닭의 살모넬라 감염증(感染症)에 관한 연구 (Studies on Salmonella Infection of Chicken)

  • Investigations of the prevalence of SalmoneIla organisms in chicken of Suwon area and the effect of selenite broth on the growth of Sal. pullorum were made. The results obtained were summarized as foIlowings. 1. Salmonella organisms was not isolated from fecal samples of 357 chicken from 6 poultry farms of Suwon area. 2. In selenite broth, the growth of Sal. pullorum was completely inhibited may due to the toxic effect of the medium when the inoculum contained less than $5{\times}10^3$ organisms. However, enrichment was attained overcoming the inhibitory action of the medium when $5{\times}10^3$ or more organisms were inoculated.

Microwave 조사에 의한 Salmonella enteritidis, Salmonella typhimurium 과 E. coli의 불활성에 관한 연구 (Thermal Inactivation of Salmonella enteritidis, Salmonella typhimurium and E. coli O111 in Liquid Cultures During Microwave Radiation)

  • The purpose of this study was to determine the thermal inactivation of Salmonella enteritidis, Salmonella typhimurium and E. coli O111 in liquid cultures treated with microwave energy. Furthermore, this study was to introduce new methodologies for studying nonthermal microwave effects on microorganisms, using controlled microwave energy and specially designed apparatuses. For the automatic temperature control during microwave heating, the real time data acquisition and computation system is designed with BASIC routine. The automatic temperature control system used in the experiments perform relatively stable control at the experiment temperature of 45, 50, 55 60$^{\circ}C$ and 65$^{\circ}C$ for 30 minutes. The effects of microwave heating on liquid cultures was compared with that of conventional heating, still reduces effectively the number of pathogenic bacteria in liquid cultures. While no particular differences between microwave heating and conventional heating was observed in the activation of E. coli at 45$^{\circ}C$ test, the activation of Sal. enteritidis and Sal. typhimurium was slightly reduced during the microwave treatments.

알카리 내성 Bacillus속 Promoter의 Cloning (Cloning of Promoters from Alkali-tolerant Bacillus sp.)

  • 토양에서 분리한 알카리 내성 Bacillus sp. YA-14 의 promoter를 Bacillus promoter probe vector인 pPL703을 이용하여 Bacillus subtilis내에 cloning 하였다. 얻어진 형질전환체 중 promoter 활성이 가장 높은 균주의 CAT 비활성은 8.07로 expression vector인 pPL708의 CAT 비활성보다 2.5배 이상 높았으며 대수 증식기가 끝난 이후에 그 활성이 급격하게 증가하였다. 재조합 plasmid내의 삽입 DNA 단편은 그 크기가 2.8kb이고 제한효소 BamHI, Sal I 인식부위가 각각 한군데 존재하였다.

  • 난대림 지역인 전라남도 신안군 흑산면 가거도에서 발견된 미기록 1종 푸른가막살(Viburnum japonicum (Thunb.) Sprengel)을 보고한다. 잎은 상록성으로 털이 없고 광택이 있으며 겨울눈은 나출되어 있어 이미 보고된 가막살나무속(Viburnum)의 다른 종들과 쉽게 구별된다.

  • The ultraviolet photochemistry of N,N'-bis(salicylidene)ethylenediamine copper(II), Cu(sal)2en, was investigated with low pressure mercury lamp. Redution of Cu(Ⅱ) and formation of Cl- were shown on 254 nm irradiation both for aerated and deaerated chlorinated hydrocarbon solvent such as CH2Cl2, chloroform, and 1,2-dichloroethane. Relatively long lived $({\tau}=100{\mu}sec)$ intermediate was detected by flash photolysis. Overall photo-process can be described as the formation of Cl- and new copper complex, product(1) by chlorohydrocarbon mediation, photoinduced reduction by abstraction of halogen from solvent, followed by redox induced substitution of axial ligand with chlorine. Product(1) is possibly Cu(III) chlorosalicylaldeimido complex and cyclic -CH2CH2- moiety is absent in the structure. 247nm band of Cu(sal)2en should contain ligand to metal charge transfer character.

Assessment and Optimization of Xylanase Production Using Mono-Culture and Co-Cultures of Bacillus subtilis and Bacillus pumilus

  • Xylanase is an industrially relevant enzyme used for the production of xylobiose and xylose. Various methods are used to enhance the microbial yield of xylanase. In the present study, co-culturing of Bacillus subtilis and Bacillus pumilus were investigated using submerged fermentation for xylanase production, which was markedly increased when sal, sagwan, newspaper, wheat bran, and xylan were used as single carbon sources. Maximum xylanase production was reported after 5 days of incubation in optimized media at pH 7.0 and 37℃, resulting in 2.69 ± 0.25 µmol/min by coculture. The 1:1 ratio of sal and sagwan in optimized production media was shown to be suitable for xylanase synthesis in submerged fermentation (SMF). In comparison to mono-culture using B. pumilus and B. subtilis, co-culturing resulted in an overall 3.8-fold and 2.15-fold increase in xylanase production, respectively.

Complete Genome Sequence of Salmonella Typhimurium-Specific Phage vB_SalA_KFSST3 Possessing Antibiofilm Activity

  • In a previous study, Salmonella Typhimurium-specific phage vB_SalA_KFSST3, which possess antibiofilm activity, was isolated and purified from wastewater used in slaughterhouses. This study aimed to perform bioinformatic analyses to investigate the genes associated with its antibiofilm activity. Phage genome consisted of a single chromosome of 156,555 bp with a GC content of 44.8%. Among its 202 open reading frames (ORFs), three tail spike proteins (TSPs; orf141, orf142, orf143) were identified with high confidence. All TSPs were predicted to encode putative depolymerase activities, including two endoglycosidases and one endorhamnosidase. The genome has been deposited in GenBank under the accession number PP_994976.1.

경기도 북부지역 야생멧돼지(Sus scrofa) 전염성 질병 감염 실태 조사 (A survey for prevalence of infectious diseases in wild boar (Sus scrofa) in northern Gyeonggi province, South Korea)

  • This study was conducted to investigate the infection of livestock diseases using 500 blood samples from wild boars captured in six cities and one county in northern Gyeonggi province, South Korea. We examined 239 cases of classical swine fever virus (CSFV), and each of 500 cases of foot and mouth disease virus (FMDV), porcine reproductive and respiratory syndrome virus (PRRSV), porcine circovirus type 2 (PCV2), Mycoplasma hyopneumoniae (MH), Actinobacillus pleuropneumoniae (APP), Pasteurella multocida type A (PMA), Hemophilus parasuis (HP), Salmonella (Sal.) spp. infections. Antibodies were detected against CSFV (23.4%), PRRSV (4.0%), PCV2 (60.4%), MH (3.0%), APP (69.2%), PMA (52.8%), HP (11.8%), and Sal. spp. infections (37.2%). No antibodies were detected against FMDV. As a result of antigenic analysis of 68 positive cases (13.6%) out of 500 PRRS antigen tests, 61 North American cases, 6 European cases, 1 North American-European complex case. PCV2 has 158 positive cases (31.6%) out of 500 antigen tests, and the results indicate that a considerable number of individuals are infected. To our knowledge, this is the first seroprevalence report of MH, APP, PMA, HP, and Sal. spp. infections in wild boars in South Korea.

국소 전뇌 허혈 모델 백서에서 가감정지환(加減定志丸)과 오수유(吳茱萸)가 기억증진과 신경세포보호에 미치는 효과 (Effect of Gagamjeongji-hwan and Evodiae Fructus on Memory Impairment and Neuronal Damage Induced by Focal Ischemia in the Rat)

  • This study was conducted to determine the effect of Gagamjeongji-hwan (JJH)(Jiajiandingzhi-wan) and Evoidae Fructus (EF) on learning and memory disturbance and neuronal damage induced by focal ischemia in the rat. Rats were used for testing in the following three. Morris Water Maze, Cholineacetyltransferase (ChAT) immunohistochemistry, acetylcholine esterase (AchE) histochemistry. JJH+ISCH group (ischemia-induced rats pretreated with JJH) and EF+ISCH group (ischemia-induced rats pretreated with EF) significantly reduced the latency of swimming time, compared with those of ISCH group (ischemia-induced rats) in morris water maze acquisition test. JJH+ISCH group attenuated ischemia.induced learning and memory damage in morris water maze retention test. The density of ChAT neurons of the JJH+ISCH and EF+ISCH group in the hippocampal CA1 area was increased, compared to that of SAL+ISCH group (ischemia-induced rats pretreated with SAL). The density of AchE neurons of the JJH+ISCH and EF+ISCH group in the hippocampal CA1 and CA3 area was increased, compared to that of SAL+ISCH group. These results suggest that Gagamjeongji-hwan (JJH) and Evodiae Fructus (EF) may have significant protective effects on ischemia-induced brain damage and memory impairments.
