Spatial database architecture for organizing a unified information space for manned and unmanned aviation

  • The widespread introduction of unmanned aircrafts has led to the understanding of the need to organize a common information space for manned and unmanned aircrafts, which is reflected in the Russian Unmanned aircraft system Traffic Management (RUTM) project. The present article deals with the issues of spatial information database (DB) organization, which is the core of RUTM and provides storage of various data types (spatial, aeronautical, topographical, meteorological, vector, etc.) required for flight safety management. Based on the analysis of functional capabilities and types of work which it needs to ensure, the architecture of spatial information DB, including the base of source information, base of display settings, base of vector objects, base of tile packages and also a number of special software packages was proposed. The issues of organization of these DB, types and formats of data and ways of their display are considered in detail. Based on the analysis it was concluded that the optimal construction of the spatial DB for RUTM system requires a combination of different model variants and ways of organizing data structures.

Adenovirus Vectors: Excellent Tools for Vaccine Development

  • Adenovirus was originally used as a vector for gene therapy. In recent years, with the development of the next-generation vectors with increased safety and high immunogenicity to transgene products, its utility as a vaccine vector has continued to increase. Adenovirus-based vaccines are currently being tested not only to prevent various infectious diseases but also to be applied as cancer vaccines. In this review, I discuss the innate and adaptive aspects of the immunological characteristics of adenovirus vectors and further examine the current status of advanced adenovirus-based vaccine development. Various methods that can overcome the limitations of currently used adenoviruses as vaccine vehicles are also discussed. Through this study, I hope that vaccine development using adenovirus vectors will be expedited and more successful.

Influence of Immunity Induced at Priming Step on Mucosal Immunization of Heterologous Prime-Boost Regimens

  • Background: The usefulness of DNA vaccine at priming step of heterologous prime-boost vaccination led to DNA vaccine closer to practical reality. DNA vaccine priming followed by recombinant viral vector boosting via systemic route induces optimal systemic immunity but no mucosal immunity. Mucosal vaccination of the reversed protocol (recombinant viral vector priming-DNA vaccine boosting), however, can induce both maximal mucosal and systemic immunity. Here, we tried to address the reason why the mucosal protocol of prime-boost vaccination differs from that of systemic vaccination. Methods: To address the importance of primary immunity induced at priming step, mice were primed with different doses of DNA vaccine or coadministration of DNA vaccine plus mucosal adjuvant, and immunity including serum IgG and mucosal IgA was then determined following boosting with recombinant viral vector. Next, to assess influence of humoral pre-existing immunity on boosting $CD8^+$ T cell-mediated immunity, $CD8^+$ T cell-mediated immunity in B cell-deficient (${\mu}K/O$) mice immunized with prime-boost regimens was evaluated by CTL assay and $IFN-{\gamma}$-producing cells. Results: Immunity primed with recombinant viral vector was effectively boosted with DNA vaccine even 60 days later. In particular, animals primed by increasing doses of DNA vaccine or incorporating an adjuvant at priming step and boosted by recombinant viral vector elicited comparable responses to recombinant viral vector primed-DNA vaccine boosted group. Humoral pre-existing immunity was also unlikely to interfere the boosting effect of $CD8^+$ T cell-mediated immunity by recombinant viral vector. Conclusion: This report provides the important point that optimally primed responses should be considered in mucosal immunization of heterologous prime-boost regimens for inducing the effective boosting at both mucosal and systemic sites.

HIV-l 유래 렌티바이러스 벡터의 복제가능 바이러스 검출과 역가측정 분석방법 비교 (Comparison of Analysis Methods for Detection of Replication Competent Virus and Functional Titers of HIV-l Based Lentivirus Vector)

  • Human Immunodeficiency Virus type 1 (HIV-l) based lentivirus vector has demonstrated great potential as gene therapy vectors mediating efficient gene delivery and long-term transgene expression in both dividing and nondividing cells. However, for clinical studies it must be confirmed that vector preparations are safe and not contaminated by replication competent lentivirus (RCL) related to the parental pathogenic virus, HIV-l. In this study, we would like to establish the method for titration and RCL detection of lentivirus vector. The titration was determined by vector expression containing the green fluorescent protein, GFP in transduced cells. The titer was $1{\times}10^7$ Transducing Unit/ml in the GFP expression assay and $8.9{\times}10^7$ molecules/ml in the real-time PCR. Also, for the detection of RCL, we have used a combination method of PCR and p24 antigen detection. First, PBS/psi and VSV-G region in the genomic DNA of transduced cells was detected by PCR assay. Second, transfer and expression of the HIV-1 gag gene was detected by p24 ELISA. In an attempt to amplify any RCL, the transduced cells were cultured for 3 weeks (amplification phase) and the supernatant of amplified transduced cell was used for the second transduction to determine whether a true RCL was present (indicator phase). Analysis of cells and supernatant at day 6 in indicator phase were negative for PBS/psi, VSV-G, and p24 antigen. These results suggest that they are not mobilized and therefore there are no RCL in amplification phase. Thus, real-time PCR is a reliable and sensitive method for titration and RCL detection of lentivirus vector.

Strain demand prediction of buried steel pipeline at strike-slip fault crossings: A surrogate model approach

  • Significant progress in the oil and gas industry advances the application of pipeline into an intelligent era, which poses rigorous requirements on pipeline safety, reliability, and maintainability, especially when crossing seismic zones. In general, strike-slip faults are prone to induce large deformation leading to local buckling and global rupture eventually. To evaluate the performance and safety of pipelines in this situation, numerical simulations are proved to be a relatively accurate and reliable technique based on the built-in physical models and advanced grid technology. However, the computational cost is prohibitive, so one has to wait for a long time to attain a calculation result for complex large-scale pipelines. In this manuscript, an efficient and accurate surrogate model based on machine learning is proposed for strain demand prediction of buried X80 pipelines subjected to strike-slip faults. Specifically, the support vector regression model serves as a surrogate model to learn the high-dimensional nonlinear relationship which maps multiple input variables, including pipe geometries, internal pressures, and strike-slip displacements, to output variables (namely tensile strains and compressive strains). The effectiveness and efficiency of the proposed method are validated by numerical studies considering different effects caused by structural sizes, internal pressure, and strike-slip movements.

  • The prospect of live vaccines consisting of genetically modified vaccinia virus expressing foreign genes is exciting, but important issues concerning safety and efficacy need to resolved. Vaccinia virus (VV) is an efficient expression vector with broad host range infectivity and large DNA capacity. This vector has been particularly useful for identifying target antigens for humoral and cell-mediated immunity. The WHO smallpox eradication program, involving the extensive use of VV vaccines, resulted in the late 1970s in the elimination of one of the world's most feared diseases. This achievement is a triumph for preventive medicine and for international collaboration in public health. In 1980, WHO recommended that the routine use of smallpox vaccine should be stopped. Against this background, the prospect of li ve vaccines consisting of genetically modified VV expressing foreign antigens arising from the work of Moss, and Paoletti and their colleagues in 1982 has been greeted with enthusiasm. These investigators have shown that genes coding for immunogenic proteins can be inserted into VV DNA without impairing the ability of the virus to grow in cell culture. Moreover experimental animals infected with VV recombinants containing genes coding for a variety of immunizing proteins have been shown to be protected against challenge infection with the corresponding infectious agent. In this communication, I describe current progress in the construction of a novel plasmid vector that facilitate the insertion and expression of foreign genes in VV as well as the selection of recombinants.

FUZZY SUPPORT VECTOR REGRESSION MODEL FOR THE CALCULATION OF THE COLLAPSE MOMENT FOR WALL-THINNED PIPES

  • Since pipes with wall-thinning defects can collapse at fluid pressure that are lower than expected, the collapse moment of wall-thinned pipes should be determined accurately for the safety of nuclear power plants. Wall-thinning defects, which are mostly found in pipe bends and elbows, are mainly caused by flow-accelerated corrosion. This lowers the failure pressure, load-carrying capacity, deformation ability, and fatigue resistance of pipe bends and elbows. This paper offers a support vector regression (SVR) model further enhanced with a fuzzy algorithm for calculation of the collapse moment and for evaluating the integrity of wall-thinned piping systems. The fuzzy support vector regression (FSVR) model is applied to numerical data obtained from finite element analyses of piping systems with wall-thinning defects. In this paper, three FSVR models are developed, respectively, for three data sets divided into extrados, intrados, and crown defects corresponding to three different defect locations. It is known that FSVR models are sufficiently accurate for an integrity evaluation of piping systems from laser or ultrasonic measurements of wall-thinning defects.

움직임 벡터를 이용한 낙상 감지 시스템 (Fall Detection System Using Motion Vector)

  • 본 논문에서는 움직임 벡터를 이용한 낙상 감지 시스템에 관해 기술한다. 두드러진 움직임을 위한 가중치 차영상 기법, 움직임 벡터를 이용하여 인간이라고 판단되는 블랍을 검출하고, 추출된 움직임 벡터를 이용하여 낙상 여부를 판단한다. 기존의 영상 기반 낙상 감지 시스템의 경우 특정 방향으로 낙상이 발생하는 경우에만 낙상 감지에 성공하였지만 제안 시스템의 경우 다양한 각도에서 낙상이 발생하여도 상황 판단이 가능하다는 장점이 있다. 실험을 위해서 150개의 상황을 연출하였으며, 약 85% ~ 97.1% 낙상 상황 판단 성공률을 보였다.

  • Although there are still many technical difficulties to be overcome, recent advances in the molecular and cellular biology of gene transfer have made it likely that gene therapy will soon start to play an increasing role in clinical practice. However. safety issues are raised from vector system. It is not clear whether it is safe to incorporate genes into nuclear DNA. Little is known about the antigenicity of gene product which the immune system is encountering. In this review, some safety-related topics are introduced and discussed.

A Study on Evaluation of Whole-Body Vibration from Vehicle for Different Road Surfaces

  • The purpose of this study is the measurement of whole-body vibration for different road surfaces. Experimental measurements were taken on asphalt, cement, and off-road surfaces as defined by ISO 2631-1. Each experiment was conducted under the same set of conditions (measurement duration, times, speed, vehicle type). Measurement duration was 10 minutes and 3 separate measurements were taken on each road surface. Vehicle speed was 60km/h. In accordance with ISO 2631-1, an acceleration sensor is set up between the driver's seat and the human body. For evaluation, RMS(root-mean-square) values were taken as suggested by ISO 2631-1. The results suggest "health guidance caution zones", and the evaluation was based on obtaining the vector sum with "health guidance caution zones".
