다양한 유제품에 이용될 수 있는 녹차(Camellia sinensis)에서 추출한 EGCG(Epigallocatechin gallate)의 Cronobacter spp.와 Salmonella spp.에 대한 항미생물 활성 효과에 관한 연구: 예비실험연구 (Antimicrobial Activity of Epigallocatechin Gallate (EGCG) extracted from Green Tea (Camellia sinensis) against Cronobacter spp. and Salmonella spp. in Various Dairy Products: A Preliminary Study)

  • The main constituent of tea catechins, EGCG [(-)-Epigallocatechin-3-gallate], could inhibit the growth of various microorganisms and differently affect gram-positive and gram-negative bacteria. Antimicrobial activity of EGCG, a compound from green tea (Camellia sinensis) extract, against Cronobacter spp. and Salmonella spp. was studied to evaluate the possibility of using EGCG as a natural food additive in various dairy products. In pure TSB culture, the growth of Cronobacter spp. was suppressed below the detection limit (1 log CFU/mL) depending on EGCG concentration ($600{\sim}800{\mu}g/mL$), after 5~16 days at $4^{\circ}C$. Similarly, the growth of Salmonella spp. was suppressed below the detection limit (1 log CFU/mL) depending on EGCG concentration ($400{\sim}800{\mu}g/mL$), after 5~16 days at $4^{\circ}C$. Therefore, these results suggest that EGCG could be used as an effective additive to inhibit the growth of Cronobacter spp. and Salmonella spp. in various dairy products, such as yoghurt, cheese, dried infant powder, and so on.

포장돈육 중 Salmonella Typhimurium과 Salmonella spp.에 오염된 돈육으로부터 식품과 식품접촉면으로의 오염 부착율 (Adherence Rates of Salmonella Typhimurium and Salmonella spp. in Pork Meat Contaminated during Processing)

  • 표준균주 S. Typhimurium과 분리균주 Salmonella spp.에 오염된 돈육을 $10^{\circ}C$에서 다른 돈육(삼겹살, 안심), 컨베이어벨트와 스테인리스 스틸, 도마에 일정시간(2, 4, 24시간)동안 접촉시킨 후 부착율을 조사하였다. Salmonella 표준균주와 분리균주에 오염된 돈육으로부터 삼겹살에 6시간 접촉 시 부착율은 4.21%, 26.87%를 나타내었고, 안심부위에 16.40%, 27.48%를 각각 나타내었지만, 24시간 후에는 두균 모두 삼결살과 안심부위에 100%의 부착율을 보였다. S. Typhimurium에 오염된 돈육으로부터 컨베이어벨트로의 부착율은 접촉시간 2시간 후에는 1.34%을 증가율을 나타냈으나, 6시간 후에는 0.60%로 감소하였고 분리균주 Salmonella spp. 는 접촉 6시간에 부착율이 5.14%의 부착율을 보였다. 스테인리스 스틸의 경우, 표준균주는 6시간 접촉 후에도 부착율이 전혀 없었으며, 분리균주는 접촉 6시간 후에 5.01%의 부착율을 보였다. 그러나 항균도마에는 표준균주와 분리균주 모두 전혀 부착하지 않았다. 본 연구결과, S. Typhimurium과 Salmonella spp.에 오염된 돈육에서 다른 돈육 또는 식품접촉면으로의 부착율은 접촉시간에 따라 영향이 컸으며 분리균주가 표준균주에 비하여 현저하게 높은 부착율을 나타내었다.

비살균 원유로 제조된 다양한 Gouda 치즈의 미생물 안전성 (Microbiological Safety of Various Gouda Cheeses Produced from Raw Milk)

  • 본 연구에서는 비살균 원유로 제조되고 숙성실에서 60일 이상 숙성된 7가지의 다양한 Gouda 치즈의 미생물학적 안전성을 실험적으로 검증하기 위해서 저장기간 동안에 다양한 미생물학적 변화를 조사하였다. Cheese 1과 2는 전통적인 비살균 Godua 치즈에 있어서는 60일 저장기간 동안 Salmonella spp.는 전혀 검출되지 않았으며, E. coli는 20 day부터는 검출되지 않았다. 대장균군은 Cheese 1에서 40 day부터 검출되지 않았지만, Cheese 2에서는 60 day까지 검출되었다. Cheese 1과 Cheese 2는 비살균 Gouda 치즈이기에 약간의 차이는 있었지만 전반적으로 비슷한 경향을 보였다. Cheese 3(원유에 인위적으로 Salmonella spp.를 오염시킨 후 제조된 비살균 Gouda 치즈), Cheese 4(Cheese 3에서 kefir에서 분리한 DH5 유산균 첨가)와 Cheese 5(Cheese 3에서 kefir에서 분리한 DN1 유산균 첨가)에 있어서는 60일 저장기간 동안 Salmonella spp.의 저해가 관찰되었다. 특히 Cheese 4와 Cheese 5에 추가적으로 첨가된 Kefir에서 분리한 유산균 DH5와 DN1은 Cheese 3보다 유의적으로 Salmonella spp.의 저해를 보였다. Cheese 6(유산균첨가 없이 제조된 비살균 Gouda 치즈)에서는 저장기간이 길어질수록 유산균수가 다른 Cheese들에 비해서 유의적으로 낮게 나타났다. 그리고 Cheese 7(살균한 원유로 제조한 전통적인 Gouda 치즈)에 있어서는 대장균 및 대장균군, Salmonella ssp.는 거의 검출되지 않았고, 유산균은 다른 Cheese들보다 약간 높게 나타났다. 60일 숙성된 Cheese 1부터 Cheese 7까지의 모든 샘플에 대한 Listeria monocytogenes, Staphylococcus aureus, Clostridium perfringens, E. coli O157:H7 검사에서 모두 음성을 보였다. 향후 kefir에서 분리한 유산균 DH5와 DN1의 비살균 원유를 이용한 숙성치즈의 제조에 관한 연구가 필요하겠다. 본 연구에서 Salmonella spp.를 인위적으로 접종한 Cheese 4와 5에서 보여준 Salmonella spp.의 저해 효과는 비살균 원유를 이용한 숙성치즈의 제조에 보다 큰 안전성을 부여할 것이라 사료된다.

제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사 (Investigation of Microbial Contamination the Level in Fish Farms of Jeju East Coast.)

  • 본 연구는 2007년 6윌부터 10월까지 제주도에서 양식장이 가장 많이 밀집되어 있는 제주도 동부 연안에 위치한 양식장 4개소를 대상으로 하여 사육 용수, 양어 사료, 양식 넙치의 미생물학적 오염도를 조사하였다. 식품병원성 미생물 중 일반세균과 총대장균군 그리고 Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus를 조사 하였다. 이번 연구를 수행하는 동안 동부지역은 집중적인 호우로 인해 전년보다 400 mm 이상의 많은 강우량을 나타내었다. 사육 용수의 검사결과 V. parahaemolyticus, Salmonella spp., S. aureus, B. cereus가 빈번하게 검출되었고, 양어 사료의 경우 Salmonella spp, S. aureus, B. cereus는 각각 $0{\sim}4.3{\times}l0^3$ CFU/ml, $0{\sim}2.7{\times}l0^4$ CFU/ml, $0{\sim}7.6{\times}l0^3$ CFU/ml 검출 되었다. 또한 양식 넙치의 미생물 검사 결과 적은 양이지만 Salmonella spp., S. aureus, B. cereus가 검출되었고, 총대장균군과 E. coli Ol57:H7은 검출되지 않았다. 이번 조사 결과로 보아 미생물학적 위해요소는 양식장의 작업 환경이나 주변 환경 등의 환경적 요인이 크게 작용하는 것으로 사료되며, 양식장의 위생관리에 작업자들은 주의를 기울여야 할 것으로 생각된다. 앞으로 이와 관련하여 수산물 위생에 대한 좀 더 체계적인 시스템을 도입하여 적용한다면 수산물에서 야기되는 문제점들이 해결되어 제주도의 청정 브랜드의 이미지를 더욱 부각시킬 수 있으며, 안전하고 깨끗한 수산물의 생산이 가능할 것으로 생각된다.

  • Salmonella spp. is the most common cause of gastrointestinal food poisoning worldwide, and human salmonellosis is mostly caused by the consumption of contaminated food. Therefore, the development of rapid detection methods for Salmoenlla spp. and rapid identification of the source of infection by subtyping are important for the surveillance and monitoring of food-borne salmonellosis. Therefore, this review introduces (1) History and nomenclature of Salmoenlla spp., (2) Epidemiology of Salmoenlla spp., (3) Detection methods for Salmoenlla spp. - conventional culture method, genetic detection method, molecular detection methods, and aptamer, and (4) Subtyping methods for Salmoenlla spp. - pulsed-field gel electrophoresis and repetitive sequence-based polymerase chain reaction (PCR).

양돈 생산성에 따른 주요 질병 분포 조사 (Correlation between Disease Prevalence and Production Performance in Korean Swine Farms)

  • Currently, various diseases reside in Korean swine farms and affect production performance of the farms greatly. These damages from disease are further aggravated by the concurrent infection of other disease. In this study, y investigating the distribution of porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Salmonella spp. in farms, correlation between the damage and the prevalence of disease was analyzed. Ten selected Korean swine farms that uses PCV2 vaccine were tested for presence of antibody and antigen of PRRSV, PCV2, Salmonella spp. per ages of pigs, 4weeks, 7weeks, 11weeks and 17weeks, respectively. The results were analyzed by dividing the farms in to groups with MSY above 19, and that with MSY below 19. Then calculating the distribution of disease each ages of pigs. Farms with MSY below 19 showed high prevalence of disease by PRRSV, PCV2 and Salmonella spp.. In this group, the detection rate of PCV2 and Salmonella spp. was increased by the activation/viremia of PRRSV in the young ages of pigs. The results are proved that the correlation between disease prevalence and production performance in Korean swine farms were very significant. The prevalence of PRRSV is more important index which influence to the productivity in current prevalence of diseases.

경북지방 돼지유래 salmonella 속균의 약제내성과 plasmid profile (Antibiotic resistance and plasmid profile of salmonella spp isolated from swine in Kyoungbuk province)

  • This study was conducted to investigate the antibiotic resistance and plasmid profiles of 58 salmonella spp isolated from mesenteric lymphnodes of slaughter pigs in Kyoungbuk province during the period from September 1997 to June 1998. The results obtained are as follow that all isolates were susceptible to amikacin, gentamicin, ciprofloxacin and enrofloxacin, and the majority of isolates were highly susceptible to norfloxacin, colistin, nalidixic acid and apramycin while they were moderately susceptible to kanamycin, cephalothin, chloramphenicol, ampicillin, trimethoprim and penicillin. The majority of isolates were over 90% resistant rates to lincomycin, sulfadimethoxine, vancomycin, methicillin and erythromycin. The plasmid profiles of 58 salmonella spp are developed 1 to 4 fractions, 0.9 to 29.5 Kb molecular range sizes and U strains (45.5%) were showed plasmid profiles by agarose gel electrophoresis. 5 derby harbored 29.5 Kb and 7 Kb, and S schwarzengrund had 14 Kb and 0.9 Kb harboring sizes. Four of 10 S agona and 2 of 4 S typhimurium were harbored 3.1 Kb and n.5 Kb, respectively. Thirty-five untypable strains are developed variable size fractions its showed small size plasmid profile less than 6 Kb and 22 (62.8%) of them had no detectable plasmids.

닭 분변유래 E. coli 및 Salmonella spp.의 항생제 내성패턴 (Antibiotic resistance pattern of E. coli and Salmonella spp. isolated from chicken feces)

  • The purpose of this study was to investigate the antibiotic resistance pattern of E. coli and Salmonella spp. isolated from chicken feces. One hundred and forty-seven E. coli isolates showed resistance to tetracycline (95.2%), erythromycin (89.2%), ampicillin (70.1%), streptomycin (59.2%), cephalothin (56.5%), sulfamethoxazole/trimethoprim (53.7%), ciprofloxacin (57.1%), enrofloxacin (59.2%) and norfloxacin (57.1%). The multiple resistance was seen in 144 isolates (97.9%) and the rate of five, six and seven drugs resistance pattern were 20.4%, 18.4% and 16.3%, respectively. Also, the multiple resistance of E. coli to twelve drugs were seen in 1 isolates (0.7%). Fourteen Salmonella spp. showed resistance to ampicillin (50.0%), streptomycin (57.1%), erythromycin (64.3%) and tetracycline (57.1%) and the rate of two and three drugs resistance pattern were 4 isolates (28.6%), respectively. The prevalence of resistant organisms in Korea probably reflects lack of proper antibiotic policy resulting in prolonged and indiscriminate use of antimicrobial agents.

Serovars distribution and antimicrobial resistance patterns of Salmonella spp. isolated from the swine farms and slaughter houses

  • Salmonella spp. is an important pathogen to both public and swine industry. The aim of this study was to investigate the distribution of Salmonella serovar and antibiotics susceptibility of the isolates from Korean swine producing systems. A total of 63 (5.28%) Salmonella spp. was isolated from 1,194 samples (977 fecal materials and 67 organ samples). The predominant Salmonella (S.) enterica serotype and serovar was group B (69.8%) and S. Typhimurium (47.6%), S. Derby (20.6%) and S. Heidelberg (1.6%). But S. Cholerasuis which is characterized host specific by septicemia and enteritis to pigs was not isolated. Antimicrobial susceptibility of the isolates varies as follows: Norfloxacine (75%), Ciprofloxacin (67.5%), Amikacin (60%), Colistin (60%), Enrofloxacin (55%). All of isolates were resistant to Erythromycin, Penicillin, Tetracycline and Lincomycin. The results of this study provided useful information regarding antimicrobial susceptibility and resistance patterns to treat salmonellosis and to prevent emergence of multidrug resistance Salmonella.

  • Salmonella spp. is of increasing public health concern as causative pathogens of food poisoning. The aim of this study was to investigate the serotypes and antimicrobial resistance pattern of Salmonella spp. isolated from duck farms in Daegu-Gyeongbuk province. Also, S. Enteritidis and S. Typhimurium isolates were further examined for plasmid analysis, phage typing and pulsed-field gel electrophoresis (PFGE). A total of 34 Salmonella spp. (16.4%) were isolated from duck farms and ten serotypes were identified. The predominant serotypes were S. Typhimurium (23.5%) S. Fyris (17.6%) and S. Haardt (11.8%), S. Agona and S. Enteritidis (respectively 8.8%). Of 34 Salmonella isolates, 15 (44.1%) isolates were resistant to at least one antimicrobial agent and multiple resistance (resistance to more than 4 drugs) was observed in 9 strains (26.5%). The high resistance was found to streptomycin (32.4%), tetracycline (29.4%), ampicillin, kanamycin and nalidixic acid (respectively, 26.5%), all Salmonella isolates were susceptible to cefoxitin, cefotaxime, gentamicin, amikacin and ciprofloxacin. All S. Enteritidis and S. Typhimurium isolates were found to contain only one plasmid (ca. 54 or 55kb, respectively). Among the S. Enteritidis isolates, two phage types were found, PT32a and PT1c, respectively, one isolates did not react with any of the phages used. Whereas, all S. Typhimurium isolates were RDNC (reacts but does not conform). PFGE showed to be a useful typing method better than plasmid analysis and phage typing for discrimination of isolates especially, S. Typhimurium isolates. Our results indicated that the serotypes of Salmonella isolates are widely distributed in duck farms, further epidemiological studies should be carried out.
