Identification of Salmonella Enteritidis and S. Typhimurium by multiplex polymerase chain reaction (Multiplex PCR 기법을 이용한 Salmonella Enteritidis와 S. Typhimurium의 특이적 검출에 관한 연구)

  • Salmonella species are the most important etiologic agents of food-borne acute gastroenteritis. The most common serotypes isolated from humans are Salmonella enterica serotype Typhimurium (S. Typhimurium) and S. Enteritidis. Traditional detection methods for Salmonella are based on cultures using selective media and characterization of suspicious colonies by biochemical and serological tests. These methods are generally time-consuming and not so highly sensitive. Recently, the polymerase chain reaction (PCR) has been used as a highly sensitive, specific, and rapid test for the presence of pathogenic bacteria. In this study, a multiplex PCR (m-PCR) was used to detect S. Typhimurium and S. Enteritidis. We selected m-PCR target genes, which were the spv (virulence plasmid specific for S. Enteritidis) and sefA (S. Enteritidis fimbrial antigen) genes, fliC (H1-i antigen specific for S. Typhimurium) and a randomly cloned sequence specific for the genus Salmonella. With m-PCR, random sequence was detected from all strains of Salmonella spp, spv and sefA were detected from all strains of S. Enteritidis (100%), and fliC was detected from all strains of S. Typhimurium (100%). This assay indicate that the specificity of the m-PCR make them potentially valuable tools for detection of S. Typhimurium and S. Enteritidis.

Antimicrobial Effect of Oregano (Origanum majorana L.) Extract on Food-borne Pathogens (오레가노 추출물이 식중독세균에 대한 항균효과)

  • This study was performed to investigate the antimicrobial effects of Origanum majorana L. ethanol extract against food-borne pathogens. The antimicrobial activity of Origanum majorana L. extract was determined using a paper disc method. The extract exhibited growth inhibiting activities in a concentration dependent manner on 10 species microorganisms. The extract of Origanum majorana L. showed the highest antimicrobial activity against Salmonella enteritidis. The growth inhibitory effects of Origanum majorana L. extract on food poisoning microorganisms were determined against Salmonella typhimurium and Listeria monocytogenes, gram negative and positive bacteria, respectively. The extract of Origanum majorana L. had strong antimicrobial activity against Salmonella typhimurium and Listeria monocytogenes at the concentration of $700 mg{\cdot}L^{-1}$. At this concentration, the extract of Origanum majorana L. inhibited the growth of Salmonella typhimurium and Listeria monocytogenes up to 60 and 36 hours, respectively. The results in the present study demonstrate antimicrobial effects of Origanum majorana L. ethanol extract against food-borne pathogens, suggesting that Origanum majorana L. could be an effective natural antibacterial agent in food.

Nontyphoidal Salmonella Meningitis in an Immunocompetent Child

  • Salmonella meningitis is rare yet poses causes significant neurological morbidity in children. Infants, especially those under 3 months of age, and those with immunocompromised states, such as malignancy, malaria, and human immunodeficiency virus infection, are at increased risk for developing Salmonella meningitis. Herein, we describe a case of Salmonella meningitis in a previous healthy 8-year-old girl who presented with high fever, vomiting, and altered mental status. Group D Salmonella species were isolated in cerebrospinal fluid culture, and no abnormal findings were noted in brain magnetic resonance imaging. Immunoglobulin levels and lymphocyte subset counts were within the normal ranges, and no genetic mutation responsible for primary immunodeficiency disease was detected by next-generation sequencing. The patient's condition improved rapidly with third-generation cephalosporin, and no complications or sequalae developed. Nontyphoidal Salmonella can cause meningitis in immunocompetent children and can be successfully treated with early administration of antibiotics.

Distribution and Drug Resistance of Salmonella Causing Enteric Fever in Taegu Area of Korea (대구지방에서의 장열성 Salmonella의 분포와 항균제내성)

  • Salmonella strains isolated from blood in the Dong-san hospital, Taegu during the period from 1971 to 1986 were studied for species distribution, drug resistance, and R plasmids. The number of strains was 2,527 and all of them were classified into S. typhi and S. paratyphi A. Approximately 300 strains were isolated in the period from 1974 to 1976 and 1978, 268 in 1982, and 204 in 1983, but the numbers isolated in the 1980's have a tendency to decrease as compared with those of the 1970's. S. typhi occupied 85% or more of strains isolated until 1976, but the isolation frequency decreased yearly with some variation, and S. paratyphi A increased gradually from 1974. Only 4 strains of S. paratyphi A were resistant to some drugs, and the resistance was not transferred to E. coli by conjugation. S. typhi resistant to drugs were 15 in 1971 through 1973, 24 in 1974, and 13 in 1975, but afterwards only few resistant strains were isolated. These strains were resistant to two or more drugs; chloramphenicol(Cm), tetracycline(Tc), streptomycin(Sm), sulfisomidine(Su), ampicillin(Ap), and kanamycin(Km) and no strain resistant to other drugs tested was found. Strains resistant to 3 or less drugs didn't transfer the resistance to E. coli by conjugation. There were 15 strains resistant to four or more drugs, and were isolated in years from 1972 to 1976. These strains transferred the resistance to E. coli, and the resistance was considered to be mediated by R plasmids. Transfer frequency was higher at $25^{\circ}C$ than at $37^{\circ}C$ and patterns of transferred resistance were Cm, Tc, Sm, Su; Ap, Km; Cm. R plasmids having markers of Cm, Tc, Sm and Su were classified into Inc H1.

  • In order to observe the bactericidal effect of Ranunculus species on pathogenic bacteria, the minimal inhibitory concentration and bactericidal effect was tested Throughout the studies, the following experimental results were obtained and summarized. 1. Minimal inhibitory concentration of Ranunculus species extracts on E. coli was observed in the medium in which 1% Ranunculus species extracts added to brain heart infusion agar. 2. Minimal inhibitory concentration of Ranunculus species extracts on Salmonella species observed in the medium in which 1% Ranunculus species extracts added to brain heart infusion agar. 3. Minimal inhibitory concentration of Ranunculus species extracts on Staphylococcus and Streptococcus was observed in the medium in which 1.5% Ranunculus species extracts added to brain hrart infusion blood agar. 4. The Bactericidal effect of Ranunculus species extracts on E. coli and S. typhi was observed in 30 minutes. 5. The Bactericidal effect of Ranunculus species extracts on staphylococcus aureus was obserded in 40 minutes.

  • We developed a geno-chromatographic assay system based on hybridization between target DNA (e.g., amplified product from PCR) and a capture probe immobillized on a predetermined site of membrane. This system can offer a rapid detection of a gene sequence specific to a certain type of cells as well as simplicity of the analytical procedure. Such performances were demonstrated by utilizing Salmonella typhimurium as model analyte.

  • Salmonella is closely related with human health of modern society which has concern increased in livestock goods consumption as well as give economic damage throughout the chicken industry such as farm, hatchery, slaughter house and processing plant. From 2007 to 2011, this study investigated Salmonella prevalence from 200 old hen delivery trucks which deliver old egg-laying hens and broiler breeders in Korea. The prevalence of Salmonella species was 38.0% in old hen delivery truck. Serogroup C1 was the most frequently detected serogroup of Salmonella, followed by the serogroups D1, C2 and B. A total of 25 serotypes were identified and Salmonella infantis was the most frequently isolated serotype. In addition, we applied disinfectant to old hen delivery truck for the reduction of Salmonella contamination. The disinfectant consists of formaldehyde, glutaladehyde and quaternary ammonium compound was applied to the trucks. Salmonella isolation rate was significantly decreased after disinfection from 38.0% to 7.5%. Disinfectant could not effectively reduce Salmonella contamination at a dilution of 1:200 which is recommended by manufacturer, but Salmonella isolation rate was significantly decreased at a dilution of 1:50. Since old hen delivery truck could be a potential vector to carry Salmonella into farms and abattoirs, chicken delivery truck should be disinfected thoroughly and sufficiently to control contamination of farms and abattoirs.

Epidemiology of pullorum disease and characteristics of Salmonella pullorum isolates in Korea (국내 추백리 발생역학 및 감염계로부터 분리한 Salmonella pullorum의 특성)

  • Pullorum disease caused by Salmonella pullorum, has been considered as one of the most important diseases in both clinically and economically in poultry industry since it had been firstly reported in 1925 in Korea. This disease is still problem in the industry in this country even though several attempts have been made to eradicate the disease. As one of the trials to solve the problem, we investigated the pattern of the outbreak of the disease, isolated the causative agent, S pullorum and tested biochemical properties, antimicrobial susceptibility and plasmid profiles of the isolates. Outbreak of the disease based on the species was the highest in layer followed by in Korean native chick, and broiler. Daily mean mortality in vertical transmission (0.90%) was higher than that in horizontal (0.14%). There was no seasonal difference in the outbreak. Also, biochemical properties and antimicrobial susceptibility pattern of the isolates were same. However four different plasmid profiles of the isolates were observed. These results suggested that S pullorum isolates were different in the genotype while they were same in phenotypes.

Detection of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes in Kimchi by Multiplex Polymerase Chain Reaction (mPCR)

  • We developed an mPCR assay for the simultaneous detection, in one tube, of Escherichia coli O157:H7, Salmonella spp., Staphylococcus aureus and Listeria monocytogenes using species-specific primers. The mPCR employed the E. coli O157:H7 specific primer Stx2A, Salmonella spp. specific primer Its, S. aureus specific primer Cap8A-B and L. monocytogenes specific primer Hly. Amplification with these primers produced products of 553, 312, 405 and 210 bp, respectively. All PCR products were easily detected by agarose gel electrophoresis, and the sequences of the specific amplicons assessed. Potential pathogenic bacteria, in laboratory-prepared and four commercially available kimchi products, were using this mPCR assay, and the amplicons cloned and sequenced. The results correlated exactly with sequences derived for amplicons obtained during preliminry tests with known organisms. The sensitivity of the assay was determined for the purified pathogen DNAs from four strains. The mPCR detected pathogen DNA at concentrations ranging from approximately 0.45 to $0.05\;pM/{\mu}l$. Thus, this mPCR assay may allow for the rapid, reliable and cost-effective identification of four potentially pathogens present in the mixed bacterial communities of commercially available kimchi.

In vivo Antagonistic Effect of Lactobacillus helveticus CU 631 against Salmonella enteritidis KU101 infection

  • In vivo antagonistic effect of Lactobacillus helveticus CU 631 and Lactobacillus spp. against typical enteritis causing pathogen Salmonella enteritidis KU 101 have been determined, which showed an increase in survival rate and the decline in viable cell numbers of pathogen in liver and spleen at sacrifice. A signifcant difference in the antagonistic effect against KU 101 were observed, which was species and/or strain dependent of Lactobacillus (p<0.01), the survival rate of the mice in the Salmonella infection by feeding L. helveticus CU 631 has been shown to be 157%, whereas those of L. rhamnosus GG ATCC 53103, L. acidophilus ATCC 4356, L. johnsonii C-4 were 137%, 132%, 119% respectively on the basis of lactobacilli non-associated control KU101 fed mice to be 100%. Viable cells of S. enteritidis KU101 in the liver and in the spleen at sacrifice were decreased in Lactobacillus spp. fed group with no significant difference. The higher level of total secretory IgA concentration in the intestinal fluid of lactobacilli fed mice than control mice have been observed. In vitro antagonistic activity of Lactobacillus spp. against KU101 have been determined, a prominent antagonistic activity of CU 631 against KU 101 were demonstrated.
