Characterization of Mucilage Produced from the Solid-state Fermentation of Soybean Grit by Bacillus firmus

  • Mucilage containing ${\gamma}-polyglutamic$ acid (PGA) was efficiently generated by the solid-state fermentation (SSF) of soybean grit by Bacillus firmus NA-1. B. firmus NA-1 was shown to be a glutamate-dependent strain for PGA production. The SSF of soybean grit was optimized in order to produce mucilage with a fortification of 5% glutamate, resulting in higher levels of mucilage production (6.14%) and a higher consistency index ($1.1\;Pa\;sec^n$). The sticky mucilage was comprised of 38% PGA, 7% levan, and some biopolymers. With regard to the viscoelastic properties of the mucilage solution, the viscous modulus (G") obtained from soybean grit fortified with 5% glutamate was approximately 64 times higher titan that of the mucilage solution obtained without glutamate. Although the addition of glutamate in the SSF of soybean grit influenced the rate of PGA production, the molecular weight of PGA remained unaltered, and was detected in a range between 1,400-1,440 kDa.

Genome Shuffling of Mangrove Endophytic Aspergillus luchuensis MERV10 for Improving the Cholesterol-Lowering Agent Lovastatin under Solid State Fermentation

  • In the screening of marine mangrove derived fungi for lovastatin productivity, endophytic Aspergillus luchuensis MERV10 exhibited the highest lovastatin productivity (9.5 mg/gds) in solid state fermentation (SSF) using rice bran. Aspergillus luchuensis MERV10 was used as the parental strain in which to induce genetic variabilities after application of different mixtures as well as doses of mutagens followed by three successive rounds of genome shuffling. Four potent mutants, UN6, UN28, NE11, and NE23, with lovastatin productivity equal to 2.0-, 2.11-, 1.95-, and 2.11-fold higher than the parental strain, respectively, were applied for three rounds of genome shuffling as the initial mutants. Four hereditarily stable recombinants (F3/3, F3/7, F3/9, and F3/13) were obtained with lovastatin productivity equal to 50.8, 57.0, 49.7, and 51.0 mg/gds, respectively. Recombinant strain F3/7 yielded 57.0 mg/gds of lovastatin, which is 6-fold and 2.85-fold higher, respectively, than the initial parental strain and the highest mutants UN28 and NE23. It was therefore selected for the optimization of lovastatin production through improvement of SSF parameters. Lovastatin productivity was increased 32-fold through strain improvement methods, including mutations and three successive rounds of genome shuffling followed by optimizing SSF factors.

Ethanol Production from Seaweed, Enteromorpha intestinalis, by Separate Hydrolysis and Fermentation (SHF) and Simultaneous Saccharification and Fermentation (SSF) with Saccharomyces cerevisiae

  • Ethanol productions were performed by separate hydrolysis and fermentation (SHF) and simultaneous saccharification and fermentation (SSF) processes using seaweed, Enteromorpha intestinalis (sea lettuce). Pretreatment conditions were optimized by the performing thermal acid hydrolysis and enzymatic hydrolysis for the increase of ethanol yield. The pretreatment by thermal acid hydrolysis was carried out with different sulfuric acid concentrations in the range of 25 mM to 75 mM $H_2SO_4$, pretreatment time from 30 to 90 minutes and solid contents of seaweed powder in the range of 10~16% (w/v). Optimal pretreatment conditions were determined as 75 mM $H_2SO_4$ and 13% (w/v) slurry at $121^{\circ}C$ for 60 min. For the further saccharification, enzymatic hydrolysis was performed by the addition of commercial enzymes, Celluclast 1.5 L and Viscozyme L, after the neutralization. A maximum reducing sugar concentration of 40.4 g/L was obtained with 73% of theoretical yield from total carbohydrate. The ethanol concentration of 8.6 g/L of SHF process and 7.6 g/L of SSF process were obtained by the yeast, Saccharomyces cerevisiae KCTC 1126, with the inoculation cell density of 0.2 g dcw/L.

  • The technology of Semi-Solid Forging (SSF) has been actively developed to fabricate near-net-shape products using light and hardly formable materials. Generally, the SSF process is composed of slug heating, forming, compression -holding and ejecting step. After forming step in SSF, the slug is compressed during a certain holding time in order to be completely filled in the die cavity and be accelerated in solidification rate. The compression holding time that can affect microstructural characteristics and shape of products is important to make decision, where it is necessary to find overall heat transfer coefficient properly which has large effect on heat transfer between slug and die. This paper presents the procedure to predict compression holding time of obtaining the final shaped part with information of temperature and solid fraction for a cylindrical slug at compression holding step in closed-die compression process using heat transfer analysis considering latent heat by means of finite element method. The influence of the predicted compression holding time on microstructural characteristics of products is finally investigated by experiment.

섬유질계 동시당화발효를 위한 내열성 융합 효모, Kluyveromyces marxianus CHY1612의 개발 (Development of Thermostable Fusant, CHY1612 for Lignocellulosic Simultaneous Saccharification and Fermentation)

  • To develop thermostable ethanol fermentative yeast strain for lignocellulosic simultaneous saccharification and fermentation, high ethanol producing yeast, Saccharomyces cerevisiae CHY1012 and thermostable yeast, Kluyveromyces marxianus CHY1703 were fused by protoplast fusion. The thermostable fusant, CHY1612 was identified as a Kluyveromyces marxianus by phenotypic and physiological characteristics, as well as molecular analysis based on the D1/D2 domains of the large subunit (26S) rDNA gene and the internally transcribed spacer (ITS) 1 + 2 regions. For lignocellulosic ethanol production, AFEX pretreated barley straw at $150^{\circ}C$ for 90 min was used in a simultaneous saccharification and fermentation (SSF) process using thermotolerant CHY1612. The SSF from 16% pretreated barley straw at $43^{\circ}C$ gave a saccharification ratio of 90.5%, a final ethanol concentration of 38.5 g/L, and a theoretical yield of 91.2%. These results show that K. marxianus CHY1612 has potential for lignocellulosic ethanol production through simultaneous saccharification and fermentation with further development of process.

캇사바전분의 무증자당화에 의한 에타놀발효에 관한 연구(I I) (Ethanol Fermentation of Raw Cassava Starch (II))

  • 증자하지 않은 cassava 생전분의 ethanol 발효생산을 위한 동시당화 -발효의 최적조건을 검토하였다. 생전분의 분해력이 가장 좋은 당화효소의 선별에서는 Asp. shirousami27이 가장 좋았으며 발효효모는 균주에 따라 큰 차이가 없는 것으로 나타났다. 동시당화-발효는 pH3.6에서 가장 높았으며 발효전 6$0^{\circ}C$에서 당화효소에 의한 전당화와 발효시 교반등이 발효속도 및 발효율에 효과가 있었고, 분쇄처리도 효과가 있었다. cassava 생전분의 동시당화-발효에서 5일간의 긴 발효 기간을 요구하는데 발효중 발효액의 성분 조사 결과 중간물인 glucoserk 축적되지 않는 것으로 당화과정이 속도결정단계임을 알 수 있다.

Optimization of Solid State Fermentation of Mustard (Brassica campestris) Straw for Production of Animal Feed by White Rot Fungi (Ganoderma lucidum)

  • The objective of the experiment was to determine the optimum cultural [moisture levels (55, 60 and 70%), days of fermentation (7, 14 and 21), temperature (25 and $35^{\circ}C$) of incubation)] and nutritional parameters (urea addition (0 and 2%) and variable levels of single super phosphate (0.25 and 0.50% SSP)) for bio-processing of the mustard (Brassica campestris) straw (MS) under solid-state fermentation (SSF) system. The performance of SSF was assessed in terms of favorable changes in cell wall constituents, protein content and in vitro DM digestibility of the MS. Sorghum based inoculum (seed culture) of Ganoderma lucidum to treat the MS was prepared. The 50 g DM of MS taken in autoclavable polypropylene bags was mixed with a pre-calculated amount of water and the particular nutrient in the straw to attained the desired levels of water and nutrient concentration in the substrate. A significant progressive increase in biodegradation of DM (p<0.001), NDF (p<0.01) and ADF (p<0.05) was observed with increasing levels of moisture. Among the cell wall constituents the loss of ADF fraction was greatest compared to that of NDF. The loss of DM increased progressively as the fermentation proceeded and maximum DM losses occurred at 28 days after incubation. The protein content of the treated MS samples increased linearly up to the day $21^{th}$ of the incubation and thereafter declined at day $28^{th}$, whereas the improvement in in vitro DM digestibility were apparent only up to the day $14^{th}$ of the incubation under SSF and there after it declined. The acid detergent lignin (ADL) degradation was slower during the first 7 days of SSF and thereafter increased progressively and maximum ADL losses were observed at the day $28^{th}$ of the SSF. The biodegradation of DM and ADL was not affected by the variation in incubation temperature. Addition of urea was found to have inhibitory effect on fungal growth. The effect of both the levels (0.25 and 0.50) of SSP addition in the substrate, on DM, NDF, ADF, cellulose and ADL biodegradation was similar. Similarly, the protein content and the in vitro DM digestibility remain unaffected affected due to variable levels of the SSP inclusion in the substrate. From the results it may be concluded that the incubation of MS with 60 percent moisture for 21 days at $35^{\circ}C$ with 0.25 percent SSP was most suitable for MS treatment with Ganoderma lucidum. Maximum delignification, enrichment in the protein content and improvement in in vitro DM digestibility were achieved by adopting this protocol of bioprocessing of MS.

  • Trichoderma sp. KI 7-2와 내열성 효모를 이용하여 볏짚에서 ethanol을 생산하기 위한 동시당화-발효의 최적조건을 검토하였다. 발효생산시 고체배양에서의 배지는 밀기울과 볏짚을 3 : 2 로 혼합하고 pH를 4.5로 조절한 수분함량 50%의 고체배지에서 7일간 배양한 koji를 사용했을 때 가장 높은 ethanol이 생산되었다. 기질의 전처리는 ethanol 생산성에 영향을 미치지 않는 것으로 나타났다. 볏짚의 동시당화-발효를 위해서는 효소량은 볏짚 1g당 2.47units가 요구되며, 발효액중 초기 효모 농도가 2.5$\times$$10^{7}$ cell/$m\ell$이면 충분하였다. 동시당화-발효 과정을 위한 최적 pH와 온도는 각각 4.5와 4$0^{\circ}C$였다. 또한 동시당화-발효액에 다시 기질 또는 효소와 기질을 첨가하여 최종 ethanol 농도를 높일 수 있었다.

에탄올 향상을 위한 탈아세틸화 백합나무 당화액의 발효저해물질 제거와 semi-동시당화발효 (Improved Ethanol Production from Deacetylated Yellow Poplar (Liriodendron tulipifera) by Detoxification of Hydrolysate and Semi-SSF)

  • 본 연구에서는 백합나무의 아세틸기 제거를 위해 전처리 전에 수산화나트륨을 이용하여 탈아세틸화를 수행하였다. 0.8%의 수산화나트륨을 첨가하여 $60^{\circ}C$에서 80분 동안 반응시켜 헤미셀룰로오스로부터 대부분의 아세틸기를 제거하였다. 탈아세틸화 처리된 바이오매스를 옥살산 전처리에 이용하였으며, 전처리된 바이오매스 투입량(10, 12.5, 15%) 및 효모 투입시간(0, 6, 12, 24시간)에 따라 동시당화발효 및 semi-동시당화발효를 수행하였다. 최대 에탄올 수율은 바이오매스 투입량 10%에서 효모를 당화시작과 동시에 첨가했을 때 120시간 후 26.73 g/L의 에탄올을 생산하였으며 이것은 88.14%의 에탄올 수율에 해당하였다. 바이오매스 투입량 12.5%와 15% 조건에서는 효모 투입시간 6시간 조건에서 각각 32.34 g/L, 27.15 g/L의 에탄올을 생산하였고, 이는 각각 85.58%와 59.87%의 에탄올 수율에 해당하였다. 옥살산 전처리 후 얻어진 액상 가수분해산물로부터 발효저해물질의 제거를 위해 수산화칼슘을 처리하였으며 발효 72시간 후 5.28 g/L의 최대 에탄올을 얻었다.

SSFNet 기반 사이버 공격 및 탐지를 위한 네트워크 시뮬레이터의 구현 (Implementation of a Network Simulator for Cyber Attacks and Detections based on SSFNet)

  • 가상 공격을 수행하고 이에 따른 네트워크의 행동 변화를 시뮬레이션하기 위하여는 네트워크 구성요소들의 특성을 시뮬레이션 모델에 반영할 수 있어야 하며, 다양한 사이버 공격과 이를 방어하는 시스템들의 특성을 표현할 수 있어야 한다. 본 연구에서는 사이버 공격시 네트워크의 부하가 어떻게 변하는지를 실험하기 위하여, 프로세스 기반 사건 중심 시뮬레이션 시스템인 SSF[9, 10]를 확장 구현하였다. 사이버 공격을 시뮬레이션하기 위해 보안 관련 클래스인 방화벽과 공격용 프로그램을 작성하기 위한 도구들의 모임인 패킷 조작기를 SSF의 구성요소인 SSFNet에 새로이 추가하였다. 이는 보안 체계를 가진 네트워크를 시뮬레이션 가능하게 할 뿐 아니라, 기존 사이버 공격 프로그램을 쉽게 이식하여 시뮬레이션에 적용할 수 있는 장점을 제공한다. 추가된 클래스들의 작동을 검증하기 위하여 가상 네트워크를 구성한 후, 대표적인 서비스-거부 공격인 smurf 공격을 시뮬레이션하고, 이 때의 네트워크의 행동 변화를 관찰하였다. 실험 결과 본 연구에 의하여 개발된 방화벽이나 패킷 조작기가 정상적으로 작동됨을 확인할 수 있었다.
