Particulate leaching method for the preparation of porous PLLA scaffolds was carried out and especially, the effect of PLLA/$CHCl_3$ solution concentration on the salt leaching rate and the pore structure of PLLA scaffolds were considered. It was found that maintaining lower PLLA/$CHCl_3$ concentration and higher $CHCl_3$ evaporation temperature in the preparation of PLLA/NaCl mixtures resulted in the enhancement of salt leaching rat e and higher porosity. This is understood that those conditions could minimize the formation of dense PLLA layer on the surface of PLLA/NaCl mixture as well as introducing better porosity on the surface. Higher salt leaching temperature accelerated the salt leaching rate but it seems that there is no influence on the porosity of PLLA scaffolds.
This laboratory study was carried out in order to produce fundamental data for analyzing salt movement and desalinization effects, using samples of silt loam soil collected in Gyehwado and Daeho reclaimed tidelans, and samples of silty clay loam soil collected in Kimie tideland. Desalinization experiments with gypsum treatment were performed to analyze changes of the hydraulicc conductivity with changes of the soil property and the salt concentration during the desalinization of reclaimed tideland soils by leaching through the subsufface drainage, and correlations between factors infl uencing the reclamation of salt affected soils were analyzed by the statistical method. The results were summarized as follows: 1. The reclaimed tideland soils used in this study were saline-sodic soils with the high exchangeable sodium percentage and the high electrical conductivity. 2. Changes of the hydraulic conductivity with the amount of leaching water and the leaching time elapsed were affected by the amount of gypsum except exchangeable sodium and clay contents. The regression equation between the depth of water leached per unit depth of soil (Dw / Ds : X) or the square root of the leaching time elapsed (T $^1$$^2$ : X) and the relative hydraulic conductivity (HCr:Y) could be expressed in Y=a . bx. 3. The more exchangeable sodium and clay contents regardless of the amount of gypsum, the more the leaching time was required until a given volume of water was leached through the soil profile. The regression analysis showed that the relationship between the depth of water leached per unit depth of soil(Dw /Ds:X) and the square root of the leaching time elapsed(T$^1$$^2$ :Y) could be described by Y=a . Xb. 4. The hydraulic conductivity was influenced to a major degree by the salt concentration provided that the electrical conductivity was below 10 mmhos / cm during the desalinization of reclaimed tideland soils. The regression equation between the relative electrical conductivity ( ECr : X) and the relative hydraulic conductivity (HCr:Y) could be expressed in Y=a + b . X-$^1$. 5. In conclusion, the hydraulic conductivity, leaching requirements and the leaching time elapsed can be estimated when the salt concentration decreases to a certain level during the desalinization of reclaimed tidelands, and the results may be applied to the analysis of salt movement and desalinization effects.
Highly open porous polymer matrices are required for high density cell seeding, efficient nutrient, and oxygen supply to the cells cultured in the three dimensional matrices. However, there are severe problems of mass transfer limitations within the cell/scaffolds culture system. Thus we hypothesize that continuos-flow culture conditioning of cells with the scaffolds may improve the cell viability and the differentiated function. In this study, we fabricated porous PLGA scaffolds by using gas-foaming/salt-leaching method as previous described. Viscous PLGA gel paste contains ammonium bicarbonate particulates, acting as a gas-foaming agent as well as a salt-leaching porogen, were cast into Teflon mold and dried. Ammonium bicarbonate salt upon contact to an acidic aqueous solution evloves gaseous ammonia and carbon dioxide by itself. And we conjugated galactose moiety [AGA; $N-(aminobuty1)-O-{\beta}-D-galactopyranosyl-(1{\rightarrow}4)-D-glucoamide]$ to the terminal end group of a PLGA to increase the cell adhesion and matain the differentiated function of hepatocytes. Cell-seeded scaffolds were secured in a flow bioreactor chamber and exposed to continuous flow at 5 ml/min. As a result of our study, the high yield of hepatocytes attachment was accomplished by increasing the concentration of PLGA-AGA conjugate in polymer scaffolds and cells in the scaffolds under continuos flow condition maintained a high level of viability and albumin secretion rate of cultured hepatocytes showed a higher level that of control groups.
This study was carried out in order to determin optimum irrigation requirements and water management practices for normal growth of dry field crops in reclaimed tidelands, and apply m planning of the irrigation projects. Desalinization experiments were performed by water management practices in the experimental field with high salt concentration, and growth experiments were conducted by irrigation point treatments using tomato and beet with relatively high salt tolerance. The results obtained from this study were summarized as follows : 1. Leaching or rinsing-leaching method was found to be effective in desalinizing the reclaimed tideland with rather high permeability. In this case, the water requirement for desalinizing the root zone layer of 40cm in depth, was estimated to be 1,200mm in depth. 2.The gypsum treatment in the desalinization of reclaimed tidelands, was ineffective in water requirements ; however, it could produce the desired effect in the facility of desalinization and the shortening of desalinization period with the sustaining permeability, in case of the desalinization by leaching method. 3.The optimum irrigation point which maintains the salt concentration within salt tolerance and maximizes the crop yield in reclaimed tidelands of silt loam soil, was found to be pF 1.6 in tomato and pF 1.8 in beet. The interval of irrigation date within 2 days was proved to he effective in both cases. 4.The optimum irrigation requirement and the water reguirement for the prevention of salt rise during the growing period after transplanting, were estimated to be 602mm(6.7mm/day) and 232mm for tomato, respectively. 5.The optimum irrigation requirement and the water requirement for the prevention of salt rise during the growing period after transplanting, were estimated to be 261mm(3.7mm/day) and 66mm for beet, respectively.
조직공학은 생명과학, 의학, 공학의 기본개념 및 기술을 바탕으로 생체조직을 대체할 수 있는 인공조직 및 장기를 제작하여 이식함으로써 생체의 기능을 유지, 향상 또는 복원하는 것을 목표로 하는데 여기에 사용되는 기본 재료가 장기나 조직의 형태를 만들도록 돕는 scaffold이다. Scaffold를 만드는데 있어서 Solvent-casting과 Particulate leaching 기법은 다공성 폴리머 scaffold의 제조에서 널리 쓰이는 방법인데 여기 쓰이는 particle에는 소금과 젤라틴 둥이 사용되고 있다. 소금은 얻기가 쉽고 다루기에도 편리하다는 장점 때문에 가장 일반적으로 사용되고 있으며 젤라틴은 소금에 비하여 세포의 초기 접착과 증식에 유리하다는 이유로 최근에 많이 사용되고 있으나 이에 관한 비교실험은 아직 보고 된 바 없다. 본 연구에서는 소금과 젤라틴으로 만들어진 두 가지 scaffold를 비교해 보았으며 그 결과 젤라틴 scaffold가 초기상태의 세포 접합과 증식에 있어서 좋은 결과를 보였고 같은 공극율일 때 공극의 연결 상태가 훨씬 더 우수한 결과를 보였다.
This study was performed in order to produce the basic data for developing prediction techniques of desalinization to be applicable to reclaimed tidal lands at the beginning stage. the desalinization experiments were carried out by two water management practices, namely, the leaching method by subsurface drainage and the rinsing method by surface drainage. The 5 soil samples used in this study were collected in 4 tidal land reclamation projects. Regression equations were obtained in order to investigate the changes of electrical conductivity during the desalinization of reclaimed tidal lands and to estimate water requirements for desalinization. The results obtained from this study were summarized as follows: 1. According to USDA Salinity Laboratory classification system of salt affected soils the reclaimed tidal land soils used in this study were saline-sodic soils with the high electrical conductivity and the high exchangeable sodium percentage. 2. With the increase of the water requirements for desalinization the electrical conductivity was decreased with high degree of correlationships and the desalinization effects were remakable in both the leaching method and the rinsing method. 3. In case of the leaching method the electrical conductivity had been reduced below the classification value of salt affected soils when the depth o water leached per unit depth of soil (Dwl/Ds) was 0.3 and the desalinization effects showed a tendency to be much the same in each treatment.
3D 프린터를 이한 인공지지체 제작 기술은 손상된 골 조직 재생을 위해 개발되고 있다. 골 조직 재생에 적하기 위해 인공지지체는 생체적합성, 생분해성 그리고 적절한 기계적 특성을 지녀야 하며, 분한 양의 공극과 내부 연결성을 지닌 구조로 제작되어야 한다. 본 연구에서는 3 차원 이중 공극 인공지지체를 제작하기 위해서 열 해 적층법(fused deposition modeling, FDM) 기반의 폴리머 적층 시스템을 이하였다. 사된 재료는 폴리카프로락톤(polycaprolactone, PCL)과 알긴산 나트륨(sodium alginate, SA)이다. 제작된 3 차원 형상의 인공지지체에 이중 공극을 갖기 위해 염 침출법을 이하였다. 완성된 인공지지체는 주사 전자 현미경과 X 선 검출 분광기(scanning electron microscope-energy dispersive spectroscopy, SEM-EDS)를 통해 관찰하였으며, MG-63 세포를 이하여 in-vitro 평가를 하였다.
In this study, we used a polymer deposition system, based on fused deposition modeling, to fabricate the 3D scaffold and then fabricated micro-pores on a 3D scaffold using a salt leaching method. Materials included polycaprolactone (PCL) and sodium chloride (NaCl). The 3D porous scaffolds were fabricated according to blending ratio such as PCL (70 wt%)/NaCl (30 wt%) and PCL (50 wt%)/NaCl (50 wt%). The 3D porous scaffolds were observed by scanning electron microscopy. The results showed that 3D porous scaffolds had a deposition width of $500{\mu}m$, contained a pore size of $500{\mu}m$ and below $100{\mu}m$. To evaluate the 3D porous scaffolds for bone tissue engineering, we carried out the cell proliferation experiment using a CCK-8 and a mechanical strength test using a universal testing machine. In summary, the 3D porous scaffold was found to be suitable for cancellous bone of human in accordance with the result of in-vitro cell proliferation and mechanical strength. Thus, a 3D porous scaffold could be a promising approach for effective bone regeneration.
조직 공학에서는 전통적인 인공지지체 제작 방식인 가스 발포, 염 침출, 스폰지 복제 그리고 동결주조 법 등이 이용되고 있다. 하지만 다양한 공극 형태 및 크기를 가지고 있어서 세포 상호 작용 효과 및 충분한 기계적 특성에 한계가 있다. 그러나 열 용해 적층 법은 조직공학에서 폴리머 재료를 이용하여 다양한 3차원 인공지지체를 제작할 수 있는 가장 적절한 기술이다. 따라서 본 연구에서는 PCL 몰드를 제작하고 실리카와 알긴산 나트륨 염을 포함하는 세라믹 슬러리를 제조하여 몰드에 주입시켰으며, 1일 동안 자연 건조를 시켰다. 제작된 3차원 슬러리 몰드는 PCL 몰드의 제거 및 슬러리를 경화시키기 위해 $100^{\circ}C$의 오븐에서 2시간 열처리 되었고, 열처리 후에 $1100^{\circ}C$에서 소결되었다. 제작된 인공지지체는 주사전자현미경을 통해 관찰되었고, 압축 시험을 통해 알긴산 나트륨 염의 혼합량에 따른 인공지지체의 기계적 특성은 평가되었다.
Polycaprolactone(PCL)은 생분해성 고분자로 인장강도, 신장률, 충격강도 등의 기계적 물성이 우수하다. $TiO_2$ (titanium dioxide) nanoparticle은 친수성으로 밀도가 높고 생체적합성이 우수하다. 본 연구에서는 PCL과 $TiO_2$(titanium dioxide) nanoparticle을 이용하여 salt-leaching방법으로 3차원 다공성 지지체를 제작하였다. 제작한 지지체를 FESEM, FTIR, TGA, 압축강도 측정 등을 통해 물성을 분석하였다. $TiO_2$ nanoparticle에 의해 물흡수도와 팽윤도는 감소하였으나 압축강도는 증가하였다. CCK-8 assay를 통해 세포의 증식률을 확인한 결과, $TiO_2$ nanoparticle에 의한 세포 독성은 없는 것으로 확인되었다. 이러한 연구결과는 PCL/$TiO_2$ nanoparticle 지지체의 생체재료로 사용가능성을 제시하였다.
