Lactococcus lactis subsp. cremoris ATCC 11602-A1의 Bacteriophage 저항성 기작에 관한 연구 (Characteristics of the Bacteriophage Resistance Mechanism of Kactococcus lactis subsp.cremoris ATCC 11602-A1)

  • The characteristics of the bacteriophage resistant Lactococcus lactis subsp. cremoris ATCC 11602-A1, the phage-resistant mutant of Lactococcus lactis subsp. cremoris ATCC 11602, was examined. Electron microscopic study of phage adsorption to A1 revealed that after 10 min. incubation of the host-phage mixture, A1 did not show phage adsorption, and after 60 min. did not show a real burst and the release of new phage particles which could be detected in the mixture of its parent strain and phage. However, the phage adsorption rate of A1 after SDS treatment increased to 98%. Moreover, when the cell walls from A1 and parent strain, and the polysaccharide(PS) and peptidoglycan(PG) of their cell wall were mixed with phage and incubated for 15 min., PS and PG from A1 did not bind phage, but only SD-treated cell wall bound phage, and the cell wall and PS of parent strain bound phage. Both A1 and parent strain treated with 0.2 N HCl-and 5% TCA(100$$C) did not bind phage. The results suggest that the phage receptor is still present in the cell wall of the A1, but a cell wall constituent hydrolyzed by SDS blocks phage adsorption by masking the phage receptor. It also suggests that the phage receptor of parent strain is associated with PS of the cell wall.

  • 탄소나노튜브는 유연 전도체로서 투명전극, 유연 히터, 투명 스피커 분야에 활발히 응용되고 있다. 본 연구에서는 PET 유연 모재의 전면적에 다중벽 탄소나노튜브(MWCNT)를 도포하는 스프레이법 이용하여, 투명하고 전도성 있는 MWCNT/PET 복합 박막을 제조하였다. 분산제로 사용된 SDS (sodium dodecyl sulfate)의 농도를 조절함으로써 MWCNT 의 분산도를 조절하였으며, 분산도가 조절된 시험편의 인장실험을 통해 기계적인 변형 하에서 MWCNT/PET 전도성 박막의 전기-기계적인 거동과 분산제의 효과를 평가하였다.

Cloning of the gense coding for extracellular proteases from alkalophilic xanthomonas SP. JK311

  • The alkalophilic bacterium, Xanthomonas sp. JK311, producing extracellular proteases, was isolated from soil. Xanthomonas sp. JK311 produced five extracellular proteases that are all metalloproteases. Four of them were resistant against 1% SDS. Chromosomal DNA of the Xanthomonas sp. JK311 was digested with BamHI and cloned into PUC19. Among E. coli strain HB101 transformants, a clone secreting the proteases was screened through halo formation on skim-milk agar plate and by Southern blot analysis. It had the recombinant plasmid pXEP-1 containing the 7.5 kb-BamHI DNA fragment and produced three extacellular proteases. Their protease properties corresponded to those of Xanthomonas sp. JK311.

국내에서 분리된 포도상구균의 Vancomycin 내성빈도 및 특성 (Characterization and Frequency of Vancomycin Resistance in Staphylococcus aureus Isolated in Korea)

  • Vancomycin은 세포벽의 합성을 억제하여 세균에 대한 항균효과를 나타내는 glycopeptide 계 항생물질로서 그람양성세균으로 인한 감염치료에 광범위하게 사용되며, 특히 methicillin 내성 포도상구균의 선택적 치료제로 쓰이고 있다. 그러나 최근 임상검체에서도 중등도의 내성을 가지는 포도상구균 (Mu50: MIC 8 $\mu\textrm{g}$/ml)이 나타나기 시작하였고 여러 가지 여건상 국내에서도 내성균주가 분리될 가능성이 높다고 사료되어 임상검체 중 methcillin 내성 포도상구균을 대상으로 vancomycin 감수성 및 내성빈도 조사를 실시하고 이에 따른 내성기전을 알아보고자 하였다. 본 실험 결과 107주 (株)의 methicillin 내성균주 중 23.3%가 vancomycin에 대하여 내성을 보였으며 vancomycin 내성을 나타내는 표준균주인 Mu50과 Mu3의 중간 정도의 내성빈도를 보였다. 중합효소 연쇄반응을 통해 장구균의 vancomycin 내성에 관여하는 vanA, vanB, vanC1, vanC2, vanH 특이 유전자는 증폭되지 않았다. SDS-PAGE를 실시하여 81 kDa, 58 kDa, 33 kDa, 28 kDa 등의 주요 단백 분획을 확인하였고, Mu50에서 45 kDa의 특징적인 단백 분획을 관찰하였다. LDH enzyme assay에서는 한 개의 검체가 Mu50과 함께 높은 LDH 활성을 보였다.

Cispatin 내성인 사람 위암 세포주 SNU-1의 복합약제내성 및 Verapamil의 효과 (Multidrug Resistance and Cytotoxicity of Anticancer Drug by Verapamil in Cisplatin Resistant Human Stomach Cancer Cell)

  • 복합약제내성암세포주의 출현은 암의 화학 요법에 있어서 중요한 문제점중의 하나이며 이 복합내성 암세포의 출현에 대한 정확한 기전은 아직 밝혀져 있지 않다. 본 실험에서는 약제내성 암세포의 다른 항암제에 대한 복합내성 형성정도와 calcium 길항제인 verapamil에 의한 내성극복 정도를 비교하고 내성 암세포의 세포막 단백질의 변화를 관찰하였다. 사람의 위암 세포주 SNU-1을 cisplatin 농도 $0.001{\mu}M$에서 시작하여 $10{\mu}M$까지 증가시켜 내성세포 SNU-1/$Cis_5$를 얻었으며, MTT assay로 세포성장을 관찰한 결과 doubling time은 SNU-1이 약 29 시간, SNU-1/$Cis_5$는 약 38시간으로 내성세포가 감수성세포보다 성장속도가 느린 것을 관찰하였다. 약제 감수성 검사를 위해서 4일간의 MTT assay로 대조군에 대한 50% 세포 생존시의 약제농도 $IC_{50}$를 비교하여 상대적 내성도 (Relative Resistance, RR)를 측정하였는데 cisplatin, 81.4; vinblastine, > 43.0; epirubicin, 22.9 ; dactinomycin, 16.0 ; etoposide, 15.0 ; vincristine, 9.2 ; adriamycin, 5.7 ; aclarubicin, 5.3으로 관찰되었고 그외의 약제인 cyclophosphamide, 5-fluorouracil, methotrexate, daunorubicin에서는 낮게 나타났다. $10{\mu}M$의 verapamil에 의한 내성 억제 효과는 vincristine, 13.1 ; epirubicin, 10.0 ; etoposide, 6.3 ; vinblastine, 4.4 ; dactinomycin, 3.6 ; daunorubicin, 2.4로 나타났다. Radioiodination을 이용한 SDS-PAGE로 SNU-1/$Cis_5$에서 항암제 내성과 관련된 것으로 여겨지는 51,400와 81,300 dalton의 막단백질의 발현을 관찰하였다.

Isolation, Molecular Characterization and Antibiotic Susceptibility Pattern of Vibrio parahaemolyticus from Aquatic Products in the Southern Fujian Coast, China

  • Vibrio parahaemolyticus is a major gastroenteritis-causing pathogen in many Asian countries. Antimicrobial resistance in V. parahaemolyticus has been recognized as a critical threat to food safety. In this study, we determined the prevalence and incidence of antimicrobial resistance in V. parahaemolyticus in the southern Fujian coast, China. A total of 62 isolates were confirmed in retail aquatic products from June to October of 2018. The serotype O3:K6 strains, the virulence genes tdh and trh, antibiotic susceptibility and molecular typing were investigated. Then plasmid profiling analysis and curing experiment were performed for multidrug-resistant strains. The results showed that the total occurrence of V. parahaemolyticus was 31% out of 200 samples. Five strains (8.1%) out of 62 isolates were identified as the V. parahaemolyticus O3:K6 pandemic clone. A large majority of isolates exhibited higher resistance to penicillin (77.4%), oxacillin (71%), ampicillin (66.1%) and vancomycin (59.7%). Seventy-one percent (44/62) of the isolates exhibited multiple antimicrobial resistance. All 62 isolates were grouped into 7 clusters by randomly amplified polymorphic DNA, and most of the isolates (80.6%) were distributed within cluster A. Plasmids were detected in approximately 75% of the isolates, and seven different profiles were observed. Seventy-six percent (25/33) of the isolates carrying the plasmids were eliminated by 0.006% SDS incubated at 42℃, a sublethal condition. The occurrence of multidrug-resistant strains could be an indication of the excessive use of antibiotics in aquaculture farming. The rational use of antimicrobial agents and the surveillance of antibiotic administration may reduce the acquisition of resistance by microorganisms in aquatic ecosystems.

삼세기(Shaggy sea raven, Hemitripterus villosus)의 carbonic anhydrase III에 관한 연구 (Presence of Carbonic Anhydrase III-like Protein in Shaggy Sea Raven, Hemitripterus villosus)

  • 본 논문에서는 삼세기(Shaggy sea raven, Hemitripterus villosus)를 실험 재료로 선택하여 Carbonic Anhydrase Isozymes (CAs) 중의 하나인 CA III에 대한 연구를 SDS-PAGE, Isoelectric Focusing (IEF), Western blot analysis의 방법을 통하여 진행하였다. SDS-PAGE와 Western blot 결과 삼세기 아가미, 혈액, 장, 간, 신당, 근육, 심장조직에서 CA III의 분자량인 30 kDa의 band가 확인되었다. 삼세기의 근육과 아가미에 대한 등전점 전기영동(IEF)과 Western blot analysis 결과 pI 7.0 부근에서 band가 형성되는 것을 확인할 수 있었다. 특히 SDS-PAGE와 IEF 실험결과 삼세기의 아가미 조직에서 CA III의 발현량이 다른 조직들에 비하여 우세하게 나타났다. 이는 아가미가 다른 조직들과 달리 어류의 생체기관 중 유일하게 외부와 직접 접촉이 가능한 조직으로서 활성산소에 대한 손상을 최소화하기 위한 것으로 사료된다.

Impedance and Thermodynamic Analysis of Bioanode, Abiotic Anode, and Riboflavin-Amended Anode in Microbial Fuel Cells

  • Understanding exoelectrogenic reactions of the bioanode is limited due to its complexity and the absence of analytics. Impedance and thermodynamics of bioanode, abiotic anode, and riboflavin-amended anode were evaluated. Activation overpotential of the bioanode was negligible compared with that of the abiotic anode. Impedance spectroscopy shows that the bioanode had much lower charge transfer resistance and higher capacitance than the abiotic anode in low frequency reaction. In high frequency reaction, the impedance parameters, however, were relatively similar between the bioanode and the abiotic anode. At open-circuit impedance spectroscopy, a high frequency arc was not detected in the abiotic anode in Nyquist plot. Addition of riboflavin induced a phase angle shift and created curvature in high-frequency arc of the abiotic anode, and it also drastically changed impedance spectra of the bioanode.

계면활성제를 이용한 합성고분자 수용액의 마찰저항감소 및 퇴화 특성 향상 연구 (The Study on Drag Reduction Rates and Degradation Effects in Synthetic Polymer Solution with Surfactant Additives)

  • The turbulent flow resistance of water solution with polymer is reduced as compared with that of pure water. This effects is named th drag reduction and offers the significant reduction of the pumping power and the energy consumption. But the intense shear forces and the high temperature experienced by the polymer solution when passing through the pipes cause the degradation a loss of drag reduction effectiveness. Especially, the degradation behavior is found to be strongly dependent on temperature. This mechanical and thermal degradation can be avoided by adding materials such as surfactant to the polymer solution, which enhance the bonding force between molecules. In the present study, Copolymer and SDS were utilized and they were mixed in 10 different mixture ratios, while total concentration was fixed as 100wppm. Degradation of Copolymer-SDS mixture solutions was investigated experimentally in closed loop at the temperature of $10^{\circ}C\; and\; 80^{\circ}C$ with various flow average velocities of 1.5 m/sec, 3.0m/sec, and 4.5m/sec. Degradation characteristics of polymer solution without surfactant show a radical loss of drag reduction effectiveness at high temperature. Degradation alleviation ability of surfactant is especially effective at high temperature. Consequently, this results show that the addition of surfactant to the polymer solution can control unfavorable degradation phenomena for high temperature systems.
