Chemical Composition and Pharmacological Activities of Vietnamese Ginseng, Panax viehamensis

  • From the underground part of Panax vietnamensis Ha et Grushv., commonly known as Vietnamese ginseng, 50 triterpene glycosides including 24 new dammarane saponins named visa-ginsenosides-Rl-24 were isolated and identified. The structure of the new saponins was elucidated based on chemical and spectroscopic evidence. The saponin composition of Vietnamese ginseng is almost similar to that of ginseng (Panax ginseng C.A. Meyer) and other cultivated Panax spp. However, the content of ocotillos-type saponins, especially that of the major saponin, majonoside-R2 (5.29% yield), was surprisingly very high. The pharmacological activities of Vietnamese ginseng are essentially similar to those of ginseng. In addition, it has marked antibacterial activity against pathogenic Streptococcus app. and is effective in treatment of granular angina. The chemical composition and pharmacological activities have made Vietnamese ginseng an interesting member of Panax spp. from chemotaxonomical and pharmacological points of view,

Inhibitory Effects of Ginseng Total Saponins on Hypoxia-induced Dysfunction and Injuries of Cultured Astrocytes

  • The effects of ginseng total saponins (GTS) on hypoxic damage of primary cultures of astrocytes were studied. Hypoxia was created by placing cultures in an air tight chamber that was flushed with 95% $N_2/5%CO_2$ for 15 min before being sealed. Cultures showed evidence of significant cell injury after 24 h of hypoxia (increased lactate dehydrogenase (LDH) content in the culture medium, cell swelling and decreased glutamate uptake and protein content). Addition of GTS (0.1, 0.3 mg/ml) to the cultures during the exposure to hypoxic conditions produced dose-dependent inhibition of the LDH efflux. GTS (0.1, 0.3 mg/ml) also produced significant inhibition of the increased cell volume of astrocytes measured by $[^3H]$ O-methyl-D-glucose uptake under the hypoxic conditions. Decreased glutamate uptake and protein content was inhibited by GTS. These data suggest that GTS prevents astrocytic cell injury induced by severe hypoxia in vitro.

  • Saponins in leaf and stem of Gynostemma pentaphyllum that was collected from Jeju Island were extracted by the method for ginseng saponin. Comparison by overlapping chromatogram(HPLC) of pentaphyllum on that of ginseng and cochromatogram and ginseng although ginsenoside Rg2, Rg1 and Rf might be in common with rare possibility. It seems to be little difference in the kind of saponin glycosides between leaf and stem of pentaphyllum. Saponin content in leaf of pentaphyllum was higher than in stem, and much higher than those in ginseng. The kind of saponin glycoside in pentaphyllum appeared to be less than 22 and greater than those in ginseng. There was almost no change in saponins of pentaphyllum in methanol for 3 years at room temperature.

  • Saponins of Gynostemma pentaphyllum that collected from Geochang region(Korea) and Dokujima(Japan) were extracted by the method for ginseng saponin. Comparison by retention time in chromatogram(HPLC) of G. pentaphyllum to that of ginseng showed that it is hardly to find out a common saponin between penta-phyllum and ginseng saponins. Saponin content extracted from G. pentaphyllum growing in Geochang region was higher than that growing in Dokujima region.

  • 고양이 말초혈액 다형핵백혈구(PMNC)의 유주성에 있어서 인삼 saponins(ginseng total ssponin(GTS), ginseng PT saponin(GfT) 및 ginseng PD saponin(GPD))의 면역증강 효과를 검토하였다. PMNC에 대한 유주성을 hoyden chamber 변법으로 측정한 결과, GTS, GPT, GPD는 PMNC에 대해 직접적인 유주활성을 나타내지 않았다. 또한 인삼 saponins을 첨가하여 배양한 PMNC 배양상충액의 경우게는 미약한 유주활성을 나타내었다. 그러나 GPf및 GPD를 첨가하여 배양한 말초혈액 단핵구세포(MNC)배양상충액에서는 PMNC의 유주활성이 현저하게 증가하였다. PMNC에 대해 유주활성이 인정된 GPI)로 배양한 MNC 배양상충액을 이용하여 checkerboard assay를 실시한 결과 배양상충액의 농도차에 의존하는 진의 유주활성임을 알 수 있었다. 이상의 결과로부터 인삼 saponine 중 GPT 및 GPD가 고양 이 말초혈액 다형핵백혈구의 유주성을 중강시키는 효과가 있으며, 이것은 이들 saponins에 의해 활성화된 단핵구세포에서 분비되는 유주성인자에 의해 다형핵백혈구의 유주활성이 증 강되는 것으로 사료되었다.

  • Crude ginseng saponin fraction reduced the production of thromboxane $A_2$in the lipopolysaccharide-stimulated macrophages. Several kinds of crude saponins showed variant potency that might be caused by the compositional difference of ginseng saponins. From the metabolic labeling experimental data, this reduction of thromboxane $A_2$formation, at least in part, resulted from the reduction of protein synthesis of inducible isozyme of cyclooxygenase(COX-2). This activity may be resulted from the fact that ginseng saponins have steroidal moiety in their structures.

  • Two new triterpenoid saponins, named segetoside D and E, have been isolated from the seeds of Vaccaria segetalis. On the basis of chemical reactions and spectral data, structures of segetoside D and E have been established as: $28-O-[{\beta}-D-xylopyranosyl-(1{\rightarrow}4)-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}2)]-[5-O-acetyl-{\alpha}-arabinofuranosyl(1{\rightarrow}3)]-[4-O-acetyl-{\beta}-D-fucopyranosyl]-quillaic\;acid-3-O-[{\beta}-D-galactopyranosyl(1{\rightarrow}2)]6-O-methyl\;ester-{\beta}-D-glucuronopyranoside$ and $28-O-[{\beta}-D-xylopyranosyl-(1{\rightarrow}4)-{\alpha}-L-rhamnopyranosyl-(1{\rightarrow}2)]-[5-O-acetyl-{\alpha}-arabinofuranosyl(1{\rightarrow}3)]-[4-O-acetyl-{\beta}-D-fucopyranosyl]-quillaic\;acid\;-3-O-[{\beta}-D-galactopyranosyl(1{\rightarrow}2)]-6-O-n-butyl\;ester-{\beta}-D-glucuronopyranoside$, respectively.

Effect of Crude Saponins from Soybean Cake on Body Weight and Glucose Tolerance in High-Fat Diet Induced Obese Mice (대두박 사포닌 보충이 식이성 유도 비만마우스의 체중과 내당능에 미치는 영향)

  • This study investigated the beneficial effects of crude saponins from soybean cake on body weight and glucose tolerance in high-fat (37% calories from fat) diet fed C57BL/6 mice. The mice were supplemented with three doses of saponins (0.5%, 1.0%, and 1.5%, wt/wt) and 1.0% Garcinia cambogia (wt/wt), positive control for 9 weeks. The body weight, visceral fat weight and epididymal adipocyte area were significantly reduced in the saponin supplemented groups in a dose dependent manner compared to the high-fat group. Saponins did not significantly affect food intake; however, cambogia significantly lowered food intake compared to the high-fat fed control group. The crude saponins from soybean cake supplement significantly lowered plasma leptin, triglyceride and total cholesterol levels, whereas they significantly elevated the fecal excretion of triglyceide in a dose dependent manner compared to the high-fat group. Cambogia did not affect the fecal excretion of lipid in the diet-induced obese mice. Supplementation of 1.5% saponin reduced the hepatic triglyceride content compared to the high-fat group. High-fat induced glucose intolerance with the elevation of blood glucose levels compared to the normal group; however, the saponins supplement significantly improved postprandial glucose levels. After 9 weeks of being fed a high-fat diet, the mice presented with significantly increased activities of hepatic fatty acid synthase and fatty acid ${\beta}$-oxidation; however, saponins and cambogia normalized these activities. These results indicate that saponins from soybean cake exhibit a potential anti-obesity effect and may prevent glucose intolerance by reducing body weight and plasma lipids, increasing fecal lipid excretion and regulating hepatic lipid metabolism in high-fat fed mice.

  • This study was carried out to investigate effects of fat-soluble solvents on the purification against nan-saponin substances such as chlorophylls and other pigments and on the yields of saponins in separating saponins from ginseng root, leaf and stem. Ginseng root saponins were effectively purified by various fat-soluble solvents while ginseng leaf stem saponins were by chloroform. And alternative extractions of ethyl acetate, ethyl ether, chloroform and benzene there more effective for ginseng leaf stem saponins than that by any single solvent. Contents of crude saponin fractions and total ginsenosides in ginseng leaf were $18.5{\sim}19.5%\;and\;10.8{\sim}11.4%$, which were very high compared with $4.6{\sim}5.1%\;and\;2.0{\sim}2.6%$ in ginseng root or $2.2{\sim}2.5%\;and\;0.63{\sim}0.67%$ in ginseng stem. Therefore, ginseng leaf is good resources for total saponin or $ginsenosides-Rg_1,\;.Re,\;-Rc,\;-Rd,\;-Rb_2\;and\;-Rf$.

Effects of Ginseng Saponins on the Induction of Differentiation in Mammary Epithelial Cells and Mammary tumor Cells (홍삼 사포닌에 의한 유선상피 및 유선암세포의 분화 유도 효과 연구)

  • Using Ginseng saponins (crude saponin and total saponin) and ginsenoside Rbl Rb2, Rc, Rd, Re, Rhl, and Rh2 in this study, we have examined the effects of the compounds on the induction of differentiation in normal rat mammary epithelial cells and 7,12-dimethylbenz[a]anthracene (DMBA)-induced mammary tumor cells in culture. When normal rat mammary organoids were cultured in 100-mm culture plates in the presence or absence of ginseng saponins, there were four different cell colonies after two weeks in culture: cobble stone, spindle, honey comb, and senescence type colonies. Ginseng saponins showed different effects on the development of each colonies. Scrape-loading dye transfer tech-nique was performed to measure the effects of total saponin, Rhl, and Rh2 on intercellular junctional communication. Intercellular communication was not observed at short cultilral time, e.g., four or seven days, but when it cultured it up to two weeks, cell to cell communication was observed in saponin-treated cells. Reconstituted basement membrane, Matrigel, supported the growth and development several different multicellular structures from normal mammary organoids (e.g., ductal, webbed, stellate, and squamous colonies) or DMBA-induced mammary tumor (e.g., alveolar unit, foamy alveolar unit, squamous metaplasia, lobule-ductal, stellate, and webbed colony). In ginseng saponin-treated groups, webbed colonies were more and squamous colonies were less than control group. Moreover, the ductal colonies, marker tructure of well-differentiate mammary epithelial cells, were developed more in saponin-treated group than in control group. In conclusion, ginseng saponins affected on the differentiation of normal rat mammary epithelial cells and DMBA-induced mammary tumor cells in culture.
