Effect of Medicinal Herbs' Extracts on the Growth of Lactic acid bacteria isolated from Kimchi and Fermentation of Kimchi (한약재 추출물이 김치 관련 유산균의 성장과 김치의 숙성에 미치는 효과)

  • This studies were carried out to investigate antimicrobial activity of 35 kinds of medicinal herbs against lactic acid bacteria isolated from home-made Kimchi. The most isolated lactic acid bacteria were inhibited by ethanol extract of various medicinal herbs such as Schizandra chinensis (SC), Salvia miltiorrhiza (SM), Glycyrrhiza uralensis (GU), Lithospermum erythrorhizon (LE) and Sophrora flavescens AITON (SF). But Cnidium monnier, Pinus densiflora, Paeonia suffruticosa, Acanthopanax sessiliflorum and Rhus chinensis inhibited the growth of only a few isolated lactic acid bacteria. The pH of Kimchi containing 1% of medicinal herbs extracts such as SM, GU, LE and SF was higher than that of control during fermentation for 25 days at $10^{\circ}C$, respectively. Titratable acidity and viable cells of total bacteria and lactic acid bacteria of the Kimchi were lower than that of control during fermentation. The sensory quality (taste, flavor and overall acceptability) of SM, GU, and LE added Kimchi was similar to that of control at 10th day of fermentation. But Sophrora flavescens AITON added Kimchi decreased significantly its sensory quality compared with control(P<0.05).

Effect of retinoic acid on the radiosensitivity of normal human oral keratinocyte (Retinoic acid가 사람 정상 구강각화세포의 방사선감수성에 미치는 영향에 관한 연구)

  • Purpose : To evaluate the effect of all-trans-retinoic acid (ATRA) on the radiosensitivity of normal human oral keratinocyte (NHOK). Materials and methods: Relative cell survival fraction including SF2 (survival fraction at 2 Gy) was calculated on the basis of colony formation assay. Data were fitted to the linear-quadratic model to establish the survival curve and calculate α and β values. Using flow cytometry at 1, 2, 3, 4, and 5 days after exposure to 2 and 10 Gy irradiation, cell cycle arrest and apoptosis were analysed. To understand the molecular mechanism of the radiosensitization of ATRA on NHOK, proteins related with apoptosis and cell cycle arrest were investigated by Western blot analysis. Results: Treatment with ATRA resulted in a significant decrease of SF2 value for NHOK from 0.63 to 0.27, and increased α and β value, indicating that ATRA increased radiosensitivity of NHOK. ATRA increased LDH significantly, but increasing irradiation dose decreased LDH, suggesting that the radiosensitizing effect of ATRA is not directly related with increasing cell necrosis by ATRA. ATRA did not induce appotosis but increased G2 arrest after 10 Gy irradiation, implying that the increased radiosensitivity of NHOK may be due to a decrease in mitosis casued by increasing G2 arrest. ATRA inhibited the reduction of p53 at 3 days after l0Gy irradiation and increased p21 at 1 day after 10 Gy irradiation. Further study is required to determine the precise relationship between this effect and the radiosensitizing effect of A TRA. Conclusion: These results suggested that ATRA increase radiosensitivity by inhibiting mitosis caused by increasing G2 arrest.

  • This study investigates convergent influence on Self Factors(SF), Stress Factors(STF), Resilience & Control Factors(RCF), Test Anxiety(TA), Learning Flow(LF) and academic burnout among Health College Students in TOEIC class(HCST). The survey was administered to 291 HCST from 1 college located in J area during the period from April 3, 2017 to April 14, 2017. The structured self-administered questionaries were used. With the analysis of covariance structure, we could confirm relationship among the six factors such as SF, STF, RCF, TA, LF and academic burnout. The results of the study indicate that the efforts, to manage these factors, are required to decrease the academic burnout of HCST. Squared multiple correlations, which explain the academic burnout related with the stress from economic pressure and job seeking, test anxiety, learning flow and self factors, were 98.8%. The results are expected to be useful for the development of TOEIC learning curriculum and course to decrease the academic burnout of HCST. In the following study, the analysis about additional factors of influence on academic burnout will be needed.

Preventive effect of fermented red ginseng on cisplatin-induced nephrotoxicity mouse (Cisplatin으로 유도된 신손상 마우스 모델에 대한 발효홍삼의 예방효능)

  • Red ginseng is known to have many beneficial effects. Cisplatin, an effective antineoplastic drug, can cause many side effects like irreversible sensorineural hearing loss and serious tinnitus in humans. This study is aimed to reduce a cisplatin's side effect, nephrotoxicity by fermentated korean red ginseng. Korea ginseng was produced by steaming and dring and fermentation. And mice were divided into 4 groups- (A) normal mice, (B) Vehicle treated cisplatin mice, (C) RG0F0-treated cisplatin mice, (D) RG8F3-treated cisplatin mice. C and D groups were feed each material 200 mg/kg/day during 4 days. And cisplatin 20 mg/kg injected to B, C, and D groups as abdominal injection. After 24 h, blood sample was collected. The kidneys were harvested for histological, immuno histochemical and western blot analysis. 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging activity was depended on steaming hours. RG0F0 and RG8F3 (ginseng-8 h steamed and fermented by Saccharomyces cerevisiae) were showed antioxidants effect in DPPH and ABTS radical scavenging activity. Component amounts according to steaming hours. 8 h steamed red ginseng had the most ingredients of ginsenoside. Treatments with RG8F3 reduced cisplatin-induced nephrotoxicity in the mice resulting in increase of GSH and decrease of ROS, BUN, creatinine, and inflammatory mediators. This result seems to be involved with the restriction of the inflammation in the kidney. Therefore, fermented red ginseng might have therapeutic efficacy in reduce kidney injury induced by cisplatin treatment.

Assessment of the possibility as Cosmetics Materials by Brown algae from Jeju Island Using Supercritical Fluid System (초임계 유체 기술을 이용한 제주산 갈조류의 화장품 소재로서의 가능성 평가)

  • Although various biological extracts derived from the resources from Jeju are currently being developed for cosmetic materials, few studies using Brown algae from Jeju have been conducted. This study identified materials with high antioxidant levels and examined their whitening effects to assess their feasibility as potential cosmetic resources. Brown algae from Jeju (Undaria pinnatifida (Harvey) Suringar, Laminaria japonica, Ecklonia cava, Hizikia fusiforme (Harvey) Okamura (HF), and Sargassum fulvellum (Turner) C. Agardh (SF) were processed using a supercritical fluid system at 50℃ and 400 bar, and their total phenol content, antioxidant activity, tyrosinase inhibition, and primary skin irritation were measured. Of all the samples, only SF using the supercritical fluid system (SFS) displayed concentration-dependent behavior for the total phenol content (p<0.05). Among the samples, the antioxidant activity of SFS was significantly higher (p<0.05) and the one increased at higher concentrations (p<0.05). On the other hand, tyrosinase was inhibited to a greater extent with HF than the others. The primary skin irritation tests showed that none of the samples were skin irritants. This study confirmed the feasibility of using Phaeophyceae from Jeju as potential cosmetic materials, where the samples can be selected and utilized based on the required physicochemical properties.

Association between nutritional status, sarcopenia, and frailty in rural elders (농촌 지역 노인의 영양 상태, 근감소증과, 노쇠의 연관성)

  • Objectives: Frailty and sarcopenia are recent important concepts in elder health care. Sarcopenia is the most important factor influencing frailty, and exercise and nutritional status are known to affect sarcopenia. The purpose of this study was to identify the relationship between nutritional status, sarcopenia, and frailty. Methods: This study was a cross-sectional design. The subjects of this study were 411 elderly people aged 65 or older from 10 villages in Gyeongnam. The survey tools were the K-FRAIL for frailty, the GDS-SF for depression, the SARC-F questionnaire for sarcopenia, and the DETERMINE for nutritional status. Frequency analysis, the chi-square test, and multiple logistic regression analysis were performed using the SPSS 25.0 program. Results: As a result of the chi-square test, there was a significant difference in the nutritional status and the frailty proportion (p<0.001), and there was a significant difference between frailty and suspected sarcopenia (p<0.001). After adjustment, nutritional status was significantly associated with sarcopenia (OR=2.946, p<0.001). In addition, nutritional status was significantly associated with frailty (OR=2.958, p<0.001), and sarcopenia also had a significant effect on frailty (OR=5.898, p<0.001). Finally, even after including sarcopenia, nutritional status had a significant effect on frailty (OR=2.246, p=0.002). Conclusions: Nutritional status can have both a direct effect on frailty and an indirect effect through sarcopenia, and it was found that sarcopenia also affects frailty. Therefore, it is necessary to evaluate sarcopenia and nutritional status and to evaluate their levels in the elderly and to take appropriate interventions.

Relationship Analysis between the Box Office Performance and Sentimental Words in Movie Review (영화의 흥행 성과와 리뷰 감정어휘와의 관계 분석)

  • This study aims to understand distribution of the sentimental words on each genre and find relationship between box office performance and sentimental words in movie review using 673 movies that have more than 1,000 reviews. For the analysis, crawling movie reviews and made data was composed movie genre, movie name, sales, attendance, screen, normal attendance, 7 sentimental words. For analysis results, we used correlation analysis and Parallel coordinates. As a results, First, the highest box office value of the genre is comedy and the lowest box office value of the genre is horror through analyze box office on each genre. Secondly, Movie genre of fantasy feel a lot of boring emotion and Movie genre of SF feel a lot of anger emotion even if 'Happy' and 'Surprise' have highest sentiment value on every genre. Third, We found 'Anger' increase sentimental value when 'Disgust' increase sentimental value and 'Surprise' decrease sentimental value when 'Happy' increase sentimental value through analyze correlation relationship between sentimental words using total data. Fourth, We found 'Happy' have linear relationship between box office and 'Fear' have non-linear relationship between box office through analyze sentimental words according to box office performance.

The Content and Risk Assessment of Heavy Metals in Herbal Pills (유통 환제의 유해 중금속 함량 및 위해도 평가)

  • The objective of this study is investigation of contamination levels and assessment of health risk effects of heavy metals in herbal pills. 31 Items and 93 samples were obtained for this investigation from major herbal medicine producing areas, herbal markets and on-line supermarkets from Jan to Jun in 2010. Inductively coupled plasma mass spectrometer method was conducted for the quantitative analysis of Pb, Cd and As. In addition, the mercury analyzer system was conducted for that of Hg without sample digestion. The average contents of heavy metals in samples were as follows : 0.87 mg/kg for Pb, 0.08 mg/kg for Cd, 2.87 mg/kg for As and 0.16 mg/kg for Hg, respectively. In addition, the average contents of heavy metals in different parts of plants, including cortex, fructus, herba, radix, seed, algae and others were 0.63 mg/kg, 3.94 mg/kg, 1.42 mg/kg, 1.05 mg/kg, 0.16 mg/kg, 22.31 mg/kg and 10.17 mg/kg, respectively. After the estimations of dietary exposure, the acceptable daily intake (ADI), the average daily dose (ADD), the provisional tolerable weekly intake (PTWI) and the relative hazard of heavy metals were evaluated. As the results, the relative hazards compared to PTWI in samples were below the recommended standard of JECFA as Pb 3.1%, Cd 0.9%, Hg 0.5%. Cancer risks through slope factor (SF) by Ministry of Environment Republic Korea and Environmental Protection Agency was $4.24{\times}10^{-7}$ for Pb and $3.38{\times}10^{-4}$ for As (assuming that the total arsenic content was equal to the inorganic arsenic). Based on our results, possible Pb-induced cancer risks in herbal pills according to parts used including cortex, fructus, herba, radix, seed, algae and others were $1.95{\times}10^{-7}$, $1.45{\times}10^{-6}$, $2.14{\times}10^{-7}$, $6.27{\times}10^{-7}$, $1.99{\times}10^{-8}$, $3.61{\times}10^{-7}$ and $9.64{\times}10^{-8}$, respectively. Possible As-induced cancer risks in herbal pills by parts used including cortex, fructus, herba, radix, seed, algae and others were $1.54{\times}10^{-5}$, $7.24{\times}10^{-5}$, $1.23{\times}10^{-4}$, $2.02{\times}10^{-5}$, $3.25{\times}10^{-6}$, $2.18{\times}10^{-3}$ and $5.67{\times}10^{-6}$ respectively. Taken together, these results indicate that the majority of samples except for some samples with relative high contents of heavy metals were safe.

Mid-IR Luminosity Functions of Local Galaxies in the North Ecliptic Pole Field

  • We present the mid-infrared (MIR) luminosity function (LF) of local (z < 0.3) star forming (SF) galaxies based on the AKARI's NEP-Wide Survey data. We utilized a combination of the NEP-Wide point source catalogue containing a large number (114,000) of infrared (IR) sources distributed over the wide (5.4 sq. deg) field and spectroscopic redshift (z) data for 1790 selected targets obtained by optical follow-up surveys with MMT/Hectospec and WIYN/Hydra. The AKARI's continuous $2{\sim}24{\mu}m$ wavelength coverage and the spectroscopic redshifts for sample galaxies enable us to derive accurate spectral energy distributions (SEDs) in the mid-infrared. We carried out SED-fit analysis and employed 1/Vmax method to derive the mid-IR (e.g., $8{\mu}m$, $12{\mu}m$, and $15{\mu}m$ rest-frame) luminosity functions. Our results for local galaxies from the NEP region generally consistent with various previous works for other fields over wide luminosity ranges. The comparison with the results of the NEP-Deep data implies the luminosity evolution from higher redshifts towards the present epoch. We attempted to fit our derived LFs to the double power-laws and present the resulting power indices. We also examined the correlation between mid-IR luminosity and total IR luminosity.

Isolation and Structure Determination of Metabolites from Cultures of Aspergillus protuberus (Aspergillus protuberus 배양물로부터 대사체 분리 및 구조 결정)

  • Marine-derived microbes have yielded a variety of metabolites so far. In the course of the project to find metabolites from marine microbes, an isolate of Aspergillus protuberus (SF 5767) was selected for chemical investigation. A large scale culture of this strain in PDA media was extracted with an organic solvent and the extract was fractionated by silica gel column chromatography. Repeated reverse phase HPLC of the fractions led to the isolation of three metabolites. Their chemical structures were elucidated as deoxybrevianamide E (1), brevianamide V (2), and ergosterol peroxide (3) on the basis of spectroscopic data including MS, NMR, and UV. To the best of our knowledge, chemical investigation of A. protuberus was conducted for the first time in this study.
