Preparation of Edible Films from Soybean Meal (대두박을 이용한 가식성 필름의 제조에 관한 연구)

  • Effect of extraction pH on mechanical properties such as tensile strength (TS) and elongation (E) and on water vapor permeability (WVP) of soybean protein isolate (SPI) edible films extracted from soybean meal was investigated. Five pHs, acidic range (pH 2.0 and pH 3.0), neutral range (pH 7.0) and alkalic range (pH 10.0 and 12.0), were used to extract SPI. TS of the film extracted at pH 7.0 was the lowest, and WVP of $SPI_3$ (SPI extracted at pH 3) film was the lowest value among the films. The WVP of $SPI_3$ films was $3.349\;{\times}\;10^{-10}\;g{\cdot}m/m^2{\cdot}s{\cdot}Pa$ and increased to $3.871\;{\times}\;10^{-10}\;g{\cdot}m/m^2{\cdot}s{\cdot}Pa$ as film thickness increased from $55\;{\mu}m$ to $72\;{\mu}m$ thickness. Three different plasticizers (glycerol, polyethylene glycol and propylene glycol) were used for $SPI_2$ (SPI extracted at pH 2) film. TS of $SPI_2$ films was 12.297 MPa and decreased to 1.356 MPa for glycerol and showed the same trend in other two plasticizers. The SPI films extracted at acidic range were shown higher mechanical properties and lower water vapor permeabilities than those of extracted at neutral and alkalic ranges. The difference of SPI film properties seemed to be attributed by 11S/7S ratio as well as protein content.

Hfq and ArcA Are Involved in the Stationary Phase-Dependent Activation of Salmonella Pathogenicity Island 1 (SPI1) Under Shaking Culture Conditions

  • In Salmonella enterica serovar Typhimurium, many genes encoded within Salmonella pathogenicity island 1 (SPI1) are required to induce intestinal/diarrheal disease. In this study, we compared the expression of four SPI1 genes (hilA, invF, prgH, and sipC) under shaking and standing culture conditions and found that the expression of these genes was highest during the transition from the exponential to stationary phase under shaking conditions. To identify regulators associated with the stationary phase-dependent activation of SPI1, the effects of selected regulatory genes, including relA/spoT (ppGpp), luxS, ihfB, hfq, and arcA, on the expression of hilA and invF were compared under shaking conditions. Mutations in the hfq and arcA genes caused a reduction in hilA and invF expression (more than 2-fold) in the early stationary phase only, whereas the lack of ppGpp and IHF decreased hilA and invF gene expression during the entire stationary phase. We also found that hfq and arcA mutations caused a reduction of hilD expression upon entry into the stationary phase under shaking culture conditions. Taken together, these results suggest that Hfq and ArcA regulate the hilD promoter, causing an accumulation of HilD, which can trigger a stationary phase-dependent activation of SPI1 genes under shaking culture conditions.

Evaluation of Short-Term Drought Using Daily Standardized Precipitation Index and ROC Analysis (일 단위 SPI와 ROC 분석을 이용한 단기가뭄의 평가)

  • The Standardized Precipitation Index (SPI) is widely applied to evaluate for meteorological droughts. However, the SPI is limited to capture a drought event with a short duration, expecially shorter than one month. In this study, we proposed a daily SPI (DSPI) as a way to overcome the limitation of the monthly SPI for drought monitoring. In order to objectively assess the ability of the drought reproduction of the DSPI, we performed a receiver operating characteristic (ROC) analysis using the quantified drought records from official reports, newspapers, etc. The results of ROC analysis showed that the DSPI has an ability to reproduce short-term drought compared with other indices. It also showed that the main cause of historical droughts was the shortage of rainfall accumulated during the time period less than 90 days compared with the rainfall of normal years.

Quality Characteristics of SPI and Na-Caseinate Substituted Sausage for Meat Protein (분리대두단백 및 카세인 대체 소시지의 품질 특성)

  • Meat emulsions containing 0, 15, 30 and 45% of soy protein isolate(SPI), Na-caseinate(Na-CN) and their mixtures were prepared in order to determine the effect of these non-meat proteins on the physical properties and their sensory quality in emulsion type sausage. It was found that SPI was better fat stabilizer and better binder than Na-CN. The mixtures of SPI and Na-CN didn't exert any significant effect on emulsion stability. From the texture profile analysis by using Instron two-cycle compression tests, decrease in the substitution levels and increase in the ratio of SPI/Na-CN resulted in a significant increase in the textural values of hardness, adhesiveness, gumminess, chewiness. The finished products showed that the substituted product for 15 % meat protein had higher textural values than the unsubstituted product. The sensory quality evaluated for the final products showed no significant difference between the SPI substituted product for 15 % meat protein and the unsubstituted product. However, all of the substituted products for 15 % meat protein and some of those for 30 % substitution with SPI and 67 % SPI received higher scores than average.

  • 본 연구에서는 한강유역에 발생한 가뭄 분석을 위해 표준강수지수를 활용하였으며, 가뭄분석을 위해 한강유역에 실제로 발생한 가뭄사상 중에 한강유역에 조금 더 많은 피해를 미친 것으로 조사된 2000 - 2001년에 대해 가뭄과 수량 스트레스 관계를 살펴보았다. 수량 스트레스값은 강수에만 영향을 받고 있는 값보다는 실제 가뭄 피해의 영향정도를 살펴보기 위해 수량 스트레스의 결과값과 상호 비교하였다. 이는 실제 발생한 기상학적 가뭄으로 인한 피해가 수량 스트레스 변화에 어떠한 영향을 미치는지를 상호 비교함으로써 보다 정확한 가뭄 피해 발생 시기를 확인하고자 함이다. 표준강수지수 SPI3, SPI6, SPI12을 분석하기 위해 2000년 1월 - 2001년 12월까지의 데이터를 구축하였다. 이와 함께 수량 스트레스의 경우 월별 최대 NDI 값을 추출하여 월단위 자료를 구축하였다. 표준강수지수와 NDI 값을 상호 비교한 결과 2000 - 2001년에 발생한 가뭄 사상의 경우, 실제 피해가 발생한 시점은 2001년 3월 - 5월에 발생한 동아시아 지역의 가뭄의 영향이 큰 것으로 나타났다. 한강유역의 대부분에서 수량 스트레스가 발생하는 시기와 SPI3로 인한 가뭄 발생시기가 유사한 패턴으로 나타났다. SPI6와 비교분석해본 결과 기존 SPI3에서 나타낸 2001년 5월에 발생된 가뭄 발생은 실제 수량 스트레스가 발생하는 2001년 5월 이후와 유사한 패턴을 보이는 것으로 나타났지만, 실제 수량 스트레스 발생 시기와는 상관성이 떨어지는 것을 확인할 수 있었다. SPI12에서는 대부분의 유역에서 2000년에는 가뭄이 발생하지 않은 것으로 분석되었다. 이는 한강유역에 발생한 가뭄 피해를 생각해보면, 2001년에 발생한 가뭄 자체가 단기적인 가뭄의 영향이 아닌 장기적인 가뭄의 영향을 띄고 있는 것으로 해석 할 수 있으며, 2000년대 초반 극심한 가뭄 피해를 입힌 2001년 가뭄은 지속적인 가뭄 동반한 장기 적 가뭄 피해임을 확인할 수 있었다. 즉, 수량 스트레스와 가뭄을 연계하여 가뭄 해석에 활용한다면, 추후에 좀 더 정확한 가뭄 피해를 사전에 예측할 수 있을 것으로 판단된다.

Comparison of Surface Water and Groundwater Responses to Drought using the Standardized Precipitation Index (SPI) (표준강수지수(SPI)를 이용한 가뭄에 대한 지표수와 지하수 반응 비교)

  • A correlation analysis was performed to investigate differences in the response of surface water and groundwater to drought using the Standardized Precipitation Index (SPI). Water level data of 20 agricultural reservoirs, 4 dams, 2 rivers, and 8 groundwater observation wells were used for the analysis. SPI was calculated using precipitation data measured at a nearby meteorological station. The water storage of reservoirs and dams decreased significantly as they responded sensitively to the drought from 2014 to 2016, showing high correlation with SPI of the relatively long accumulation period (AP). The responses of rivers varied greatly depending on the presence of an upstream dam. The water level in rivers connected to an upstream dam was predominantly influenced by the dam discharge, resulting in very weak correlation with SPI. On the contrary, the rivers without dam exhibited a sharp water level rise in response to precipitation, showing higher correlation with SPI of a short-term AP. Unlike dams and reservoirs, the responses of groundwater levels to precipitation were very short-lived, and they did not show high correlation with SPI during the long-term drought. In drought years, the rise of groundwater level in the rainy season was small, and the lowered water level in the dry season did not proceed any further and was maintained at almost the same as that of other normal years. Conclusively, it is confirmed that groundwater is likely to persist longer than surface water even in the long-term drought years.

Synthesis and Properties of Siloxane Containing Copolyimides (실록산이 함유된 폴리이미드의 합성과 물성)

  • Siloxane containing copolyimide (SPI) was synthesized from 3, 3', 4, 4'-benzophenonetetracarboxylic dianhydride(BTDA), 4, 4'-methylene dianiline(MDA), 4, 4'-oxydianiline(ODA) and amine-terminated polydimethyl-siloxane(PDMS). Homopolyamic acid(HPAA) in tetrahydrofuran(THF) was reacted with PDMS to obtain siloxane containing polyamic acid(SPAA) followed by the thermal curing to manufacture SPI. SPAA and HPAA exhibited inherent viscosity value of 0.35~0.48dl/g. Glass transition temperature of SPI ranged in $258^{\circ}C{\sim}264^{\circ}C$. SPI had a lower $T_g$ than that of HPI. ODA based HPI and SPI showed slightly higher $T_g$ values, thermal stability, and water content.

Effects of Heat-oxidized Soy Protein Isolate on Growth Performance and Digestive Function of Broiler Chickens at Early Age

  • This study was conducted to investigate effects of heat-oxidized soy protein isolate (HSPI) on growth performance, serum biochemical indices, apparent nutrient digestibility and digestive function of broiler chickens. A total of 320 1-day-old Arbor Acres chicks were randomly divided into 4 groups with 8 replicates of 10 birds, receiving diets containing soy protein isolate (SPI, control group) or the same amount of SPI heated in an oven at $100^{\circ}C$ for 1, 4, or 8 h, for 21 days, respectively. The results indicated that compared with the control group, body weight gain and feed intake of birds fed diet containing SPI heated for 8 h were significantly lower (p<0.05). Serum urea nitrogen concentration was higher in the broilers fed diet containing SPI heated for 4 or 8 h at d 21 (p<0.05). In contrast, serum glucose content was decreased by HSPI substitution at d 21 (p<0.05). The relative pancreas weight in HSPI groups was higher than that in the control group at d 21 (p<0.05). Meanwhile, the opposite effect was observed for relative weight of anterior intestine and ileum in broilers fed a diet containing SPI heated for 8 h (p<0.05). Birds fed diets containing SPI heated for 4 or 8 h had a decreased lipase activity in anterior intestinal content at d 14 and 21 (p<0.05), respectively. In addition, the same effect was also noted in broilers given diets containing SPI heated for 1 h at d 21 (p<0.05). Similarly, amylase, protease and trypsin activity in anterior intestinal content were lower in broilers fed diets containing SPI heated for 8 h at d 21 (p<0.05). The apparent digestibility of dry matter (DM) from d 8 to 10 and DM, crude protein (CP), and ether extract from d 15 to 17 were lower in broilers fed diets containing SPI heated for 8 h (p<0.05). Besides, birds given diets containing SPI heated for 4 h also exhibited lower CP apparent digestibility from d 15 to 17 (p<0.05). It was concluded that HSPI inclusion can exert a negative influence on the growth performance of broilers, which was likely to result from the simultaneously compromised digestive function.

Volatile Compound, Physicochemical, and Antioxidant Properties of Beany Flavor-Removed Soy Protein Isolate Hydrolyzates Obtained from Combined High Temperature Pre-Treatment and Enzymatic Hydrolysis

  • The present study investigated the volatile compound, physicochemical, and antioxidant properties of beany flavor-removed soy protein isolate (SPI) hydrolyzates produced by combined high temperature pre-treatment and enzymatic hydrolysis. Without remarkable changes in amino acid composition, reductions of residual lipoxygenase activity and beany flavor-causing volatile compounds such as hexanol, hexanal, and pentanol in SPI were observed after combined heating and enzymatic treatments. The degree of hydrolysis, emulsion capacity and stability, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity, and superoxide radical scavenging activity of SPI were significantly increased, but the magnitudes of apparent viscosity, consistency index, and dynamic moduli (G', G") of SPI were significantly decreased after the combined heating and enzymatic treatments. Based on these results, it was suggested that the enzymatic hydrolysis in combination with high temperature pre-treatment may allow for the production of beany flavor-removed SPI hydrolyzates with superior emulsifying and antioxidant functionalities.

Assessment of Smoke Risk of Combustible Materials in Fire (화재 시 가연성 물질의 연기 위험성 평가)

  • The smoke hazard assessment of building materials focusing on smoke performance index-II (SPI-II) and smoke growth index-II (SGI-II) was investigated. The test species used were Japanese cedar, spruce, lauan, and red pine. The smoke characteristics of wood specimen were investigated using a cone calorimeter (ISO 5660-1). SPI-II was measured after the combustion reaction increased by 1.31~2.15 times based on red pine. The fire risk by SPI-II increased in the order of spruce, lauan, Japanese ceda, and red pine. SGI-II increased by 1.18~2.55 times compared to that of Japnese ceda. The fire risk caused by SGI-II increased in the order of Japanese ceda, spruce, lauan, and red pine. COmean concentrations were ranged from 58 to 133 ppm, which was higher than permissible exposure limits of the occupational safety and health administration (OSHA), 50 ppm. Therefore, woods such as red pine containing various volatile organic substances, were considered to be highly smoke hazardous due to low SPI-II and high SGI-II.
