A Case of Single-step Mutations at Two Short Tandem Repeat loci (D13S317 and DXS10148) among Three Generations of a Korean Family

  • The DNA profiling of short tandem repeat (STR) markers is a powerful tool for forensic identification and forensic paternity testing. However, STR loci are susceptible to mutation that cause mismatches between parents and children when paternity is tested. Herein, we examined paternity disputes with 23 autosomal STR loci using two commercial human identification kits and revealed successive mismatches at the D13S317 locus across three generations of a Korean family. Additionally, we investigated 12 X-chromosomal STRs and discovered an inconsistency at the DXS10148 locus between the father and daughter of the same Korean family. Furthermore, we confirmed STR genotypes at the D13S317 and DXS10148 loci of the family using sequencing analysis. Consequently, we identified a successive single-step mutation at the D13S317 locus and one single-step mutation at the DXS10148 locus in three generations of the Korean family. Therefore, this case study may be useful for interpreting and understanding forensic paternity tests.

Effects of Ginseng Extract on the Acid Production and Growth of Yoghurt Starter (인삼(人蔘)Extract가 Yoghurt Starter의 산생성(酸生成) 및 증식(增殖)에 미치는 영향(影響))

  • In order to investigate the effects of ginseng extract on the acid production and bacterial growth in the skim milk medium, ginseng extract was added to skim milk medium for 0.3 to 2.4% and the medium was fermented by Str. thermophilus, L. bulgaricus and mixed strain with both bacteria. The acidity and bacterial number were examined by incubation time. The results were summarized as follows: 1. The acidity of medium without ginseng extract after 12 hours incubation by Str. thermophilus was 11.1% and that by L. bulgaricus was 1.01%, whereas that of medium with ginseng extract was 1.08~0.61% for Str. thermophilus and 0.99~0.49% for L. bulgaricus. Therefore, acid production was inhibited by ginseng extract. 2. The number of bacteria in the medium without ginseng extract after 12 hours incubation was $5.2{\times}10^8/m{\ell}$ for Str. thermophilus and $3.2{\times}10^8/m{\ell}$ for L. bulgancus, whereas that in the medium with ginseng extract was $3.6{\times}10^8/m{\ell}{\sim}1.3{\times}10^8/m{\ell}$ for Str. thermophilus and $2.9{\times}10^8/m{\ell}$ and $1.4{\times}10^8/m{\ell}$ for L bulgancus. Therefore, the number of bacteria was decreased by the amount of ginseng extract increased. 3. However, when skim milk medium was fermented by mixed strains, ginseng extract was not affected on the acid production and bacterial number. On the contrary, in some case, the number of bacteria was increased by addition of ginseng extract.

Preparation of Antibiotic-Resistant Bradyrhizobium japonicum and Its Inoculation Effects on Soybean [Glycin max(L.) Merr] (항생물질(抗生物質) 표식(標識) 근류균(根瘤菌)의 조제(造製)와 그 접종효과)

  • This study was carried out to evaluate the fate of inoculant Bradyrhizobium japonicum and the inoculation effect on soybean in complex soil environment. To moniter Rhizobium strains from the root, streptomycine and streptomycine and nalidixic acid resistant marker strains were prepared by spontaneous mutagenesis. The characteristics and properties of antibiotic marked strains were not altered by the mutagenesis. The comparison of properties of wild type and antibiotic resistant Bradyrhizobium strains are summarized as follow : 1) The strains of USDA110K-$STR^r$, USDA110N-$STR^r$ and R318-$STR^r$ showed weak tolerance to pH 9.0. The utilization of carbon sources by fast growing group was different from that of slow growing group. The marked strains of R214-$STR^rNAL^r$, USDA110K-$STR^r$ and USDA110N-$STR^r$ was doubtful in utilization of sorbitol and R138-$STR^rNAL^r$ was doubtful in utilization of xylose as a carbon source. 2) By examining the agglutination reaction of serogroups, the strains used were identified as different ones. There were no differences between wild type and marked strains in agglutination titer values. 3) The plasmid size of fast group was slightly greater than that of slow group. However, there was no differences in plasmid size between the wild type and antibiotic resistant strains. This result indicates that the antibiotic resistance was not encoded in plasmid. 4) The recovery of the inoculated strains was up to 12.5 % in soybean cultivated soil and was up to 25 % in soybean uncultivated soil. 5) When the wild type or marked strains were inoculated. there was no significant effect on soybean plant, whereas the inoculation effect was pronounced in soybean uncultivated soil. The inoculation effect seemed to be more pronounced in wild type strains than antibiotic resistant strains. however, the difference was not significant.

  • Background: Matrix metalloproteinases(MMP) are essential enzymes for tumor invasion and metastasis. Among the MMP family, elevated MMP-9 and stromelysin-3(STR-3) expression have been reported to be poor prognostic factors in lung cancer patients. To evaluate the possibility of a molecular diagnosis of lung cancer using peripheral blood, the mRNA expression level of MMP-9 and STR-3 was measured using a reverse transcriptase-polymerase chain reaction (RT-PCR) in patients with lung cancer. Methods : Ninety six patients(44 patients with lung cancer, 19 pulmonary infection, and 33 control) were included. To detect MMP-9 and STR-3 mRNA expression, RT-PCR was performed in peripheral blood mononuclear cells. ELISA was also used to measure the serum level of MMP-9. Results : MMP-9 was expressed more frequently in patients with a pulmonary infection(18/19, 94.7%) compared to lung cancer patients(26/44, 59.1%) or the controls (23/33, 69.7%) (p=0.018). On the other hand, STR-3 expression was observed more frequently in patients with lung cancer(37/44, 84.1%) compared to the lung infection patients(8/19, 42.1%) or control(20/33, 60.6%) (p=0.003). Among the lung cancer patients, MMP-9 was expressed more frequently when a tumor invaded the lymph nodes(17/24, 70.8%) compared to when a tumor did not(3/13, 23.1%) (p=0.005). The MMP-9 and STR-3 expression levels had no relationship with age, sex, tumor size, distant metastasis, or tumor histology. The serum MMP-9 concentration was not higher in lung cancer patients compared to patients with a pulmonary infection or the control subjects. Conclusion : STR-3 may be used as a diagnostic marker in the peripheral blood of lung cancer patients using RT-PCR. Further studies to evaluate the clinical significance of elevated STR-3 expression in lung cancer patients is recommended.

  • The optimal cell concentration and dilution rate for maximum ethanol productivity were obtained using dynamic simulation in cell recycled continuous bioreactor. The good control performance was observed using rule-based STR (self-tuning regulator) compared to conventional STR. Rule-base contained the scheme to implement the STR in an efficient on-off way and the scheme for the controlled variable to reach the optimal value in a short time. Since a mathematical model was used to analyze and estimate the changes of the state variables and the parameters, it was possible to understand the physical meaning of the system.

  • In human chromosome, a short sequence of DNA has been repeated a number of times. These repeats are called variable number of tandem repeat(VNTR) or short tandem repeat(STR) which has short repeat core. VNTR and STR are used in the field of forensic science, evolution, and anthropology. In this work, we examined allele frequencies of 3 VNTR(YNZ22, NeuR, D21S11) and one STR(Humth01) in a Korean population sample by polymerase chain reaction(PCR) followed by high-resolution polyacrylamide gelelectrophoresis(PAGE) with silver staining. Subsequently, the polymorphism information content(PIC) was calculated : the highest PIC was observed for the NeuR locus(0.95680) and lowest for the Humth01 locus(0.75809).

  • On the basis of pigment production on brain heart infusion agar medium containing crocin, differentiation of mastitic streptococcus was investigated. Intensive crocin reaction was showed in braille heart infusion agar when maximum volume of gardenia powder was 5.0 mg/ml and dark-violet pigmentation of Str. uberis and Group D (Str. facalis) Streptococcus were differentiated from negative strains of Str. dysgalactiae. Also, although most of Str. agalactiae was producted pigment, a few was not affected with the crocin reaction.

Pathogenic effects of porcine reproductive and respiratory syndrome virus isolates in swine tracheal ring culture

  • Pathogenic effects of 29 different porcine reproductive and respiratory syndrome(PRRS) virus isolates were investigated in swine tracheal ring(STR) cultures by examining their effects on the ciliary activity of STR. Inhibition of ciliary movement and destruction of the tracheal epithelium were seen between 72 and 96 hours postinoculation(PI). Virus replication was demonstrated by examining viral infectivity of the supernatants from the STR cultures. PRRS virus antigen in macrophages was detected by a streptavidin-biotin complex(ABC) immunoperoxidase method. Of the 29 PRRS virus isolates, 8 isolates were classified into pathogenic, and the remaining 21 isolates were determined as mildly pathogenic or apathogenic viruses. These results suggest that STR examination may be used as a method for predicting pathogenic variability of PRRS virus isolates.
