Whole genome sequencing of Luxi Black Head sheep for screening selection signatures associated with important traits

  • Objective: Luxi Black Head sheep (LBH) is the first crossbreed specialized for meat production and was developed by crossbreeding Black Head Dorper sheep (DP) and Small Tailed Han sheep (STH) in the farming areas of northern China. Research on the genomic variations and selection signatures of LBH caused by continuous artificial selection is of great significance for identifying the genetic mechanisms of important traits of sheep and for the continuous breeding of LBH. Methods: We explored the genetic relationships of LBH, DP, and several Mongolian sheep breeds by constructing phylogenetic tree, principal component analysis and linkage disequilibrium analysis. In addition, we analysed 29 whole genomes of sheep. The genome-wide selection signatures have been scanned with four methods: heterozygosity (HP), fixation index (FST), cross-population extended haplotype homozygosity (XP-EHH) and the nucleotide diversity (𝜃π) ratio. Results: The genetic relationships analysis showed that LBH appeared to be an independent cluster closer to DP. The candidate signatures of positive selection in sheep genome revealed candidate genes for developmental process (HoxA gene cluster, BCL2L11, TSHR), immunity (CXCL6, CXCL1, SKAP2, PTK6, MST1R), growth (PDGFD, FGF18, SRF, SOCS2), and reproduction (BCAS3, TRIM24, ASTL, FNDC3A). Moreover, two signalling pathways closely related to reproduction, the thyroid hormone signalling pathway and the oxytocin signalling pathway, were detected. Conclusion: The selective sweep analysis of LBH genome revealed candidate genes and signalling pathways associated with developmental process, immunity, growth, and reproduction. Our findings provide a valuable resource for sheep breeding and insight into the mechanisms of artificial selection.

The Effects of Injinchunggan-tang(Yinchenqinggan-tang) on DMN-induced Liver Damage by Applying Proteomics (인진청간탕(茵蔯淸肝湯)이 DMN 유발 간섬유화와 단백질 발현에 미치는 영향)

  • Objectives : The purpose of this study was to investigate the effects of Injinchunggan-tang (Yinchenchinggan-tang) on DMN-induced liver damage by applying proteomics. Materials and Methods : Sprague-Dawley rats were used in this experiment and were divided into the normal group (normal saline), the control group (DMN) and the sample group (DMN+IJCGT). DMN was injected i.p. once a day three times a week for 3 weeks in the control group. Normal saline instead of DMN was administered to the normal group. In the sample group, Injinchunggan-tang (Yinchenchinggan-tang) extract was orally administered once a day for 10 days after DMN was induced. The livers of each group were processed and analyzed by histology, Western blot, $Oxyblot^{TM}$, CBB and 2-dimensional electrophoresis. Results : In the histological findings of the liver, IJCGT reduced collagen deposition and liver damage in DMN-induced hepatic fibrosis. IJCGT increased MMP-13 protein production assessed by western blot. Protein oxidation induced by DMN treatment was decreased by IJCGT. In the 2-dimensional electrophoresis finding, the level of the increased proteins induced by DMN treatment such as GRP 75, 58kDa glucose regulated protein and heat shock 70kDa protein 5 were decreased by IJCGT. IJCGT was considered to have the protective effects on hepatotoxicity induced by DMN. In the 2-dimensional electrophoresis finding, the level of increased oxidized proteins such as heat shock 70 protein, mitochondrial malonyltransferase, calreticulin precursor, actin, NADP-isocitrate dehydrogenase, ankyrin repeat and SOCS box protein 11 were decreased by IJCGT. IJCGT was considered to have protective effect on the protein production induced by DMN treatment. Conclusion : Injinchunggan-tang (Yinchenchinggan-tang) exerts an inhibitory effect against the fibrosis and protein oxidation induced by DMN treatment in the rat liver. IJCGT was considered to have protective effects on the hepatotoxicity and protein production induced by DMN treatment.

Development of Index of Park Derivation to Promote Inclusive Living SOC Policy (포용적 생활 SOC 정책 추진을 위한 공원결핍지수 개발 연구)

  • In order to resolve the imbalances in the supply of living SOCs according to socio-economic status, location, and population groups, the discussions on inclusive city policies are expanding. The purpose of this study is to propose an Index of Park Derivation (IPD) as an alternative indicator for the promotion of an inclusive urban park policy that can be applied in the 7 major metropolitan cities to select a region with a relatively high park needs. The main research results are as follows. First, the concept of an inclusive urban park policy is defined as "a policy to supply to manage high-quality park services with priority given to areas with low socio-economic and environmental status, such as a large amount of elderly, children, low-income families, areas vulnerable to disasters, such as heat and fine dust, and population groups." Second, we developed the index of park derivation (IPD), which is a combination of 17 variables including park service level, demographic characteristics, economic and educational level, health level, and environmental vulnerability. The variables that constitute the index of park deprivation (IPD) can be applied to SOC policies outside the parks, such as sports facilities, daycare centers, kindergartens, and public libraries. Third, applying index of park deprivation (IPD) to 1,148 Eup/Myeon/dong areas of the 7 metropolitan cities resulted in areas with relatively high park service needs. This study implies that the central and the local government suggest an alternative index to promote an inclusive urban park policy based on statistical and geographical information and data that can be easily accessed and utilized.

Study on Predicting Changes in Traffic Demand in Surrounding SOCs Due to Road SOC Construction Using Big Data - Centered Around the Connecting Road between Incheon Yeongjong International City and Cheongna International City (3rd Bridge) - (빅데이터를 활용한 도로 SOC건설에 따른 주변 SOC 교통수요 변화 예측 연구 - 인천 영종국제도시~청라국제도시 간 연결도로(제3연륙교)를 중심으로 -)

  • Purpose: Currently, the only routes that enter Yeongjong Island are Yeongjong Bridge and Incheon Bridge, which are private roads. The purpose of this study is to predict and study changes in transportation demand for new routes and two existing routes according to the plan to open the 3rd Bridge, a new route, in December 2025. Method: The basic data for traffic demand forecast were O/D and NETWORK data from 2021.08, KOTI. In order to examine the reliable impact of Yeongjong Bridge and Incheon Bridge on the opening of the 3rd Bridge, it is necessary to correct the traffic distribution of Yeongjong Island and Incheon International Airport to suit reality, and in this study, the trip distribution by region was corrected and applied using Mobile Big Data. Result: As of 2026, the scheduled year of the opening of the 3rd Bridge, two alternatives, Alternative 1 (2,000 won) and Alternative 2 (4,000 won), were established and future transportation demand analysis was conducted, In the case of Alternative 1, which is similar to the existing private road toll restructuring, the traffic volume of the 3rd Bridge was predicted to be 42,836 out of 199,101 veh/day in the Yeongjong area in 2026, and the traffic volume reduction rate of the existing road was analyzed as 21.5%. Conlclusion: As a result of the review (based on Alternative 1), the proportion of convertted traffic on the 3rd Yanji Bridge was estimated to be 70% of Yeongjong Bridge and 30% of Incheon Bridge, and 21.5% of the predicted traffic reduction on the existing road when the 3rd Yanji Bridge was opened is considered appropriate considering the results of the case review and changes in conditions. It is judged that it is a way to secure the reliability of the prediction of traffic demand because communication big data is used to reflect more realistic traffic distribution when predicting future traffic demand.
