Effects of Stireed Tank Bioreactor Scale-up on Cell Growth and Alkaloids Production in Cell Cultures of Eschscholtzia californica (탱크 교반형 생물반응기의 scale-up이 Eschscholtzia californica 세포생장 및 알칼로이드 생성에 미치는 영향)

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

  • In this paper, to draw out the parameter of synchronization for ADSL(Asymmetric Digital Subscriber Line) chip design, we analyze the performance of STR(Symbol Timing Recovery) and frame synchronization with computer simulation. We analyze and design PLL(Phase Lock Loop) loop for ADSL. As a result, we obtained the optimum parameter of STR to design ADSL chip. Also, when performed frame synchronization with several algorithm, we analyzed the performance of FER(Frame Error Rate) and the effect of frame offset with computer simulation.

  • Clinically isolated Salmonella Enteritidis strain has a multi drug resistance plasmid, which confers ampicillin, chloramphe-nicol, sulfonamide, streptomycin and tetracycline, named pCAST2. We cloned a 7 kb Sacl fragment of pCAST2 which has sulfonamide, streptomycin and tetracycline resistance genes. The 7 kb SacI fragment showed the organization of sulII-strA-strB-tetR-tetA gene cluster which is different from the other clusters reported previously. In this study, we presented the method to detect this cluster by PCR analysis and showed that this cluster was found in Salmonella strains occurred sporadically at Kyungpook province in 2002.

Characteristics of the antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium (Lactobacillus casei subsp. 및 Streptococcus faecium이 생산한 항균성물질의 성상)

  • Antibacterial substances produced by Lactobacillus casei subsp. and Streptococcus faecium were examined for its antibacterial effects against some pathogenic bacteria. They were partially purified with ammonium sulfate precipitation, methanol-acetone extraction, G-50 gel filtration and examined its characteristics. When L casei subsp. and Str faecium were cultivated in MRS broth, stationary phase of L casei is until 24 hours and Str faecium is 20 hours. pH change of the cultured medium was both decreased after 12 hours and then constant at pH 4.5~4.6 after 28 hours. MRS broth culture fluids of L casei subsp. and Str faecium appeared the antibacterial effects by the spot-on-the-lawn method against ETEC, Sal pullorum and Sta aureus. Culture filtrates of L casei subsp. and Str faecium also appeared the antibacterial effects by the disc diffusion method. Culture filtrates of L casei sub. rhamnosus 7469 produced 0.032M of lactic acid and 0.01M of acetic acid. Str faecium 27273 also produced 0.027M of lactic acid and 0.01M of acetic acid. Protein concentrations of culture filtrates produced by L casei sub rhamnosus 7469 and Str faecium 27273 was $495{\mu}g/m{\ell}$ and $594{\mu}g/m{\ell}$, respectively. Antibacterial substances which are partially purified by ammonum sulfate precipitation, methanol-acetone extraction and G-50 gel filtration inhibit the growth of ETEC, Sal pullorum and Sta aureus. Characteristics of purified antibacterial substances was examined. Its molecular weight was about 31Kd, stabilized at $100^{\circ}C/20min.$ and some of proteolytic enzyme treatment.

  • VNTR and STR DNA typing are typical genetic analysis methods which are widely used in DNA-fingerprinting for forensic science and other genetic research purposes. In this study, genomic DNA of different constitutions(Taeun, Soyang and Soum) were analyzed by VNTR and STR DNA typing to provide scientific and objective references for Sasang Medicine. It was found out in this study that VNTR-MCT118 and YNZ22 loci showed too many different variation of allele distribution and numbers for each constitution. Therefore, it is thought that VNTR typing can not used for genetic classification study for Sasang Constitution which classifies human body into 4 groups. However, vWA locus, one of the STR loci investigated in this study, showed slight difference in allele distribution for each different constitution.

Tree-based Group Key Agreement Protocol using Pairing (Pairing을 이용한 트리 기반 그룹키 합의 프로토콜)

  • Secure and reliable group communication is an increasingly active research area prompted by the growing popularity of many types of group-oriented and collaborative applications. The central challenge is secure and efficient group key management. While centralized methods are often appropriate for key distribution in large multicast-style groups, many collaborative group settings require distributed key agreement techniques. Most of prior group key agreement protocols have been focused on reducing the computational costs. One exception is STR protocol that optimizes communicational cost. On the other hand, it requires O(n) number of modular exponentiations. In this paper, we propose a new group key agreement protocol that modifies STR protocol by utilizing pairing based cryptography. The resulting protocol reduces computational cost of STR protocol while preserving the communication cost.

An Optimality-Based Analysis of Relative Positioning of Wh-related Prepositions in English

  • In this paper, we discuss the relative positioning of Wh-related English prepositions in a Wh-interrogative construction within the Optimality Theory [1-2]. By employing the two key constraints such as *Prep-Str and Align which are developed for the positioning of Wh-related prepositions from Romance languages such as French and Italian [3] and for the positioning of Wh-related prepositions from the middle English prose from 1500 to 1900 [4-6], and by slightly modifying the constraint hierarchy of *Prep-STR >>Align into **PrepSTR <<>>Align, Choi argues that his new theory can properly explain the unique behaviors of English Whrelated prepositions being able to take two 'optional' operations such as pied-piping and stranding to find legitimate landing sites in a Wh-interrogative construction [7]. However, this new analysis again reveals the following critical problems: (1) Unlike the 'light' English Wh-related prepositions which can two optional operations for legitimate landing sites in a Wh-interrogative construction, 'heavy' Wh-related English prepositions are not allowed to have such two options: they take just one option of pied-piping only. Thus, (2) his argumentation based on the existing constraints and the modified constraint hierarchy is neither general enough nor proper to explain the issue of the relative positioning for all English Wh-related preposition cases. To include such exceptional syntactic property of the 'heavy' preposition cases within the Optimality Theory, we suggest a new constraint of *HPrep-STR ranked at the highest position of the constraint hierarchy to disallow a 'heavy' or multi-syllabic Wh-related English preposition to stay alone at the end of a sentence. The new final hierarchy of constraints we suggest to explain the exceptional positioning of 'heavy' Wh-related prepositions together with the other 'light' Wh-related prepositions in English Wh-interrogative construction will be as follows: *HPrep-STR>>Align<<>>*Prep-STR.
