  • In this study, the antioxidative activity, inhibitory effects on tyrosinase and elastase, and active components of Quercus salicina Blume extracts were investigated. Q. salicina Blume was extracted using 50 % ethanol, from which ethyl acetate and aglycone fractions were prepared. The DPPH (1,1-diphenyl-2-picrylhydrazyl) scavenging activity ($FSC_{50}$) of Q. salicina Blume aglycone fraction was the highest ($8.25\;{\mu}g$/mL). The luminol-dependent chemiluminescence assay showed that reactive oxygen species (ROS) scavenging activities ($OSC_{50}$) of Q. salicina Blume aglycone fraction on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system was the most prominent at $0.039\;{\mu}g$/mL. The protective effects of extract/fractions of Q. salicina Blume against the rose-bengal sensitized photohemolysis of human erythrocytes were increased in a concentration dependent manner ($1{\sim} 25\;{\mu}g$/mL). Especially, ${\tau}_{50}$ of aglycone fraction in $10 \;{\mu}g$/mL concentration showed the most protective effect at 259.9 min. The inhibitory effects ($IC_{50}$) on tyrosinase and elastase of Q. salicina Blume extracts were higher at aglycone fraction (respectively, $21.82 \;{\mu}g$/mL, $41.18\;{\mu}g$/mL). Active component analysis by TLC and HPLC showed quercetin, keampferol, catechin, quercitrin, isoquercitrin, hyperoside, and etc. These results indicate that Q. salicina Blume extract has strong antioxidative activity and can be used as antioxidant. Particularly, aglycone fraction of Q. salicina Blume showed superior antioxdative activity and high inhibitory effect on tyrosinase and elastase. Therefore, aglycone fraction of Q. salicina Blume could be applicable to new functional cosmetics.

  • In this study, the antioxidative effects and component analysis of 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from Annona muricata leaf were investigated. Free radical scavenging activities were performed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay, total antioxidant capacities were estimated using luminol-dependent chemiluminescence assay and $^1O_2$ quenching effects were estimated. Free radical scavenging activities ($FSC_{50}$) of 50% ethanol extract, ethyl acetate fraction and aglycone fraction were 45.6, 29.8 and $18.0{\mu}g/mL$, and total antioxidant capacities ($OSC_{50}$) were 4.4, 1.1 and $2.8{\mu}g/mL$, respectively. As a result of $^1O_2$ quenching experiment, ethyl acetate and aglycone fraction showed similar activities to L-ascorbic acid used as a comparative control. The cellular protective effects of 50% ethanol extract on the $^1O_2-induced$ cellular damage of human erythrocytes were exhibited at concentration-dependent ($5-50{\mu}g/mL$). TLC and HPLC were used to analyse components in the ethyl acetate fraction and aglycone fraction of Annona muricata leaf. In ethyl acetate fraction, rutin (quercetin-3-O-rutinoside), kaempferol-3-O-neohesperidoside, nicotiflorin (kaempferol-3-O-rutinoside), p-coumaric acid were identified. In aglycone fraction, kaempferol was identified. These results suggest that the extracts of Annona muricata leaf have the applicability as antioxidant cosmeceutical ingredients.

  • Chrysanthemum is a cut flower species that normally lasts for 1 to 2 weeks, in some cases 3-4 weeks. This has been attributed to low ethylene production during senescence. Reduction in cut flower quality has been attributed to the formation of air embolisms that partially or completely blocks the water transport from the vase solution to the rest of the cut flower stem, increasing hydraulic resistance which may cause severe water stress, yellowing, wilting of leaf, and chlorophyll degradation. Standard type chrysanthemum can be harvested when buds were still tightly closed and then fully opened with the simple bud-opening solution. Standard type chrysanthemum can also be harvested when the minimum size of the inflorescence is about 5-6 cm bud which opened into the first flower full-sized flower. While spray varieties can be harvested when 2-4 most mature flowers have opened (40% opening). Cut flowers are sorted by stem length, weight, condition, and so on. Standard chrysanthemum is 80 cm length for standard type and 70cm for spray type. Pre-treatment with a STS, plant regulator such as GA, BA, 1-MCP, chrysal, germicide, and sucrose, significantly improved the vase life and quality of cut flowers. It is well established that vase solutions containing sugar can improve the vase life of cut chrysanthemum. Chrysanthemum is normally packed in standard horizontal fiberboard boxes. Chrysanthemum should normally be stored at $5{\sim}7^{\circ}C$. Precooling resulted in reduction in respiration, decomposition, and transpiration activities as well as decoloration retardation. There was significant difference between "wet" storage in 3 weeks and "dry" storage in 2 weeks. In separate pulsing solution trials, various germicides were tested, as well as PGRs to maintain the green color of leaves and turgidity. Prolonging vase life was attained with the application of optimal solution such as HQS, $AgNO_3$, GA, BA and sucrose. This also retarded senescence in leaves of cut flower stems. Fresh cut chrysanthemum can be transported using a refrigerated van with $5{\sim}7^{\circ}C$. Increasing consumption and usage of cut chrysanthemum of various cultivars would require efficient transport system, and effective information exchange among producer, wholesaler, and consumer.

  • An attempt was made to utilize the enzyme produced by Asp. oryzae as meat tenderizer. The production, purification, and various properties of proteinase produced by Asp. oryzae were investigated. Results obtained are as follow; 1. A strain which had the highest proteolytic activity was selected among 9 Aspergillus species. 2. Culture medium consisted of wheat bran 10g, 2% glucose, 0.03% urea and 0.1% $MgSO_4$ (pH 6.5). Mold was incubated at $30^{\circ}C$ for 3 days. 3. Enzyme extract from culture medium were fractionated with ammonium sulfate and purified by Sephadex G-75 column chromatography. 4. When pH of reaction mixture was controlled, maximal activity of proteinase by Asp. oryzae was obtained at pH 3, pH 6.6, $8.4{\sim}8.5$ and pH 10.0 to 10.5. Those results were interpreted to show that enzyme consists of acid proteinase, neutral proteinase and alkaline proteinase. Enzyme was stable at pH 6 to 10. 5. Opt. temperature for proteinase activity was $50^{\circ}C$, but enzyme was stable up to $40^{\circ}C$. 6. The proteinase was inhibited by $Ag^+$. It was also inhibited by EDTA. 7. When myofibrillar proteins were treated by proteinase from Asp. oryzae, ATPase activities of myofibrillar proteins changed remarkably. Accordingly, it was concluded that proteinase produced by Asp. oryzae were able to be used as meat tenderizer.

  • This pot experiment was conducted to investigate the effects of combined micronutrient application($T_1$;control, $T_2$; Fe, $T_3$; Fe+Mn, $T_4$; Fe+Mn+Cu, $T_5$ ; Fe+Mn+Cu+Zn, $T_6$ ; Fe+Mn+Cu+Zn+Mo, $T_7$ ; Fe+Mn+Cu+Zn+Mo+B) on forage performance of pure and mixed cultures of orchardgrass and white clover. The third part was concerned with the changes in the contents and yields of N-compounds (crude/pure protein and soluble N-compounds) in forages. The results obtained are summarized as follows: 1. The contents of N-compounds(crude/pure protein and soluble N-compounds) were generally different according to the forage species, whether it was a pure or mixed culture, and additional fertilization, especially N. In orchardgrass, these contents were relatively low at the $T_3$ and $T_6$ in both pure and mixed cultures. In white clover, these contents were relatively decreased by the $T_1$, $T_3$, and $T_6$ in mixed culture. 2. The treatments influenced relatively more on the yields of crude/pure protein than on the dry matter yields of forages, and this tendency was more significant in white clover than in orchardgrass. 3. In white clover, the great differences in the yields of crude protein by the treatments occurred especially in mixed culture and at 5th cut without no additional fertilization. In white clover, the positive effects of optimum treatments on the yields of crude protein seemed to be decreased by the additional fertilization, especially N. In mixed culture, the favorable growth of white clover by the optimum treatments tended to be positively related to the favorable contents and yields of N-compounds. The changes in the yields of pure protein were similar to the tendency of crude protein

This 4th part was related to the changes in the contents of micronutrients(Fe, Mn, Cu, Zn, Mo, and B) in forages. The results obtained are summarized as follows: 1. General differences have been showed in the contents of micronutrients based on the treatments, forage species, pure/mixed culture, cutting order, and additional fertilization, especially N. Compared to pure culture, orchardgrass showed relatively high contents of Mn and Zn, and low contents of B and Fe in mixed culture. White clover, however, tended to be exactly opposed to the above trends. The contents of Cu and Mo did not show any differences between pure and mixed cultures. 2. In relative comparison, the $T_7$ influenced negatively on the contents of Cu, Zn, and Mo in orchardgrass. The $T_7$ also influenced negatively on the contents of Mo in white clover. However, the $T_7$ influenced positively on the contents of Mn in orchardgrass, and also influenced positively on the contents of Fe, Mn, and Cu in white clover. Because of the antagonism between Fe and Mn, the Fe contents in both forages were significantly decreased by the $T_3$. Under the various conditions, the differences among Fe contents tended to be more significant in white clover than in orchardgrass. 3. At the $T_6$ and $T_7$, the Mo contents in both forages tended to be relatively high. The Mo contents, however, were somewhat decreased by the $T_7$ 7/. The Mo-toxicity, which was caused by the high Mo-contents, tended to be diminished, and was likely to be prevented by the optimum B/Mo ratio and B application($T_7$ ).

  • Pinosylvin is a stilbenoid found in the Pinus species. Pinosylvin at ~pM to ~nM concentrations induces cell proliferation, cell migration and anti-inflammatory activity in endothelial cells. However, it was recently reported that pinosylvin at high concentrations (50 to 100 μM) induces cell death in bovine aortic endothelial cells. In this study, we conducted a series of experiments to discover how pinosylvin at a high concentration (50 μM) induces endothelial cell death. Pinosylvin at the high concentration was shown to induce endothelial cell apoptosis through enhancing caspase-3 activity, flip-flop of phosphatidyl serine, and nuclear fragmentation. We found that pinosylvin at the high concentration additively increased caspase-3 activity enhanced by serum-starvation or treatment with 100 μM etoposide. We also determined that pinosylvin at the high concentration promoted activations of c-Jun N-terminal kinase (JNK) and endothelial nitric oxide synthetase (eNOS). We further ran a series of experiments to find out which signaling molecule plays a critical role in the pinosylvin-induced apoptosis. We finally found that SP-600125, a JNK inhibitor, had an inhibitory effect on the pinosylvin-induced endothelial cell death, but L-NAME, an eNOS inhibitor, had no effect. These data indicate that JNK is involved in the pinosylvin-induced apoptosis. Collectively, pinosylvin at high doses induces cell apoptosis via JNK activation.

  • Investigation on health conditions of Thai koi (Anabas testudineus) were carried out through clinical and histopathological observations from different farms of Mymensingh district for seven months during August 2006 to February 2007. Fish sampling and water quality parameters (temperature, dissolved oxygen and pH) were monitored on a monthly basis. Clinical examination of fishes was also carried out for any kind of abnormalities at monthly intervals. Samples of skin, muscle, gill, liver and kidney were observed by histological technique. Among the water quality parameters the values of water temperature, dissolved oxygen were found to be at unfavorable level for fish during the colder months in the farms. Clinically it was observed that fishes were more affected from December to January and almost normal in appearance during August to September and February. Different clinical symptoms like scale loss, dermal lesion, ulcer and loss of caudal fin were noticed in December and January. In histopathological study, structures of fish organs were normal from August to September. In the months of October and November, minor pathologies were found to be started. Marked pathological changes like necrosis, pyknosis, inflammation, hemorrhage, hypertrophy, hyperplasia, missing of primary and secondary gill lamellae were observed in the months of December and January. Whereas, in the month of February the pathological condition of fish gradually reduced. Again when considered individual fish farm, fishes of Reliance Aqua Farm were more affected than Sotota Matshya Hatchery. The study showed that severity of clinical and pathological changes were increased in December and January. During the period EUS and protozoan diseases were noticed in A. testudineus of the investigated farms.

  • A microorganism producing carboxymethylcellulase (CMCase) was isolated from seawater and identified as Bacillus atrophaeus. This species was designated as B. atrophaeus LBH-18 based on its evolutionary distance and the phylogenetic tree resulting from 16S rDNA sequencing and the neighbor-joining method. The optimal conditions for rice bran (68.1 g/l), peptone (9.1 g/l), and initial pH (7.0) of the medium for cell growth was determined by Design Expert Software based on the response surface method; conditions for production of CMCase were 55.2 g/l, 6.6 g/l, and 7.1, respectively. The optimal temperature for cell growth and the production of CMCase by B. atrophaeus LBH-18 was $30^{\circ}C$. The optimal conditions of agitation speed and aeration rate for cell growth in a 7-l bioreactor were 324 rpm and 0.9 vvm, respectively, whereas those for production of CMCase were 343 rpm and 0.6 vvm, respectively. The optimal inner pressure for cell growth and production of CMCase in a 100-l bioreactor was 0.06 MPa. Maximal production of CMCase under optimal conditions in a 100-l bioreactor was 127.5 U/ml, which was 1.32 times higher than that without an inner pressure. In this study, rice bran was developed as a carbon source for industrial scale production of CMCase by B. atrophaeus LBH-18. Reduced time for the production of CMCase from 7 to 10 days to 3 days by using a bacterial strain with submerged fermentation also resulted in increased productivity of CMCase and a decrease in its production cost.
