A Sesquiterpene, Dehydrocostus Lactone, Inhibits the Expression of inducible Nitric Oxide Synthase and TNF$\alpha$ in LPS- Activated Macrophages

  • An enhanced formation of nitric oxide (NO) is an important mediator of hypotention, peripheral vasodilation and vascular hyporeactivity to vasoconstrictor agents in endotoxaemia. And tumor necrosis factor (TNF${\alpha}$), as a primary mediator of circulatory shock has been known to induce inducible nitric oxide synthase (i-NOS), leading to excessive production of NO. We isolated two sesquiterpene lactone compounds from Saussurea lappa and their structures were elucidated as dehydrocostus lactone and costunolide. These compounds inhibit the production of both NO and TNF${\alpha}$ by LPS (1 $\mu\textrm{g}$/$m\ell$)-activated Raw 264.7 cells. NO was measured spectropho-tometrically as nitrite by the Griess reagent and TNF${\alpha}$ by ELISA. Dehydrocostus lactone (IC$\sub$50/ : 3.0 ${\mu}$M) and costunolide (IC$\sub$50/ : 4.5 ${\mu}$M) inhibited the production of NO in LPS-activated Raw 264.7 cells by suppressing inducible nitric oxide synthase enzyme expression. These compounds also decreased the TNF${\alpha}$ levels in LPS-activated system in vitro and in vivo.

Quantitative Change of Aristolochic Acid Contents by Processing Methods on the Plants of Aristolochiaceae (쥐방울과 한약의 수치에 따른 aristolochic acid 함량변화)

  • Aristolochic acid (AA) included in the plants Aristolochiaceae have been well known to be nephrotoxic and carcinogenic inducer and to cause renal disease such as Chinese Herb Nephropathy (CHN). In this study, we used a high performance liquid chromatopaphy-mass spectrometry (HPLC-MS) under the positive ion detection mode for the quantitative change of aristolochic acid-I and-II (AA-I and AA-II) in Aristolochiaceae (Aristolochia contorta Bunge, Aristolochia debilis Sieb. et Zucc., Aristolochia fangchi Wu), some related plants (Cocculus trilobus De candolle, Inula helenium Linne, Saussurea lappa Clarke), and its prescriptions (防己茯笭湯, 定喘散) with or without processing. Here, the processing methods and prescriptions in oriental medicine were generally used to alleviate toxicity or alter property of herbal medicines. However, the concentrations of AA-I and AA-II were highly determined in processed material extracts rather than unprocessed those, not measured in some related plants. Also, the concentrations of AA-I and AA-II even at the prescriptions mixed the plants of Aristolochiaceae were detected to range from 0.73 to 2.53 ppm. Thus, the present results suggest that the content of AA-I and AA-II contained to plants of Aristolochiaceae was not reduced by the processing methods or prescriptions which can induce the physico-chemical change and pharmacological transformation in traditional herbal medicines.

  • Object: Show the treatment about Jungkijeung (Zhongqizheng) by distinguishing Apoplexy(Zhongfeng), yujungfung(Leizhongfeng), gualjung(Juezheng), and observation of cause, process, quality. Method: Researched definition, cause, process, treatment and herb med through chinese & korean medical publication refered to Jungkijeung(Zhongqizheng) Result: 1. Jungkijeung(Zhongqizheng) is simmilar to Apoplexy(Zhongfeng) that spiritual shock occurred to syncope, dysarthria, trismus, quadriplegia. But the symptom of Jungkijeung(Zhongqizheng) is coldness, no-sputum, sink-pluse; that of Apoplexy(Zhongfeng) is warmness, much secretion, float-pulse. 2. Jungkijeung(Zhongqizheng) is mainly caused by the serious anger and the reverse movement of spints by the seven emotional factor. The process of Jungkijeung(Zhongqizheng) are "Fire and Fever(huore)" "Weatness and sputum(shitan)", and most importantly "Weakness of vital-qi(qixi)" 3. The treatment of Jungkijeung(Zhongqizheng) is adjustment and circulation of jiao. In early stage, don't use of Apoplexial- Med. 4. As following herb-med are used for Jungkijeung (Zhongqizheng). Sohaphang-won(Suhexiangyan) is 18 times; Palmisungi-san(Baweishunqisan) is 13 times; Kang-tang(Jiangtang) is 8 times: Mokhyangsungi san(Muxiangshunqisan) is 6 times. 5. Atractylodes macrocephala KOIDZ(Baishu) is used for 40 times most frequently, Saussurea lappa CLARKE (Muxiang), Cyperus rotundus L (Xiangfuzi), Citrus unshiu MARCOR(Chenpi), Glycyrrhiza uralensis FISCH (Gancao), poria cocos WOLF (Furing), Panax ginseng NESS (Renshen) etc are orderly used. 6. Acupoints same as GV20(Baihui), LI4(Hegu), Liv2(Xingjian), 12 Junghyul(Jingxue) is used for acupunture. And CV8(Shenque), CV4(Guanyan) is used for moxibustion. Conclusion: As Jungkijeung(Zhangqizheng) is differed from Apoplexy(Zhongfeng), yujungfung(Leizhongfeng), gualjung(Juezheng), we must also cure Jungkijeung (Zhongqizheng) to the other disease. It is need to distinguishment Jungkijeung(Zhongqizheng) from neurotic, psychotic disease, though similar to conversation neurosis.

The effects of herbs on the radiation-induced apoptosis in intestinal crypt cells (방사선 조사 마우스에서 소장움세포의 Apoptosis 발생에 미치는 생약의 효과)

  • This study was performed to determine the effect of several herbs on radiation-induced apoptosis in jejunal crypt cells. Longyanrou(Euphoris longana), Suanzaoren(Zizyphus vulgaris), Yuanzhi(Polygala tenuifolia), Rensan(Panax ginseng), Fuling(Poria cocos), Muxiang (Saussurea lappa), Chuanxiong(Cnidium officinale), Baishaoyao(Paeonia lactifolia), Shengma (Cimicifuga heracleifolia), Chaihu (Bupleurum falcatum) and Dongchongxiacao ( Paecilomyces japonica) reduced the frequency of radiation-induced apoptosis(p<0.05). Although the mechanisms of this effect remain to be elucidated, these results indicated that Longyanrou, Suanzaoren, Yuanzhi, Rensan, Fuling, Muxiang, Chuanxiong, Baishaoyao, Shengma, Chaihu and Dongchongxiacao might be useful inhibitors of apoptosis, especially since these are relative nontoxic natural products.

The Radioprotective Effect of Kuei-Pi-Tang as a prescription of Traditional Chinese Medicine in Mice (생쥐에서 귀비탕 및 구성단미의 방사선 방호효과)

  • In this study, we evaluated the radioprotective effects of Kuei Pi Tang and its ingredients, as a prescription of traditional Oriental medicine. The jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells were investigated in mice irradiated with high and low dose of ${\gamma}$ rays. In the mice administered with Kuei Pi Tang extract before irradiation, the jejunal crypts were protected significantly(p<0.005) and the frequency of radiation induced apoptosis was reduced(p<0.05). In the experiment on the effects of the ingredients of Kuei Pi Tang, the results indicated that the extract of Dangui(Angelica sinensis), Suanzaoren(Zizyphus valgaris), Rensan (Panax ginseng), Fuling(Poria cocos) and Muxiang(Saussurea lappa) might have a major radioprotective effect. Although the mechanisms of this inhibitory effect remain to be elucidated, these results indi cated that Kuei Pi Tang might be a useful radioprotector, especially since it is a relatively nontoxic natural product.

Anti-Inflammatory Effects of Volatile Flavor Extracts from Cnidium officinale and Angelica gigas (천궁(Cnidium officinale) 및 당귀(Angelica gigas) 휘발성 향기추출물의 항염증효과)

  • The effects of volatile flavor extracts of eight different herbal medicines, Juniperus rigida (JR), Saussurea lappa SL), Cnidium officinale (CO), Angelica gigas (AG), Eugenia caryophyllata (EC), Angelica tenuissima (AT), Mentha arvense (MA), and Artemisiae argyi (AA), were investigated on LPS-stimulated inflammation using Raw 264.7 cells. The volatile flavor extracts of CO and AG considerably inhibited LPS-stimulated NO, $PGE_2$, IL-6, and TNF-${\alpha}$ (except AG) production, as well as iNOS expression. Major volatile components of CO were identified as ligustilide and of ${\beta}$-eudesmol as AG by GC-MS analysis. Thus, these results suggest that the volatile extracts of CO and AG may be useful as potential therapeutic agents for inflammation-associated disorders.
