Chrysanthemum zawadskii (CZ) is a perennial herb belonging to the Asteraceae family. CZ is used medicinally to treat inflammatory and uterine diseases in Asia. CZ was extracted with 50% ethanol and CZ extract (CZE; at 125, 250, and 500 mg/kg body weight) was administered orally every day for 5 or 6 weeks to investigate the anti-diabetic effects in streptozotocin (STZ)-induced rats and STZ + high-fat diet (HFD)-fed mice. CZE significantly decreased fasting blood glucose levels in STZ- and STZ + HFD-induced diabetic models. In addition, glucose tolerance and insulin tolerance were improved in the STZ + HFD + CZE group by increasing insulin levels and decreasing hemoglobin A1c (HbA1c) levels in serum. Furthermore, CZE supplements decreased components of the serum lipid profile such as triglyceride, total cholesterol, and low-density lipoprotein cholesterol levels. These results suggest that CZE may be a potential candidate for controlling hyperglycemia.

Streptozotocin (STZ) is a naturally occurring nitrosoamide used extensively to produce diabetes in experimental animals. Our previous study has demonstrated that i.v. administraton of streptozotocin induces significant red blood cell hemolmysis in rats. Since it has been reported that the highest concentration of STZ is found in the liver, the effect of STZ in freshly isolated rat hepatocytes has been investigated. STZ treatment (10 mM) did not cause significant loss of viability throughout 4 hour incubation, while high dose of STZ (300 mM) to hepatocytes resulted in complete cell death within 3 hours. Addition of 40 mM glucose to incubation medium did not potentiate STZ-induced hepatotoxicity, suggesting that STZ-induced hyperglycemia in vivo did not affect its hepatotoxicity. To investigate the mechanixm of the toxicity, intracellular total glutathione level was determined. Tratment with 10 mM STZ which was not toxic to hepatocytes led to complete depletion of intracellular glutathione level within 1 hour incubation. These results suggest that STZ-induced hepatotoxicity may be independent on the intracellular glutathione depletion.
Objectives : This study aimed to evaluate the hypoglycemic, hepatoprotective, and nephroprotective effects of 'Saengjinyanghyul-tang($Sh\={e}ngj\={i}ny\v{a}ngxu\`{e}-t\={a}ng$ : SJYHT)' in streptozotocin (STZ)-induced diabetic SD rat model. Methods : SJYHT extracts were once a day dosed for 28 days at a dosage 1000, 500, and 250mg/kg/5ml from 25 days after STZ-dosing, and the changes on blood glucose levels, liver and kidney weight, serum AST, ALT, BUN and creatinine levels were observed with histological changes on the liver and kidney. Results : Increased blood glucose levels, and diabetic hepatopathy & nephropathy including increased serum AST, ALT, BUN and creatinine levels, histological changes after STZ-dosing were dose-dependently reduced by SJYHT extract-dosing. Conclusions : 'SJYHT' water extracts have favorable hypoglycemic, hepatoprotective, and nephroprotective effects on the STZ-induced diabetic SD rats. Therefore, it is expected that 'SJYHT' has potential for use in the management of diabetes and diabetic complications.
Previous report has suggested that the albumin levels were reduced in the peripheral blood mononuclear cells (PBMCs) and consequently oxidative stress was elevated in streptozotocin (STZ)-induced diabetic rats as albumin is the predominant antioxidant in plasma. In this study, we suggest that the levels of pro-inflammatory cytokine such as interleukin-6 (IL-6) and tumor necrosis factor-${\alpha}$ ($TNF-{\alpha}$) were increased by approximately 3.4- and 2.9-fold, respectively, in the serum of STZ-diabetic rats, compared to those of normal rat. In addition to the cytokines, the levels of C-reactive protein (CRP) were also about 3.6-fold higher, indicating that STZ induced a pro-inflammatory response in rat blood. However, when purified rat albumin was externally co-administrated with STZ through the tail vein, the serum levels of IL-6, $TNF-{\alpha}$, and CRP were markedly reduced, although the values were still higher than those of normal (non-diabetic) rats. Albumin administration also decreased STZ-induced oxidative stress in serum and PBMCs. Moreover, the decrease in cytokine and CRP levels was dependent on the dose of injected albumin. These results suggest that STZ-induced pro-inflammation and oxidative stress in rat blood might be attenuated by treatment with exogenous albumin.
The present study was designed to investigate the hypoglycaemic and hypolipidaemic activities of Qurs-e-Ziabetus 16 (QZ-16) in Streptozotocin (STZ) induced diabetic rats. QZ-16, a polypharmaceutical herbomineral formulation developed on the principles of Unani medicine is used for non-insulin dependent diabetes mellitus (NIDDM). The elevated levels of fasting blood glucose, serum levels of cholesterol, triglycerides and urea observed in rats treated with STZ (55 mg/kg body wt.) were significantly reduced by the treatment of QZ-16 (240 mg/kg, p.o.) and gliclazide (30 mg/kg, p.o.). The reduced HDL cholesterol levels were also increased by the QZ-16 and gliclazide treatments in the STZ induced diabetic rats. These data show that QZ-16 has hypoglycaemic, hypolipidaemic properties in STZ induced diabetic rats.

The purpose of this study was to investigate the effects of green tea catechin on lipid metabolism in streptozotocin(STZ)-induced diabetic rats. Male Sprague-Dawley rats weighing 150$\pm$10gm were randomly assigned to one normal and three STZ-induced diabetic groups. Diabetic animals were fed catechin free diet(DM-0C group), 0.5% catechin diet(DM-0.5C group) and 1% catechin diet(DM-1C group). Diabetes was experimentally induced by intravenous injection of 55mg/kg body wt of STZ in citrate buffer(pH 4.3) after feeding of three experimental diets for 4 weeks. Animals were sacrificed at the 6th day of diabetic states. Levels of blood glucose were three fold higher in all three STZ-induced diabetic groups than that of the normal group. The levels of plasma insulin were markedly lower in three STZ-induced diabetic groups than that of the normal group. The levels of plasma cortisol were increased in DM-0C group compared with that of the normal group. Triglyceride, total-cholesterol and LDL-cholesterol levels in serum were increased in DM-0C groups compared with the normal group but were not significantly different between catechin diet groups and normal group. Serum HDL-cholesterol levels were reduced in DM-0C and DM-0.5C groups by 38% and 25%, respectively and had similar tendency in the DM-1C group compared with that of control group. Atherogenic index have shown same pattern as the result of total cholesterol. Activities of 3-hydroxy-3-methylglutaryl Co enzyme A(HMG-CoA) reductase were higher in DM-0C groups than those of the normal group but were not significantly different between catechin diet groups and the normal group. It is concluded that dietary catechins can modulate lipid levels of serum and liver HMG-CoA reductase activity in diabetic rats.
This research was performed to investigate the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on blood lipid profiles and tissue differentiation in streptozotocin (STZ)-induced diabetic rats. The animal groups consisted of a normal-control group, a STZ-control group, three STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 combined with exercise; eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for a 10 week experimental period. The MF supplementation group showed a significant reduction in levels of serum total cholesterol and triglyceride compared to the STZ-control group. HDL-cholesterol levels were significantly increased in the MF supplementation group compared to STZ-control group. The ratio of HDL-cholesterol to total cholesterol was significantly higher in MF supplementation group compared to the STZ-control group. The Atherogenic Index (AI) values in the MF supplementation groups were found to be significantly lower than in the STZ-control group. Serum AST and ALT levels were significantly reduced in the MF-supplementation groups compared to the STZ-control group. Total cholesterol level in the liver tissue was significantly decreased in the MF-360 group and in the MF-360 + exercise group compared to the STZ-control group. In immunohistochemical staining of the pancreatic islets of the MF-supplemented groups, a significantly higher number of insulin-immunoreactive cells were observed compared to the STZ-control group. In the MF supplementation groups, Bowman's capsules were clearly observed as hypertrophy of the glomeruli was not obvious. In the MF supplementation groups, a relative reduction in the hypertrophy of the basement membrane of the glomeruli and a significant reduction in the mesangium were observed compared to the STZ-control group. The results of this study suggest that supplementation of MF has beneficial effect in improving plasma lipid and tissue metabolism in streptozotocin-induced rats.
In this study, the effect of Cheonggukjang powder were investigated on the regulation of blood glucose and inflammatory in STZ-induced diabetic rats. The experimental diet used this study were three kinds of Cheonggukjang, which were soybean Cheonggukjang, Yakkong Cheonggukjang and black foods such as black rice, black sesame seeds, and sea tangle added Yakkong Cheonggukjang powder. The experimental animals were divided into 5 groups and fed experimental diets for 7 weeks; non-diabetes with normal diet group (C), diabetes with normal diet (DC), diabetes with soybean Cheonggukjang (DS), diabetes with Yakkong Cheonggukjang (DY), and diabetes with Yakkong black foods added Cheonggukjang (DYB). Blood glucose and insulin resistance of STZ-induced diabetic groups were were significantly higher than C group. But insulin levels and insulin secertory of STZ-induced diabetic groups were significantly lower than C group. However, supplementation of Yakkong or black foods added Yakkong Cheonggukjang were proven to regulation them. In diabetic group, free fatty acid level was significantly increased than C group, but this contents was significantly decreased supplementation of soybean Cheonggukjang. Leptin and adiponectin levels were significantly decreased in STZ-induced diabetic groups.
This research was conducted to study the effects of the supplementation of multi-extracts of mori folium (MF) and of exercise on plasma insulin and glucose levels in streptozotocin (STZ)-induced diabetic rats. Eight male Sprague-Dawley rats, 4 weeks old, were assigned to each experimental group and were raised in the laboratory for 10 weeks. The animal groups consisted of a normal-control group, a STZ-control group, 3 STZ-induced diabetic groups supplemented ad libitum with various amounts of MF extracts (MF-720, MF-360, and MF-180 groups), and a STZ-induced diabetic group supplemented with MF-360 along with exercise. In the normal-control group, glucose tolerance tests resulted in the peak blood glucose level being achieved in 15 minutes and a fasting blood glucose level being achieved in 60 minutes. In the STZ-control group, the peak blood glucose level was reached after 60 minutes and, even after 90 minutes, blood glucose shown at a significantly higher level compared to the fasting levels. In the groups supplemented with MF extracts, the blood glucose level peaked after 30 minutes of glucose challenge, and returned to the fasting level after 90 minutes; the MF-360 and MF-360+exercise groups showed the best levels of glucose tolerance. Blood glucose levels in the STZ-induced diabetic groups were significantly higher compared to the normal-control group. However, after 7 weeks of supplementation with MF extracts, a significant lowering of blood glucose levels was observed in all groups supplemented with the MF extract. The best effect was observed in the group given MF extract combined with exercise. Compared to the normal-control group, blood insulin levels were significantly lower in all STZ-induced diabetic groups; however, a significantly higher level of insulin was observed in the groups given MF extracts compared to the STZ-control group. This study shows that the supplementation of MF extracts in STZ-induced diabetic rats resulted in increased blood insulin levels and lower blood glucose levels.
Objectives : The present study was carried out to investigate the beneficial effect of Coicis Semen extract(CSe) on streptozotocin(STZ)-induced diabetic nephropathy rats. CSe was given to rats with oral administration. Methods : The experimental animals were divided into 3 groups : normal group of rats, control group of STZ-induced diabetic rats, sample group with CSe treatment. Experimental diabetes was induced by the injection of STZ(60 mg/kg) to the rat via the peritoneum. The effects of CSe on STZ-induced diabetic nephropathy were observed by measuring the serum level of creatinine, BUN and uric level of glucose. Kidney level of lipid peroxidation and the activities of reduced glutathione(GSH) were also examined Results : STZ-induced increase of serum creatinine was lowered by CSe treatment, but BUN and uric level of glucose did not show significant changes. CSe oral administration showed statistical decrease of lipid peroxidation in renal cortical tissues, but it has no effect on the activities of GSH. Conclusion : CSe treatment showed protective effect on rat diabetic nephropathy model, but action mechanism of the effect was still not dear. We thought to be concerned with anti-oxidative stress.
