Mycelial Growth and Fruiting Body Formation of Hericium erinaceum in Sawdust and Agricultural By-product Substrates (톱밥 및 농업부산물 이용 배지상에서 노루궁뎅이버섯(Hericium erinaceum)의 균사생장 및 자실체형성)

  • This study was carried out to investigate the suitability of various agricultural by-products as basal substrates for the mycelial growth and fruiting body formation of Hericium erinaceum. For this aim, oak sawdust, cotton waste, sugarcane bagasse, Job's tears, rice hull, Chinese cabbage, and coconut waste were used as sole or mixed substrate(s). Corn waste and rice bran were used as nutrient supplements. The growth and density of mycelium, yield of fruiting body, and biological efficiency were compared among tested substrates colonized by Hericium erinaceum. The best measurement of mycelial growth and density, yield of fruiting body, and biological efficiency in a laboratory test was found in a spawn substrate composed with oak sawdust 80% and rice bran 20%. The suitability of this spawn substrate composition for Hericium fruiting body production was testified through practical tests in plastic bottles (850 ml) in a mushroom farm which had bottle cultivation facility. However, test in a mushroom farm which had plastic bag cultivation facility, best production of Hericium fruiting body (520 g per one bag) was observed in a spawn substrate composed of cotton waste 40%, saw dust 40%, corn waste 10%, and rice bran 10%.

  • Mushroom is very important to rural dwellers due to the various economic, health and nutritional benefits that can be derived from its consumption, sales and utilization. Hence, this experiment was carried out with a view to evaluating the influence of locally available substrate i.e., sawdust of different wood species on the cultivation of Pleurotus ostreatus (oyster mushroom). Pleurotus ostreatus was grown on sawdust (indigenous and exotic species) using two pretreatment methods, which included hot water treatment to remove (a water-soluble extract) and non-extracted sawdusts (substrate). The result of this study showed that there was a significant difference in the effect of different wood species selected. Parkia biglobosa produced a better yield (71.06 g), followed by Anogeissus leiocarpus (53.17 g) and Gmelina arborea (51.39 g) in a non-treated unextracted states. In the treated samples, Parkia biglobosa has the highest yield (50.61 g) followed by Anogeissus leiocarpus (49.28 g) and Gmelina arborea (45.00 g). Thus, Parkia biglobosa and Anogeissus leiocarpus produced a higher yield of mushroom either in a treated or non-treated state and they could be recommended for growing oyster mushroom (Pleurotus ostreatus).

  • For artificial cultivation of Phellinus pini (Thore. Fr.) Ames, we conducted some study on mycelium growth and optimum condition for fruitbody formation. The optimum condition for mycelial growth was $25{\sim}30^{\circ}C$ at pH $5.0{\sim}6.0$. Optimum sawdust media were oak sawdust+willow sawdust+rice bran (4.5:4.5:1, V/V) and oak sawdust+pine sawdust+rice bran (4.5:4.5:1, V/V) and the optimum spawn incubation period was about $33{\sim}34$ days. Mycelial growth in the inner portion of oak log was 40 mm after 60 days and duration for first fruitbody primordia formation was about 110 days after inoculation.

Effects of Aeration of Sawdust Cultivation Bags on Hyphal Growth of Lentinula edodes

  • The effects of aeration through lid filters on the hyphal growth of Lentinula edodes (oak mushroom) in sawdust cultivation bags were investigated. The aeration treatment levels were traditional 27 mm hole cotton plugs, cotton balls and combinations of seven hole sizes ${\times}$ two hole positions (up and under) in the lids covering plastic bags containing 1.4 kg sawdust medium at 63% moisture that had been autoclaved for one hour and inoculated with sawdust spawn of L. edodes strain 921. Aeration treatment effects were measured based on the $CO_2$ concentration at the 15th wk, as well as the hyphal growth rate and degree of weight loss of bags every 14 days for 15 wk. In bags with traditional cotton plugs, the $CO_2$ concentration was $3.8{\pm}1.3%$, daily mean hyphal growth was $2.3{\pm}0.6mm$ and daily mean weight loss was $0.84{\pm}0.26g$. In the bags with 15 mm diameter holes, the $CO_2$ concentration was $6.0{\pm}1.6%$, daily hyphal growth was $2.8{\pm}0.2mm$ and daily weight loss was $0.86{\pm}0.4g$. The bags with 15 mm holes had a higher $CO_2$ concentration and lower water loss than bags with other hole sizes, but the hyphal growth was not significantly different from that of other bags. The weight loss of bags increased proportionally relative to the lid hole sizes. Taken together, these results indicate that traditional cotton plugs are economically efficient, but 15 mm hole lids are the most efficient at maintaining hyphal growth and controlling water loss while allowing $CO_2$ emissions.

Availability of Sikhae Factory Wastewater as a Submerged Culture Medium for Lentinula edodes

  • Sikhae is a Korean traditional beverage of saccharified rice. Its factory waste(SFW) is usually thrown away instead of being used. We developed a cheap substrate of SFW for use in liquid spawn that is known for its higher fruit body yields than grain spawn in sawdust cultivation. Mycelia of Lentinula edodes ASI 3046, which is regarded as the most suitable strain for sawdust cultivation, were cultured on six kinds of previous known media and SFW. As the seven kinds of media were applied, a Sikhae Factory Waste(SFW) was most excellent in growth. The dried mycelial weight in SFW was almost four times as much as that in the other media. In the flask culture, optimum culture conditions for the mycelial growth were obtained after 13 days of cultivation at media volume of 100 ml, 100 rpm, initial pH 4.5, and $25^{\circ}C$. The best mycelial growth was observed when $MgSO_4{\cdot}7H_2O$ and D-sucrose were added as a supplement in SFW. SWM must be a remarkable medium for L. edodes because of its simple preparation and low cost.

Culture conditions of liquid spawn and the growth characteristics of Pleurotus ostreatus (느타리버섯의 액체종균 배양 조건과 생육 특성)

  • To improve the productivity of Pleurotus ostreatus, different conditions of liquid spawn culture were tested. The optimum culture conditions were potato dextrose broth, incubation temperature of $22^{\circ}C$, and pH 6. Fifteen different media ('A' to 'O') containing 0.3% soybean meal (SM) were prepared by varying sugar and glucose contents. The cultures were propagated in SM media for 14 days, at $22^{\circ}C$, and pH 6. Their absorbances were higher after 14 days of incubation in the media containing both sugar and glucose. In particular, the absorbance of media containing 5 to 20% of glucose alone ('C', 'D', 'E', and 'F') tended to increase in the incubation period. Dry cell weight was lower in media containing less than 20% sugar or glucose alone than in media containing 30% sugar ('A') or 30% glucose ('B') alone. In sawdust media, in 900 mL-bottle, the optimum inoculation volume of liquid spawn was 15 to 20 mL. The texture of the mushroom cultivated with the liquid spawn was superior to that cultivated in the solid spawn.

Optimum mixing rate of used media for saving the production cost of Flammulina velutipes (팽이버섯(Flammulina velutipes) 생산비 절감을 위한 폐배지 적정 혼합비율)

  • These experiments were conducted to find the optimum mixing rate of used media for saving the production cost of Flammulina velutipes. The materials for media formation was used needle-leaf tree sawdust, media that finished 1th cultivation of F. velutipes, corncob meal, and rice bran, wheat bran as nutrition source. We inoculated the F. velutipes in 14 kinds of different media types and checked the spawn growth speed, fruit body quality and quantity. Two nutrition agents, which is rice bran and wheat bran, did not affected the incubation period, but the effective stem number, quality and quantity of fruit body was better at rice bran than wheat bran. The quality of fruit body produced at mixed 20% of used media (needle-leaf tree sawdust 60% + used media 20% + rice bran 20%) was similar to control plot (needle-leaf tree sawdust 80% + rice bran 20%), but the yield was improved 10% than control plot 130g. According as the used-media mixing amount increases, quality and quantity of fruit body became low remarkably. Therefore, the optimum mixing amount of used-media was 20% and it increased 10% of fruitbody yield.

Effect of Rice Bran Added at Spawn-making on the Cultivation of Oyster Mushroom, Pleurotus spp. (느타리버섯 종균의 미강함량이 재배에 미치는 영향)

  • This experiment were carried out to investigate the effect of the rice bran added into spawn various amounts on the cultivation. Our results show that 10 to 20 percent addition of the rice bran as a supplement results in a good mycelial growth and density. However we didn't find a significant variance among the different species of oyster mushroom using poplar sawdust as a medium. When it inoculated spawn with various amounts of rice bran on the medium of rice straw, the mycelial density was increased according to the increase of the added supplement, while there was no significant in the mycelial growth among the treatments. Through the field test it was showed that 15 to 20 percent addition of the supplement results in the highest yield, the shortest days from spawing to pinhead, and the lowest infection rate.

  • Armillaria isolates (KNU-A110, KNU-A234, KNU-A1022 and KNU-A1030) were excellent isolates for producing Gastrodia tuber in farm cultivation. Depth of soil between $10\;cm{\sim}19\;cm$ was favorable for producing Gastrodia tuber and rhizomorph at cultivated area. Eighty nine isolates were collected from 5 countries; 16 from Japan, 22 from USA, 26 from France, 4 from Africa and 21 from Korea. Mycelial fan of most isolates were better formed on basal medium with lemon extract than without lemon.A. mellea (KNU-A997) were strongly pathogenic to Gastrodia but A. gallica (KNU-A110) were excellent symbiotic to Gastrodia tuber. Mycelial growth were good on basal medium containing $0.25%{\sim}0.5%$ ethanol and sawdust spawn added wheat bran and corncob. Liquid culture inoculation were not only fast growth of mycelium but also reduction of contamination.

  • The effect of punching treatment on bags containing sawdust medium for the stable production of fruiting bodies of Lentinula edodes 'Hwadam' was investigated. After spawn inoculation, the sawdust medium temperature and CO2 concentration reached 27.0-28.8℃ on the 23rd day of incubation, and 14.0-15.6% on the 16th day of incubation, respectively, decreasing thereafter. The O2 concentration showed an opposite pattern to the CO2 concentration. As the number of punching treatments on the medium increased during the incubation period, the lightness value decreased, and conversely, the color difference and weight loss rate increased. There was no difference in the cultivation period according to the punching treatment. As the punching treatment increased on the sawdust medium, the fruiting body yield increased; a three-time punching treatment produced the best yield with 605.7 g. Thus, proper punching treatment on a sawdust medium during the incubation period can increase the number of fruiting bodies and yield.
