양파 음료 제조를 위한 기능성 성분 추출 최적화 (Extractive Optimization of Functional Components for Processing of Onion Health Promotion Drink)

  • 복합음료의 재료인 양파, 솔잎, 뽕잎, 당귀, 감초, 대추를 물과 methanol로 상온에서 각각 추출하여 전자공여능, thiosulfinate함량, ascorbic acid 함량, SOD유사황성 및 pH 1.2와 3.0에서 아질산 소거능 등을 조사하였다. SOD 유사활성은 모든 시료에서 methanol 추출이 우수하였고, 아질산 소거능은 물 추출이 우수하였고 다른 기능성 항목은 재료에 따라 추출방법에 대한 우열의 일정한 경향을 보이지 않았다. 그러나 3 hr동안 hard boil한 물추출이 thiosulfinate의 함량을 제외한 모든 기능성을 상온에서 물추출보다 증가 시켰고, 특히 상온에서 methane이 물 추출보다 높은 값을 보였던 양파, 뽕잎, 대추의 전자공여능과 당귀의 ascorbic acid 함량은 methanol 추출보다도 높은 추출 효율을 보였으므로 hard boil에 의한 물추출이 효과적인 것으로 나타났다. 단 휘발성인 thiosulfinate는 온도의 상승에 따라 그 함량이 급격한 감소를 보였으므로 이 성분은 저온에서 추출하여 혼합하는 방법을 모색해야 할 것으로 여겨진다. 아울러 hard boil에 의한 적정한 추출시간을 조사한 결과, 6 hr이면 thiosulfinate를 제외한 기능성분은 거의 용출 되어지는 것으로 밝혀졌다. 저장중의 변화를 조사한 결과, 저온저장시 ascorbic acid만이 약간의 손실이 더 있는 편이고 다른 기능성 성분은 상온 저장과 거의 같거나 저온저장이 유리하였다. 이상의 결과로 양파의 기능성 강화 복합음료의 제조는 thiosulfinate를 제외하고 6 hr 물로 고와서 추출하고 저온 저장하여야 한다는 것을 알 수 있었다.

백화사설초(白花蛇舌草) 추출물(抽出物)의 항균실험(抗菌實驗) 및 SOD류사활성(類似活性), 전자공여능(電子供與能)에 관(關)한 연구(硏究) (Study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$)

  • In order to study on SOD like activity and Electric donor ability of Hedyotis diffusa $W_{ILLD}$, testing antibacterial effect on E. coli O157 which causes infectious inflammatory disease; measunng inhibitory effect on tyrosinase which stimulates melanin formation; and measuring the influence of it to SOD like activity, DPPH and TBARS which are related to ageing and carcinogenesis. The results of these experiments, are as follows. 1. In the antibacterial experiment with the extract of Hedyotis diffusa $W_{ILLD}$ on E. coli O157, there was no change in proliferation from the beginning of culture to two hours after, but proliferation inhibiting effect on E. coli O157 was detected from three hours after the beginning of culture. 2. Tyrosinase inhibitory effect was measured as $0.39{\pm}0.026%$. Compared with the control group, the effect was very slight. 3. The SOD like activity of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 21.33${\pm}$ 4.264%. Compared with the extracts of several other herbs, it was much more significant. 4. The DPPH of the extract of Hedyotis diffusa $W_{ILLD}$. was $53.3{\pm}0.91%$ when 0.02g was used and $83.5{\pm}1.82% $ when 0.05g was used. The result when 0.05g was used was more significant than 0.02g was used. 5. The TBARS of the extract of Hedyotis diffusa $W_{ILLD}$. was measured as 0.724 ${\pm}$0.O04MDAppm. Compared with the extracts of several other herbs, the result was more significant. From these results, we found that Hedyotis diffusa $W_{ILLD}$. can be used in therapy of dysentery with bloody stool and fever which is caused by infection with E. coli O157 and that it can be also used effectively as age resister or anticarcinogen.

Physiological Activities of Rheum undulatum and Rheum palmatum Extracts as Affected by Solvents

  • Rheum undulatum and Rheum palmatum have been widely used as food material as well as medicinal ingredients for their therapeutic effects in oriental countries. Many kinds of herbs are being used in the manufacture of functional foods. The objective of this study was to determine polyphenol content, superoxide dismutase (SOD)-like activity, tyrosinase activity, and electron donating ability of R. undulatum and R. palmatum. Total polyphenol content were most effective in 50 and 100% ethanol extracts from R. undulatum and R. palmatum. SOD-like activities of R. undulatum extracts were higher than those of R. palmatum extracts, and water extracts of samples were highest. EDAs of R. undulatum extracts were higher (26.76-44.46%) than those of R. palmatum extracts, while those of both extracts were lower than 1.0 and 0.1% L-ascorbate. And these suggest that the extracts of R. undulatum and R. palmatum can be used as a material in functional food.

Saponin-SOD가 SNUC-4 Cell에 의해 유발편 대장암 Nude Mice의 암 성장 억제 효과 (Tumor-Suppressing Effects of Saponin-SOD in Nude Mice)

  • This study was performed to measure the antioxidant effects of red ginseng extracts which antioxidation had been promoted through enzyme hydrolysis. In order to observe their tumor-suppressing effects, an anti-cancer medicine and Saponin-SOD, which was a highly antioxidant beverage made from red ginseng saponin adding SOD-like rice (with embryo buds) extracts, were administered to nude mice with large intestine cancer induced. There was a significant increase in the content of phenolic compounds as the enzyme was added. The red ginseng extracts showed a high electron-donating ability with the passage of time. The electron-donating ability was particularly high in the enzyme-treated red ginseng extract, and also observed as high in Saponin-SOD. The lipid-peroxide generation was inhibited depending on the concentration of Saponin-SOD added; the addition of 0.625% Saponin-SOD served to decrease the inhibition level up to 65% compared with the case of no addition (100%). As a result, it could be assumed that Saponin-SOD would strongly inhibit the oxidation of ghost membrane. After the cancer was induced in nude mice through the injection of SNUC-4 cell, there was a significant inhibition in the growth of tumors in nude mice into which Saponin-SOD were injected; the growth of tumors was gradually decreasing with the passage of time after the cancer induction. In particular, when Saponin-SOD was administered together with an anti-cancer medicine, the synergic effect was observed. In conclusion, Saponin-SOD, when used with an anti-cancer medicine, is expected to reduce the amount of free radical and lipid peroxide, which are known to cause harmful effects occurring from the internal application of medicine.

김천산 자두 후무사의 화학적 성분과 항산화 활성 (Chemical Composition and Antioxidant Activities of Prunus salicina Formosa Produced in Gimcheon)

  • 본 연구에서는 김천산 후무사의 일반성분, 유리당, 유기산, 총 페놀 화합물, 플라보노이드 등 화학적 성분을 조사하고 DPPH radical 소거능, superoxide dismutase(SOD) 유사활성, 아질산염 소거능, 환원력 등의 항산화 활성을 평가하였다. 유리당 함량은 fructose>glucose>sucrose>maltose 순이었으며, 유기산 함량은 malic acid가 가장 높았다. 총 페놀 함량과 플라보노이드 함량은 각각 60% acetone 추출물, 80% ethanol 추출물에서 높았다. DPPH radical 소거능, 환원력, 아질산염 소거능은 60% acetone 추출물에서 가장 높은 활성을 보여주었으나, SOD 유사활성은 농도나 추출용매, 온도에 따른 차이가 거의 없었다.

수순(水醇)추출법으로 조제된 오공 약침액의 생리활성 효과 (Biological Activities of Scolopendrid Pharmacopuncture)

  • Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical increases with age, the reactive oxygen species(ROS) such as hydrogen peroxide($H_2O_2$), nitric oxide(NO). Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. Our reserch objective was to examine the in vitro biological activity of Scolopendrid Pharmacopuncture, including the total poly-phenol content, DPPH radical scavenging, ABTS radical scavenging, Superoxide dismutase(SOD)-like activity, Nitrite scavenging ability. The total poly-phenol contents of Scolopendrid Pharmacopuncture was 35.859mg/L. Elctron donation ability on DPPH was 36.82%. The 2,2'-azinobis-3-ehtlbezothiazoline-6-sulfonic acid radical decolorization (ABTS) was 84.7%. The superoxide dismutase (SOD)-like activities of Scolopendrid Pharmacopuncture was 44.33%. The nitrite scavenging effects were pH dependent, and were highest at pH 1.5(45.2%) and lowest at pH 6.0(11.3%). We conclude that Scolopendrid Pharmacopuncture may be useful as potential sources of antioxidant.

신이화 (Magnoliae Flos)의 항산화 활성에 관한 연구 (Study on the Anti-oxidant Activity of Extracts from the Magnoliae Flos)

  • Objectives : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. Methods : The effects on electron donating ability (EDA), SOD-like activity and xanthine oxidase inhibition have been investigated the anti-oxidant activity for utilization as a natural material of cosmetics. Results : This study was investigated to analyze the anti-oxidant activity of water and ethanol extracts from the Magnoliae Flos. In the electron donating ability test, water and ethanol extract from Magnoliae Flos showed an effect of 75.6% and 83.5% at 1,000 ppm concentration, respectively. In the superoxide dismutase (SOD)-like activity, 1,000 ppm of water extract showed an effect of 45.2% and ethanol extract showed an effect of 64.2%. Xanthine oxidase showed a meaningful inhibitory effect of 68.8% in water extract and 23.2% at 1,000 ppm in ethanol extract. Conclusion : From these results, Magnoliae Flos showed the development possibility as a natural material of anti-oxidant cosmetics.

호부추와 실부추 추출물의 생리활성 효과 (Physiological Activities of Leek Extracts from Allium tuberosum and Allium senescens)

  • 호부추(Allium tuberosum)와 실부추(Allium senescens)를 열수, 50% 에탄올 및 100% 에탄올 등의 추출용매를 사용하여 건물 중량의 10배에 해당되는 부피(w/v)일 때, 추출물의 생리활성을 탐색하였다. 전자공여작용의 경우 호부추의 50% 에탄올추출물에서 41.68%의 높은 전자공여능을 나타내었으며, SOD 유사활성을 측정한 결과 호부추의 열수추출물이 36.33%로 다른 추출물에 비해 높은 활성을 보여주었다. Tyrosinase 저해활성의 경우 두 종류 모두 열수추출물에서 높은 저해활성을 보여주었으며, 호부추가 실부추보다 다소 높은 활성을 나타내었다. 특히 비교물질인 0.01% L-ascorbate와 유사한 활성을 보여주었다. 총 폴리페놀 함량의 경우 두 종류 모두 열수추출물보다 50%, 100% 에탄올추출물에서 높은 함량을 나타내었다. 아질산염 소거작용을 측정한 결과 pH 1.2일 때 호부추와 실부추 모두 소거능이 높게 나타났다. 이와 같은 결과는 부추의 생리활성을 밝혀 기능성 소재로써 이용도가 크게 증가할 것으로 판단된다.

연수 추출물과 Kaempferol의 항산화 및 항암작용에 관한 연구 (Study on the Antioxidant and Anticancer Effects of Extract of Stamens of Nelumbo nucifera and Kaempferol)

  • Objectives : The present study was performed to clarify the antioxidant and anticancer effects of extract of stamens of Nelumbo nucifera and Kaempferol. Methods : Antioxidant effect was measured by DPPH-radical scavenging activity for electron donating ability (EDA), superoxide dismutase (SOD)-like activity for SOD and lipid peroxidation. Anticancer effect was assessed by MTT absorbance for cytotoxicity. Results : Vitamin E, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera increased SOD-like activity and DPPH-radical scavenging activity dose-dependently. On the contrary, lipid peroxidation was time-dependently decreased. Furthermore, Kaempferol and ethyl acetate extract of stamens of Nelumbo nucifera significantly decreased the growth rate of C6 glioma cells. Conclusions : These results suggest that ethyl acetate extract of stamens of Nelumbo nucifera may be a putative antioxidant or anticancer substance.

복분자 딸기의 생리활성 (Physiological Activities of Rubus coreanus Miquel)

  • 복분자딸기 열매의 미숙과와 완숙과 그리고 잎을 사용하여 추출방법에 따른 생리활성을 측정한 결과는 다음과 같다. 즉, 총 폴리페놀 함량은 추출용매에 의한 차이는 보이지 않았고 전체적으로 복분자딸기의 잎에서 높은 총 폴리페놀 함량을 나타내었으며, 특히 75% 아세톤으로 추출한 경우 잎에서 건물기준으로 100 g당 11.38 g으로 가장 높은 값을 나타내었다. 복분자딸기의 전자공여능은 추출 용매에 따른 차이는 보이지 않았으며, 전체적으로 미숙과가 완숙과나 잎에 비하여 90%이상의 높은 전자공여능을 나타내었다. 복분자딸기의 아질산염 소거능을 pH 1.2, 4.2, 6.0의 조건으로 조사한 결과 pH 1.2일 때 가장 높은 아질산염 소거율을 나타내었다. pH 1.2의 조건에서 미숙과의 아질산염 소거율은 80% 메탄올 추출물이 63.24%, 75% 아세톤 추출물은 60.56%로 완숙과나 잎에 비하여 높은 값을 나타내었고, 열수 추출물에서는 잎이 49.83%로 미숙과와 잎에 비하여 높은 아질산염 소거율을 나타내었다. 복분자딸기의 SOD유사활성은 완숙과가 미숙과나 잎에 비하여 모든 추출조건에서 높은 SOD유사활성을 나타내었다. 복분자딸기 추출물의 항균활성을 조사하기 위하여 E. coli, S. typhimurium, B. cereus, S. aureus를 시험균주로 하여 agar diffusion test를 실시한 결과 모든 추출물이 B. cereus에서만 높은 항균활성을 나타내었다.
