Detection of Laminariaceae Species Based on PCR by Family-specific ITS Primers

  • To analyze nucleotide sequence encoding internal transcribed spacer (ITS) regions specific to the Laminariaceae family, genomic DNA was isolated from six brown algae species distributed along the east coast of Korea. These included three species from the Laminariaceae family (Agarum clathratum Dumortier, Costaria costata [C. Agardh] Saunders, and Saccharina japonica Areschoug) and two species from the Alariaceae family (Undaria pinnatifida [Harvey] Suringer and Ecklonia cava Kjellman), both in the order Laminariales, and one species from the family Sargassaceae in the order Fucales (Sargassum serratifolium). Based on a sequence analysis of ITS-1 and ITS-2 for A. clathratum, C. costata, and E. cava, oligonucleotides were designed from the regions that showed sequence conservation in Laminariaceae. Following polymerase chain reaction using three sets of primers, amplification of ITS-1 and ITS-2 was detected in reactions using genomic DNA isolated from the species belonging to Laminariaceae, but not from the species belonging to the other families. The results indicate that this method can be used for the detection and identification of Laminariaceae species.

국내산 다시마, 미역포자엽, 톳, 모자반 정제 fucoidan의 구성당 결합 특성 (Structural Characterization of Purified Fucoidan from Laminaria religiosa, Sporophylls of Undaria pinnatifida, Hizikia fusiforme and Sagassum fulvellum in Korea)

  • 국내산 주요 갈조류인 미역, 다시마, 톳, 모자반 정제 fucoidan의 기본 결합 구조 및 황산기 결합 위치를 확인하기 위하여 정제 fucoidan및 이를 methanolysis하여 얻은 고분자 획분을 각각 methylation 및 FT-IR 측정을 하였다. 다시마, 톳, 모자반, 미역포자엽 정제 fucoidan의 주 결합은 $(1\rightarrow3)$ linked fucose였으며 미역포자엽에는 $(1\rightarrow3)$ linked galactose도 상당량 존재하였다. 황산기의 대부분은 fucose와 galactose의 C-4의 위치에 결합되어 있는 것으로 추정된다.

  • This research dealt with the nutrient value of the seaweed, Sargessum horneri (Turner) C. Ag. which showed the highest nutrient value in the previous exporiment (Park 1968). Its objective was to find the most effective mixing ratio of ordinary chicken feed used for pullet. In order to investigate the most effective mixing ratio, seven kinds of sample feed with vary-ing amounts of the seaweed as shown in table 2 were prepared and were fed to seven groups of chickens, each group consisting of twenty, four week old, pullets. In addition, the effect of sodium glutaminate mixed in the feed was also investigated, and the following result was obtained. The average increase in body weight of each group during the experiment period was determined. The group fed with the feed containing $10\%$ powdered Sargassum horneri (Turner) C. Ag. showed the highest body weight increase. In this experiment the body weight increase was accelerated by mixing $0.5\%$ sodium glutaminate In the above feed.

해조류 및 해양저질에서 항 염증성의 방선균 선발 (Screening Anti-inflammatory Actinomycetes Isolated from Seaweeds and Marine Sediments)

  • The anti-inflammatory activities of marine actinomycetes were surveyed. In total, 363 strains were isolated from marine sediments, seaweed tissue, and seaweed rhizosphere. Of these, strains 16 and 291-11 showed the most potent anti-inflammatory activity in phorbol-ester-induced mouse ear edema and erythema assays. Strains 16 and 291-11 were isolated from the rhizosphere of the brown seaweeds Sargassum thunbergli and Undaria pinnatifida, respectively, and were identified as Streptomyces macrosporeus and St. praecox, respectively, using 165 rDNA sequence analysis.

Growth Enhancement of the Microalga Tetraselmis suecica by an Extract of the Green Alga Monostroma nitidum

  • Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.

Biodiversity and Community Structure of Marine Benthic Organisms in the Rocky Shore of Dongbaekseom, Busan

  • Species composition, community structure and biodiversity of marine benthic community were studied in the rocky shore of Dongbaekseom, Busan. A total of 82 species of marine algae including 4 Cyanophyta, 11 Chlorophyta, 20 Phaeophyta and 47 Rhodophyta are listed. The dominant algal species were Ulva pertusa, Chondria crassicaulis, Corallina spp. and Melobesioidean algae. Sargassum thunbergii, Chondracanthus intermedia, Gelidium divaricatum and Ralfsia verrucosa were subdominant in cases of different seasons and vertical layers. Chthamalus challengeri, Littorina brevicula and Mytilus edulis were dominant zoobenthic species upper-middle layer of the intertidal zone. The community structure of this area seemed to be controlled by spatial competition with benthic marine algae. The species diversity indices estimated from different sources were quite different. Indices from coverage were 1.87, 3.98 from frequency, 2.26 from the average of total frequency and coverage and 2.15 from importance value. The similarity indices on the present study showed decreasing trends comparing to the previous benthic algal composition researches.

남서해안 신안군 무인도서의 하계 해조식생 (Summer Marine Algal Vegetation of Uninhabited Islands in Sinangun, Southwestern Coast)

  • The objective of this study was to clarify marine algal flora and community structure of 16 uninhabited islands in Sinangun area, south western coast of Korea. As a result, a total of 63 species - 11 greens, 16 browns and 36 reds - was identified. The functional groups of marine algal species were filamentous 14.3%, foliose 20.5%, corticated 45.3%, leathery 13.7%, and articulated calcareous algae 6.2%. The (R+C)/P value was calculated as 2.9 showed temperate flora. The result of DCA showed that 16 uninhabitated islands tend to be distributed eastern and western parts. The dominant species was Ulva pertusa, while the subdominats were Gelidium divaricatum, Myelophycus simplex. The vertical distribution pattern of intertidal marine algae represented three distinct zones; Gloiopeltis furcata, Myelophycus simplex, Gelidium divaricatum - Ulva pertusa, Enteromorpha spp., Ishige okamurae - Ulva pertusa, Sargassum thunbergii, Symphyocladia latiuscula.

  • 갈조 모자반속 식물의 괭생이모자반은 1년생으로 조하대에 생육하는 해중림 구성 해조로서 울릉도와 제주도를 포함 우리나라 동ㆍ남ㆍ서해에 널리 분포하는 종이다(Kang, 1966). 이종의 생활사에 관하여는 Uchida(1993)가 실내배양을 통하여 밝힌 바 있으며, Okuda(1987)에 의해 자웅동주개체와 가을철의 성숙에 관한 생리. 생태가 밝혀졌으며, 실내배양을 통하여 가을에 성숙하는 조체의 난방출, 배발생 및 광합성속도의 계절변화가 밝혀졌고(Honda와 Okida, 1989), 곡구ㆍ산전(1988)에 의해서 마츠시마만에서의 괭생이모자반 군락의 주년변화와 생산력에 관한 생태가 밝혀지는 등 특히 이종을 식용으로 하는 일본을 중심으로 다양한 분야에 걸쳐서 많은 연구가 이루어져 왔다. 해조류 및 생약의 Tyrosinase 억제활성 검색 (Screening of the Tyrosinase Inhibitors from Marine Algae and Medicinal Plants)

  • We have tested tyrosinase inhibitory activity on the methanol extracts of 23 species marine algae and 23 species traditional medicinal plant. Among them, four medicinal plants, Ephedra sinica, Atractylodes japonica, Pinnelia ternata, and Citrus aurantium, showed strong inhibition potency over 90% at concentration of 0.33 mg/mL. Also, two marine algae, Enteromorpha compressa and Sargassum singgildianum showed mild inhibition potency over 50% at concentration of 0.33 mg/mL.

Vertical Distribution and Seasonality of Intertidal Macroalgae on the Coast of Hawon-Pando, Southwestern Korea

  • An overview of the spatial and seasonal patterns of phytobenthic communities is described from the intertidal zone at Hawon-Pando on the southwestern coast of Korea based on quantitative and qualitative estimates of macroalgae. There were considerable variations of macroalgal diversity and cover value with the intertidal levels. Tn general, the number of species decreased with increasing intertidal height. The upper region was dominated by Sargassum thunbergii. S. thunbergii and crustose red algae were dominant in the mid intertidal habitat. In the lower intertidal Corallina pilulifera and crustose red algae were the conspicuous dominants. Macroalgal community structure at Hawon-Pando is discussed in the context of functional-form groups. The Hawon-Pando intertidal is characterized as consisting of a relatively high cover of species assigned to the thick leathery, crustose and jointed calcareous groups, and is distinguished by a paucity of the filamentous group.
