Electrochemical Properties of Conducting Polymer for Supercapacitor (Supercapacitor용 도전성 고분자의 전기화학적 특성)

  • The purpose of this study is to research and develop conducting polymer(CP) composite electrode for supercapacitor. Supercapacitor cell of CP composite electrode with 1M LiClO$_4$/ PC bring out good capacitor performance below 4V. The radius of semicircle of CP composite cell with PAn composite electrode adding 30wt% acetylene black was absolutely small. The total resistance of supercapacitor cell mainly depended on internal resistance of the electrode. The discharge capacitance of supercapacitor cell with PAn composite electrode adding 30wt% acetylene black in 1st and 50th cycles was 27F/g and 31F/g at current density of 1mA/$\textrm{cm}^2$. Supercapacitor cell with PAn composite electrode adding 30wt% acetylene black performed a good cycliability.

  • The purpose of this study is to research and develop conducting polymer(CP) composite electrode for supercapacitor. Electrochemical capacitor(supercapacitor) cell of CP composite electrode with 1M LiClO$_4$PC bring out good capacitor performance below 4V. The radius of semicircle of CP composite cell with PAn composite electrode adding l5wt%SP270(PAnS15) and PT composite electrode adding 50wt%SP270 (PTS50) was absolutely small. The total resistance of supercapacitor cell mainly depended on internal resistance of the electrode. The discharge capacitance of supercapacitor cell with PTS50(+)/PAnS15(-) in 1st and 20th cycles was 38F/g and 28F/g at current density of 1mA/cm$^2$. Supercapacitor cell with PTS50(+)/PAnS15(-) showed good capacitance and stability with cycling.

  • Objectives & Methods : The purpose of this study is to observe the effects of Gentianae macrophyllase Radix herbal-acupuncture solution(GR-HAS) at Joksamni(ST36) on Collagen-induced arthritis(CIA) in DBA/1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II , change of immunocyte count, histological change of CIA mouse joint related with RA. Results : 1. In the GR-HA group, the arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In GR-HA group, there were no weight loss and similarly maintained as normal group. Spleen size, adhesion rate and the edema and transformation of knee joint were low and similarly maintained as normal group. 3. In the GR-HA, cytokine level, IgG, IgM and anti-collagen II were significantly decreased. 4. In the GR-HA, the change of immunocyte count were similarly maintained as normal group. 5. In the histological changes of the CIA mouse joint, the cartilage destruction and synovial cell proliferation were decreased in the GR-HA(Hematoxylin and eosin stain). The collagen fiber expressions in the GR-HA were similar with that of the normal group(Masson's Trichrome stain) Conclusions : These results suggest that GR-HA at ST36 has an effect to control immune reaction and suppress inflammation, synovial cell proliferation and cartilage destruction in rheumatoid arthritis.

1H-NMR-Based Metabolic Profiling of Cordyceps militaris to Correlate the Development Process and Anti-Cancer Effect

  • The study of metabolomics in natural products using the diverse analytical instruments including GC-MS, LC-MS, and NMR is useful for the exploration of physiological and biological effects and the investigation of drug discovery and health functional foods. Cordyceps militaris has been very attractive to natural medicine as a traditional Chinese medicine, due to its various bioactive properties including anti-cancer and anti-oxidant effects. In this study, we analyzed the metabolite profile in 50% ethanol extracts of C. militaris fruit bodies from three development periods (growth period, matured period, and aging period) using $^1H-NMR$, and identified 44 metabolites, which are classified as 16 amino acids, 10 organic acids, 5 carbohydrates, 3 nucleotide derivatives, and 10 other compounds. Among the three development periods of the C. militaris fruit body, the aging period showed significantly higher levels of metabolites including cordycepin, mannitol (cordycepic acid), and ${\beta}-glucan$. Interestingly, these bioactive metabolites are positively correlated with antitumor growth effect; the extract of the aging period showed significant inhibition of HepG2 hepatic cancer cell proliferation. These results showed that the aging period during the development of C. militaris fruit bodies was more highly enriched with bioactive metabolites that are associated with cancer cell growth inhibition.

Analysis if Somatic Cell Counts of Raw Milk in Korea -Recommendation to Payment for Milk on the Basis of Quality- (체세포수(Somatic Cell Counts)를 주로한 원유질의 평가 -원유등급제도에 의한 유질향상과 산유량 증가방안-)

  • The somatic cell counts SCC and bacteria counts were done by D milk plant, P milk plant, S milk plant and Inch'${\v{\times}}n$ Vet. Serv. Lab from 1987 to 1990 with Coulter counter, Fossomatic 90, Bactoscan, Rolling ball viscometer and Resazurin reduction test. The results were summarized as follows 1. In the distribution of SCC of the bulk herd milk, D milk plant from Nov. 1989 to Oct. 1990 remarks 80.2% on the range below 500, 000, 14.5% ranging from 1, 000, 000 to 1, 500, 000, 1.2% ranging from 1, 500, 000 to 2, 000, 000, 0.69% ranging from 2, 000, 000 to 3, 000, 000, 0.71% on the range over 3, 000, 000. P milk plant remarks 237, 000 in the first half year and 251, 000 in the second half year in 1990 year. S milk plant remarks annual average of 335, 000 in 1987, 273, 000 in 1988 and 262, 000 in 1989. The individual record of Inch'${\v{\times}}n$ Vet. Serv Lab. remarks 79.35% and 80.2% below 500, 000 8.30% and 7.40% from 500, 000 to 1, 000, 000, 2.37% and 3.2% from 1, 000, 000 to 1, 500, 000, 2.77% and 2.30% from 1, 500, 000 to 2, 000, 000, 1.67% and 2.00% from 2, 000, 000 to 3, 000, 000, 5.53% and 4.40% over 3, 000, 000 in 1989 and 1990, respectively. The grade distirbution of SCC is as follows: D milk plant shows 1st grade-80.20%, 2nd grade-l6.5% and 3rd grade-3.30%. And P milk plant shows all 1st grade. S milk plant shows 87.30%, 8.6% and 4.1% in 1987 and 91.90%, 6.1% and 2.0% in 1988, and 92.40%, 6.1% and l.5% in 1989 on the 1st, 2nd and 3rd grade respectively. 2. The distribution of bacteria P milk plant reached 15.123 in 1st half year and 21.515 in 2nd half year. Also, S milk plant reached 81.5%, 12.5%, 6.0% in 1987, and 86.20%, 9.70%, 4.1% in 1988, and 86.2%, 10.8%, 3.0% in 1989 respectively for 1st, 2nd and 3rd grade. 3. The regional SCC distribution in D milk plant shows 1, 540, 000 in three regions and 714, 000 in one region. And monthly SCC distribution shows 671, 000 in December and 1, 165, 000 in June. 4. As a result of the individual SCC test, 9 times for 16 cows in “I”farm(1986-1988), and 6 times for 13 cows in“D”farm(1987-1988) No.3, 5, 9, 14 cows in“I”farm showed the high SCC beyond 1, 000, 000 over 4-5times. 5. If the SCC over 300, 000 reach 40%, the national producing quality of milk can be reduced by 87, 600M /I annually and in the sum of money, it should be about 35.5 billion Won. 6. The difference between high group and low group for SCC in D milk plant reached over 1, 000, 000. In case that the difference reaches 1, 000, 000 in the farm bulk milk at a farm breeding 20 cows which produce 20kg milk per day, it was estimate that the annual difference of producing quantity and sum of money respectively should be reached 26, 280kg in milk and 10, 643, 400 Won in income.

Changes in the Low Latitude Atmospheric Circulation at the End of the 21st Century Simulated by CMIP5 Models under Global Warming (CMIP5 모델에서 모의되는 지구온난화에 따른 21세기 말 저위도 대기 순환의 변화)

  • Projections of changes in the low latitude atmospheric circulation under global warming are investigated using the results of the CMIP5 ensemble mean. For this purpose, 30-yr periods for the present day (1971~2000) and the end of the $21^{st}$ century (2071~2100) according to the RCP emission scenarios are compared. The wintertime subtropical jet is projected to strengthen on the upper side of the jet due to increase in meridional temperature gradient induced by warming in the tropical upper-troposphere and cooling in the stratosphere except for the RCP2.6. It is also found that a strengthening of the upper side of the wintertime subtropical jet in the RCP2.6 due to tropical upper-tropospheric warmings. Model-based projection shows a weakening of the mean intensity of the Hadley cell, an upward shift of cell, and poleward shift of the Hadley circulation for the winter cell in both hemispheres. A weakening of the Walker circulation, which is one of the most robust atmospheric responses to global warming, is also projected. These results are consistent with findings in the previous studies based on CMIP3 data sets. A weakening of the Walker circulation is accompanied with decrease (increase) in precipitation over the Indo-Pacific warm pool region (the equatorial central and east Pacific). In addition, model simulation shows a decrease in precipitation over subtropical regions where the descending branch of the winter Hadley cell in both hemispheres is strengthened.

Serum Amyloid A as an Independent Prognostic Factor for Renal Cell Carcinoma - A Hospital Based Study from the Western Region of Nepal

  • Objective: The objective of our present study was to assess the role of serum amyloid A (SAA) in stages and prognosis of renal cell carcinoma. Material and Methods: It was a hospital based retrospective study carried out in the Department of Medicine and Biochemistry of Manipal Teaching Hospital, Pokhara, Nepal between $1^{st}$ January 2008 and $31^{st}$ December 2011. The variables collected were SAA, CRP. Approval for the study was obtained from the institutional research ethical committee. Quantitative analysis of human SAA and C-reactive protein (CRP) was performed by radial immune diffusion (RID) assay for all cases. Results: Of the 422 total cases of renal cell carcinoma, 218 patients had normal and 204 abnormal SAA. SAA levels were grossly elevated in T3 stage ($122.3{\pm}SD35.7$) when compared to the mean for the T2 stage ($84.2{\pm}SD24.4$) (p value: 0.0001). Similarly, SAA levels were grossly elevated in M1 stage ($190.0{\pm}SD12.7$) when compared to the M0 stage ($160.9{\pm}SD24.8$) (p: 0.0001). There was no significant association with elevated CRP levels ($209.1{\pm}SD22.7$, normal $199.0{\pm}SD19.5$). Conclusion: The validity of SAA in serum as being of independent prognostic significance in RCC was demonstrated with higher levels in advanced stage disease.

Effects of Amino Acid in In-vitro Maturation Medium on Nuclear Maturation and Embryo Development of Korean Native Cow (체외성숙 배지에 아미노산의 첨가가 한우 난포란의 핵성숙과 배발달에 미치는 영향)

  • The objective of this study was to investigate the effects of amino acid supplementation of oocyte maturation medium on 1st polar body(PB) extrusion, embryo development and blastocsyt cell number. In experiment 1, Cumulus oocyte complexes(COCs) were matured in in vitro maturation(IVM) medium supplemented with 1, 2, or 4-fold of 10 $\mu$l/ml MEM non-essential amino acid(NEAA) and 20 Park, $\mu$ l/ml BME essential amino acid(EAA). The PB extrusion rate of oocytes matured in 1-fold amino acid group was significantly higher than that matured in medium without amino acid (p<0.05), but it was decreased by the increase of the dosage of amino acid. There were no difference in the percentage of embryos reaching 2-cell, 8-cell and blastocyst in all treatments. The number of trophectoderm(TE) cells and total cell number of blastocysts were highest in 2-fold amino acid group, and the number of inner cell mass(ICM) cells was increased by the increase of the dosage of amino acid. In experiment 2, COCs were matured in IVM medium with 1, 5, or 10 mg/ml lactalbumin hydrolysate(LAH). The PB extrusion rate of oocytes matured in medium with 5 mg LAH was significantly higher than that matured in medium with 1 mg LAH (p<0.05). The development rate to the blastocyst stage was significantly higher in non-supplement and 1 mg LAH group than in 5 mg and 10 mg LAH group (p<0.05). The number of TE cells and total cell number did not differ among treatment groups, but the number of ICM cells was increased by the increase of LAH supplement. These results suggested that the supplement of certain group of amino acid in IVM medium effective on the quality of blastocyst, and further studies will be accompany with the search of new sources of amino acid used for the use of in vitro embryo production.

Comparison of Different Thawing Methods on Cryopreserved Aorta (냉동 보존된 대동맥의 해동방법)

  • The studies on cryopreserved arterial allograft have been focused on cooling methods, pre-treatment, cryoprotectant agents, and preservation temperature. But recently, several studies have reported that thawing methods also play an important role in the occurrence of macroscopic and microscopic cracks. This study was designed to investigate the cell injury after thawing, using a rabbit model to clarify the effect of thawing methods on cryopreserved arteries. Material and Method: Segments of the rabbit aorta were obtained and divided into 3 groups (n=60) according to whether the specimens were fresh (control, n=20), cryopreserved and rapidly thawed (RT) at 37$^{\circ}C$ (n=20), or cryopreserved and subjected to controlled, automated slow thawing (ST)(n=20). Cell damage was established using the TUNEL method and the morphological changes were also evaluated. Result: In the group that was rapidly thawed, the expression of TUNEL (+) cells increased significantly more than in the slowly thawed group. In addition, the endothelial denudation, microvesicles and edema were significant in the rapidly thawed group compared with those changes in the slowly thawed group. Conclusion: Our study suggests that the rapid thawing method may be one of the major causes of cellular damage and delayed rupture in cryopresewed arterial allografts. The expression of TUNEL (+) cells and structural changes were significantly low in the slowly thawed group, which might have contributed to the improvement of graft failure after transplantation.

Safety assessments of recombinant DTaP vaccines developed in South Korea

  • Purpose: Pertussis bacteria have many pathogenic and virulent antigens and severe adverse reactions have occurred when using inactivated whole-cell pertussis vaccines. Therefore, inactivated acellular pertussis (aP) vaccines and genetically detoxified recombinant pertussis (rP) vaccines are being developed. The aim of this study was to assess the safety profile of a novel rP vaccine under development in comparison to commercial diphtheria-tetanus-acellular pertussis (DTaP) vaccines. Materials and Methods: The two positive control DTaP vaccines (two- and tri-components aP vaccines) and two experimental recombinant DTaP (rDTaP) vaccine (two- and tri-components aP vaccines adsorbed to either aluminum hydroxide or purified oat beta-glucan) were used. Temperature histamine sensitization test (HIST), indirect Chinese hamster ovary (CHO) cell cluster assay, mouse-weight-gain (MWG) test, leukocytosis promoting (LP) test, and intramuscular inflammatory cytokine assay of the injection site performed for safety assessments. Results: HIST results showed absence of residual pertussis toxin (PTx) in both control and experimental DTaP vaccine groups, whereas in groups immunized with tri-components vaccines, the experimental tri-components rDTaP absorbed to alum showed an ultra-small amount of 0.0066 IU/mL. CHO cell clustering was observed from 4 IU/mL in all groups. LP tests showed that neutrophils and lymphocytes were in the normal range in all groups immunized with the two components vaccine. However, in the tri-components control DTaP vaccine group, as well as two- and tri-components rDTaP with beta-glucan group, a higher monocyte count was observed 3 days after vaccination, although less than 2 times the normal range. In the MWG test, both groups showed changes less than 20% in body temperature and body weight before the after the final immunizations. Inflammatory cytokines within the muscle at the injection site on day 3 after intramuscular injection revealed no significant response in all groups. Conclusion: There were no findings associated with residual PTx, and no significant differences in both local and systemic adverse reactions in the novel rDTaP vaccine compared to existing available DTaP vaccines. The results suggest that the novel rDTaP vaccine is safe.
