Synthesis, Antitumor Activity and Release Rate of Polymers Containing Anionic Group and 5-Fluorouracil

  • Poly(exo-3,6-epoxy-1,2,3,6-tetrahydrophthalic anhydride)s [poly(ETA)s] and poly($\alpha$-ethoxy-exo-3,6-epoxy-1,2,3,6-tetrahydrophthaloyl-5-fluorouracil)s [poly(EETFU)s ] with various average molecular weights were prepared by photopolymerizations. The number average molecular weights of the fractionated poly(ETA)s and poly(EETFU)s determined by GPC were in the range of 3,600∼21,000 and 3,600-33,400, respectively. The release rate of 5-FU from poly(EETFU) decreased with increasing average molecular weight. The in vitro cytotoxicity of poly(ETA) against a normal cell line was lower than that of 5-fluorouracil(5-FU), The in vivo antitumor activities of the synthesized samples at dosage of 0.8 mg/kg against mice bearing sarcoma 180 tumor cell line decreased in the following order: poly(EETFU) > poly(ETA) > EETFU > ETA > 5-FU. The antiangiogenic activities of the poly(ETA)s were better than those of 5-FU.

Anti-cancer Effects of Kamiboa-tang and some other Traditional Medical Prescriptions (가미보아탕(加味保我湯) 및 수종(數種) 한방처방의 항암효과에 대한 연구)

  • Objectives : Anticancer and immune-modulating effects of several Korean medical prescriptions including Yukgunja-tang, Bohwa-tang, Sogam-Won, and Kamiboa-tang were investigated. Methods : In vitro anti-cancer effects were measured by cytotoxicity MTT assay using SNU-1 gastric cancer cell lines, In vivo anti-cancer effects were measured by increased life span of S-180 sarcoma-injected ICR mouse. Immune-modulating effects were analyzed by measuring hemagglutinin titer, appearance of rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice. Results : In vitro assay showed that only Sogam-won showed cytotoxic effect with $IC_{50}$ of 87.9 ${\mu}g/ml$. All other prescriptions showed no cytotoxic effects against SNU-1 gastric cancer cell line. However, in vivo assay showed that Sogam-won showed lowest anti-cancer effects in contrast to its highest cytotoxic effects, Kamiboa-tang, which showed no cytotoxic effect, showed the highest in vivo anticancer effects, with increased life span of 140%. Kamiboa-tang showed significant immune-enhancing activities by significantly increasing rosette forming cells, lymphocyte proliferation, and phagocytic index in methotrexate-pretreated mice (P<0.05). Conclusion : The anticancer effect of Kamiboa-tang is not mediated by direct inhibition of cancer cells but is mediated by improving immune reactions against cancer cells.

Studies on the Efficacies of Water Extract of Propolis (프로폴리스 물추출물의 약효연구)

  • Propolis is a lipophilic, natural product prepared by mixing the exudates collected from various plants by honeybees with beeswax for the purpose of using to seal hive walls and to strengthen the borders of combs. Because of its versatile bioactivities, propolis has been attracting many investigators'interest. But the pharmacological studies on propolis has, to date, been exclusively performed for an alcohol extract, there is few information of water extract. Therefore, in this study, we investigated the various effects of water extract of Chinese propolis. The water extract of propolis and its fractions of organic solvents showed strong antioxidative activities, especially ether and ethylacetate fractions, and reduced the lipid peroxidation of rat liver in viro. Additionally the ether fraction of propolis (10 mg/ml) inhibited the activity of hyaluronidase by 50%. In vivo, the water extract of propolis considerably decreased s-GOT, s-GPT and s-LDH activities which represent for the hepatotoxicity induced by $CCl_4$ in rats, and prolonged the MST (Medium revival tinge) and ILS (Increasing in MST over control) by 18% in mice which inoculated with sarcoma 180 ascites cells. These results suggest that the water extract of propolis has various bioactivities as well as the alcohol extract.

  • To find pharmacologically active hybrids among the inter-order protoplast fusants of Lentinula edodes and Ginoderma lucidam the antitumor test was performed and the fusant P22 was selected among them. The hot water and alkaline extracts from the cultured mycelia of P22 were purified and separated into four fractions by DEAE-cellulose anion exchange chromatography. When a dose of 20 mg/kg/day of each fractions was injected into ICR mice by i.p., the tumor inhibition ratio of Fr. IV against solid sarcoma 180 was the higher than any other fraction. Fr. IV was a protein-bound polysaccharide which was composed of 69. 12% polysacchafide and 9.76% protein and the molecular weight of Fr. IV was $6.7{\times}10^4$ dalton.

Antitumor and Immunopotentiating Effects of Manda Enzyme

  • This study was to evaluate the antitumor and immunopotentiation effects of Manda Enzyme (ME). Oral administration of ME (0.2ml/mouse) to tumor bearing mice significantly prolonged survival rate compared to the control group with the prolongation ratio of 40%. The inhibition ratios for the first and the second experiments were 51.8% and 26.4%, respectively. Only the spleen index was significantly increased in the MEF-treated group, but not in the control group. Gamma globulin level of the MEF-treated group was elevated when mice were injected with sarcoma-180 cells on the left groin. Activities of natural killer (NK) and lymphokineactivated killer (LAK) cells were observed by $^{51}Cr-release$ method. Activities of NK cell against YAC-1 cells were significantly increased in the MEF treated group. And LAK cell activities against P815 cells were also significantly increased in the experimental group. These observations, therefore, suggest that ME may have an anticancer effect and immunopotentiating effect in vivo.

Augmentation of Immune Responses by Oral Administration of Gynostemma pentaphyllum Ethanol Extract (돌외 에탄올 추출물의 생체방어력 증진효능)

  • The immunomodulatory activities of the ethanol extract of Gynostemma pentaphyllum, termed hereafter as GPE, were examined in immunosuppressed mice as well as in normal mice in the present study. Oral administration of GPE into mice prevented dexamethasone (DEX)-induced immunosuppression as determined by the mitogen-induced proliferation of the splenocytes and the the cytokine production (TNF-$\alpha$, IL-$1{\beta}$) in the whole blood culture. In addition, oral administration of GPE increased antitumor host defense in mice implanted with sarcoma-180 tumor cells. The immunoaugmenting activity of orally administered GPE was also confirmed in mice immunized with ovalbumin (OVA). Mice that were orally administered with GPE generated much more potent OVA-specific cytotoxic T lymphocyte (CTL) responses upon intravenous OVA injection compared to the untreated controls. These results demonstrate that oral administration of the ethanol extract of Gynostemma pentaphyllum could be useful to increase host defense in immunocompromised situations such as stress- or tumor-induced immunosuppression.

  • To investigate antitumor constituents of higher fungi, the carpophores of Polyporus giganteus Pers. ex. Fr. (81 g, dry weight) which were collected in Indiana, U. S. A. were examined for antitumor activity. Two protein-bound-polysaccharide fractions (I and II) were prepared from the hot water extract and one fraction (III) from the 0.1 N NaOH extract of the carpophores. The antitumor effect of each fraction was tested against sarcoma 180 implanted subcutaneously in female ICR mice. Of three fractions, Fraction II showed 85.2% inhibition ratio at the dose of 20 mg/kg/day for 10 days and was named gigantan. Gigantan was found to contain 59% polysaccharide and 27% protein. Its polysaccharide moity was a heteroglycan that consisted of mainly glucose (89.3%), galactose (7.7%), minaose (2.0%) and fructose (1.0%).

  • The crude polysaccharide from Panax ginseng prepared by hot water extration and precipiation with ethanol was further fractionated into neutral and acidic fractions by DEAE- cellulose ion exchange chromatography. The chemical compositions were 85.0% carbohydrorate and 15.0% protein for the neutral fraction, and 28.4% carbohydrate, 10.0% protein and 29.0% uronic acid for the acidic fraction. The acidic fraction was more effective in increasing of the ratio of spleen to body weight, the number of antibody secreting cells to SRBC and phagocytic activity of reticuloendothelial system, as well as antitumor activity against the solid form of sarcoma 180 in ICR mice than the neutral fraction. All polysaccharide fractions were mitogenic to cultured spleen cells of C57BL/6 mice. However, FA was different from FN in the co-mitogenicities with lectin mitogens. Both crude and acidic fractions potentiated remarkably the mitogenic activity of PHA-P or LPS in dose-dependent manner but neutral fraction enhanced only that of LPS. Three polysaccharide fractions had no effect on that of Con A. These results suggest that the acidic fraction may stimulate B and Td cells as well as macrophages while the neutral fraction may simulate only B cells and macropages.

  • To find anititumor metabolites in Korean basidiomycetes, the shake-cultured mycelia of eight of the higher fungi were extracted with hot water and the extracts, after being partially purified, were subjected to in vivo antitumor test. When administered i. p. at the dose of 30mg/kg/day for ten consecutive days into the female ICR mice, which had been implanted with $1{\times}10^{6}$ / cells of sarcoma 180 twenty four hours before the first injection, the extracts of Agaricus campestris, Lyophyllum decastes, Lyophyllum ulmarium, Armillaria Tabescence and Calvatia exipuliformis respectively showed inhibition ratios of 64.1%, 65.45, 60.-%, 53.0 and 49.3%. These five species were selected for further study, whereas the extracts of Phallus impudicus, Coprinus comatus and Pholiota squarrosa whih showed the inhibition ratios of 31.2%, 33.5% and 19.0% were discontinued.

Immunoactivities of the Protein-Polysaccharides of the Tips of the Growing Carpophores of Ganoderma lucidum (영지버섯(Ganoderma lucidum) 생장점으로부터 분리한 단백다당류의 면역활성)

  • Two protein-polysaccharide fractions, GLA and GLB, respectively, were prepared from the pileus of the fully grown carpophores and the tips of the growing carpophores of Ganod erma lucidum. At a dose of 100mg/kg/day ip, GLA and GLB inhibited the growth of sarcoma 180 solid tumor in ICR mice by 56.3% and 81.8%, respectively. In a flow cytometric (FCM) analysis, GLA and GLB enhanced the formation of lymphoblasts of BALB/c, splenic leukocytes at a concentration of 100 ${\mu}$g/ml, by 38.3% and 61, 3%, respectively. When ip injected into ICR mice, GLB exerted anti-leukopenic effect against cyclophospamide (75mg/kg, ip) in that the leukocyte counts of the peripheral blood of the normal and the cyclophosphamide-treated mice. respectively. was (11.1 ${\pm}$ 3.8) ${\times}$ 10$^3$ and (4.0 ${\pm}$ 1.8) ${\times}$ 10$^3$, while the GLB-cyclophosphamide treated mice showed a leukocyte count of (10.8 ${\pm}$ 5.1) ${\times}$ 10$^3$ CELLS/${\mu}$l. These results suggest that GLB is a promising candidate for an effective, cancer immunotherapeutic agent.
