Experimental Studies on Detection of Salmonellae in Animal-origin Foods by Means of Dirct Fluorescent Antibody Technique (직접형광항체법(直接螢光抗體法)에 의한 축산식품중(畜産食品中)의 Salmonella 균(菌) 검출(檢出)에 관한 실험적(實驗的) 연구(硏究))

  • The experiment was performed in order to investigate the applicability of the rapid detection of salmonellae in various animal-origin foods by means of the direct fluorescent antibody technique. Egg, sausage and chicken were inoculated with various concentrations of Sal.paratuphi A, Sal. paratyhi B and Sal. thompson, and the fluorescent antibody technique was applied and compared with the conventional cultura method for the sensitivity of detection of the organisms. Two methods were employed in the fluorescent antibody technique; the direct smear method in which the smear being made directly from the specimens, and the enrichment smear method in which the smear being made from the enrichment broth. The effect of various enrichment time (1,5,8,11 and 13 hours) in tetrathionate broth on the detection of salmonellae in the fluoresent antibody technique was also studied. The results obtained were summarized as followings; 1. Of the three methods, the enrichment smear method of fluorescedt antibody technique was highly effective as cultural method for the detection of salmonella organisms. 2. Direct smear method of fluorescent antibody technique was effective as two other methods $5{\times}10^4$ organisms presented in 50 g(ml) of specimens. This method may not be applicable when the specimens contained $5{\times}10^2$ or less organisms. 3. Of the three specimens, the recovery rate of Salmonella organisms from egg was slightly higher than that of sausage and chicken. 4. In fluorescent antibody technique and cultural method, the specimens inoculated with Sal. thompson were found to be higher detection rate than the specimens inoculated with Sal. paratyphi A, 5. The optimum enrichment time of Salmonella organisms in tetrathionate broth on the detection by fluorscent antibody technique was found to be 11 hours or longer when the specimens of egg, sausage and chicken were inoculated with approximately 500 organisms. The longer enrichment time was the higher detection rate up to 11 hours tested.

  • Molecular cloning and restriction mapping on ATPase $\alpha$-subunit gene (atpA) were carried out to obtain genomic information concerned with the gene structure and organization in Korean ginseng mitochondria. Two different clones containing the homologous sequence of atpA gene were selected from SalI and PstI libraries of mitochondrial DNA (mtDNA) of Korean ginseng. The sizes of mtDNA fragments inserted in SalI and PstI clones were 3.4 kb and 13 kb, respectively. Southern blot analysis with [$^{32}P$] labelled Oenothera atPA gene probe showed that atpA gene sequence was located in 2.0 kb XkaI fragment in PstI clone and in 1.7 kb XbaI fragment in SalI clone. A partial sequening ascertained that the SalI clone included about 1.2 kb fragment from SalI restriction site to C-terminal sequence of this gene but about 0.3 kb N-terminal sequence of open reading frame was abscent. The PstI fragment was enough large to cover the full sequence of atpA gene. The same restriction pattern of the overlapped region suggests that both clones include the same fragment of atiA locus. Data of Southern blot analysis and partial nucleotide sequencing suggested that mtDNA of Korean ginseng has a single copy of atpA gene. Key words ATPase a-subunit, mitochondrial DNA, Panax ginseng.

Shear-induced microstructure and rheology of cetylpyridinium chloride/sodium salicylate micellar solutions

  • In this article, we considered shear-induced microstructure and rheological behavior of micellar solutions of cationic surfactant, cetylpyridinium chloride (CPC) in the presence of a structure-forming additive, sodium salicylate (NaSal). Shear viscosity, shear moduli and flow birefringence were measured as functions of the surfactant and additive concentrations. In the presence of NaSal, the micellar solution exhibited the non-linear rheological behavior due to the formation of supramolecular structures when the molar ratio of NaSal to CPC exceeded a certain threshold value. Flow birefringence probed the change in micelle alignment under shear flow. At low shear rates, the flow birefringence increased as the shear rate increased. On the other hand, fluctuation of flow birefringence appeared from the shear rate near the onset of shear thickening, which was caused by shear-induced coagulation or aggregation. These results were confirmed by the SEM images of in situ gelified micelle structure through sol-gel route.

  • The isolation and charateriation of a type II restriction endonuclease from Xanthomonas citri IFO 3835 were described. This enzyme (Xci I endonuclease) is an isoschizomer of Sal I endonuclease recognizing 5'-GTCGAC-3' and cleaving at the site indicated by the arrow. Unlike Sal I endonuclease, Xci I endonuclease requries a NaCl concentration of 50mM for its maximum activity.

A Study on the Natural Hybridization between M. lagowskii and M. owcephalus (버들개와 버들치의 자연잡종에 관한 연구)

  • 버들치, 버들개의 공서지역에서 2종 사이의 종문제를 명확히 구명하기 위해 883개체를 채집하여 형태형 질과 유전자분석을 실시하여 생식적 격 리기작 여부를 조사한 결과는 다음과 같다. 2종 사이 에 잡종개체가 약 12%( n = 104) 발견되었다. 형 태분류형 질인 측선상부린수( SAL)의 경우 버들개와 버들치가 각각 SAL = 24.25$\pm$1, 32, 16.93 $\pm$ 0.92로 뚜렷한 차이를 나타내었으나 잡종개체는 평균 SAL = 19.56$\pm$1.80으로 중간값을 나타내었다. 잡종개체 중 F2 이상의 잡종개체 비율은 약 2.7%로 Fl(9"yp)에 비해 현저히 낮고, 잡종개체의 출현률이 기 대치보다 낮은 것으로 보아(P < 0.001) 2종간에 자유교배가 되지 않으며 따라서 Predating isolating mechanism이 작용한다고 보며 또한 잡종개체의 치사율이 높은 점으로 보아 이들사이에는 Postdating isolating mechanism도 동시에 작용한다고 사료되며 따라서 이들 2종은 비교적 최근에 분화된 별종임이 재확인 되었다. 되었다.

Study on Formation and Properties of Dioxomolybdenum Complexes (디옥소몰리브덴 착물의 합성과 그 성질에 대한 연구)

  • Dioxobis(sub.-salicylaldiminato) molybdenum (VI) complexes, $MoO_2\;(X-sal-N-R)_2,\;(X=H,\;5-CH_3,\;R=C_6H_5,\;p-F-C_6H_4,\;m-Cl-C_6H_4,p-I-C_6H_4\;and\;p-C_2H_5-C_6H_4)$, have been prepared by reactions of dioxobis(sub.-salicylaldehydato) molybdenum (VI), $MoO_2(X-sal)_2$ with primary amines, in which $MoO_2(X-sal)_2$ complexes were obtained by acidification of a mixture solution of ammonium paramolybdate in water and appropriate salicylaldehyde in methanol. All these complexes show two strong Mo=O stretching imodes in the 900-940$cm^{-1}$ and p.m.r. spectra exhibited only one signal for the azomethine group. These results confirmed that the complexes are six-coordinated octahedron with a $cis-MoO_2$ group and the geometrical configurations of the complexes possess a C2 axis of symmetry. From the mass analyses of the complexes, it found that the composition ratios of $MoO_2$ : ligand are 1 : 2. The charge transfer transition corresponding to N-Mo, and O-Mo occured at 29,000$cm^{-1}$ and 32,000$cm^{-1}$ respectively. Where, the complexes were found to be non-ionic materials by conductivity measurements in dimethylformamide.

Evidences that Suggest the Spread of Multiple-Antibiotic-Resistance (mar) Operon of Escherichia coli Mutants among Gram-Negative Bacilli (Mar (Multiple-Antibiotic-Resistance) Operon 돌연변이 대장균의 그람음성 세균들간 전파 가능성에 대한 근거)

  • To evaluate the spreading possibilities of the marRAB mutation of E. coli Mar mutant among gram-negative bacilli, chromosomal marRAB mutations of Mar mutants were transduced by $\lambda$placMu9 into pUC19 (Lac$^{+}$, Ap$^{r}$) cloning site in another strains of E. coli or onto the chrmosome of S. typhimurium and P. aeruginosa, selected for transduction by Mar phenotype, Lac$^{-}$, or Ap$^{r}$, and tested for their antimicrobial resistance with or without addition of salicylate (SAL). Compared with wild type strains of JM109, NM522, harboring pUC19 or not, respectively, all strains of JM109 or NM522 carrying pUC19::marRAB mutation showed higher levels of antimicrobial resistance and SAL induction of Mar phenotype than those of wild type. However, in contrast to the original Mar mutants, there were some tendencies of decreased antimicrobial resistance of JM109 or NM522 harboring pUC19::marRAB mutation with SAL induction against chlorarnphenicol (Cm) and tetracycline (Tc), or Tc and ciprofloxacin (Cp), respectively. Almost the same results, as shown as the cases of E. coli JM109 or NM522, were obtained from all transductants of S. typhimurium and P. aeruginosa, except Cp, against which increased antimicrobial resistance with SAL induction was shown. This study, employed the methods of transformation or transduction among intercellular gene transfer methods between gram-negative bacteria, shows the evidences that suggest indirectly the spreading possibilities of marRAB mutation among gram-negative bacilli.

Rapid Detection of Salmonella spp. in Fresh-Cut Cabbage by Real-Time PCR (Real-Time PCR을 이용한 신선편이 양배추에서 Salmonella spp.의 신속검출)

  • This study was conducted to find out the minimal time needed for detection of Salmonella spp. which exist at very low concentration in foods by using real-time PCR. The sal-F and sal-R sequences were used as primers and sal-P was used as a probe. The detection limit of Salmonella spp. was $3.77{\times}10^2\;cfu/mL$ in buffered peptone water (BPW). Microbial growth was monitored after artificially inoculated Salmonella spp. into BPW. The obtained growth curve was well fitted with the equation, y=$0.0127x^2$+0.5927x-0.4317 ($R^2$=0.99), if assuming that 1 cell exists in 25 g sample (0.04 cfu/mL). The microbial concentration will be reduced to 10 fold by adding BPW during sample treatment, so actual initial concentration at the starting point of enrichment is 0.004 cfu/mL. At this condition, real-time PCR detection would be possible only when microbial concentration increase occurs to exceed the detection limit (377 cfu/mL). The time needed for microbial increase was calculated from the growth curve equation as 7 hours and 20 minutes. Therefore the total time required for detection was less than 10 hours including the PCR operating time.

  • Sal, typhi, Ty 2 菌體에 電離放射線을 照射하여 放射線 照射時 隋半되는 菌體의 化學的 性狀을 比較 分析하였다. 放射線 2.5 Mrad, 5.0 Mrad 照射로서 酸性 Amino acid는 增加하였으나 中性 및 鹽基性 Amino acid는 若干 減少하였다. 5.0 Mrad 照射로서 菌體의 總 Carbohydrate 量은 49.7%, 總 Nitrogen 量은 44.6%, Lipid 에 있어서는 Sterol 40%, Glyceride 30%, Phospholipid 14%의 減少를 各各 나타내었다. Polyacrylamide gel 電基泳動結果 放射線 照射群에서는 對照群에 比하여 Cell membrane protein의 變性을 認知할 수 있었다.
