Anti-hyperlipidemic Effect of Vitex doniana Ethanol Extract in Poloxamer Induced Hyperlipidemia

  • To determine the effect of Vitex doniana (leaves stem and root bark) ethanolic extracts on lipid profiles of Poloxamer 407 (P407) induced hyperlipidemic and normal rats. Fifty four mixed sex rats weighing 100-200g were divided into nine groups comprising six animals per group. At the end of the 21 day, the animals were sacrificed and blood sample were collected for determination of serum levels of: Total cholesterol (TC), Triacylglycerides (TAG), High-density lipoprotein cholesterol (HDL-c) and Low-density lipoprotein cholesterol (LDL-c). The studies showed that all induced treated groups significantly (P<0.05) lower serum levels of TC, TAG, LDL-c and significantly (P<0.05) increased HDL-c when compared to the P407 induced hyperlipidemic control. The normal treated groups showed no significant (P>0.05) difference in the serum levels of TC, TAG, LDL and HDL when compared to the normal control group. Calculation of atherogenic risk predictor indices of the induced treated groups showed that all the extracts significantly (P<0.05) lowered the LDL-c/HDL-c, log (TAG/HDL-c) and significantly (P<0.05) increased HDL-c/TC ratio when compared to the P407 induced hyperlipidemic control group. The atherogenic risk predictor indices of normal treated groups showed no significant difference (P>0.05) in LDL-c/HDL-c, Log (TAG/HDL-c) and HDL-c/TC ratio when compared to the normal control group. The study demonstrates the phytotherapeutic effect of Vitex doniana (leaves, stem and root bark) ethanolic extract in poloxamer 407 induced hyperlipidemia.

Study on the Eruption Age of the First Molar (제1대구치의 맹출 연령에 관한 분석 연구)

  • This study was undertaken to obtain the data age determination following the eruption of individual cusps of the first molars in the point of forensic odontology. 532 children (294 male and 238 female, born between April, 1989 and March, 1986) from a kindergarten and a primary school in a reesidential district in Seoul were studied. The eruption state of the first molar was divides into 6 stages according to the degree of exposure of individual cusps, and correlation between the stage and age was stastically analysed. The results of the study lead to following conclusions : 1. The most frequently seen eruption sequence of cusps for the upper first molar was mesibuccal cusp, mesiopalatal cusp, distobuccal cusp, mesiodistal cusp, distopalatal cusp. As for the lower first molar : mesiobuccal cusp, mcsiolingual cusp, distobuccal cusp, distolingual cusp. 2. The time of eruption was earlier in the female ethan in the male, but it was stastically remarkable only in upper and lower parts of the right and the left teeth of subjects age between 6.0 and 6.5 and upper part of the right and the left teeth of subjects age over 7.5 3. The eruption of the lower first molar was comparatively earlier than that of the upper first molar and there was no significant stastical difference between the right and the left first molars in the time of eruption. 4. There was no noticeable difference in the eruption sequence of cusps, irrespective of sex and side. 5. The eruption of the upper first molar is started at the age of about 6.4 and complete the age of about 7.1 and as for the lower first molar, it is from 6.3 to 7.0

  • Purpose: This study investigated the possible correlation between the mineralization of the second molars and the chronological age of a sample population of the state of Paraiba, Brazil. Materials and Methods: One thousand eight hundred fifty-four digitized panoramic images using a scanner of a private dental radiology service were obtained in six months. Of these, 457 were selected from patients aged 4.6 to 16 years who met certain criteria. The images were assessed twice by a radiologist with 5 years experience. A minimum interval of 30 days between the evaluations of the same patient was established. To analyze the relationship between chronological age, calcification level proposed by Demirjian et al in 1973, gender, and tooth, a multiple linear regression model was adjusted, taking age as the response variable (p<0.05). Results: The gender and calcification level were significantly correlated with the age of the patients (p<0.05). There was a significant difference between the average age of the patients' upper teeth compared to the lower ones in both genders (p<0.05). The dental development occurred earlier in female individuals than in male ones, and there was no significant difference between the right and left second permanent molars. Conclusion: It was observed that ethnic variables are related to certain parameters of age and sex identification in the Brazilian population, providing important information for forensic evaluations.

Accuracy of an equation for estimating age from mandibular third molar development in a Thai population

  • Purpose: This study assessed the accuracy of age estimates produced by a regression equation derived from lower third molar development in a Thai population. Materials and Methods: The first part of this study relied on measurements taken from panoramic radiographs of 614 Thai patients aged from 9 to 20. The stage of lower left and right third molar development was observed in each radiograph and a modified Gat score was assigned. Linear regression on this data produced the following equation: Y=9.309+1.673 mG+0.303S (Y=age; mG=modified Gat score; S=sex). In the second part of this study, the predictive accuracy of this equation was evaluated using data from a second set of panoramic radiographs (539 Thai subjects, 9 to 24 years old). Each subject's age was estimated using the above equation and compared against age calculated from a provided date of birth. Estimated and known age data were analyzed using the Pearson correlation coefficient and descriptive statistics. Results: Ages estimated from lower left and lower right third molar development stage were significantly correlated with the known ages (r=0.818, 0.808, respectively, $P{\leq}0.01$). 50% of age estimates in the second part of the study fell within a range of error of ${\pm}1year$, while 75% fell within a range of error of ${\pm}2years$. The study found that the equation tends to estimate age accurately when individuals are 9 to 20 years of age. Conclusion: The equation can be used for age estimation for Thai populations when the individuals are 9 to 20 years of age.

A Study on Diphtheria Antibody Titer of Residents in Seoul Area (서울 지역에 거주하는 일부 주민들의 디프테리아 항체가에 관한 연구)

  • Diphtheria was the world-spread, respiratory infectious disease in the past. But after developing the diphtheria toxoid, diphtheria vaccine, composed of diphtheria toxoid, is generalized, so prevalence of diphtheria is sharply dropped. But diphtheria is come back in NIS(New Independent States) in 1990s. It is believed that recurrence of diphtheria is mainly due to shortage of protective diphtheria antibody titer in adults. And it is related to 70% prevalence in adults, not in children. Considering this changing environment, this study was designed to research the diphtheria antibody titer of whole population and existence of titer difference by sex. Also it was studied whether ELISA has fitness as determination method of diphtheria antibody titer compared with neutralization method on microcell culture. This study was done to 277 samples of three hospitals in Seoul area, and sera was tested by neutralization method on microcell culture and ELISA method. The results of this study were as follows 1) Of this studied population, 31.05% had an antibody titer below the protective level (<0.01 IU/ml), 40.79% had a titer of a relative degree of protection (0.01-0.09 IU/ml) and 28.16% had a reliable degree of protection ($\geq$0.1 IU/ml). Therefore, 68.95% had a basic protective antibody titer level. 2) 20-40 age group showed the lowest diphtheria antibody titer among the studied population. 3) GMT of diphtheria antibody titer was the highest in the children. But, after child period, GMT was fallen. Standard deviation value was the lowest in older group. 4) Protective level of diphtheria antibody titer of male was slightly higher than female (70.94% vs 66.66%). But this difference was not statistically significant. 5) Diphtheria antibody titer by ELISA method was wholly higher than by neutralization method on microcell culture. Compared the two results by ELISA and neutralization, the regression coefficient was 0.38. And in titer, which was obtained by ELISA method, false-positive results was abundant.

  • Radioactive phosphorus(P-32) was injected to the chicken in the purpose of determination of the uptake and distribution, as related to sex and hour differences of the various organs of the body. $2{\mu}Ci$ of P-32 were injected to each chicken and the distribution of P-32 was observed at 1 hr, 6 hrs, 12 hrs, 24 hrs and 48 hrs after injection. In this experiment 34 heads of chicken were used(30 chicken for P-32, 4 chicken for control group) and the results obtained as follows: 1. The uptake of P-32 per gram of various organ in g. mm, femur(1 hr), liver, femur, tibia(24 hrs) and tibia(48 hrs) exhibited higher in the male than the female. 2. The uptake of P-32 per gram of various organ in heart, kidney, ovary(1 hr), kidney, brain(24 hrs) and kidney(48 hrs)exhibited higher in the female than the male. 3. The uptake ratio of brain, spleen, g. mm and tibia were increased gradually by the 12 hrs after injection of P-32, but decreased in liver, heart and kidney by the 24 hrs. 4. The uptake ratio of the femur was increased gradually by the 24 hrs, but testis and ovary was increased after 24 hrs. 5. The organs showed an uptake of P-32 per gram of various organ, with the following sequence : femur, tibia, testis or ovary, spleen, liver, kidney, heart, g. mm and brain.

  • Calcineurin is a $Ca^{2+}$/Calmodulin activated Ser/Thr phosphatase that is well conserved from yeast to human. It is composed of catalytic subunit A (CnA) and regulatory subunit B (CnB). C. elegans homolog of CnA and CnB has been annotated to tax-6 and cnb-1, respectively and in vivo function of both genes has been intensively studied. In C. elegans, calcineurin play roles in various signaling pathways such as fertility, movement, body size regulation and serotonin-mediated egg laying. In order to understand additional signaling pathway(s) in which calcineurin functions, we screened for binding proteins of TAX-6 and found a novel binding protein, HLH-11. The HLH-11, a member of basic helix-loop-helix (bHLH) proteins, is a putative counterpart of human AP4 transcription factor. Previously bHLH transcription factors have been implicated to regulate many developmental processes such as cell proliferation and differentiation, sex determination and myogenesis. However, the in vivo function of hlh-11 is largely unknown. Here, we show that hlh-11 is expressed in pharynx, intestine, nerve cords, anal depressor and vuvla muscles where calcineurin is also expressed. Mutant analyses reveal that hlh-11 may have role(s) in regulating body size and reproduction. More interestingly, genetic epistasis suggests that hlh-11 may function to regulate serotoninmediated egg laying at the downstream of tax-6.

Association of Thigh Muscle Mass with Insulin Resistance and Incident Type 2 Diabetes Mellitus in Japanese Americans

  • Background: Skeletal muscle plays a major role in glucose metabolism. We investigated the association between thigh muscle mass, insulin resistance, and incident type 2 diabetes mellitus (T2DM) risk. In addition, we examined the role of body mass index (BMI) as a potential effect modifier in this association. Methods: This prospective study included 399 Japanese Americans without diabetes (mean age 51.6 years) who at baseline had an estimation of thigh muscle mass by computed tomography and at baseline and after 10 years of follow-up a 75-g oral glucose tolerance test and determination of homeostasis model assessment of insulin resistance (HOMA-IR). We fit regression models to examine the association between thigh muscle area and incidence of T2DM and change in HOMA-IR, both measured over 10 years. Results: Thigh muscle area was inversely associated with future HOMA-IR after adjustment for age, sex, BMI, HOMA-IR, fasting plasma glucose, total abdominal fat area, and thigh subcutaneous fat area at baseline (P=0.033). The 10-year cumulative incidence of T2DM was 22.1%. A statistically significant interaction between thigh muscle area and BMI was observed, i.e., greater thigh muscle area was associated with lower risk of incident T2DM for subjects at lower levels of BMI, but this association diminished at higher BMI levels. Conclusion: Thigh muscle mass area was inversely associated with future insulin resistance. Greater thigh muscle area predicts a lower risk of incident T2DM for leaner Japanese Americans.

A Study of Growth and Age Structure for Chub Mackerel, Scomber japonicus Caught by a Large Purse Seine in the Korean Waters (한국 주변해역에서 대형선망으로 어획한 고등어(Scomber japonicus)의 성장과 연령구조 연구)

  • We examined the growth and age structure for chub mackerel caught by a large purseseine in the Korean waters from January to December 2015. A total of 459 specimens were used for otolith analysis, ranging from 19.6 cm to 46.0 cm in fork length. Translucent zone was regarded as an annual mark, and age was counted using the information of the number of translucent zone, capture date, edge type of the otolith and nominal birthdate of 1 January. Annuli in otoliths were mainly formed in May, coinciding with the spawning season. Estimated ages were 0~6 years, and the von Bertalanffy growth models were not significantly different between male and female. Sex-combined growth model was obtained as FLt=39.3×{1-exp[-0.90×(t+0.033)]}. Among the chub mackerel caught in 2015, the age 2 group had the highest proportion (30.9%), and the age 0 to 2 group accounted for 88.5% of the total catch.

Development of a Molecular Marker Linked to the A4 Locus and the Structure of HD Genes in Pleurotus eryngii

  • Allelic differences in A and B mating-type loci are a prerequisite for the progression of mating in the genus Pleurotus eryngii; thus, the crossing is hampered by this biological barrier in inbreeding. Molecular markers linked to mating types of P. eryngii KNR2312 were investigated with randomly amplified polymorphic DNA to enhance crossing efficiency. An A4-linked sequence was identified and used to find the adjacent genomic region with the entire motif of the A locus from a contig sequenced by PacBio. The sequence-characterized amplified region marker $7-2_{299}$ distinguished A4 mating-type monokaryons from KNR2312 and other strains. A BLAST search of flanked sequences revealed that the A4 locus had a general feature consisting of the putative HD1 and HD2 genes. Both putative HD transcription factors contain a homeodomain sequence and a nuclear localization sequence; however, valid dimerization motifs were found only in the HD1 protein. The ACAAT motif, which was reported to have relevance to sex determination, was found in the intergenic region. The SCAR marker could be applicable in the classification of mating types in the P. eryngii breeding program, and the A4 locus could be the basis for a multi-allele detection marker.
