  • 한국에서 자생하는 삼백초(Saururus Chinensis)의 지상부에서 Quercetin 배당체인 Hyperin, Isoquercitrin, Quericitrin, Rutin등 4종의 Flavonoid들이 고성능 액체 크로마토그래피(HPLC), 기체 크로마토그래피(GC) 및 얇은막 크로마토그래피(TLC)를 이용하여 분리, 확인되었으며 주요 Flavonoid 성분으로 알려져 있는 Avicularin은 검출되지 않았다.

Isolation of Dineolignans, Saucernetin-7 and -8, with Nitric Oxide Inhibitory Activity and NMR assignment from Saururus chinensis

  • Two dineolignans (1,2) with nitric oxide inhibitory activities were isolated from Saururus chinensis (Saururaceae) using silica gel column chromatography. Although the structures, saucernetin-7 (1) and -8 (2), have been already reported, NMR assignment of the two compounds was completed aided by 2D-NMR spectroscopy including $^1H-^1H$ COSY, $^1H-^{13}C$ COSY, HMBC and NOESY NMR spectra. Compounds 1 and 2 exhibited significant nitride oxide inhibitory activity in LPS-induced RAW 264.7 cells with $IC_{50}$ values of $11.3{\mu}M\;and\;7.1{\mu}M$, respectively.

삼백초와 식물 2종의 지역개체군별 RAPD 분석 (RAPD Analyses on the Regional Populations of Two Species of Saururaceae in Korea)

  • 삼백초과식물 2속 2종에 대한 30개 지역별 개체군의 유전적 상관관계를 알아보기 위해 RAPD 분석을 실시하였다. PCR 과정을 통해 증폭된 RAPD 절편들은 300bp에서 2,000bp 사이의 구간에서 관찰되었다. 총 16개의 oligoprimer를 이용한 효소중합반응에서 156개의 유효한 polymorphic band makers를 확인하였고 이러한 자료에 근거하여 2종의 종내 개체군 군집에 대한 UPGMA phenogram을 도출하였다. 이 결과 유집형태에 근거하여 재배지 개체군과 자연집단 개체군과의 구분이 가능하였고 또한 지역별 개체군들끼리 유집되는 결과를 보였다. 2종 모두에서 지역별 개체군중 경남의 개체군과 제주도의 개체군이 전남의 개체군에 비해 유연관계가 밀접한 것으로 밝혀졌다. 따라서 RAPD 분석은 삼백초과 식물의 개체군별 유연관계를 분석하거나 재배집단과 자연집단을 분석하는데 유용한 실험적 방법으로 생각된다.

삼백초와 약모밀의 내부형태 비교 연구 (A Comparative Anatomical Study of Saururus chinensis (Lour.) Baill. and Houttuynia cordata Thunb. (Saururaceae))

  • Objectives : Saururus chinensis and Houttuynia cordata (Saururaceae) are perennial herbs using for medicinal purposes in Korea. The objectives of this study are to compare anatomical key characters between two medicinal plants and to provide fundamental information for the identification of two herbal medicines by using anatomical features. Methods : Cross-sections of root, rhizome, stem, petiole, and leaf for each species were observed in this study. Materials were analyzed through dehydration, paraffin embedding and micro-sectioning, and double staining with Safranin O and Fast-Green FCF. Observations of permanent preparation were conducted using light microscope. Results : S. chinensis and H. cordata were distinguished with anatomical differentiations; Idioblasts with essential oil were scattered in the parenchyma cell of cortex, pith, and phloem of S. chinensis, on the other hand, in H. cordata, idioblasts were distributed ring-shaped in the cortex of the root. S. chinensis had two cycles of vascular bundles in the stem while H. cordata had one cycle. Hypodermis layer was conspicuous in a stem of H. cordata, crystals were observed the only parenchyma in a stem of S. chinensis, and epidermal oil cells were developed in the epidermis of H. cordata. S. chinensis had air cavity at the cortex and pith of the stem. The shape of cross-section was polygonal in the stem of S. chinensis and was circular in the stem of H. cordata. Conclusions : We investigated anatomical study of Korean S. chinensis and H. cordata. To identify two herbal medicines, we considered main anatomical features and provided identification key here.

삼백초의 진통성분 (Analgesic Constituent from the Herba of Saururus chinensis ($L_{OUR.})B_{AILL.}$)

  • For the investigation of bioactive natural products with analgesic activity, we have evaluated various extracts of Saururi Herba (Saururaceae), which has been used in traditiona l medicine for edema, beriberi, jaundice, turbid urine, carbuncle and furuncle. The diethylether extract of this plant was found to show a significant analgesic effect on writhing syndrome in mice. Using bioactivity-guided separation of the diethylether extract, analgesic constituent, (8S, 8`R, 7R, 6`R)-2'-oxo-4,5: 4',5'-bis(methylenedioxy)-${\Delta}^{1,3,5,3'}$-8.8', 7.6', 2.O.5'-neolignan(sauchinone) was isolated and structurally identified by physico-chemical properties and spectroscopic evidences. This compound has good analgesic activity with lower toxicity, as compared to other antipyretic-analgesic drug(acetaminophen).

Inhibitory Effects of Extracts of Houttuynia cordata Thumb (Saururaceae) on Cytokine Release of Human Monocytic THP-1 Cells and Human Eosinophilic EoL-1 Cells

  • In the present study, we investigated whether Houttuynia cordata Thumb (Saururaceae; HC) extracts have an anti-inflammatory effect in human monocytic THP-1 cells and human eosinophilic EoL-1 cells. The dried and powdered whole plants of HC were extracted with 80% EtOH. The combined extract (HC-1) was concentrated under reduced pressure. The residue was diluted with water, and then successively partitioned with n-hexane, EtOAc, and BuOH to produce the n-hexane (HC-2), EtOAc (HC-3), BuOH (HC-4), and the water-soluble fractions (HC-5), respectively. HC extracts have no cytotoxicity on THP-1 cells and EoL-1 cells at a high concentration of $10\;{\mu}g/ml$ for 24 h, except HC-2 extract ($10\;{\mu}g/ml$). Interleukin-6, Interleukin-8 and Monocyte chemoattractant protein-1 in THP-1 cells were increased after the treatment with the extract from house dust mite or LPS. The increase of cytokine production was strongly suppressed by HC-3 extract, in comparision with other extracts. HC-3 also had inhibitory effect on Interleukin-6 production increased by mite extract and LPS in EoL-1 cells. However, HC-3 extract increased Interleukin-8 production induced by mite extract and LPS in EoL-1 cells. These results suggest that HC extracts may be used as useful agents for treating allergic disorders such as asthma and atopic dermatitis.

  • In our search for bioactive natural products with antitumor activity, we have valuated various extracts of Saururi Herba (Saururaceae), which has been used in traditional medici ne for edema, beriberi, jaundice, turbid urine, carbuncle and furuncle. The hexane extract of the aerial part of this plant was found to show a potent cytotoxicity against several kinds of cultured human solid tumor cell lines (AGS, A549, HCT15, SKOV3, HEP-3B) in vitro. Using cytotoxicity-guided chromatographic separation of the hexane extract, cytotoxic constituent: 10-aminomethyl-3-hydroxy-4-methoxyphenanthrene-carboxylic acid lactam, was isolated and structurally identified by physico-chemical properties and spectroscopic evidences.
