Generation of Newly Discovered Resistance Gene mcr-1 Knockout in Escherichia coli Using the CRISPR/Cas9 System

  • The mcr-1 gene is a new "superbug" gene discoverd in China in 2016 that makes bacteria highly resistant to the last-resort class of antibiotics. The mcr-1 gene raised serious concern about its possible global dissemination and spread. Here, we report a potential anti-resistant strategy using the CRISPR/Cas9-mediated approach that can efficiently induce mcr-1 gene knockout in Escherichia coli. Our findings suggested that using the CRISPR/Cas9 system to knock out the resistance gene mcr-1 might be a potential anti-resistant strategy. Bovine myeloid antimicrobial peptide-27 could help deliver plasmid pCas::mcr targeting specific DNA sequences of the mcr-1 gene into microbial populations.

건축공사 안전관리비 비율 산정 모형에 관한 연구 (A Study on the Estimating Rate of Safety Management Cost in Building Work)

  • Standard safety management costs can not be applied to each site with same rate, it is very difficult, because it depends on the experience, work method, work kind, work progress schedule, and hazard level of each construction company. Therefore, this study is to find out hazard level of each work kinds through questionnaire and interview and investigate analyze the status which standard safety management costs have been used. Also, this study is to show reasonable rates of standard safety management costs in construction industry and to set up countermeasures against those problem after reviewing its status in korea with in Japan and Europe. This study is to investigate eleven project kinds of domestic system, first, and to investigate eleven items of apartment bldg, office. Also this study is to investigate and analyze performed costs of presently processing worker finished work so that it shows a new reasonable rate against standard safety management costs in construction industry, in order to make basical data and material to be systemized.

The domestic system of standard safety management costs is not considered in the foreign country, while only related subjective items have been investigated and evaluated for this study. This study is to investigate eleven project kinds of domestic system, first, and to investigate eleven items of apartment bldg, office, civil work such as bridge, tunnel, dam, plant etc, secondly. Additionally, three items of gymnasium, railway, particular steel tower are investigated in this study. Also this study is to investigate and analyze performed costs of presently processing worker finished work so that it shows a new reasonable rate against standard safety management costs in construction industry, in order to make basial data and material to be systemized.

  • Korea gas industrial because of received LNG in 1987 was converted by the Gas Safety Management System in relation to city gas, LPG and LNG. Gas accident were caused by treatment problem on the supplier and user, on technical and use management mater, on facilities and goods, and included problem with gas safety connection system. This study is present standard application plan of each department to use planning gas technical standard and gas utilization standard of introduction model than statute for change of self-regulation management system and complicated regulation of examination center and order.

  • There are many reason for unsafety action without safety operation. The reason is no standard safety operation in company or the workers never get a enough education. Standard safety operation is prepared by paper for accurate directions and orders and operators must read the paper to prevent an error of action. Also an essential particular is safety of equipment and machine must be assumed to establish standard safety operation then safety of operations will be possible.

  • In this paper, we conduct a study on the design that can secure the safety of the EPLA system by performing safety activities based on the ISO 26262 standard for vehicle functional safety. In the case of a company developing a detailed system, it is responsible for verification through hardware design and safety analysis in the overall flow of safety activities, and safety analysis according to the ASIL safety level must be properly performed. At this time, there are cases where the safety goal quantitative metric value suggested by the ISO 26262 standard cannot be satisfied only by the hardware design of the basic function, so it is necessary to design and install the safety mechanism. Based on ISO 26262 safety activities, it is possible to derive an effective design plan through hardware safety analysis.

조립식 가설기자재의 완성품에 대한 안전인증 타당성 연구 (A Feasibility Study of Safety Cetification for Finished Product of Febricated Temporary)

  • Recently, the using of prefabricated temporary equipments is increasing in the industrial field because it can reduce the installation and dismantling period. The various types of prefabricated temporary equipments are manufacturing with various materials in domestic and overseas countries. However, those equipments should win the safety certificate according to the "Governmental Notification about Safety Certification" which regulate the member-based safety certification. It tends to cause over performance of temporary equipments because it couldn't consider structural benefit of the prefabricated products. As the result of this study, it is concluded that the establishment of new safety certification standard for the prefabricated temporary equipments is resonable and the movable scaffold is appropriate for those prefabricated temporary equipment. The movable scaffolds are using as single structure and cannot be expands horizontally. Other types of temporary equipments are using as complex structure which can freely expand horizontally according to the main structure. From the results of the study, the standard of vertical compression performance of prefabricated movable scaffolds should be more than double the performance of single main frame. Other test items of safety certification standard should be fixed when the Notification is amended. The prefabricated temporary equipments which are adopted for the safety standard on assembled structure should be used, moved and kept in assembled state and not separated discretionally. The establishment of new standard for the assembled structure based on this study leads to the development of various types of temporary equipments using structural efficiency of prefabricated structure.

병원간호사의 감염예방 표준주의지침 지식, 인식, 안전환경 및 수행도에 관한 연구 (Knowledge, Perception, Safety Climate, and Compliance with Hospital Infection Standard Precautions among Hospital Nurses)

  • Purpose: The purpose of this study was to examine knowledge, perception, safety climate and compliance with hospital infection standard precautions and to identify the factors influencing nurse's compliance with standard precautions. Methods: Using the structured survey, data were collected from 292 nurses working at 4 general hospitals in the metropolitan city Seoul in October, 2008. Data were entered and analyzed with SPSS 12.0. Results: The majority of nurses were female with a mean age of 27.8. The mean score for knowledge of standard precautions was 21.2 out of 25. The nurses lacked knowledge on reusable gloves or gowns. Nurses perceived use of protective devices may not only increase time strain but also hinder development of therapeutic relationships with patients. Of safety climate factors, lack of time was the most frequently reported barrier to compliance with standard precautions. Knowledge, perception, and safety climate explained 16.1% variance of compliance with standard precautions. Conclusion: To improve nurses' compliance with standard precautions, provision of education and support for safety climate are necessary.

  • 경전철은 중전철에 비하여 건설비가 50% 정도 소요되며 친환경적이어서 최근 국내의 많은 지자체에서 여러 가지 형식의 경전철을 계획하고 있으며 일부 건설이 진행중인 곳도 있다. "도시철도법제22조의 2(도시철도차량의안전기준)"에서는 "구조 및 장치가 안전운행에 필요한 기준(구조 및 장치)에 적합하지 아니하면 운행하지 못한다." 도시철도차량안전기준에관한규칙 제3조에는 도시철도의 건설자 또는 운영자가 안전기준에 맞게 차량을 발주하거나 운영하도록 되어있다. 도시철도차량안전기준에관한규칙은 2000년에 제정되어 대구지하철사고이후 화재안전기준등이 강화되어 2004년에 한번 개정이 되었다. 이는 전적으로 중 대형 전동차를 위주로 작성되었다. 완전무인운전을 기본으로 하여 신호장치 등에 대한 안전기준이 다르고 연절형 대차등을 적용 및 축중등에 대한 안전기준 내용등이 달라 국내 경전철과 관련된 많은 기관 등에서 전반적인 개정요구가 많은 실정이다. 본 논문에서는 현재 국내 경전철 차량의 특징 및 개정의 필요성등을 다루어 향후 경전철 관련 안전기준의 개정 방향을 설정하는데 그 목적이 있다.

  • From the research which it sees it tried to observe from the comfort viewpoint where the passenger feels the high-speed railway car safety standard which it is see clearly from the domestic railroad safety law. It presents a minimum safety standard, the passengers being an safety most, the place where to sleep receive the service which is comfortable from the railroad safety law and they do there is a most big difference. It analyzed the correlation of the comfort where the passenger feels the vehicle efficiency standard which it is presenting from railroad safety law 59th clause and from the research which it sees consequently.
