Molecular Identification and First Morphological Description of Larvae and Juveniles of Neosalanx anderssoni (Salangidae) Collected from the Southwestern Sea of Korea (한국 서해 남부해역에서 채집된 도화뱅어, Neosalanx anderssoni (뱅어과) 자치어의 분자 동정 및 첫 형태기재)

  • During ichthyoplankton survey in the southwestern sea of Korea, we collected six individuals of noodlefish larvae and juveniles between April and May 2023. They were identified as Neosalanx anderssoni by mitochondrial DNA cytochrome c oxidase subunit I or 16S ribosomal RNA sequences, and their external morphological traits were described for the first time. All six individuals have a slender and elongated body. When preflexion and flexion larval stages (10.24 mm notochord length, NL and 15.47 mm total length, TL, respectively), oval-shaped black melanophores were distributed in a row along the ventral side of the gut. However, when postflexion larval and juvenile stages (23.58~25.90 mm TL, and 29.20~31.26 mm TL, respectively), melanophores on the ventral side of the gut were disappeared, and dark spot-shaped melanophores appeared along the dorsal side of the gut in a single row. Also, from the postflexion larval stage (23.58 mm TL), two large black spots began to appear symmetrically on the caudal fin. Our results suggest that N. anderssoni may use coastal area as spawning and/or nursery ground unlike previous study (Kim and Park, 2002).

Isolation and Nitrate Reduction Characteristics of Aerobic Denitrifier Pseudomonas sp. DN-9 (호기성 탈질균 Pseudomonas sp. DN-9의 분리 및 질산염 환원 특성)

  • From sludge of S municipal wastewater treatment plant in Busan, Korea, we isolated the denitrifier DN-9 which showed the ability of denitrification under aerobic conditionby the color change and gas formation in liquid culture with Giltay medium. The isolated strain was identified as Pseudomonas sp. DN-9 on the basis of the morphological, physiological, biochemical and nucleotide sequence analysis of l6S rRNA. The isolated strain, Pseudomonas sp. DN-9, has cytochrome $cd_1$, nirS of nitrite reductase. By the co-existance of additional ammonium and nitrate ion, the strain was not affected largely on growth in SL series broth. It seemed the result of denitrification. Although Pseudomonas sp. DN-9 has a good nitrate reduction activity under aerobic condition, the activity is less than Pseudomonas stutzeri in same cultivation condition. However, Escherichia coli had little the activity of aerobic denitrification and Pseudomonas putida showed lower activity of aerobic denitrification than Pseudomonas sp. DN-9 and Pseudomonas stutzeri in this study.

Effects of the Cell-to-Cell Communication between Oocyte and Cumulus Cells on the Quality of Oocytes (난자와 난구세포간 Cell-to-Cell Communication이 난자에 미치는 영향)

  • Production of a mature oocyte is a complex process that requires the close association between oocyte and follicular cells. The present study was conducted to investigate the difference between oocytes with and without close junctional communication with cumulus cells and the involvement of two connexins(CXs) in the interactions. Follicles at different sizes(small: 200~400 ㎛; large:>450 ㎛) were mechanically isolated from PMSG-primed mouse ovaries, and punctured to get cumulus-oocyte complex(COC). Oocytes were released themselves(denuded), with partially attached(partial), or with tightly attached(intact) cumulus cells. Maturation and fertilization capacity of the COCs were measured. Expression of CX 37 and CX 43 was examined by RT-PCR and in situ hybridization. The ratio between intact COC and denude/partial oocytes was 30%(SI) and 70%(SPD) in small follicles, while 55%(LI) and 45%(LPD) in large follicles, respectively. Maturation and fertilization rates of the released oocytes were similar among SI, LPD, LI groups, but those were significantly lower in SPD oocytes. In oocytes, CX 37 was the major CX and CX 43 was not expressed, whereas in the cumulus cells, CX 43 was the major, and CX 37 was the next. Results of the present study suggest that 1) immature oocytes from small follicles with intact cell-to-cell communication with cumulus have the similar quality to that of the oocytes from larger follicles, 2) gap junction between oocyte and cumulus cells may be the heterotypic channel, and 3) we could not explain the difference in the cell-to-cell communication between intact and partial/denuded COCs through the expression of the two CXs.
