Antiviral effects of various plant extracts against viral haemorrhagic septicaemia virus (VHSV) (바이러스성출혈성패혈증바이러스에 대한 식물 추출물의 항바이러스 효능 탐색)

  • Since viral haemorrhagic septicaemia virus (VHSV) was first reported in European rainbow trout (Oncorhynchus mykiss) in the 1930s, it has caused high prices in freshwater and saltwater fish around the world, causing enormous economic damage to the aquaculture industry. We have been seeking required countermeasures against viruses because of economic damage to the aquaculture industry. However, commercial vaccines have the limitations of being costly to use in farms and being effective to only one pathogen. The aquaculture industry these days is taking on new alternatives to vaccines, antibiotics and chemicals. In this study, the suitability of antiviral effects against VHSV was evaluated in vitro for various plant extracts to judge their effectiveness. Atriplex gmelinii, Ixeris repens, Arctium lappa, and Sargassum coreanum were tested to know the correlation between the amount of virus and the concentration of extract investigates if these extracts have antiviral effects. Virus and extracts at various concentrations were inoculated simultaneously as 1:1 ratio into EPC cell lines. There are no antiviral effects with Atriplex gmelinii, Ixeris repens and Arctium lappa. Extract of Sargassum coreanum only has the antiviral activity in a dose-dependent manner. These results show that extract of Sargassum coreanum can be used in aquaculture industry as an antiviral materials.

Changes in BOD, COD, Chlorophyll-a and Solids in Aquaculture Effluent with Various Chemical Treatments

  • Four chemical treatments with hydrogen peroxide (H2O2), copper sulfate (CuSO4), potassium permanganate (KMnO4) and chlorine (Cl2) were applied to the effluent pond water of a hybrid striped bass saltwater recirculating aquaculture system to compare their oxidation power. Four chemicals were applied at concentrations of 0 (control), 1, 5, 10 and 20 mg/l. An additional concentration of 40 mg/l was included in the chlorine treatment. Water samples from four hybrid striped bass ponds were tested with KMnO4 and Cl2. H2O2 did not reduce any of BOD, COD and chlorophyll-a, and copper sulfate was only effective on chlorophyll-a for the effluent pond. Removal efficiencies for chlorophyll-a by copper sulfate were 19.2%, 37.5%, 54.2% and 74.1% dose-dependently. Potassium permanganate effectively removed the BOD, COD and chlorophyll-a. The COD removal rates in four fish ponds varied from 15.9% to 31.6% at the concentration of 10 mg/l. Interestingly, Cl2 did not reduce the BOD and COD at all, but the BOD and COD instead increased drastically with increasing the Cl2 concentration. The pond water with the highest initial BOD and COD values among the fish ponds tested increased by 350% in the BOD and 150% in the COD at 20 mg/l. Furthermore, Cl2 did not significantly reduce any types of solid matter in this study, while KMnO4 seemed to reduce some extent volatile dissolved solid in the fish pond.

The Changes of Side Dishes in "Eumsikdimibang", "Gyuhapchongseo", "Chosunm usangsinsikyorijebub" according to the Current of the Time ("음식디미방","규합총서(閨閤叢書)","조선무쌍신식요이제법(朝鮮無雙新式料理製法)"에 수록된 시대적 흐름에 따른 부식류의 변화)

  • This study examined the changes of side dishes in "Eumsikdimibang", "Gyuhapchongseo", "Chosunmusangsinsikyorijebub" over time. As food ingredients, seaweed, red pepper, salted fish, Korean hot pepper paste have been used in "Gyuhapchongseo". The use of cattle by parts, saltwater fish, Chinese cabbage, powdered red pepper, garlic have increased in"Chosunmusangsinsikyorijebub". Cooking techniques, such as the use of wheat flour, a double boiler by the use of steam, and boiling and then roasting cuisine were mostly used in"Eumsikdimibang", but reduced in"Gyuhapchongseo". In "Gyuhapchongseo", the cooking methods are primarily aimed at a positive visual effect, and slices of dried meat seasoned with spices have increased. In"Chosunmusangsinsikyorijebub", various Tang (Guk), the taste of food changed by controlling the gravy content (Gigimi, Chigye, Chorim), meat mixing oil, vegetable and mushroom together to cook and boiling down the main food ingredients to soak the seasoning were increased. Dog-meat steamed dish using the intestine of dog in "Eumsikdimibang" was changed to small intestine of cattle steamed dish in "Gyuhapchongseo". And seasoned dog meat with choncho in "Eumsikdimibang" influenced on beef tail soup with Korean hot pepper paste in "Gyuhapchongseo", and Yookgyejang soup using Korean hot pepper paste in"Chosunmusangsinsikyorijebub". In steamed young chicken, the stuffing such as soybean paste, choncho, welsh onion, leek and flour in "Eumsikdimibang" was changed to minced beef, welsh onion, dropwort and Shiitake mushroom in"Gyuhapchongseo". The steamed young chicken in "Chosunmusangsinsikyorijebub" with stuffing was added to chicken soup using glutinous rice, and ginseng powder. Now, the chicken soup was changed to Samgyetang with glutinous rice, and ginseng. In "Chosunmusangsinsikyorijebub", various vegetable dishes were cooked with beef.

Numerical Modeling for Sedimentation Characteristics of the Lower Nakong River and Sediment Dredging Effects at the Nakdong River Estuary Barrage (낙동강 하류의 유사특성과 낙동강하구둑 준설효과에 관한 수치모의 연구)

  • The Nakdong River Estuary Barrage (NREB) was constructed in 1987 to prevent saltwater intrusion and to provide the sustainable water supply in the upstream channel. Sediment dredging has been conducted to eliminate deposited sediments in the approached upstream channel of the NREB. Fluvial changes and sedimentation problems have been continued due to urbanization and development in the watershed as well as construction of the NREB. However, the sufficient field monitoring and researches for sedimentation characteristics and bed changes have not been performed after construction of the NREB. Therefore, bed elevation changes and seasonal sediment concentration distribution were analyzed using the quasi-steady state model with historical field data in this study. The water surface elevation changes with and without sediment dredging operation were calculated using the developed quasi-steady state model and finally the sediment dredging effects were evaluated.

Application of Flux Average Discharge Equation to Assess the Submarine Fresh Groundwater Discharge in a Coastal Aquifer (연안 대수층의 해저 담지하수 유출량 산정을 위한 유량 평균 유출량 방정식의 적용)

  • Water supply is decreasing due to climate change, and coastal and island regions are highly dependent on groundwater, reducing the amount of available water. For sustainable water supply in coastal and island regions, it is necessary to accurately diagnose the current condition and efficiently distribute and manage water. For a precise analysis of the groundwater flow in the coastal island region, submarine fresh groundwater discharge was calculated for the Seongsan basin in the eastern part of Jeju Island. Two methods were used to estimate the thickness of the fresh groundwater. One method employed vertical interpolation of measured electrical conductivity in a multi depth monitoring well; the other used theoretical Ghyben-Herzberg ratio. The value using the Ghyben-Herzberg ratio makes it impossible to accurately estimate the changing salt-saltwater interface, and the value analyzed by electrical conductivity can represent the current state of the freshwater-saltwater interface. Observed parameter was distributed on a virtual grid. The average of submarine fresh groundwater discharge fluxes for the virtual grid was determined as the watershed's representative flux. The submarine fresh groundwater discharge and flux distribution by year were also calculated at the basin scale. The method using electrical conductivity estimated the submarine fresh groundwater discharge from 2018 to 2020 to be 6.27 × 106 m3/year; the method using the Ghyben-Herzberg ratio estimated a discharge of 10.87 × 106 m3/year. The results presented in this study can be used as basis data for policies that determine sustainable water supply by using precise water budget analysis in coastal and island areas.

Assessment on Saline Water Intrusion between Types of Injections of Artificial Reclaimed Water and Extractions in Artificial Aquifer (인공 하수처리수 주입과 양수 방식에 따른 인공 대수층의 해수침투평가)

  • The study with laboratory sandbox model has been carried out to address potential use of reclaimed water, as a way for artificially recharging the coastal aquifer, to effectively prevent from seawater intrusion. To do this, we assessed hydraulic and geochemical properties depending upon various extraction and recharging conditions. While solely being recharged, the intrusion could be significantly retarded than those of recharge and extraction implied together. At 0.5 to 2 for the ratio of the extraction over the recharge rate, the fresh water was exploited from the tank, where the void regime was simultaneously saturated with the recharged water. In the meantime, the saline water zone was diluted and back-tracked by the recharged water due to forming a hydraulic geochemical barrier around the injection well. However, if the ratio was being increased to greater than 4, saltwater more deeply intruded to the freshwater zone because the artificial recharge was not sufficiently supplied to timely back-fill the void space. When the aquifer water was intermittently extracted at the ratio of $0.5{\sim}2$ over the recharge rate, the value of S.M.I. decreased, but increasing it to more than 4 unlikely escalated the value of S.M.I as much as $3{\sim}47%$ indicating that the salt water intruded. It finally revealed that the proper ratio of extraction/recharge or intermittent extraction would efficiently retracted seawater intrusion while the freshwater sources could be conservatively utilized.

Spatial Characteristics of Vegetation Development and Groundwater Level in Sand Dunes on a Natural Beach (해안사구의 지하수위와 식생 발달의 공간적 특성 연구)

  • Field observations were used to study the characteristics and influence of groundwater level fluctuations on vegetation development on the natural beach of a sandy barrier island, in the Nakdong River estuary. The spatial/temporal fluctuations of the groundwater level and the interactions with the external forces (weather, ocean wave and tide) were analyzed. The results indicated that when it rains the groundwater level rises. During summer, when precipitation intensity is greater than 20 mm/hour, it rose rapidly over 20 cm. Subsequently, it fell gradually during periods of no precipitation. Seasonal characteristics indicated that the groundwater level was high during the summer rainy season and tended to fall in the winter dry season. The time-averaged groundwater level, observed from the four observations over 3 years (2012-2014), was about 1.47 m, higher than mean sea level (M.S.L.). It was shown that the average annual groundwater level rises toward the land rather than showing intertidal patterns observation. Differences in the presence or absence of a coastal sand dunes affected the progress of vegetation. In other words, in environments of saltwater intrusion where the groundwater level varies, dependent on the distance from the shoreline and bottom slope, sand dunes can be provided to affect soil conditions and groundwater, so that vegetation can be grown reliably.

An Analysis on Applicability of Geophysical Exploration Methods to Monitoring Polymer-flooding (물리탐사 기법들의 화학공법 모니터링 적용성 분석)

  • Polymer flooding for enhancing hydrocarbon production injects into a reservoir polymer solution that is viscous. It is very important to monitor the behavior pattern of the polymer solution in order to evaluate the effectiveness of polymer flooding. To monitor the distribution of polymer solution and thus fluid substitution within the reservoir, we first construct seismic and resistivity rock physics models (RPMs), which are functions of reservoir parameters such as rocks and type of fluid, fluid saturation. For the seismic and resistivity RPMs, responses of seismic and electromagnetic (EM) tomography are numerically simulated as polymer injection, using two dimensional (2D) staggered-grid finite difference elastic modeling and 2.5D finite element EM modeling algorithms, respectively. In constructing RPM for EM tomography, three different reservoir rocks are considered: clean-sand, dispersed shale-sand, and sand-shale lamination rocks. The polymer solution is assumed to have 2 wt% of polymer as normally generated, while water is freshwater or saltwater. Further, neutron logging is also considered to check its sensitivity to polymer flooding. The techniques discussed in the paper are important in monitoring not only hydrocarbon but also geothermal reservoirs.

Diversity of Endophytic Fungal Strains from Jeju Aquatic Plants (제주 수생식물에서 분리한 내생균류의 다양성)

  • Endophytic fungi are present in host plants and contribute to resistance to biotic and abiotic stress. Aquatic plants are living in aquatic environment such as saltwater or freshwater and exposed more water stress than other land plants. In this study, we investigated 4 wetlands in Jeju and collected 11 aquatic plants. Exogenous microbes were removed by preprocessing of plants and endophytic fungal strains were isolated from the plants. We isolated 126 fungal strains from Namsaengi-pond, 22 fungal strains from Sujangdong-marsh, 44 fungal strains from Yongsu-reservoir and 32 fungal strains from Gangjeongcheon. The fungal strains were identified using internal transcribed spacer (ITS) region and analyzed the phylogeny and diversity. Endophytic fungi isolated from plants of Namsaengi-pond were classified to 30 genera, 19 families, 12 orders, 7 classes and 4 phyla. Endophytic fungi of Sujangdong-marsh were classified to 11 genera, 11 families, 6 orders, 5 classes and 4 phyla. Endophytic fungi of Yongsu-reservoir were classified to 13 genera, 12 families, 7 orders, 5 classes and 4 phyla. Endophytic fungi isolated from Gangjeongcheon were classified to 9 genera, 7 families, 5 orders, 2 classes and 1 phyla. Overall, they were divided 40 genera and Alternaria, Colletotrichum and Fusarium were isolated from 4 sites in common. By investigating the endophytic fungi in aquatic plants, it is for baseline data that determination of diversity and the ecological distribution of endophytic fungi.

Salinity Distribution and Ecological Environment of Han River Estuary (한강 하구역의 염분 분포 및 생태환경특성)

  • Water quality and ecological environment in the Han River estuary was analyzed using the longterm water quality monitoring data from National Fisheries Research and Development Institute (NFRDI) and the existing data collected in this area. Based on the salinity distribution and changes of current direction in the lower Han River and its estuary, boundaries of the estuary were identified and also, distribution patterns of the phyto- and zooplankton, benthos, ichthyoplankton and fish were discussed related with the salinity changes in the macrotidal subestuary of Han River. Seasonal and spatial distribution of salinity suggested that the direct impact of freshwater be limited to the Incheon North Harbour all the year round and even extended to the southern area of Gyunggi Bay near Palmi island during limited time, usually in summer. Upper limit of salt water intrusion through the Han River is likely to be Singok underwater dam located Gimpo, Gyunggi Province, and normally limited to much lower part of the river, Jeonryuri, Gimpo. Biological boundaries of the Han River estuary exceeded the physical boundaries based on the salinity distribution. Many estuarine species in plankton and fish were found at the totally freshwater or saltwater depending on the seasons and tidal cycles. Some estuarine ichthyoplanktons showed extremely limited distributions in the estuary whereas adult fish revealed wide ranges of salinity adaptation. Critical environmental issues in the Han River estuary and its drainage basin are likely to be 1) pressure on development-promoted district for new town in the drainage area of the estuary, 2) reduction of tidal flat by reclamation, 3) pollutant input through river from municipal sewages and industrial wastes, and 4) ecological barrier between river and terrestrial systems by the military wire fence and riverside road.
