Studies on Microbial Utilization of Agricultural Wastes (Part IV) Cellulosic Waste Materials as Substrate on the Production of Cellulosic Single Cell Protein. (농산폐자원의 미생물학적 이용에 관한 연구 (제육보) 섬유소단세포단백 생산에서의 천연기질의 이용성)

  • Experiments were carried out to pursue the availability and the feasibility of utilizable cellulosic materials as substrate for the production of cellulosic single cell protein. The resuluts were obtained as fellows. 1. Effects of carbolydrates as a sole carton source on the growth of Cellulomonas flavigena KIST 321 were examined. The result showed that cellulose and xylose would be most utlizable for cell mass production. 2. Alkaline treated waste papers and clothes resulted in good growth of the organism than intact ones did. However the waste papers as substrate of cellulosic fermentation were not digestible, even if the meterial was treated with alkalies. 3. Rice straw, rape straw and panic grass appeared to be good substrates for the cell mass production. 4. Leaves were proved to be a good substrate for the cell mass production, but wood sawdust was hardly digested by merely alkaline treatment. 5. When cellulosic wastes as the substrate were examined into the concentration of alkaline solution, the result suggested that the best productivity of cell mass from cellulosic materials was obtained on treatment with 0.8∼1.0% NaOH solution. 6. The productivity of cell mass was increased by washing out with water after alkaline treatment of newspaper, pine sawdust, lime sawdust and pine leaf.

Laarvicidal Acrivities of Extracts from Domestic and Japanese Plants aginst the Pine Needle Gall Midge (Diptera: Cecidomyiidae) (국내산 및 일본산 식물체 추출물의 솔잎혹파리 유츙에 대한 살충활성)

  • Methanol extracts from 79 domestic plant species in 41 families and the crude oil of one Japanese plant species were tested for their larvicidal activities against Thecodiplosis japonensis Uchida et Inouye, using filter paperimpregnated and insect-dipping methods. In a test with 10 mgtpaper, strong larvicidal activity (>90% mortality) was obtained from the extracts of roots from three domestic plants belonging to Ranunculaceae (Aconitum pseudo- leave var. eretum, A. carmichaeli and Clematis florida) and the crude oil of sawdust from a Japanese plant, Thujopsis dolabrata var. hondai (Cupressaceae). However, only the crude oil from the Thujopsis sawdust revealed strong larvicidal activity when tested with 5,000 ppm by insect-dipping method. It is concluded that the Thujopsis sawdust-derived materials could be useful as a new control agent against T. japonensis.

Characteristics of Charcoal from Wood Pellet (목질펠릿으로 제조한 탄화물의 특성)

  • The objective of this research is to develop the technique for carbonization of wood pellet and analyze a possibility for the utilization of carbonized wood pellet. The properties of wood pellet charcoals, such as density, yield, elemental composition, higher heating value, and methyleneblue adsorption, were analyzed. Wood pellet was made of sawdust of Hyunsasi-poplar, Japanese larch, Korean pine, Korean red pine, and Jolcharn-oak (serrate oak), respectively. The high density charcoal ($0.5{\sim}0.7g/cm^3$) was yielded from densified wood pellet. The carbon contents and calorific values of wood pellet charcoals were increased with the increase of carbonization temperature. The methyleneblue adsorptivity of wood pellet charcoal was similar to that of wood charcoal.

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of Agrocybe cylindracea. Sawdust spawn of media composition for optimal growth was found to be pine sawdust combination of 30% wheat bran and poplar sawdust combination of 20% corn bran were the best of the optimal combination. The optimal concentration of white sugar was 1.0~1.5%. The nitrogen sources was found to be yeast extract and soybean powder. Also, optimal concentration were $0.7g/{\ell}$ and $0.1g/{\ell}$, respectively. The mineral sources of optimal medium compositions were $MgSO_4{\cdot}7H_2O\;0.3g/{\ell}$, $KH_2PO_4\;0.5g/{\ell}$ and $K_2HPO\;1.2g/{\ell}$. Optimal amount of inoculum for cultivation of A. cylindracea were $20{\sim}25g/850m{\ell}$ and $25m{\ell}/850m{\ell}$ in the sawdust spawn and liquid spawn, respectively.

Cultural Characteristics of a new variety, 『Solhyang』 Neolentinus lepideus (잣버섯 신품종 '솔향'의 재배적 특성)

  • We aimed to develop a new variety of Neolentinus lepideus from about forty strains by bag culture. To this end, "Solhyang" which means pine smell in Korean was selected as a new commercial variety of N. lepideus. N. lepideus have yellow pileus and pine smell, which characteristics make a favorable impression on the far east Asia, Korean and Japanese. The optimum temperatures for mycelial growth and fruit body development ranged $26{\sim}32^{\circ}C$ and $18{\sim}20^{\circ}C$, respectively. The required periods of mycelial incubation and fruit body growth were 30 days and 7 days, respectively. The fresh weight of fruit body was 115g/kg with pine sawdust and corn meal power(9:1, v/v) substrate.

Effects of Dietary Pine Cone Meal on Growth Performance, Blood Characteristics, Carcass Quality and Fecal Noxious Gases Compounds in Finishing Pigs (잣 부산물의 급여가 비육돈의 생산성, 혈액성상, 도체특성 및 분내 유해가스 발생에 미치는 영향)

  • This study was conducted to evaluate effects of dietary pine cone meal on growth performance, blood characteristics, carcass quality and fecal noxious gases compounds in finishing pigs. The total of sixty [(Landrace×Yorkshire)×Duroc] pigs(86.01±0.25kg in average initial body weight) were used in 35 days assay. Dietary treatments included 1) T1(2% cottonwood sawdust), 2) T2(1% cottonwood sawdust+1% pine cone meal) and 3) T3(2% pine cone meal). There were three dietary treatments with five replicate pens per treatment and four pigs per pen. During the overall periods, there were no significant differences in ADG(Average daily gain), ADFI(Average daily feed intake) and gain/feed ratio among treatments (P>0.05). Also, Nutrient digestibilities and blood characteristics were not affected by dietary treatments. At the end of this experiment, a*-value of logissimus dorsi muscle color and sensory evaluation color were higher in T3 treatment than T1 treatment(P<0.05). In fatty acid contents of lean, C18:1 and total MUFA were significantly lower in T1 treatment than other treatments(P<0.05). However, total ω6 and total PUFA were higher in T1 treatment than T2 treatment(P<0.05). In fatty acid contents of fats, total SFA was significantly higher in T2 treatment than T3 treatment(P<0.05). C18:1 was higher in T2 treatment than T1 treatment(P<0.05). There were no significant differences in fecal noxious gas compounds among the treatments. In conclusion, the results of the experiment was affected by dietary supplementation of pine cone meal on meat color and fatty acid composition of pork in finishing pigs.

Acceleration of Mycelial growth of Lentinus edodes in Coniferous Sawdust (침엽수 톱밥에서 표고 균사생장 촉진에 관한 연구)

  • In Lentinus edodes(oak mushroom) cultivation, commonly are logs and sawdusts of oak and some other broadleaved tree species used. Recently oak trees have been substantially diminished due to extensive logging. Thus, to develop comparable synthetic formula using other tree species for the cultivation of Lentinus edodes, we investigated the effect of various nutrients and pretreatment on L. edodes mycelial growth in coniferous sawdusts(i.e., Pine and Larch). We found that 1.5 hr pretreatment of sawdust with hot water and adding 10% rice bran, 3% charcoal, 0.02% $NH_4CI$ and 0.5-1% lignosulfornic acid were effective for the growth of L. edodes in pine sawdust media. In larch sawdust pretreatment with acetone for one hr and adding 20% rice bran, 3% charcoal and 0.02% $NH_4CI$ increased L. edodes mycelial growth. We also analyzed the components of oak and coniferous sawdusts and found oak has higher content of xylose and lower content of lignin, arabinose and mannose than conifers. Rice bran, compared with BITEL(HOKKEN Co.) known for better commercial substitute for rice bran, has lower content of xylose and galactose, but the similar C/N ratio.

  • Using woodceramics made from sawdust board of pine thinning logs, changes in surface temperature were investigated, by the heating rate and keeping time at maximum temperature. The heating rate of $2^{\circ}C/min$ and keeping time at maximum temperature 1 hour, were the highest in surface temperature. Also, it was found that woodcermics maintained heat for a long time because the descending velocity of their surface temperature was slower than that of the heater.

Effect of the Addition of Binders on the Fuel Characteristics of Wood Pellets (바인더의 첨가가 목재 펠릿의 연료적 특성에 미치는 영향)

  • This work was conducted to investigate the effect of the addition of binders, such as rapeseed flour, coffee waste, bark, pine cone and lignin powder, on the fuel characteristics of the pellets fabricated with larch and tulip tree sawdust. Moisture content, bulk density and higher heating value of most pellets fabricated with the binders exceeded the 1st-grade pellet standard designated by Korea Forest Research Institute, but ash content of the pellets fabricated with rapeseed flour or bark of 10 wt% on the dry weight basis of sawdust was satisfied with just the 2nd- or 3rd-grade standard. The durability of tulip tree-pellets was positively influenced by the addition of rapeseed flour, coffee waste or lignin powder and increased with increasing the amount of the binders. For larch-pellets, the increase of binders did not greatly affect the durability, and even the durability reduced with the increase of bark or pine cone. From the microscopic observation, the obvious feature of pellet surfaces was not identified by the type of binder but by the addition amount of the binder. In summary, the addition of binders contributed to the fuel characteristics of wood pellets, and particularly the characteristics of wood pellets fabricated with coffee waste improved greatly. Therefore, if the binders are secured sufficiently with a reasonable cost, it might be possible to commercialize wood/binder pellets, which have better fuel characteristics than conventional wood pellets.

Studies on the Compositional Change of Media During Oyster Muchroom Cultivation (느타리버섯 재배기간중 배지의 성분변화에 관한 연구)

  • In order to investigate the compositional change of the lauan and pine sawdust media during the coltivation of oyster mushroom (Pleurotus ostreatus) chemical components of the two media and mushroom were analized periodically from spawning to ending. The results are summerized as follows: 1) The total yield of mushroom from the lauan sawdust media was 6.39 kg/$m^2$, and from the Pine saw-dust media 6.31 kg/$m^2$, 94% of which was produced from the first and second cropping period. 2) In both media ash content increased gradually, while organic matter content decreased as the cultivation progressed. Moisture content, which was about 79% at the time of spawning, decreased a little during the priod of mycelial propagation, after which no change was observed. 3) Total nitrogen content of the two media decreased gradually as the cultivation progressed, and total loss of insoluble nitrogen was greater than that of soluble nitrogen. Amino nitrogen content tended to increase continually. 4) In both media pentosan, $\alpha$-cellulose, lignin and C/N ratio decreased gradually, while the content of reduced sugar, trehalose and mannitol continued to increase. 5) In both media phosphorus, potassium, manganese and zinc decreased, and magnecium, calcium, iron and copper showed irregular changes. 6) CMC-saccharifying and liquefying activity gradually increased from after mycelial propagation to the second cropping, after which it decreased in both media. Xylanase activity rapidly and greatly increased during the second cropping period rather than the first period. At the start of the third cropping period the activity decreased slowly. Protease activity was highest after mycelial propagati, after which it gradually decreased. The pH of the two media decreased as the cultivation progressed. 7) In both media little change was observed in the content of the components of mushroom cropped from the first to the third period, but slight decrease was noticed at the fourth cropping.
