Physiological Activities of Mistletoe Extracts from Viscum album L. (추출용매에 따른 겨우살이 추출물의 생리활성 효과)

  • Physiological activities of mistletoe extracts were examined. Total polyphenol contents, electro-donating ability, SOD-like activity, nitrite-scavenging ability and tyrosinase of mistletoe extracted with water, 50%, and 100% ethanol were determined. Total polyphenol contents of powder extracts were higher than slice extracts. EDAs showed over 90% at powder extracts. Especially mistletoe extracts with 50% ethanol were higher than water extracts. SOD-like activities of water, 50% and 100% ethanol extracts of all samples were $23.71{\sim}33.4%$ lower than those of 1.0% and 0.1% L-ascorbate solutions. Nitrite-scavenging activities at pH 1.2 were the most effective in water, 50% and 100% ethanol extracts. The results will be useful for understanding the physiolosical activities of mistletoe extracts.

  • Lactic acid bacteria (LAB) are typical probiotic microbes which are used in various industries including fermented foods, feed additives, and pharmaceuticals. The purpose of this study was to compare the ability of antioxidative activities of 23 strains of LAB isolated from cheese, kimchi, yogurt, and etc. LAB were tested for antioxidative activities such as 2,2'- azino-bis(3-ethyl-benzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities and superoxide dismutase (SOD)-like ability. Antioxidative activities were examined different media such as MRS, M17, and 10% skim milk. Culture supernatant in M17 medium showed a higher antioxidative activity than these in MRS and 10% skim milk. Comparison of ABTS radical scavenging ability by the LAB showed a wide variation with a range from 9~93% and SOD-like ability of LAB ranged from 70~78%, respectively. Therfore, some of LAB can play important roles in the protection for oxidative stress and CPP-producing activity of LAB may be a promising material for application in the dairy industry.

  • Superoxide dismutase (SOD) is an important enzyme which catalyzes superoxide radicals to hydrogen peroxide. A Cu, Zn sod-like gene was found in Bombyx mori nuclear polyhedrosis virus encoding 151 amino acids. To demonstrate its function, a recombinant virus named dsBmNPV with deleted sod gene was constructed. It was discovered that the sod gene was not essential for viral replication. Studies on growth of budded virus in BmN cells and superoxide dismutase and catalase activities in vivo after dsBmNPV infection showed that the titer of dsBmNPV decreased obviously comparing to wild type BmNPV, the sod gene was effective on genomic DNA replication of baculovirus, the peak of SOD activity of silkworm infected with wt-BmNPV appeared between 36 and 48 hrs post infection, and with dsBmNPV, it did not appear. And the changes of CAT activity after infection were similar to SOD activity.

The Effect of Chrysanthemum morifolium L. Extract on Cultured Neuroglial Cells Damaged by Glucose Oxidase

  • To clarify the oxidative stress of reactive oxygen species (ROS) and the effect of Chrysanthemum morifolium L. (CM) flower extract on the cultured neuroglial cells (C6 glioma) damaged by ROS, cell adhesion effect was measured by colorimetric assay after cultured C6 glioma cells were treated with various concentrations of glucose oxidase (GO) for 5 hours. For the antioxidative effect of CM flower extract, cell adhesion activity (CAA), superoxide dismutase (SOD)-like activity and lactate dehydrogenase (LDH) activity were assessed against GO-induced cytotoxicity on same cultures. In this study, GO remarkably decreased CAA dose-dependently, and the $XTT_{90}$ and $XTT_{50}$ values were measured at 15 mU/mL and 50 mU/mL following the treatment of C6 glioma cells with 5~60 mU/mL of GO. The CM flower extract significantly increased cell adhesion activity damaged by GO-induced cytotoxicity, and it also showed the SOD-like activity and the decrease of LDH activity. From these results, it is suggested that GO was cytotoxic on cultured C6 glioma cells, and CM flower extract showed antioxidative effects as shown by the increased CAA, SOD-like activity and the decrease of LDH activity on GO-induced cytotoxicity on the same cultures.

Antioxidant and Antihemolytic Activity of Ethanol Extracts of Rubus coreanus Miquel (복분자 에탄올 추출물의 항산화 및 항용혈 효능)

  • To develop a natural antioxidant and anti-hemolytic agents, we investigated the effects of ethanol extracts of Rubus coreanus Miquel (crude extracts). Crude extract was extracted with ethanol. Antioxidant activity of crude extracts was evaluated by employing two different assays, 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity and super oxide dismutase(SOD)-like activity. Also, anti-hemolytic activity of crude extract was determined using $H_2O_2$-induced hemolysis in normal rat red blood cells (RBC) or plasma. The extracts obtained from crude extract dose-dependently increased the scavenging activity on DPPH-induced radicals and SOD-like activity. RBC oxidative hemolysis induced by $H_2O_2$ was significantly suppressed by the extracts of Rubus coreanus Miquel in a dose-dependent manner. These results suggest that crude extracts may have value as the potential antioxidant and anti-hemolytic medicinal plant.

Antioxidant and Anti-Inflammatory Activities of Extracts from Eugenia caryophyllata Thunb. (정향 열수 및 에탄올 추출물의 항산화 및 항염 효과)

  • The objective of this study was to investigate the antioxidant and anti-inflammatory activities of Eugenia caryophyllata Thunb. water and 70% ethanol extracts. The content of total polyphenol was significantly higher in water extract than in 70% ethanol extract. The DPPH radical scavenging activity of water extract was similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. The ABTS radical scavenging activities of water and 70% ethanol extract were similar to that of Vit. C at a concentration of $1,000{\mu}g/mL$. SOD-like activity of water extract was higher than that of 70% ethanol extract at a concentration of $1,000{\mu}g/mL$ but lower than that of Vit. C. The DPPH radical scavenging activity, ABTS radical scavenging activity, and SOD-like activity increased as concentrations of water and 70% ethanol extracts increased. Cell cytotoxicity was not observed at all concentrations except at $100{\mu}g/mL$ concentration of water extract. Inhibitory activity on NO production effect of water extract was significantly higher than that of 70% ethanol extract. These results show that E. caryophyllata Thunb. has potent biological activities, and their activities were different depending on extraction solvent.

Study on the Anti-oxidant and Cosmeceutical Activity of Taraxacum platycarpum (포공영(浦公英)의 항산화 및 화장품약리학적 활성 연구)

  • Objectives : In this study, anti-oxidant activity of Taraxacum platycarpum is confirmed to investigate cosmeceutical activity for utilization as a cosmetic ingredients. Methods : In the anti-oxidant and whitening effect experiment, the electron donating ability of Taraxacum platycarpum extracts, their SOD-like activity, and the inhibitory activity of xanthine oxidase, tyrosinase were examined. Results : Radical-scavenging activity of water and ethanol extract was examined using ${\alpha},{\alpha}-diphenyl-{\beta}-picrylhydrazyl$ (DPPH). Water and ethanol extract from Taraxacum platycarpum showed 62.9% and 54.7% at 500 ppm concentration in DPPH radical inhibition, respectively. In the test of superoxide dismutase (SOD)-like activity, water extract showed 63.4% at 1,000 ppm concentration, while ethanol extract showed 49% at 1,000 ppm concentration. In the test of xanthine oxidase inhibition, ethanol extract showed 66.8% at 1,000 ppm concentration. Tyrosinase inhibition effect related to whitening effect showed 36.3% and 54.2% in ethanol extract and water extract at 1,000 ppm concentration, respectively. Conclusion : According to these results, it is possible that the extract of Taraxacum platycarpum can be used as a new natural material of cosmetic industry.

  • Objectives : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. Methods : The effects on electron donating ability (EDA), SOD-like activity and xanthine oxidase inhibition have been investigated the anti-oxidant activity for utilization as a natural material of cosmetics. Results : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. In the electron donating ability test, water and ethanol extract from Magnoliae Flos showed an effect of 75.6% and 83.5% at 1,000 ppm concentration, respectively. In the superoxide dismutase (SOD)-like activity, 1,000 ppm of water extract showed an effect of 45.2% and ethanol extract showed an effect of 64.2%. Xanthine oxidase showed a meaningful inhibitory effect of 68.8% in water extract and 23.2% at 1,000 ppm in ethanol extract. Conclusion : From these results, Magnoliae Flos showed the development possibility as a natural material of anti-oxidant cosmetics.

  • In order to study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$, testing antibacterial effect on E. coli O157 which causes infectious inflammatory disease; measunng inhibitory effect on tyrosinase which stimulates melanin formation; and measuring the influence of it to SOD like activity, DPPH and TBARS which are related to ageing and carcinogenesis. The results of these experiments, are as follows. 1. In the antibacterial experiment with the extract of Hedyotis diffusa $W_{ILLD}$ on E. coli O157, there was no change in proliferation from the beginning of culture to two hours after, but proliferation inhibiting effect on E. coli O157 was detected from three hours after the beginning of culture. 2. Tyrosinase inhibitory effect was measured as $0.39{\pm}0.026%$. Compared with the control group, the effect was very slight. 3. The SOD like activity of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 21.33${\pm}$ 4.264%. Compared with the extracts of several other herbs, it was much more significant. 4. The DPPH of the extract of Hedyotis diffusa $W_{ILLD}$. was $53.3{\pm}0.91%$ when 0.02g was used and $83.5{\pm}1.82% $ when 0.05g was used. The result when 0.05g was used was more significant than 0.02g was used. 5. The TBARS of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 0.724 ${\pm}$0.O04MDAppm. Compared with the extracts of several other herbs, the result was more significant. From these results, we found that Hedyotis diffusa $W_{ILLD}$. can be used in therapy of dysentery with bloody stool and fever which is caused by infection with E. coli O157 and that it can be also used effectively as age resister or anticarcinogen.

Biological Activity of Panax ginseng C. A. Meyer Culture Roots Fermented with Microorganisms (미생물 처리 발효 산삼배양근의 생리활성 변화)

  • Background: This study examined the use of new bio-materials with enhanced value and functionality, which were derived from fermented wild ginseng cultures. Methods and Results: To examine the antioxidant activity associated with biological functions, radical scavenging analyses (2,2-diphenyl-1-picrylhydrazyl, DPPH and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid, ABTS) and superoxide dismutase (SOD)-like activity analyses were conducted. Furthermore, the total phenolic and flavonoid contents of wild ginseng fermented with microorganisms (Leuconostoc mesenteroides, Bacillus circulans, Bacillus licheniformis and B. subtilis subsp. inaquosorum) were evaluated to determine the antioxidant activity increment. Regarding ginseng fermented with B. licheniformis, values of $70.6{\pm}1.4%$, $44.3{\pm}1.7%$, and $88.4{\pm}1.3%$ were measured using DPPH, ABTS, and SOD-like antioxdiant activity analyses, respectively. The total phenolic content in ginseng fermented with B. licheniformis was $184.5{\pm}0.9{\mu}g{\cdot}GAE/m{\ell}$, and the total flavonoid contents was $108.5{\pm}1.8{\mu}g{\cdot}QE/m{\ell}$ in ginseng fermented with L. mesenteroides. Conclusions: Of the four types of lactic acid bacteria examined, the use of B. licheniformis to ferment ginseng resulted in greatest increase in antioxidant activity. Therefore, ginseng fermented by microorganisms might be used to produce functional bio-materials.
