Isolation and Identification of Contaminated Organisms on Dried Persimmon (곶감으로부터의 오염미생물 분리 및 동정)

  • In this study, we isolated microorganisms from dried persimmon in Sangju and obtained 15 strains of microorganisms as the basic research to prevent the quality changes during drying and storage of dried persimmon. Contaminated microorganisms were separated using seven species of medium. Viable cell counts of dried persimmon from Sangju was $5.18{\times}10^2{\sim}1.68{\times}10^7$ CFU/g. Green mold K2-1 accounted for the highest percentage in the contaminated dried persimmons and identified as a major causative microorganism. Light violet and creamy yeasts were the second largest contaminated microorganisms. Green mold K2-1 strain was identified as Penicillium sp. and fungus K-1 and K-3 were identified as Caldosporium sp. and Aspergillus sp.

A New Species of Cyprinid Fish, Zacco koreanus with Redescription of Z. temminckii (Cyprinidae) from Korea (한국산 피라미속 어류 1 신종 Zacco koreanus 기재와 갈겨니 Z. temminckii의 재기재)

  • A new chub, Zacco koreanus sp. nov., is described on the basis of 104 specimens from six localities in Korea. The new species is distinguished from other Zacco species by the following characteristics: 9~10 scales above lateral line to dorsal fin origin, yellowish anterolateral side of body, no hemicircular red blotch on the upper margin of the eyes, reddish anterior margin of pectoral fin, larger orbit diameter and narrow interorbital width. The new species is widely distributed in the most streams of Korea, except the Yeongsan R., while Zacco temminckii is restricted in the Dongjin R., Yeongsan R., Seomjin R., Tamjin R. and Nakdong R. in Korea. When the two species occurred sympatrically in the same stream, Z. koreanus sp. nov. prefered mostly rapid lotic environments while Z. temminckii inhabited lentic ones. The Korean Z. temminckii was redescribed herein. A key to the Zacco species of Korea and Japan is provided.

An Antibiotic against Bacillus amyloliquefaciens from Streptomyces sp. KM-48 (Streptomyces 속 KM-48 균주가 생성하는 Bacillus amyloliquefaciens 에 대한 항생물질)

  • One of Streptomyces sp. was screened as an antibiotic producing strain against Gram positive bacteria specifically from about one hundred Actinomycetes collections. The isolated KM-48 strain, indexed in our collection, was secreted an anbitiotic in nutrient broth culture. The active principle was extracted with acetone from the lyophilized solid of cultured filtrate. The nonadsorbed elute in aluminium oxide was isolated as a purified homogeneity which revealed a sing1e spot under ultra-violet beam on avicel thin layer chromatography. The isolated antibiotic was sensitive specifically against the Gram positive bacteria and fungi. About fifty percent inhibition of the control growth on Bacillus amyloliquefaciens was 18$\mu\textrm{g}$/$m\ell$. The antibiotic was lavile against thermotreatment and stabled in acidic condition remarkably. The KM-48 was identified as a similar strain to Streptomyces tsusimaensis on its morphological and cultural characteristics.

Identification of Sphaerulina azaleae on Korean Azalea in Korea Based on Morphological Characteristics and Multilocus Sequence Typing (형태적 특징 및 다좌위 염기서열 분석에 의한 산철쭉 모무늬병균 Sphaerulina azaleae 동정)

  • From 2008 to 2017, Korean azalea (Rhododendron yedoense f. poukhanense) showing angular, necrotic leaf spots were found in Jeju and Hongcheon, Korea. The lesions occurred frequently, detracting from the beauty of the glossy green leaves of the plant and causing premature defoliation. Therefore, to identify the fungus associated with the lesions, morphological characterization and molecular phylogenetic analysis of actin (Act), translation elongation factor 1-alpha (EF), internal transcribed spacer (ITS), 28S nrDNA (LSU), and RNA polymerase II encoding the second largest subunit (RPB2) of the two representative isolates were performed. The phylogenetic tree inferred from the neighbor-joining method showed the isolates clustering in the Sphaerulina azaleae group. Therefore, the fungus associated with the angular leaf spots on the Korean azalea was identified as Sphaerulina azaleae.

  • Two individuals ($sp_1$, $sp_2$) of purple and one individual ($sd_1$) of red hearted flower were selected from 18 years old Hibiscus syriacus trees obtained from the seeds treated with colchicine, and their morphological and physiological characteristics were investigated and following results were obtained. 1. The somatic chromosome number of the selected individuals, $sp_1$, $sp_2$, and $sd_1$ were 2n=160, while that of the check tree was 2n=80, indicating that the selected individuals, $sp_1$, $sp_2$ and $sd_1$ were tetraploid. 2. Peroxidase isoenzyme bands of high activity in selected individuals, $sp_1$, $sd_1$ and check tree were mostly in cathode, fixed band was f and v bands, and frequency of each band and their activity were not different between selected individuals, $sp_1$ and $sd_1$ and check tree. 3. The flowers of $sp_1$ individual were large in size and more dark purple than check tree's. The flowers of $sp_2$ individual were not increased in size, but they were dark purple and red heart at the base of the petal was expanded to 2/3 of the petal length. The flower of $sd_1$ individual was also large and some of the red lines from the petal base were extended to 2/3 of the petal length, which was much longer than those of the check tree. 4. Thickess of leaves, length of guard cells, diameter of pollens, wood fiber lengths and woody fiber widths were all increased in $sp_1$, $sp_2$ and $sd_1$ as compared to those of the check tree. 5. Survival percentage of cuttings was 80% with $sp_1$ and 36% with $sd_1$, and their growth performance were inferior to control in their second growing season.

Enhanced Integrated Multi-scale Retinex based on CIELAB Color Space for Improving Color Reproduction (색 재현 개선을 위한 CIELAB 색 공간 기반의 향상된 Multi -scale Retinex)

  • In this paper, we propose the digital image enhancement method including local tone reproduction and preservation of the hue. In recent studies, an integrated multi-scale retinex (IMSR) has produced great naturalness in the resulting images through enhancement of visibility in dark area in input images. However, most methods, including IMSR, work in RGB color spaces. As such, this produces hue distortion from the perspective of the human visual system, that is, hue distortion in CIELAB color space. Accordingly, this paper proposes an tone reproduction and enhancement of saturation method in a device-independent color space, CIELAB, to preserve the hue and obtain a high contrast and naturalness. First, to achieve the desired objectives, the IMSR is then applied to only the $L^*$ values in CIELAB color space, normalization, and simple mapping function, thereby preserving the balance of the color components and enhancement of visibility. Then, saturation adjustment is performed by applying the ratio of the chroma variation at the sRGB gamut boundary according to the corrected luminance. In experiments, the proposed method is shown to improve the visibility in dark shadows and bright regions in the resulting images and reduce any color distortion then preference test are performed.

A Real-Time Head Tracking Algorithm Using Mean-Shift Color Convergence and Shape Based Refinement (Mean-Shift의 색 수렴성과 모양 기반의 재조정을 이용한 실시간 머리 추적 알고리즘)

  • In this paper, we propose a two-stage head tracking algorithm adequate for real-time active camera system having pan-tilt-zoom functions. In the color convergence stage, we first assume that the shape of a head is an ellipse and its model color histogram is acquired in advance. Then, the min-shift method is applied to roughly estimate a target position by examining the histogram similarity of the model and a candidate ellipse. To reflect the temporal change of object color and enhance the reliability of mean-shift based tracking, the target histogram obtained in the previous frame is considered to update the model histogram. In the updating process, to alleviate error-accumulation due to outliers in the target ellipse of the previous frame, the target histogram in the previous frame is obtained within an ellipse adaptively shrunken on the basis of the model histogram. In addition, to enhance tracking reliability further, we set the initial position closer to the true position by compensating the global motion, which is rapidly estimated on the basis of two 1-D projection datasets. In the subsequent stage, we refine the position and size of the ellipse obtained in the first stage by using shape information. Here, we define a robust shape-similarity function based on the gradient direction. Extensive experimental results proved that the proposed algorithm performs head hacking well, even when a person moves fast, the head size changes drastically, or the background has many clusters and distracting colors. Also, the propose algorithm can perform tracking with the processing speed of about 30 fps on a standard PC.

  • The chromosomal DNA fragments of thermophilic alkalophilic Bacillus sp, K-17, a potent xylanhydrolyzing bacterium, were ligated to a vector plasmid pBR322 and transformed into Escherichia coli HB101. The plasmid pAX278, isolated from a transformant forming yellow color on the LB agar plate containing 1 mM p-nitrophenyl- $\beta$-xylopyranoside, was found to enable the transformants to produce p-xylosidase. The 5.0 kilobase insert of pAX278 had single sites for EcoRI, PstI, XbaI, and PvuII, and 2 sites for BglII. Biotinylated pAX218 was hybridized to 0.9 kb as well as 5.0 kb fragment from Bacillus sp. K-17 DNA on nitrocellulose filter. pGX718 was constructed by inserting the 5.0 kb HindIII fragment of pGX278 at the HindIII site of pGR71, E. coli and B. subtilis shuttle vector. The enzymatic properties of $\beta$-xylosidase from E. coli HB101 carrying recombinant plasmid were the same those of $\beta$-xylosidase from Bacillus sp. K-17.

Effect of Storage Temperature and Packing Method on the Fermentation Chacteristics of Kakdugi (Diced Radish Kimchi) (저장온도와 포장방법에 따른 깍두기의 발효특성)

  • Effect of storage temperature and packing method on the changes in pH, acidity, the levels of lactic acid bacteria, volume expansion of packages and the colorness during the fermentation of kakdugi (diced radish kimchi) were investigated. The fermentation characteristics were greatly influenced by the temperature. The pH of kakdugi fermented at 15$^{\circ}C$ was 4.10 at 3 days, while the pH of kakdugi fermented at $0^{\circ}C$ was 4.03 at 18 days. The acidity of kakdugi fermented at $0^{\circ}C$ very slowly increased compared to that of the kakdugi fermented at 15$^{\circ}C$, 1$0^{\circ}C$ or 5$^{\circ}C$. The required days to reach the acidity of 0.6% (optimally ripened state) were 6 days at 15$^{\circ}C$, 7 days at 1$0^{\circ}C$, 18 days at 5$^{\circ}C$ and 30 days at $0^{\circ}C$. Despite of different fermentation temperature, the numbers of Leuconostoc sp. in optimally ripened kakdugi were similar. However, Lactobacillus sp. in kakdugi fermented at $0^{\circ}C$ was the smallest in number. Also, the volume expansion of aluminum pack was the lowest in kakdugi fermented at $0^{\circ}C$. Both pH and acidity of the kakdugi fermented in different packing condition did not show any difference at 1$0^{\circ}C$ for 20 days, however, the color of the kakdugi was affected by a packing method. Aluminium laminate package in which Ca(OH)$_2$ Pack stuck inside resulted in decolorization of kakdugi compared with aluminum package without Ca(OH)$_2$ pack or plastic jar. It showed the lowest redness and the highest lightness and yellowness. These results indicated that fermentation at $0^{\circ}C$ could increase storage period of kakdugi with minimum growth of Lactobacillus sp., and plastic jar might relieve the problem of volume expansion and decolorization of kakdugi.

Changes in Quality Characteristics and Chemical Components of Sweet Potatoes Cultivated using Different Methods (재배방법에 따른 고구마의 품질특성 및 화학성분 변화)

  • Effects of cultivation methods on quality characteristics, pasting characteristics, chemical components, and antioxidant activities of sweet potatoes (Ipomoea batatas (L.) Lam) were determined. The Brix degree, hunter color value, pasting characteristics, moisture, protein, and mineral contents of the sweet potatoes showed significant differences from cultivation methods. The total polyphenol and flavonoid contents of the methanolic extracts of the sweet potato's pericarp were higher than sweet potato's sarcocarp. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the sweet potato's pericarp on the conventional culture and successful cropped hairy vetch culture was 776.38 and 715.20 mg TE/100 g sample. The 2,2'-azino-bis-3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS) radical scavenging activity of the sweet potato's pericarp on the conventional culture and successful cropped hairy vetch culture was 708.03 and 708.58 mg TE/100 g sample. Generally, there was a difference in antioxidant compound content and radical scavenging activity on the methanolic extract of sweet potato with cultivation methods.
