Effect of Mecadox in Treatment of Colibacillosis and Prevention of Experimental Salmonella Infection in Swine (자돈하리증(仔豚下痢症) 및 Salmonella 인공감염증(人工感染症)에 대한 Mecadox의 치료 및 예방효과)

  • The effect of mecadox, trade name of carbodox, in the treatment of colibacillosis and in the prevention of experimental Salmonella infection in swine was studied throughout the experimental period of four weeks using a total of 70 piglets. The results obtained are as followings. 1. Mecadox was found moderately effective as a therapeutic for colibacillosis of piglets when administered orally as pig doser or added to the feed at the ratio of 50 ppm with or without the addition of vitamin A and $D_3$. 2. In the prevention of experimental infection with Sal. choleraesuis, mecadox was found highly effective. As judged by clinical symptoms such as diarrhea and fever together with the confirmation of excretion of the causative organisms in the feces, the best result was obtained with mecadox pig doser followed by mecadox (50ppm) with vitamin A and $D_3$, and mecadox (50 ppm) without the vitamins in the descending order of effectiveness.

  • A very rare case of mycotic aneurysm in the descending thoracic aorta due to salmonellosis was treated in our hospital The patient was a 62 year-old male who 48 days before the operation was admitted to the depart,cent of internal medicine complaining of fever. nausla. vomiting, and loose stool. He was treated for 35 days and discharged. Three days after discharge, however, the patient was readmitted to the hospital comE긴ainin프 of $\ulcorner$i포ht uppe$\ulcorner$ 킥uadrant abdominal pain. Fever developed on the third hospiti린 day, and on the eighth hospital day, the patient complAined of back pain and epigastric pale. A simple chest x-ray showed evidence of hemothorax in the left plueral space, and therefore, computed tomography of the chest was done. The patient was diagnosed as a ruptured mycotic aneurysm of the descending thoracic aorta, and was transferred to our Department of Thoracic and CArdiovascular Surgery. The aneurysm and infected tissues was widely debrided, and the site was then patched with a Dacron graft. Salmonella choleraesuis was identified in the blood and aneurysm cultures, and antibiotics were administered for weeks according to the sensitivity of the organism. The patient experienced no complication thereafter and for the last three months since the operation, he has been leading a healthy and normal social life.

  • This experiment was carried out to develope the production of specific yolk antibody from laying hens immunized with antigens from Salmonella enteritidis. Antigenic protein isolated from the flagella of Salmonella enteritidis, determined by SDS-PAGE, was pure and has a molecular mass of approximately 54.6 kDa. It was observed that the antibody titers both in egg yolk and serum were performed at 2 weeks after immunization with flagella antigen to the laying hen. And the level was increased gradually to 6 weeks after immunization. At the time of 6 weeks, the antibody titer of yolk showed higher than that of serum. According to the results of specificity test(ELISA), the yolk antibody did not react with different bacterial strains(S. choleraesuis, ETEC Kl2:K99, K88,987P), but reacted only with S. enteritidis strain. The contents of immunoglobulin(IgY) in an egg yolk was 106mg approximately. By the isolation procedure of IgY from the egg yolk, 88.3 percent of IgY content was recovered in this study.

Application of mix LPS-ELISA for monitoring of antibodies to major serogroups of Salmonella in animal (Mix LPS-ELISA법을 이용한 살모넬라균 복합감염항체 동시 모니터링)

  • Salmonellosis caused by a number of serotypes of Salmonella is an infectious, acute or chronic, zoonotic disease and characterized by enteritis and diarrhea, septicemia in animal. In these studies we investigated the prevalent serotypes of Salmonella causing animal salmonellosis in Korea and the 71 strains of Salmonella spp. were isolated from materials such as mesenteric lymph nodes, fecal samples from slaughtered animal. With the identification test results, the most prevalent serotypes were, in order, S stanley 31 strains (43.7%), S typhimurium 19 strains (26.8%) and S montevideo 11 strains (15.5%), respectively. And we could establish the method for detection of antibodies to broad variety of Salmonella serotypes. Lipopolysaccharide(LPS) antigen extracted from Salmonella was more sensitive and specific than outer membrane protein antigen from that for detection of Salmonella antibody by using an indirect ELISA. The optimal concentration of antigen was 100ng/ml of LPS, the dilutions of conjugate and serum were 1 : 1,000~2,000 and 1 : 200~400, respectively. The mix LPS-ELISA which was used by mixing LPS from S typhimurium (group B), S choleraesuis (group C) and S enteritidis (group D) were more rapid and effective than that used LPS from individual strain for detection of Salmonella serogroup O4, O7 and O9 antibody at the same time. We could obtain the high values of optical density ($0.73{\pm}0.32$) by mix LPS-ELISA on the farm which had occurred salmonellosis, but very low values of $0.17{\pm}0.06$ on the negative farm of salmonellosis. So, the mix LPS-ELISA may be used to monitor the serological surveillance for the presence of infection with a number of serotypes of Salmonella and would be useful for prevention and control of salmonellosis in animal.

  • To evaluate the availability of Cudrania tricuspidata Bureau as a natural source of antimicrobials, the antimicrobial activity of methanol extracts of harvested parts was investigated using the paper disc diffusion method. The extracts from leaves and root bark had broad antimicrobial activity against various bacteria, including Bacillus subtilis, Bacillus cereus, Staphylococcus aureus, Streptococcus mutans, Listeria monocytogenes, Enterobacter aerogenes, Enterococcus faecalis, Salmonella choleraesuis subsp. choleraesuis, Vibrio vulnificus, and Pseudomonas aeruginosa and inhibited Bacillus cereus, Staphylococcus aureus, and Listeria monocytogenes, agents of food poisoning especially well. The extract from ripe fruit had a very high antimicrobial activity against Staphylococcus aureus with a 20.2 mm of clear zone at 50 mg/mL sample concentration. These results indicated that Cudrania tricuspidata could be used as new source for developing natural antimicrobial agents.

Studies on the Antimicrobial Effect of Extracts of Propolis (프로폴리스 추출물의 항균 활성에 대한 연구)

  • Approved chemical preservatives have been widely used to preserve foods and increase their shelf life. There are increasing demends of the partial or complete removal of chemical preservatives from foods vecause of adverse health effect of chemicals. In this study, the possibility of natural antimicrobial compounds, Korean propolis as food preservatives are investigated. Propolis samples were extracted on various concentration of ethanol. Propolis extracts extracted with 100% ethanol showed the highest inhibitory effect aginst food spoilage microorganisms. The 100% ethanol extracts of propolis were selected and the antimicrobial activites of 100% ethanol extracts of proplis against several food spoilage microorganisms were examined. Bacillus subtilis, Micrococcus luteus, Escherichia coli, Staphylcoccus aureus, Bacillus cereus, Shigella sonnei, Salmonella choleraesuis, Erwinia rhapontici, and Vibrio parahaemolyticus as food poisoning microorganisms were chosen for the examination. The Propolis extracts had antimicrobial activity against food spoilage microorganisms. When the microorganisms were treated with propolis extracts, the population of food spolige microorganisms were decreased by 1~9 log.

Inhibition of Pathogenic Bacteria by Pediococcus pentosaceus Strain SH-10 Isolated from Hard Clam Meretrix meretrix Sikhae (백합(Meretrix meretrix) 식해에서 분리한 Pediococcus pentosaceus SH-10에 의한 병원성 세균의 억제 기작)

  • In this study, we investigated the mechanism of inhibition of pathogenic bacteria by Pediococcus pentosaceus strain SH-10 isolated from hard Clam Meretrix meretrix sikhae. When P. pentosaceus SH-10 was co-cultured in MRS broth with pathogenic bacteria, including Bacillus cereus, Listeria monocytogenes, Salmonella choleraesuis and Staphyloccus aureus, no viable pathogenic cells were detected after 18 h of incubation. However, pediocin or a pediocin-like bacteriocin was not detected in cultures of P. pentosaceus SH-10 by the agar diffusion method. Organic acids were produced in MRS broth in proportion to the incubation time of P. pentosaceus SH-10. These results indicate that P. pentosaceus SH-10 inhibited the growth of pathogenic bacteria by lowering the pH of the growth medium through the production of organic acids, including sodium lactate, sodium acetate, and sodium citrate.

Characteristics of the Antibacterial Substance Produced by Paenibacillus polymyxa JB115 (Paenibacillus polymyxa JB115로부터 생산된 항균물질의 특성)

  • The culture broth of a ${\beta}$-glucan-producing bacterium, Paenibacillus polymyxa JB115, was confirmed to show the antibiosis against pathogenic bacteria of livestock disease. The antibacterial substance produced by P. polymyxa JB115 exhibited strong bactericidal or bacteriostatic effect on the growth of livestock pathogenic bacteria including Staphylococcus aureus, Salmonella choleraesuis, Escherichia coli and Pseudomonas aeruginosa. This antibacterial substance also showed high stabilities in broad pH range (pH 3-11) and in broad temperature range $(40-121^{\circ}C)$, which is good enough to apply spray-dry method for the formulation of culture broth. It was also found that the antibacterial substance was very stable in artificial gastric fluid and bile acid, which implies the anticipated antibacterial activity against gastrointestinal bacteria harmful for livestocks. In conclusion, the culture broth of P. polymyxa JB115 can be developed as a multifuctional feed additive containing immune-enhancing ${\beta}$-glucan as well as antibacterial agent against livestock pathogenic bacteria.

Regulation of stf Operon Expression and Its Virulence (살모넬라가 발현하는 stf 오페론의 조절과 병원성 인자로서의 기능)

  • The stf (Salmonella typhimurium fimbriae) operon consisting of stfA(CDEFG assumes to encode putative fimbriae. The complete stf operon is existed in S. typhimurium and S. choleraesuis, whereas it is absent in S. typhi. Analyses of the amino acid residues between major subunit StfA of the Stf fimbriae and those of known other fimbriaes suggested that Stf belongs to class I type fimbriae. Through comparison of StfD chaperone with the other fimbrial chaperones, and of C-terminus in subunits of Stf fimbriae, it belongs to FGS (with a short Fl-G1 loop) subfamily. In order to investigate the expression of stf operon, we have constructed a Salmonella strain containing a chromosomal stfA::lacZYA transcriptional fusion, resulting in S. typhimurium $_X8532$. The strain $_X8532$ lacked the expression of \beta-galactosidase$ under normal culture conditions. However, with longer incubation time of the S. typhimurium $_X8532$, we have isolated 21 individual strains exhibiting $Lac^+$ phenotype. $Lac^+$ phenotype was appeared as approximately 0.03 frequency per generation. All isolates expressed lacZ constitutively in the various environmental conditions. Various global regulatory proteins including RpoS, OmpR, and CpxR were not involved in the regulation of the stf operon. A S. typhimurium $_X8661$ mutant lacking stfAC function attenuated 6.7 folds more than that of wild type $_X3761$ in the mouse virulence test, suggesting in the somehow involved in the Salmonella pathogenesis.

Antibacterial Activity of Essential Oils from Zanthoxylum piperitum A.P. DC. and Zanthoxylum schinifolium

  • This study was carried out to investigate the potential use of Zanthoxylum schinifolium and Zanthoxylum piperitum A.P. DC. as a source of antimicrobial agents against food borne pathogens. Essential oils of Z. schinifolium and Z. piperitum A.P. DC. were collected by steam distillation and analyzed by GC-MS. The antimicrobial activity of the essential oils was examined using the agar diffusion and micro-dilution assays. The effectiveness of Z. schinifolium essential oil was greater against Bacillus cereus, Staphylococcus aureus, and Vibrio parahaemolyticus than other pathogens, and the minimal inhibitory concentration (MIC) values were 1.25, 2.5, and 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Z. piperitum A.P. DC. essential oil was the most effective against all pathogens tested except for Escherichia coli O157:H7, and the MIC values against B. cereus, Salmonella choleraesuis, and V. parahaemolyticus were 1.25, 2.5, and $1.25\;{\mu}g/mL$, respectively. Limonene, the major component of Z. piperitum A.P. DC. essential oils, had the highest inhibitory activity toward V. parahaemolyticus with a MIC value of $0.15\;{\mu}g/mL$. Meanwhile, citronellal and geranyl acetate, major components of both essential oils, displayed antibacterial activity against only B. cereus with MIC values of 1.25 and $5\;{\mu}g/mL$, respectively. Therefore, these essential oils could be useful as antimicrobial agents against foodborne pathogens.
