Tertiary Structure of PreSl(21-47) of Hepatitis B Virus Studied by NMR Spectroscopy

  • To design more efficient peptide antagonist against the HBV, preSl(21-47) which carries the HBV receptor binding site for hepatocytes was synthesized and the solution structure of preSl(21-47) was investigated using CD spectroscopy and NMR spectroscopy in membrane-mimicking environments. According to CD spectra, preSl(21-47) has a random structure in aqueous solution, while conformational change was induced by addition of TFE and SDS micelle. Tertiary structures as determined by NMR spectroscopy shows that preSl(21-47) has a very flexible structure even in SDS micelle.

Rate Enhancement by Micelle Encapsulation for Oxidation of L-Glutamic Acid in Aqueous Media at Room Temperature

  • Oxidation of glutamic acid is performed in aqueous acid media at $30^{\circ}C$ under the kinetic condition [glutamic acid]$_T{\gg}[Cr(VI)]_T$. Effect of combination of micellar catalyst (SDS, TX-100) and promoter (PA, bpy, phen) has been studied. Among the promoters phen accelerates the reaction most in aqueous media. But the rate acceleration is small in the case. Combination of promoter and catalyst produces much better result. Maximum rate enhancement occurs in presence of the combination of bpy and SDS.

  • Cyclic voltammetric method is used to survey microscopic environments which take place at the surfactant-modified carbon electrode when the hydrophobic and hydrophilic environments of $Ru(ph){_3}^{2+}$(tris 1,10-phenanthroline ruthenium(II) chloride) is created by the addition of anionic surfactant, sodium dodecyl sulfate(SDS). Critical micelle concentration(CMC) of SDS in $Ru(ph){_3}^{2+}$ measured by cyclic voltammetry(CV) is in aggrement with that by surface tensiometry. Influence of the concentration of supporting electrolyte at surfactant-modified carbon electrode is investigated.

Study on the Solubilization of 4-ethylaniline in the aqueous solutions of mixed surfactants (혼합계면활성제의 수용액에서 4-ethylaniline의 가용화에 관한 연구)

  • The critical micelle concentration (CMC) values of the mixed surfactant systems and the solubilization conatant (Ks) values of 4-ethylaniline in those solutions were measured and analyzed by the UV-Vis method. As a result, the mixed surfactant systems of TTAB/LSB and TTAB/TX-100 did not deviate significantly from ideal mixed micellization. However, the mixed systems of SDS/LSB and SDS/TX-100 showed great negative deviations from ideal mixed model. These differences showed that the intensity of the interaction between two components in the mixed micelle was different for each mixed system and that these differences greatly influenced the solubilization of 4-ethylaniline by a mixed surfactant system. Among pure surfactants, an anionic surfactant such as SDS showed a greater Ks value than other ionic surfactants, and the Ks value by each surfactant system decreased in the order of SDS≫TTAB≧LSB>TX-100. In addition, the Ks values of all the mixed surfactant systems were higher than those of the pure surfactants constituting the mixed systems.

Effects of Additives on Soil Washing Efficiency for Mixed Surfactants (혼합 계면활성제에 적용된 각종 첨가제가 토양세척 효율에 미치는 영향)

  • To enhance the washing efficiency of soil polluted by hydrophobic organic compounds, the effects of electrolytes and monomeric organic additives on micelle formation and washing efficiency of mixed surfactant solutions were investigated in this study. The surface tensions and critical micelle concentrations(CMCs) of the single and mixed surfactant solutions[$POE_5$/SDS] supplemented by NaCl were measured to investigate the effects on washing efficiency, and the composition ratios of surfactants and NaCl were optimized for the efficient soil washing system. As the mixing ratio of $POE_5$/SDS was increased to 80%, the mixed surfactant with 0.01M NaCl showed more proportional increase of washing efficiency than the mixed surfactant without any salts. The 3% solution of $POE_5$ and SDS(80%/2o%) with 0.01M NaCl showed the washing efficiency of 90%. However, the washing efficiency was not enhanced by NaCl addition to the single surfactant solution of $POE_5$. The CMC of SDS(0.049%) was higher than that of $POE_5$(0.016%), but the CMCs of mixed surfactants were decreased as the mixing ratio of $POE_5$ was increased. Alcohols having longer chain and branched carbon chain were found to be desirable for the soil washing additives.

  • The interaction between the cationic dye, thionin(Th) and the anionic surfactant, sodium dodecyl sulfate (SDS) has been investigated by absorption spectra. As the temperature of surfactant solution was increased in premicellar range(S/D=10, 80, and 160) which was much lower than the critical micelle concentration(CMC), the increment or decrement of the molar extinction coeffecient ratio appeared. It was found that the most stable temperature range of the oligomer aggregate in Th-SDS system at S/D=160 was below $60^{\circ}C$. With increasing the concentration of inorganic salt and organic solvents in Th-SDS system, ${\alpha}$-band was increased, but ${\gamma}$=band or J-band was decreased. The orders of ${\alpha}$-band increasing power were $Cl^-$>$ClO{_4}{^-}$>$SO{_4}{^{2-}}$>$NO{_3}{^-}$ and 2-propanol>ethanol>methanol>ethylene glycol.

  • In the presence of SDS(sodium dodecyl sulfate), effect of $Ca^{2+}$on the interaction of NP-40EO[nonylphenol-(ethylene oxide)40] with iodine in aqueous solution were investigated by UV-visible spectrophotometer. In the presence of SDS, the intensity of interaction peaks were decreased and markedly increased by the addition of $Ca^{2+}$. Decrease of interaction peaks is caused by decrease of EO number to be interact with iodine per unit surface area of mixed micelle to the insertion of SDS and increase is attributed to the compactness of micelle in the presence of $Ca^{2+}$. These phenomena may be explained by the fact that the linear EO (ethylene oxide) chains, to be free in aqueous solution, could form a pseudo-crown ether structures of forming with $Ca^{2+}$ion.

  • Electrochemistry of 1.0 mM tris(2,2'-bipyridyl)ruthenium(II) ($Ru(bpy)_3{^{2+}}$) in 300 mM $H_2SO_4$ solution with and without sodium dodecyl sulfate (SDS) is studied. In the presence of SDS, $E_{pa}$ of $Ru(bpy)_3{^{2+}}$ shifts to positive direction compared to the SDS free case. The intersection of two lines on ${\Delta}E_p$ vs. -log[SDS] plot is measured as a critical micelle concentration (CMC), which is 3.67 mM SDS.

  • Electrochemical behavior of 1.0mM bis(2,2'-bipyridyl)copper( II )(${Cu(bpy)_2}^{2+}$) in 100mM NaCl+27mM $MgCl_2$ solution with and without sodium dodecyl sulfate(SDS) is investigated. In the presence of SDS, $E_{pa}$ and $E_{1/2}$ of ${Cu(bpy)_2}^{2+/1+}$ shifts to the positive direction compared to the SDS free. Teh intersection of two lines on ${\Delta}E_p$ vs. -log[SDS] plot for 1.0mM $Cu(bpy)_2(NO_3)_2$ in 100mM NaCl+27mM $MgCl_2$ solution is a critical micelle concentration(CMC). Reduced form ${Cu(bpy)_2}^{1+}$ in the presence of $MgCl_2$ is more easily associated to dodecyl sulfate anion($DS^-$) than oxidized ${Cu(bpy)_2}^{2+}$. As the ionic strength is increased by the addition of multiply charged cations($MgCl_2$), the double layer becomes more compact and micelle formation is more difficult.

  • The magnetized water is known to have a unique pattern of hydrogen bond between water molecules, thereby producing different physicochemical properties from the ordinary water. We have examined the effect of magnetized water on the change of critical micelle concentrations (CMC) of some surfactants. The CMC changes of SDS (sodium dodecyl sulfate) and CTAB (cetyltrimethylammonium bromide) dissolved in the magnetized water have been determined by the conductivity measurement at $25\;{^{\circ}C}$ and that of SDS, CTAB and Pluronic F-68 have also been examined by the surface tension method at $25\;{^{\circ}C}$. The CMC variation of SDS was examined by ITC (Isothermal Titration Calorimeter) at $25\;{^{\circ}C}$. The CMC of SDS, CTAB, and Pluronic F-68 are more decreased in the magnetized water, SDS is about $2.7{\sim}6.5$25 %, CTAB is about $2.3{\sim}3.0$%, and Pluronic F-68 is about 24.2 %, than in the control water.
