Nitrogen Adsorption Analysis of Wood Saccharification Residues

  • The objective of this study was to examine changes in the porosity and internal structure of wood as it goes through the process of saccharification (extraction of fermentable sugars). This study also examined the use of different drying methods to prepare samples for characterization of internal pores, with particular emphasis on the partially disrupted cell wall. Aspen wood flour samples after dilute acid pretreatment followed by enzymatic hydrolysis were examined for nitrogen adsorption. The resulting isotherms were analyzed for surface area, pore size distribution, and total pore volume. Results showed that freeze drying (with sample pre-freezing) maintains the cell wall structure, allowing for examination of saccharification effects. Acid pretreatment (hemicellulose removal) doubled the surface area and tripled the total volume of pores, which were mostly 10-20 nm wide. Subsequent enzymatic hydrolysis (cellulose removal) caused a 5-fold increase in the surface area and a ~ 11-fold increase in the total volume of pores, which ranged from 5 to 100 nm in width. These results indicate that nitrogen adsorption analysis is a feasible technique to examine the internal pore structure of lignocellulosic residues after saccharification. The information on the pore structure will be useful when considering value-adding options for utilizing the solid waste for biofuel production.

Corrosion and Adhesion of Electrophoretic Paint on AZ31 Magnesium Alloy Pretreated in Cerium Chemical Conversion Coating Solution

  • In this study, the corrosion resistance and adhesion of electrophoretic paint (E-paint) were studied on AZ31 magnesium alloy pretreated in cerium chemical conversion coating solutions with the addition of various ethanol concentrations. It was found that with increasing ethanol concentration from 0 to 90 percent can decrease the formation of $Mg(OH)_2/MgO$ and increase the formation of nano-crystalline cerium oxides on the coating. Both corrosion resistance and adhesion of E-painted AZ31 increased with increasing ethanol concentration. The best E-paint sample was observed on the sample pretreatment in cerium chemical conversion coating solution with the addition of 80 percent of ethanol. This sample showed an excellent adhesion without paint detached after water immersion test for 500 h at $40^{\circ}C$, and only a few blisters observed at the near scratched sites after 1000 h salt-spray test.

Extraction Characteristics and Quantitational Methods for Total Petroleum Hydrocarbons in Soil

  • Quantitation methods of total petroleum hydrocarbons to determinate oil contaminated level in soil were discussed. Extraction characteristics of several pretreatment methods and practical detection limit and reappearances in gas chromatography/mass spectrometry. with each pretreatment method were investigated. The obtained results showed that the newly adopted quantitation method and mechanical shaking extraction method using methanol with extraction solvent are more practical and applicable to real sample than the conventional methods. In applying these methods to gasoline, kerosene, fuel oil which are major source of soil contamination, the practical quantitation limit and % relative standard deviation was able to determine with range of 2.5 - 10 ppm, 5 - 7 %.

Analytical Method for Dioxin and Organo-Chlorinated Compounds : (I) Pretreatment of Milk Samples for Dioxin Analysis

  • A simple and reproducible pretreatment method was developed for the determination of dioxins in milk sample. Liquid-liquid extraction (LLE) was used for the initial extraction of the analyte from milk. For the elimination of interferences coextracted from milk, acid treatment followed by multilayer silica gel, and then alumina column clean-up were performed. The clean extract could be obtained without carbon column or high performance liquid chromatographic (HPLC) clean-up procedure. Polychlorinated biphenyles (PCBS) and dioxins were separated on neutral alumina activated at 180 ℃ for 12 hours. The final extract was analyzed by HPLC and high resolution gas chromatography/high resolution mass spectrometry (HRGC/HRMS). The recovery of dioxins spiked in milk at 75-300 ppt level was 83.3-98.9% and their relative standard deviation was 4.1-14%.

Comparison of Upgraded Methods for Detecting Pathogenic Escherichia coli in Foods Using Centrifugation or Filtration

  • In the present study, centrifugation and filtration pretreatments were evaluated to decrease sample preparation time and to improve the sensitivity and specificity of multiplex polymerase chain reaction (PCR) for the detection of low levels of pathogenic Escherichia coli in various foods. Pathogenic E. coli (E. coli NCCP11142, E. coli NCCP14037, E. coli NCCP 14038, E. coli NCCP14039, and E. coli NCCP15661) was inoculated into pork, beef, and baby leafy vegetables at 1, 2, and 3 Log CFU/g. The samples were shaken 30 times (control), then centrifuged or filtered. DNA extracts from the samples were subjected to PCR using the $Powerchek^{TM}$ Diarrheal E. coli 8-plex Detection Kit. In the pork samples, no E. coli was detected in the control samples, while E. coli were detected in 100% of 3-Log CFU/g inoculated and centrifuged samples, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In the beef samples, all control samples appeared to be E. coli-negative, while E. coli was detected in 50-75% of centrifuged samples, regardless of inoculated level, and in 100% of 2 and 3-Log CFU/g inoculated, and filtered samples. In baby leafy vegetables, E. coli were not detected in 25-50% of the control samples, while E. coli were detected in 0-25% of the centrifuged samples, and 75-100% of the filtered samples, depending on the inoculum amount. In conclusion, filtration pretreatment can be used to minimize sample preparation time, and improve the sensitivity and specificity of rapid detection of pathogenic E. coli in various foods.

저식염 속성 멸치 발효액화물 가공에 관한 연구(I) -숙성 중 유리아미노산 변화 및 품질지표- (Studies on the Processing of Rapid- and Low Salt-Fermented Liquefaction of Anchovy(Engrulis japonica) (I) -Changes in Free Amino Acids during Fermentation and Quality Indices-)

  • In order to establish the processing condition of rapid- and low salt-fermented liquefaction of anchovy (Engrulis japonica), effect of temperature on crude enzyme activity of anchovy viscera, pretreatment conditions, and the minimum content of adding NaCl were investigated. The minimum limitation of NaCl content for anchovy liquefaction was 10%. Sample A(water adding, heating, adding 10% NaCl): chopped whole anchovy adding 20% water and then heating for 9 hrs at $50^{\circ}C$ and then adding 10% NaCl and then fermented at room temperature$(8-29^{\circ}C)$ for 180 days. Sample B(water adding, heating, adding 13% NaCl): chopped whole anchovy adding 20% water and then heating for 9 hrs at $50^{\circ}C$ and then adding 13% NaCl and then fermented at room temperature for 180 days. Sample C(adding 13% NaCl): chopped whole anchovy and then adding 13% NaCl and then fermented at room temperature for 180 days. Sample D(adding 17% NaCl): whole anchovy adding 17% NaCl and then fermented at room temperature for 180 days. The content of free amino acids such as aspartic acid, serine and threonine fluctuated severely according to the pretreatment methods. Possibly they might be recommend quality indices of standardization for salt-fermented liquefaction of anchovy. As for the relation between fermentation period(X) and individual free amino acid(Y), five kinds of free amino acids such as glutamic acid, valine, glycine, lysine, and alanine showed highly significant in their coefficient of determination in most of samples. They might be recommend as quality indices for salt-fermented liquefaction of anchovy during fermentation. The difference of taste between products of the rapid- and low salt-fermented liquefaction and the traditional salt-fermented liquefaction were caused by their composition of the free amino acids ratios, in which were umami, sweet, and bitter taste in the extracts of anchovy during fermentation. The appropriate fermentation period of the sample A was shorten 30 days than the sample B and 60 days than the samples C and 90 days than the sample D in the processing of anchovy.

작약근의 박피와 시료형태가 건조시간 및 Paeoniflorin 함량에 미치는 영향 (Effects of Peeling and Sample types on Drying Time and Paeoniflorin Contents of Peony Root)

  • 작약의 양질약재 생산을 위한 기초 자료를 얻고자 건조과정중 박피시간별 절단과 무절단 그리고 열풍건조 온도를 달리하여 시험한 결과를 요약하면 다음과 같다. 박피시간이 길어질수록 건조시간은 통뿌리보다 절단한 것이 건조온도가 단축되었으며, $30^{\circ}C$에서 $60^{\circ}C$까지의 건조온도에서 건조온도가 상승할수록 온도상승의 효과는 감소하였다. 작약의 주 약용성분인 paeoniflorin 함량은 무박피 절단형태로 열풍온도 $30^{\circ}C$에서 건조한 작약이 3.48%로 가장 높은 함량을 유지하였고, 120분 박피후 통뿌리로 $60^{\circ}C$에 열풍건조한 작약은 2.29%로 건조과정중 많은 양이 손실되어 가장 낮게 나타났다. 건조작약의 외부색택은 통뿌리로 $60^{\circ}C$에서 열풍건조했을 때 흑갈색으로 변하는 정도가 가장 심했다. 따라서 작약 수확후 건조과정 중 건조시간 단축과 약효성분 그리고 색택면에서 건조전의 처리 방법은 무박피 절단형태가 장시간 박피 원형형태보다 우수한 것으로 판단된다.

오령산(五?散)이 CCl4 투여(投與)로 인한 흰쥐의 손상간(損傷肝)에 미치는 영향(影響) (Effect of the Oryungsan on the injured liver in CCl4-treated rats)

  • This study was investigated to define effect of pretreatment of Oryungsan on hepatotoxicity induced by $CCl_4$ in rat. The experimental anmals were divided into normal group, control group, sample group, and sample group were divided sample1, sample2, sample3. Control group were injected diluted carbon tetrachloride and olive oil, $1m{\ell}/kg$ to intraperitoneal after 8th day. Sample group were injected Oryungsan water extracts, $0.33m{\ell}/kg/day$ to intraperitoneal of sample group1, $0.66m{\ell}/kg$ day to sample group2, $1.32m{\ell}/kg/day$ to sample group 3 each other for 7day, and injected the dosage of $CCl_4$ was $1.0m{\ell}/kg$ diluted 1 : 1 with olive oil after 8th day. The following results were made by observation GOT, GPT, Albumin, Globulin, Bile acid, hepatic tissue. 1. The serum GOT and GPT level of sample group were decreased. Sample group 1 were decreased significantly. 2. The serum A/G ratio of sample group was increased as compared with normaI group. 3. The serum bile acid of sample group was decreased significantly. 4. The destruction of hepatic tissue were repaired as compared with control group. These results demonstrated that oryungsan can be attributed to recovery from hepatotoxicity.

갈색거저리(Tenebrio molitor) 유충의 냉장 저장 중 산화 안정성에 관한 연구 (Studies on Oxidative Stability of Tenebrio molitor Larvae During Cold Storage)

  • The purpose of this study was to evaluate the changes on the characteristics of the oxidative stability of Tenebrio molitor larvae during cold storage at $4^{\circ}C$. Pretreatment for T. molitor larvae was designed into three methods: raw (R), freeze-dried (F.D.), and pan-fried (P.F.). The water content of the raw sample (61.46%) was higher than those of other samples (F.D.: 5.02%, P.F.: 3.67%) and its high water content was expected to facilitate the oxidation of the raw sample. In our results, the peroxide value and the carbonyl value of all of the samples increased and the raw sample, after storage for 18 day, showed the highest value. The pan-fried sample had no significant increase in its lactic acid content, acid value, and thiobarbituric acid value; whereas those values were increased in the raw sample and the freeze-dried sample (p<0.05). The browning reaction was more progressed in the pan-fried sample than other samples at 0 day, but there was no significant change during the storage. The raw sample and the freeze-dried sample had their browning indexes increase with the increasing storage period (p<0.05). The pan-fried sample produced less oxidation products than the freeze-dried sample, indicating that the unheated sample was more susceptible to oxidation than the heated samples. In conclusion, heating treatment and low water content would be effective for improving the safety and stability of T. molitor larvae during cold storage.

자윤탕이 뇌혈류 변화에 미치는 영향 (The Effects of Jayun-tang on the Changes of Cerebral Flow)

  • Objectives : This study was designed to investigate the effects of Jayun-tang extract (JYT) on the change of cerebral hemodynamics [regional cerebral blood flow (rCBF), pial arterial diameter (PAD) and mean arterial blood pressure (MABP)] in normal and cerebral ischemic rats, na to determine the mechanisms of action of JYT. Methods : We investigated whether JYT inhibits lactate dehydrogenase activity in neuronal cells and cytokines production in serum of cerebral ischemic rats. Results : 1. JYT significantly increased rCBF and PAD in a dose-dependent manner, but MABP was not changed by injecting JYT. These results suggested JYT significantly increased rCBF by dilating PAD. 2. The JYT-induced increase in rCBF was significantly inhibited from pretreatment with indomethacin (1mg/kg, i.p.), an inhibitor of cyclooxygenase and methylene blue $(10{\mu}g/kg, i.p.)$, an inhibitor of guanylate cyclase. 3. The JYT-induced dilation in PAD was significantly inhibited from pretreatment with indomethacin, but was increased by pretreatment with methylene blue. 4 The JYT-induced increase in MABP was reduced by pretreatment with indomethacin and methylene blue. 5. JYT significantly inhibited lactate dehydrogenase activity in neuronal cells. These results suggest that JYT prevented the neuronal death. 6. Both rCBF and PAD were significantly and stably increased by JYT $(10{\mu}g/kg,\;i.p.)$ during the Period or cerebral reperfusion, which contrasted with the findings of rapid and marked increase in the control group. 7. In cytokine production in the serum drawn from femoral artery 1hr after middle cerebral artery occlusion, the sample group showed significantly decreased production of $IL-1\beta$ and $TNF-\alpha$ as well as increased production of IL-10 and $TGF-\beta$ compared with rho control group. 8. In cytokine production in the serum drawn from femoral artery 1hr after reperfusion, the sample group showed significantly decreased production of $IL-1\beta$ and $TNF-\alpha$ as well as significantly increased production of IL-10 and $TGF-\beta$ compared with the control group. Conclusions : JYT mediated by cyclooxygenase had an inhibitive effect on brain damage by inhibiting lactate dehydrogenase activity, $IL-1\beta$ and $TNF-\alpha$ production, and by accelerating IL-10 and $TGF-\beta$ production. The author feels that JYT had anti-ischemic effects through the improvement of cerebral hemodynamics and inhibitive effects on brain damage.
