Involvement of ERK1/2 and JNK Pathways in 17${\beta}-estradiol$ Induced Kir6.2 and SK2 Upregulation in Rat Osteoblast-like Cells

  • The functional expression of potassium $(K^+)$ channels has electrophysiologically been studied in bone cells from several species, however, their identity and regulation of gene expressions in bone cells are not well known. In the present study, to investigate how $K^+$ channel expressions are regulated by estrogen, we measured changes of transcript levels of various $Ca^{2+}$-activated ($K_{Ca}$) and ATP-sensitive $K^+$ channels in rat osteoblastic ROS 17/2.8 cells after treatment with estrogen. Application of 17${\beta}$-estradiol $(E_2)$ for 24 h and 48 h increased mRNA and protein expressions of inwardly rectifying $K^+$ channel (Kir) 6.2 and type 2 small conductance $K_{Ca}$ channel (SK2), respectively. Combined treatment of cells with 17${\beta}-E_2$ and ICI 182,780, a pure antiestrogen, suppressed 17${\beta}-E_2$-induced alterations of SK2 and Kir6.2 mRNA levels. In addition, treatment of cells with U0126, a specific inhibitor of extracellular receptor kinases (ERK)1/2, and SP600125, a specific inhibitor of c-jun N-terminal kinase (JNK) blocked the enhancing effects of 17${\beta}-E_2$ on SK2 and Kir6.2 protein expressions. On the other hand, blocking of p38 mitogen-activated protein kinase had no effect. Taken together, these results indicate that 17${\beta}-E_2$ modulates SK2 and Kir6.2 expressions through the estrogen receptor, involving ERK1/2 and JNK activations.

The Characterization of the Increase of Membrane Conductance after Depolarization in Single Rat Adrenal Chromaffin Cells

  • The conductance change evoked by step depolarization was studied in primarily cultured rat adrenal chromaffin cells using patch-clamp and capacitance measurement techniques. When we applied a depolarizing pulse to a chromaffin cell, the inward calcium current was followed by an outward current and depolarization-induced exocytosis was accompanied by an increase in conductance trace. The slow inward tail current which has the same time course as the conductance change was observed in current recording. The activation of slow tail current was calcium-dependent. Reversal potentials agreed with Nernst equation assuming relative permeability of $Cs^+\;to\;K^+$ is 0.095. The outward current and tail current were blocked by apamin (200 nM) and d-tubocurarine (2 mM). The conductance change was blocked by apamin and did not affect membrane capacitance recording. We confirmed that conductance change after depolarization comes from the activation of the SK channel and can be blocked by application of the SK channel blockers. Consequently, it is necessary to consider blocking of the SK channel during membrane capacitance recording.

Possible Involvement of $Ca^{2+}$ Activated $K^+$ Channels, SK Channel, in the Quercetin-Induced Vasodilatation

  • Effects of quercetin, a kind of flavonoids, on the vasodilating actions were investigated. Among the mechanisms for quercetin-induced vasodilatation in rat aorta, the involvement with the $Ca^{2+}$ activated $K^+$ ($K_{Ca}$) channel was examined. Pretreatment with NE ($5\;{\mu}M$) or KCl (60 mM) was carried out and then, the modulation by quercetin of the constriction was examined using rat aorta ring strips (3 mm) at $36.5^{\circ}C$. Quercetin (0.1 to $100\;{\mu}M$) relaxed the NE-induced vasoconstrictions in a concentrationdependent manner. NO synthesis (NOS) inhibitor, NG-monomethyl-L-arginine acetate (L-NMMA), at $100\;{\mu}M$ reduced the quercetin ($100\;{\mu}M$)-induced vasodilatation from $97.8{\pm}3.7%$ (n=10) to $78.0{\pm}11.6%$ (n=5, p<0.05). Another NOS inhibitor, L-NG-nitro arginine methyl ester (L-NAME), at $10\;{\mu}M$ also had the similar effect. In the presence of both $100\;{\mu}M$ L-NMMA and $10\;{\mu}M$ indomethacin, the quercetin-induced vasodilatation was further attenuated by $100\;{\mu}M$ tetraethylammonium (TEA, a $K_{Ca}$ channel inhibitor). Also TEA decreased the quercetin-induced vasodilatation in endothelium-denuded rat aorta. Used other $K_{Ca}$ channel inhibitors, the quercetin-induced vasodilatation was attenuated by $0.3\;{\mu}M$ apamin (a SK channel inhibitor), but not by 30 nM charybdotoxin (a BK and IK channel inhibitor). Quercetin caused a concentration-dependent vasodilatation, due to the endotheliumdependent and -independent actions. Also quercetin contributes to the vasodilatation selectively with SK channel on smooth muscle.

Mechanical Hyperalgesia Induced by Blocking Calcium-activated Potassium Channels on Capsaicin-sensitive Afferent Fiber

  • Small and large conductance $Ca^{2+}$-activated $K^+(SK_{Ca}\;and\;BK_{Ca})$ channels are implicated in the modulation of neuronal excitability. We investigated how changes in peripheral $K_{Ca}$ channel activity affect mechanical sensitivity as well as the afferent fiber type responsible for $K_{Ca}$ channel-induced mechanical sensitivity. Blockade of $SK_{Ca}$ and $BK_{Ca}$ channels induced a sustained decrease of mechanical threshold which was significantly attenuated by topical application of capsaicin onto afferent fiber and intraplantar injection of 1-ethyl-2-benzimidazolinone. NS1619 selectively attenuated the decrease of mechanical threshold induced by charybdotoxin, but not by apamin. Spontaneous flinching and paw thickness were not significantly different after $K_{Ca}$ channel blockade. These results suggest that mechanical sensitivity can be modulated by $K_{Ca}$ channels on capsaicin-sensitive afferent fibers.

Electrochemical Control of Metabolic Flux of Weissella kimchii sk10: Neutral Red Immobilized in Cytoplasmic Membrane as Electron Channel

  • Electrochemical control of the metabolic flux of W. kimchii sk10 on glucose and pyruvate was studied. The growing cell of W. kimchii sk10 produced 87.4 mM lactate, 69.3 mM ethanol, and 4.9mM lactate from 83.1mM glucose under oxidation condition of the anode compartment, but 98.9 mM lactate, 84.3mM ethanol, and 0.2 mM acetate were produced from 90.8 mM glucose under reduction condition of the cathode compartment for 24 h, respectively. The resting cell of W. kimchii sk10 produced 15.9 mM lactate and 15.2 mM acetate from 32.1 mM pyruvate under oxidation condition of the anode compartment, and 71.3 mM lactate and 3.8 mM acetate from 79.8mM pyruvate under reduction condition of the cathode compartment. The redox balance (NADH/$NAD^+$) of metabolites electrochemically produced from pyruvate was 1.05 and 18.76 under oxidation and reduction conditions, respectively. On the basis of these results, we suggest that the neutral red (NR) immobilized in bacterial membrane can function as an electron channel for the electron transfer between electrode and cytoplasm without dissipation of membrane potential, and that the bacterial fermentation of W. kimchii sk10 can be shifted to oxidized or reduced pathways by the electrochemical oxidation or reduction, respectively.

Differential Functional Expression of Clotrimazole-sensitive $Ca^{2+}$-activated $K^+$ Current in Bal-17 and WEHI-231 Murine B Lymphocytes

  • The intermediate conductance $Ca^{2+}-activated$ $K^+$ channels (SK4, IKCa1) are present in lymphocytes, and their membrane expression is upregulated by various immunological stimuli. In this study, the activity of SK4 was compared between Bal-17 and WEHI-231 cell lines which represent mature and immature stages of murine B lymphocytes, respectively. The whole-cell patch clamp with high-$Ca^{2+}$ ($0.8{\mu}M$) KCl pipette solution revealed a voltage-independent $K^+$ current that was blocked by clotrimazole (1 mM), an SK4 blocker. The expression of mRNAs for SK4 was confirmed in both Bal-17 and WEHI-231 cells. The density of clotrimazole-sensitive SK4 current was significantly larger in Bal-17 than WEHI-231 cells ($-11.4{\pm}3.1$ Vs. $-5.7{\pm}1.15$ pA/pF). Also, the chronic stimulation of B cell receptors (BCR) by BCR-ligation (anti-IgM Ab, $3{\mu}g$/ml, 8∼12 h) significantly upregulated the amplitude of clotrimazolesensitive current from $-11.4{\pm}3.1$ to $-53.1{\pm}8.6$ pA/pF in Bal-17 cells. In WEHI-231 cells, the effect of BCR-ligation was significantly small ($-5.7{\pm}1.15$ to $-9.0{\pm}1.00$ pA/pF). The differential expression and regulation by BCR-ligation might reflect functional changes in the maturation of B lymphocytes.

Performance of a DS/CDMA Packet Network in an Indoor Wireless Infrared Channel

  • In this paper, performance of a CDMA-based packet transmission system is analyzed and simulated in a wireless infrared channel. The indoor wireless infrared channel is modeled as a non-directed diffuse link. The pulse position modulation (PPM) is used as a modulation scheme. From the simulation results, it is shown that the RS coding is very effective in improving the packet-delay characteristics of the CDMA-based packet transmission system. The performance improvement is more significant especially when the infrared channel is in a worse condition.

Numerical Simulation of the Flood Event Induced Temporally and Spatially Concentrated Rainfall - On August 17, 2017, the Flood Event of Cheonggyecheon (시공간적으로 편중된 강우에 의한 홍수사상 수치모의 - 2017년 8월 17일 청계천 홍수사상을 대상으로)

  • This study identifies the cause of the accident and presents a new concept for safe urban stream management by numerical simulating the flood event of Cheonggyecheon on August 17, 2017, using rain data measured through a dense weather observation network. In order to simulate water retention in the CSO channel listed as one of the causes of the accident, a reliable urban runoff model(XP-SWMM) was used which can simulate various channel conditions. Rainfall data measured through SK Techx using SK Telecom's cell phone station was used as rain data to simulate the event. The results of numerical simulations show that rainfall measured through AWSs of Korea Meteorological Administration did not cause an accident, but a similar accident occurred under conditions of rainfall measured in SK Techx, which could be estimated more similar to actual phenomena due to high spatial density. This means that the low spatial density rainfall data of AWSs cannot predict the actual phenomenon occurring in Cheonggyecheon and safe river management needs high spatial density weather stations. Also, the results of numerical simulation show that the residual water in the CSO channel directly contributed to the accident.

Performance of a Multicode DS/CDMA Packet Radio Network with Adaptive Antenna Array

  • In this paper, performance of multicode DS/CDMA packet radio network with adaptive antenna array is analyzed and simulated in a multipath Rayleigh fading channel. The network performance is evaluated in terms of packet throughput. From the simulation results, it is confirmed that the adaptive antenna array is very effective in improving the packet throughput performance of the multicode DS/CDMA packet radio network. The results of this paper can be applied to design of IMT-2000 or future-generation mobile communication systems.

  • The multi-channel analog signal to digital signal conversion is increasing in the field of IoT and medical measurement equipments. It has chip area and power consumption constraints to use a few single or 2_channel ADC for multi_channel application. This paper described to design and implement a proposed comb filter for multi-channel, 24bit ADC. The function of proposed comb filter is verified by matlab simulation and the FPGA test board. It was fabricated using SK Hynix 0.35㎛ CMOS standard process. The performance and chip size is compared with the existing design method that uses integrator/differentiator and FIR construction. The proposed comb filter is expected to use the IoT product and medical measurement equipments that require multi-channel, low power consumption and small hardware size.
