Relation between Rainfall and Phytoplankton Community in Daechung Reservoir (대청호에서 강우와 식물플랑크톤 군집의 관계)

  • The phytoplankton community, environmental factors, and rainfall were investigated from July to October in 2001 and 2003 on Dam site in Daechung Reservoir. The monthly average rainfall in the investigated period were 91.3 and 265.3 mm in 2001 and 2003, respectively. The maximum chlorophyll a concentration was observed higher at 131.5 ㎍ L/sup -1/ in 2003 than at 45.4 ㎍ L/sup -1/ in 2001. The cyanobacterial number in 2001 was counted up to over 200,000 cells mL/sup -1/, which was much higher than the maximum number of 49,000 cells mL/sup -1/ in 2003. The relative abundance of cyanobacteria in the phytoplankton community was about 97% in 2001 and 74% in 2003. Microcystis spp. were absolutely dominant species in 2001, while a couple of cyanobacteria such as Oscillatoria spp., Phormidium spp. Chroococcus spp. and Microcystis spp. were dominant species in 2003. Consequently, it seemed that rainfall affected the diversity of phytoplankton species and decreased the density of bloom-forming cyanobacteria.

Root and Basal Stem Rot of Moth Orchid (Phalaenopsis spp.), Pung-nan (Neofinetia falcata) and Nadopung-nan (Aerides japonicum) Caused by Fusarium spp. (Fusarium spp.에 의한 호접란과 풍란류에 발생하는 뿌리 및 줄기기부썩음병)

  • Root and basal stem rot disease occurred on moth orchid (Phalaenopsis spp.), Pung-nan (Neofinetia falcata) and Nadopung-nan (Aerides japonicum) grown in the farmers' greenhouses located in Namyangju Kyonggi province, Korea during 2005 to 2006. Wilting symptoms occurred on these orchard plants at initial stage and the infected plant leaves turned yellow to red. The discolored leaves were fallen down to lead to eventual death of the entire plant. A total of 59 isolates of Fusarium spp. was obtained from roots and leaf bases of the diseased plants. The cultural and morphological characteristics of isolated Fusairum spp. were identified as Fusarium oxysporum, F. proliferatum and F. solani. F. oxysporum and F. proliferatum were isolated from all these orchard plants but F. solani was isolated only from Phalaenopsis spp. Pathogenicity of the three Fusarium spp. was confirmed by artificial inoculation. Although F. oxysporum, F. proliferatum and F. solani cusing root rot disease in Phalaenopsis spp. have been reported in Korea, the pathogens in N. falcata and A. japonicum were not reported yet. Therefore, this is the first report on the root and stem rot of N. falcata and A. japonicum caused by F. oxysporum and F. proliferatum in Korea.

A Study on the Effect of Lactobacillus spp. on the Growth and Citrinin production by Penicillium citrinum (Penicillium citrinum 생육과 citrinin 생성에 미치는 젖산균의 영향에 관한 연구)

  • ABSTRACT - This study was performed to investigate the possible effect of Lactobacillus spp. on the growth and citrinin production by Penicillium citrinum. Lactobacillus bulgaricus and Lactobacillus casei were grown with Pen. citrinum in modified APT broth containing 7% of glucose and incubated at $30^{\circ}C$ for 15 days. Four inoculation procedures were used; (a) Lactobacillus spp. and Pen. citrinum were grown alone(Pc, Lb, and Lc), (b) both organisms were added simultaneously(ST; Pc+Lb and Pc+Lc), (c) Lactobacillus spp. was grown 3 days, then conidia of Pen. citrinum were added(LbPc and LcPc), and (d) Pen. citrinum was grown 3 days, then Lactbacillus spp. was added (PcLb and PcLc). At 0, 3, 6, 9, 12, 15 days of incubation, the growth of each organism, pH and total acidity of broth, and content of citrinin were determined. Lactobacillus spp. and Pen. citrinum, when grown associatively, influenced the growth of each other. It was observed that slower growth of Pen. citrinum when in the presence of Lactobacillus spp. than when the mold grew alone. Production of citrinin by Pen. citrinum was markedly less in the mixed culture. No apparent growth and toxin production was observed when the Lactobacillus spp. was grown 3 days, then conidia of Pen. citrinum were added(LbPc and LcPc). The above results indicate that another microorganism or competing microflora in the culture can affect the behavior of Pen. citrinum.trinum.

Antibiotic Resistance Patterns of Enterococcus spp. Isolated From Commercial Frozen Foods (시판 냉동식품에서 분리한 장구균의 항생제 내성 양상)

  • From April to December in 2009, microbial investigation is accomplished for 100 frozen foods asked to microbial control team that corresponds with total aerobic viable bacteria, coliform group, Escherichia coli, Enterococcus spp. and antibiotic resistance patterns of Enterococcus spp. isolates are investigated. Average of total erobic viable bacteria numbers is $4.3{\times}10^4CFU/g$. Average of coliform group numbers is $4.3{\times}10^3CFU/g$. Average f Enterococcus spp. numbers is $1.8{\times}10^3CFU/g$. Escherichia coli from 100 frozen foods is not detected and detection ate is 0.0%. 22 Enterococcus spp. are isolated from 100 frozen foods. 12 of 22 Enterococcus spp. strains are identified as E. faecium. 7 of 22 Enterococcus spp. strains are identified as E. faecalis. 2 of 22 Enterococcus spp. trains are identified as E. gallinarum. 1 of 22 Enterococcus spp. strains is identified as E. hirae. Enterococcus spp. solates show a high resistance to erythromycin, rifampin, tetracycline, ciprofloxacin, chlorampenicol, penicillin and susceptibility to vancomycin, ampicillin, gentamicin, strepomycin, linezolid. 15 of 22 Enterococcus spp. strains are multi-resistant and the most frequent multi-resistant pattern is erythromycin-rifampin for 6 Enterococcus spp. strains.

Study Regarding Species Identification for Hanging Paintings and Hanging Painting Storage Boxes in Joseon Dynasty (조선시대 괘불탱 및 괘불궤 수종에 대한 연구)

  • Hanging paintings are used for special ceremonies in Buddhist temple. They are stored in special storage box called "Gwaebulgwae" which means storage box for a hanging painting. Studies regarding pigments, background fibers and textiles of hanging paintings are carried out with many data in the field of conservation science. However, there are only few studies for the axes of hanging paintings and storage boxes. In this study, species identification for wooden axes of hanging paintings and storage boxes from the Joseon dynasty are carried out. Hanging painting of Beopjusa temple, Magoksa temple, Sudeoksa temple, Gaeamsa temple, Sudosa temple, Bukjangsa temple, Dorimsa temple, Tongdosa temple and Daeryeonsa temple are selected. Sampling is carried out from the axes of hanging paintings and storage boxes. 81 samples are collected from hanging paintings and storage boxes. As a result, 51 hard pines of Pinus spp., 8 Tilia spp., 6 Juniperus spp., 5 soft pines of Pinus spp., 4 Tsuga spp., 3 Pseudotsuga spp., 2 Berchemia spp., 1 Pyrus spp., 1 Bambusoideae are identified. Tsuga spp. and Pseudotsuga spp. might be used in conservation and restoration works in modern times because those species do not naturally grow in Korea. Most hanging paintings and storage boxes are made of hard pine. Also, some of are made of Tilia spp., because it represents the symbol of Buddha. Some unique kinds of trees used for wooden sculpture are identified, such as Juniperus spp. and Berchemia spp. Authentic conservation and restoration works could be carried out with this data-base.

Isolation and Degradation Activity of a TBTCl (Tributyltin Chloride) Resistant Bacteriain Gwangyang Bay (광양만에서 TBTCl (Tributyltin Chloride) 내성세균의 분리 및 분해활성)

  • BACKGROUND: Tributyltin chloride is among the most toxic compounds known for aquatic ecosystems. Microorganisms are responsible for removal of TBTCl. Nevertheless, only a limited number of marine bacteria were investigated for biodegradation of TBTCl in Korea. METHODS AND RESULTS: The number of TBTCl resistant bacteria ranged from $2.5{\times}10^3$ to $3.8{\times}10^3$ cfu/mL in the seawater, and ranged from $3.2{\times}10^5$ to $9.1{\times}10^5$ cfu/g in the surface sediment, respectively. The morphological, physiological, and biochemical characteristics of TBTCl resistant bacteria were investigated by API 20NE and other tests. The most abundant species of TBTCl resistant bacteria were Vibrio spp. (19.2%), Bacillus spp. (16.2%), Aeromonas spp. (15.2%), and Pseudomonas spp. (13.1%), etc. Eleven TBTCl resistant isolates also had a resistance to heavy metals (Cd, Cu, Hg, and Zn). Among them, isolate T7 showing the strong TBTCl-resistance was selected. This isolate was identified as the genus Pantoea by 16S rRNA gene sequencing and designated as Pantoea sp. T7. In addition, this bacterium was cultivated up to the growth of 50.7% after 60 hrs at TBTCl concentration of $500{\mu}M$. TBTCl-degrading activity of Pantoea sp. T7 was measured by GC-FPD analysis. As a result of biological TBTCl-degradation at TBTCl concentration of $100{\mu}M$, TBTCl-removal efficiency of Pantoeasp. T7 was 62.7% after 40 hrs. CONCLUSION(S): These results suggest that Pantoea sp. T7 is potentially useful for the bioremediation of TBT contamination.

Serovars and Genetic Characteristic of Salmonella spp. Isolates from Jeju Island, South Korea (제주도에서 분리된 살모넬라균의 혈청형 및 유전학적 특성)

  • Salmonella spp. is among the most important water-borne and food-borne pathogens and is one of the most common causes of human gastroenteritis and diarrheal diseases globally. In this study, Salmonella spp. isolated from food, environmental samples, and patients with food poisoning or diarrhea were investigated Salmonella serovars, antibiotic resistance using Vitek2, and genetic characteristics through pulsed-field gel electrophoresis (PFGE). Salmonella spp. of 339 strains, including 26 strains from food or environmental samples and 313 strains from patients, were isolated from Jeju Island of South Korea between 2020 and 2023. The monthly number of isolated Salmonella spp. gradually increased from March, with the highest number being in August. No significant differences in Salmonella spp. isolated from patients according to gender was observed. However, Salmonella spp. was most frequently isolated from people aged 70 years or older and least frequently isolated from those between ages 10 and 19 years. Salmonella spp. isolated from food or environmental samples were distributed among eight different serovars and the main serovars were identified in the order of S. Bareilly (26.9%), S. Rissen (23.1%), and S. Thompson (19.3%). Salmonella spp. isolated from patients were distributed among 27 different serovars and the main serovars were identified in the order of S. Bareilly (31.0%), S. Typhimurium (24.6%), and S. Enteritidis (11.5%). The main cause serovars of Salmonella spp. outbreaks are S. Bareilly, S. Enteritidis, S. Thompson. Antibiotic resistance tests indicated resistance to various antibiotics and some Salmonella spp. exhibited multidrug resistance. Salmonella spp. showed various genetic correlations among the 17 serovars. These results indicate that they can be used as basic data for epidemiological investigations by predicting the appearance of Salmonella spp. and providing a scientific basis.

Contamination of Listeria spp. in Market Beef (국내 시판 쇠고기의 Listeria spp. 오염)

  • Highly lethal Listeria monocytogenes, causing bromatoxism through vegetables, dairy products, meat products and shellfish etc, was examined for possible contamination in market beef. USDA, FDA, Malthus and Modified Cold Enrichment methods were used for the detection of Listeria spp.. Samples of domestic and imported market beef were collected from local meat shopsat Seoul, Korea. Total two hundreds and six of Listeria spp. were isolated and identified from beef. Among 206 isolates, the number of L. welshimeri was one hundred and twenty-one(44.8%). The numbers of isolated L. innocua, L. murrayi, L. monocytogenes, L. grayi, L. seeligeri, and L. ivanovii were 49(18.1%), 14(5.2%), 12(4.4%), 6(2.2%), 2(0.7%), and 2(0.7%), respectively. Detection rates of Listeria spp. varied among four methods. The highest detection rate of Listeria spp. in market beef was found at USDA method and that of L. monocytogenes was found at Malthus method.

False Positive SOD Activity of Bifidobacterium spp. Grown in MRS Medium

  • The superoxide dismutase (SOD) activity of seven Bifidobacterium spp. strains was examined by an indirect SOD assay method. Some Bifidobacterium spp. showed significant levels of SOD activity. However, we could not observe any significant differences between anaerobic and aerobic cultures. Furthermore, although several Bifidobacterium spp. exhibited some degree of tolerance to paraquat which produces superoxide radicals, the apparent SOD activity of these strains was not correlated with their resistance to paraquat. In addition, when we added increasing amounts of manganese or iron to MRS medium which had been prepared without either of the metal ions, the apparent SOD activity of cell free extracts (CFEs) was increased with increasing concentration of both metal ions. To our surprise, the heat-denatured CFEs also showed nearly identical correlative patterns. Based on these results, the apparent SOD activity was likely due to a nonenzymatic dismutation. These results strongly suggest that high concentration of divalent metal ions ($Mn^{2+}$, $Fe^{2+}$) in MRS medium result in nonenzymatic dismutation which can lead to false positive SOD activities in Bifidobacerium spp.

Endophytic fungi harbored in Chinese native gramineous plants

  • $Epichlo\ddot{e}$ endophytes, including Neotyphodium spp. and $Epichlo\ddot{e}$ spp., enhance plant growth, mediate more plant tolerance or resistance to biotic and abiotic stresses, and also synthesis various biologically active compounds in their host plants, and important in many areas. In early stages, most of $epichlo\ddot{e}$ endophytes were described during surveys practiced by American, European and Oceania scientists, while fungal endophytes within native Asian plants were poorly investigated. In recent years, an $Epichlo\ddot{e}$ sp. and 4 Neotyphodium spp. were described in cool season Chinese native gramineous plants. Most of Chinese native Neotyphodium spp. were presumed as hybrids originated from members of ETC and EBY. Investigation on NRPS genes shows lack of toxic ergopeptines and potential production of peramine. Biological and ecological roles of Chinese native $epichlo\ddot{e}$ endophytes should be investigated in future, and it will be very valuable if we can have some joint projects with Korean scientists for Asian native $epichlo\ddot{e}$ endophytes.
