The mycelia browning on the substrate surface is important for the stable production. The development of methods for the rapid mycelia browning is quite required. In this study changes in intracellular enzyme during the mycelial browning were investigated to find the rapid mycelia browning. Mycelia of L. edodes was changed into brown color while it grew in agar and liquid media like sawdust substrates. Mycelia of L. edodes was started to change color at 25 days after inoculation and browning was occurred in whole mycelia colony at 30 days and browning was completed at 40 days. The activities of enzymes was evaluated in these periodically color changing mycelia. Laccase activity was highest at 15 days after inoculation on PDB, but it gradually decreased from 15 days. Tyrosinase activity drastically increased in period between 30 days and 40 days while mycelia browning was progressed. The kinds of phosphotase identified by electrophoresis were esterase, acid phosphotase, and alkaline phosphotase. Activities of phosphotase were increased before the initiation of mycelial browning but they were decreased after browning.

The effects of the processing mixture of food wastes and various organic wastes when vermicomposted on earthworm(Eisenia foefida) growth, the cast production amounts and the chemical properties of casts were evaluated. The substrates used in this experiments were cow manure, pig manure sludge, fermented pig manure with sawdust, nightsoil sludge, and sewage sludge and were respectively mixed with food wastes at a ratios of 50:50(v/v). The control consisted of food wastes alone without other wastes. All of earthworm died in the food wastes 100%, therefore the process of food wastes alone by vermicomposting was impossible in this experiment. Worm cast produced sufficiently contained quantities of available phosphorus, exchangeable potassium, exchangeable magnesium, and cation exchange capacity. The increase of earthworm's biomass occured on the mixtures of food wastes and cow manure, fermented pig manure with sawdust. Dry weight of worm cast was the highest on the mixtures of food wastes and fermented pig manure with sawdust and the proportion of cast weight after vermicomposting was significantly the highest on the mixtures of food wastes and cow manure($p{\leq}0.05$). Also, the mixtures of food wastes and cow manure, and fermented pig manure with sawdust showed a positive values of conversion rate and conversion efficiency rate of organic matter to earthworm tissue than that of other treatments. These results suggested that cow manure and fermented pig manure with sawdust are adequate to process with food wastes by vermicomposting.
A good mycelial growth of F. fraxinea was observed on CDA medium. The optimum temperature and pH for the mycelial growth of F. fraxinea was at $30^{\circ}C$ and pH 6.0, respectively. Glucose was found to be the best carbon source and arginine was favored as nitrogen source. When the basal medium was supplemented with organic acids, the best growth was shown in succinic acid and the poor growth was shown in oxalic acid. Thiamine.HCl showed the best results on the growth of this fungus on basal medium. Mycelial growth of F. fraxinea was quite good when oak tree sawdust was used to cultural substrates. The best mycelial growth was observed when 20% of rice bran was added as a supplement on sawdust substrates. Higher yield of F. fraxinea was observed on the medium with oak tree and acacia sawdust.
