The murine leukemia virus-based semi-retroviral replicating vectors (MuLV-based sRRV) had been developed to improve safety and transgene capacity for cancer gene therapy. However, despite the apparent advantages of the sRRV, improvements in the in vivo transduction efficiency are still required to deliver therapeutic genes efficiently for clinical use. In this study, we established a gibbon ape leukemia virus (GaLV) envelope-pseudotyped semi-replication-competent retrovirus vector system (spRRV) which is composed of two transcomplementing replication-defective retroviral vectors termed MuLV-Gag-Pol and GaLV-Env. We found that the spRRV shows considerable improvement in efficiencies of gene transfer and spreading in both human glioblastoma cells and pre-established human glioblastoma mouse model compared with an sRRV system. When treated with ganciclovir after intratumoral injection of each vector system into pre-established U-87 MG glioblastomas, the group of mice injected with spRRV expressing the herpes simplex virus type 1-thymidine kinase (HSV1-tk) gene showed a survival rate of 100% for more than 150 days, but all control groups of mice (HSV1-tk/PBS-treated and GFP/GCV-treated groups) died within 45 days after tumor injection. In conclusion, these findings sug-gest that intratumoral delivery of the HSV1-tk gene by the spRRV system is worthy of development in clinical trials for the treatment of malignant solid tumors.
본 논문에서는 전투체계 위협지수를 머신러닝 모델 중 Gradient Boosting Regreesor, Suppor Vector Regressor를 통해 예측하는 방법을 제시한다. 현재 전투체계는 안전성과 신뢰성이 중시되는 소프트웨어이므로 신뢰성이 보장되지 않은 AI 기술의 적용을 정책상 제한하고 있으며, 이로 인하여 전력화된 국내 전투체계는 AI 기술을 탑재하고 있지 않다. 하지만 AI의 전력화를 목표로 하는 국방부의 정책 방향에 대응하기 위하여, 전투체계의 머신러닝 적용에 필요한 기반 기술을 확보하기 위한 연구를 실시하였다. 이 연구는 위협지수 평가에 필요한 데이터를 수집한 뒤 데이터 가공 및 정제, 머신러닝 모델 선정 및 최적의 하이퍼 파리미터를 선정하여 학습된 모델의 예측 정확도를 판단하였다. 그 결과 테스트 데이터에 대한 모델 점수가 99점 이상으로 도출되었으며 전투체계에 머신러닝 모델의 적용 가능성을 확인하였다.
항로 설정은 통항 선박들의 안전을 위해 교통 흐름을 반영할 필요가 있으며, 선박들이 항로를 잘 준수하는지 지속적인 경과 분석이 필요하다. 본 연구에서는 완도항 인근해역 추천항로의 문제점을 도출하고 이에 대한 개선안을 제시하였다. 효율적인 항로 중앙선을 설정하기 위해 선박 항적을 기반으로 서포트 벡터 머신을 이용하였다. 추천항로 중앙선을 기준으로 우측으로 항해해야 하므로 통항 선박들의 항적이 2개의 군집으로 분할된다. 서포트 벡터 머신은 패턴 인식 등 많은 분야에서 이용되고 있으며, 특히 이진 분류 기능이 뛰어나다. 연구 결과 장죽수도 방향의 2.4 NM 추천항로 구간에서 동진하는 상선은 약 79.5%가 추천항로를 준수하지 않는 것으로 나타나 선박 충돌 사고 위험이 상존하는 것을 확인하였다. 추천항로를 현 위치에서 북쪽으로 약 300 m 재설정할 경우, 동진하는 상선은 항로를 역주행할 비율이 79.5%에서 30.9%로 낮아지는 것으로 나타났다. 본 연구에서 적용한 서포트 벡터 머신은 선박 항적을 두 군집으로 분류가 가능하므로 항로 중앙선을 효과적으로 설정하는데 응용할 수 있을 것으로 기대된다.
재조합 배큘로 바이러스는 배양 된 곤충 세포에서 이종 유전자를 발현하는데 널리 사용된다. 재조합 배큘로 바이러스는 광범위한 포유류 세포 유형에서 재조합 단백질의 발현을위한 유전자 전달 벡터로서 작용할 수있다. 바큘로 바이러스 시스템은 안전성, 대규모 및 높은 수준의 유전자 발현 관점에서 중요한 이점을 갖는다. 본 연구에서는 pOPINEneo-3C-GFP 벡터로부터 재구성 된 바큘로 바이러스 벡터를 사이토 메갈로 바이러스 (CMV) 프로모터, 강화 된 녹색 형광 단백질 (EGFP) 및 p53과 NcoI 및 XhoI로 재조합시켰다. 이러한 재조합 벡터를 다양한 세포 및 세포주에 감염시켰다. 이와 같이 개발 된 바큘로 바이러스 벡터는 재조합 유전자의 전이 및 발현을 통상적 인 벡터와 비교하여 분석 하였다. 이러한 결과는 바큘로 바이러스 벡터가 대조군 벡터보다 전이 및 전이에서 더 높은 효율을 갖는다는 것을 시사한다. 본 연구는 과학 기술부, 한국 정보 기술 진흥 기금 (MSIP)이 후원하는 한국 연구 재단(NRF)을 통해 중견 연구원 프로그램 (NRF-2016R1A2B4016552)을 통해 지원되었다.
최근, 컴퓨터 비전과 기계 학습 기술의 도움을 받아 효율적이고 자동적인 도시 환경에 대한 분석 방법의 개발에 대한 연구가 이루어지고 있다. 많은 분석들 중에서도 도시의 안전도 분석은 지역 사회의 많은 관심을 받고 있다. 더욱 정확한 안전도 점수 예측과 인간의 시각적 인지를 반영하기 위해서, 인간의 시각적 인지에서 가장 중요한 전역 정보와 지역 정보의 고려가 필요하다. 이를 위해 우리는 전역 칼럼과 지역 칼럼으로 구성된 Double-column Convolutional Neural Network를 사용한다. 전역 칼럼과 지역 칼럼 각각은 입력은 크기가 변환된 원 영상과 원 영상에서 무작위로 크로핑을 사용한다. 또한, 학습 과정에서 특정 칼럼에 오버피팅되는 문제를 해결하기 위한 새로운 학습방법을 제안한다. 우리의 DCNN 모델의 성능 비교를 위해 2개의 SVR 모델과 3개의 CNN 모델의 평균 제곱근 오차와 상관관계 분석을 측정하였다. 성능 비교 실험 결과 우리의 모델이 0.7432의 평균 제곱근 오차와 0.853/0.840 피어슨/스피어맨 상관 계수로 가장 좋은 성능을 보여주었다.
The conventional Kalman Filter (KF) provides a promising way for structural state estimation. However, the physical parameters of structural systems or models should be available for the estimation. Moreover, it is not applicable when the loadings applied to the structures are unknown. To circumvent the aforementioned limitations, a two-stage KF with unknown input approach is proposed for the simultaneous identification of structural parameters and unknown loadings. In stage 1, a modified observation equation is employed. The structural state vector is estimated by KF on the basis of structural parameters identified at the previous time-step. Then, the unknown input is identified by Least Squares Estimation (LSE). In stage 2, based on the concept of sensitivity matrix, the structural parameters are updated at the current time-step by using the estimated structural states obtained from stage 1. The effectiveness of the proposed approach is numerically validated via a five-story shearing model under random and earthquake excitations. Shaking table tests on a five-story structure are also employed to demonstrate the performance of the proposed approach. It is demonstrated from numerical and experimental results that the proposed approach can be used for the identification of parameters of structure and the external force applied to it with acceptable accuracy.
Performing high-fidelity computational fluid dynamics (HF-CFD) to predict the flow and heat transfer state of the coolant in the reactor core is expensive, especially in scenarios that require extensive parameter search, such as uncertainty analysis and design optimization. This work investigated the performance of utilizing a multi-fidelity reduced-order model (MF-ROM) in PWR rod bundles simulation. Firstly, basis vectors and basis vector coefficients of high-fidelity and low-fidelity CFD results are extracted separately by the proper orthogonal decomposition (POD) approach. Secondly, a surrogate model is trained to map the relationship between the extracted coefficients from different fidelity results. In the prediction stage, the coefficients of the low-fidelity data under the new operating conditions are extracted by using the obtained POD basis vectors. Then, the trained surrogate model uses the low-fidelity coefficients to regress the high-fidelity coefficients. The predicted high-fidelity data is reconstructed from the product of extracted basis vectors and the regression coefficients. The effectiveness of the MF-ROM is evaluated on a flow and heat transfer problem in PWR fuel rod bundles. Two data-driven algorithms, the Kriging and artificial neural network (ANN), are trained as surrogate models for the MF-ROM to reconstruct the complex flow and heat transfer field downstream of the mixing vanes. The results show good agreements between the data reconstructed with the trained MF-ROM and the high-fidelity CFD simulation result, while the former only requires to taken the computational burden of low-fidelity simulation. The results also show that the performance of the ANN model is slightly better than the Kriging model when using a high number of POD basis vectors for regression. Moreover, the result presented in this paper demonstrates the suitability of the proposed MF-ROM for high-fidelity fixed value initialization to accelerate complex simulation.
The lactic acid bacteria species Lactobacillus plantarum (L. plantarum) has been used extensively for vaccine delivery. Considering to the critical role of dendritic cells in stimulating host immune response, in this study, we constructed a novel CD11c-targeting L. plantarum strain with surface-displayed variable fragments of anti-CD11c, single-chain antibody (scFv-CD11c). The newly designed L. plantarum strain, named 409-aCD11c, could adhere and invade more efficiently to bone marrow-derived DCs (BMDCs) in vitro due to the specific interaction between scFv-CD11c and CD11c located on the surface of BMDCs. After incubation with BMDCs, the 409-aCD11c strain harboring a eukaryotic vector pValac-GFP could lead to more efficient expression of GFP compared with wild-type strains shown by flow cytometry analysis, indicating the enhanced translocation of pValac-GFP from L. plantarum to BMDCs. Similar results were also observed in an in vivo study, which showed that oral administration resulted in efficient expression of GFP in both Peyer's patches (PP) and mesenteric lymph nodes (MLNs) within 7 days after the last administration. In addition, the CD11c-targeting strain significantly promoted the differentiation and maturation of DCs, the differentiation of $IL-4^+$ and $IL-17A^+$ T helper (Th) cells in MLNs, as well as production of $B220^+$$IgA^+$ B cells in the PP. In conclusion, this study developed a novel DC-targeting L. plantarum strain which could increase the ability to deliver eukaryotic expression plasmid to host cells, indicating a promising approach for vaccine study.
The development of autonomous driving and Advanced Driver Assistance System (ADAS) technology has grown rapidly in recent years. As most traffic accidents occur due to human error, self-driving vehicles can drastically reduce the number of accidents and crashes that occur on the roads today. Obviously, technical advancements in autonomous driving can lead to improved public driving safety. However, due to the current limitations in technology and lack of public trust in self-driving cars (and drones), the actual use of Autonomous Vehicles (AVs) is still significantly low. According to prior studies, people's acceptance of an AV is mainly determined by trust. It is proven that people still feel much more comfortable in personalized ADAS, designed with the way people drive. Based on such needs, a new attempt for a customized ADAS considering each driver's driving style is proposed in this paper. Each driver's behavior is divided into two categories: assertive and defensive. In this paper, a novel customized ADAS algorithm with high classification accuracy is designed, which divides each driver based on their driving style. Each driver's driving data is collected and simulated using CARLA, which is an open-source autonomous driving simulator. In addition, Long Short-Term Memory (LSTM) and Gated Recurrent Unit (GRU) machine learning algorithms are used to optimize the ADAS parameters. The proposed scheme results in a high classification accuracy of time series driving data. Furthermore, among the vast amount of CARLA-based feature data extracted from the drivers, distinguishable driving features are collected selectively using Support Vector Machine (SVM) technology by comparing the amount of influence on the classification of the two categories. Therefore, by extracting distinguishable features and eliminating outliers using SVM, the classification accuracy is significantly improved. Based on this classification, the ADAS sensors can be made more sensitive for the case of assertive drivers, enabling more advanced driving safety support. The proposed technology of this paper is especially important because currently, the state-of-the-art level of autonomous driving is at level 3 (based on the SAE International driving automation standards), which requires advanced functions that can assist drivers using ADAS technology.
최근 AI 기술의 발전과 정립으로 자동화 분야에서 머신러닝 기법의 활용이 활발하게 이루어지고 있다. 머신러닝 기법의 활용에 있어 중요한 점은 데이터 특성에 따라 적합한 알고리즘이 존재한다는 점이며, 머신러닝 기법 적용을 위한 데이터세트의 분석이 필요하다. 본 연구에서는 다양한 머신러닝 기법을 기반으로 하천 하부의 토사지반을 통과하는 토압식 쉴드TBM 터널 구간의 지반정보와 굴진정보를 사용하여 토압식 쉴드TBM의 굴진율을 예측하였다. 선형회귀모델에서 모델의 통계적인 유의성과 다중공선성에서는 문제가 없었으나 결정계수가 0.76으로 나타났고 앙상블 모델과 서포트 벡터 머신에서는 0.88이상의 예측성능을 보여, 분석한 데이터세트에서 토압식 쉴드TBM 굴진성능예측에 적합한 모델은 서포트 벡터 머신임을 알 수 있었다. 현재 도출된 결과로 볼 때, 토압식 쉴드TBM의 기계데이터와 지반정보가 포함된 데이터를 활용한 굴진성능 예측 모델의 적합성은 높다고 판단된다. 추가적으로 지반조건의 다양성과 데이터양을 늘리는 연구가 필요한 것으로 판단된다.
