Aging process can be explained by many factors. In this study, we counted complete Blood Cells (CBC) such as WBC, Lymphocytes, Monocytes, Granulocytes, RBC, HGB, and HCT of both SAM P6 and SAM P1 during the aging process. Plasma albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea concentrations were also measured at the same time. In addition to these, plasma concentrations of cortisol, total T3, and total T4 were analyzed by Chemiluminescent Immunoassay. There were no change in CBC counts of SAM R1 and SAM P6 during this study. Plasma concentratins of albumin and glucose decreased significantly in SAM R1. However, plasma alkaline phosphatases and creatinine concentration in SAM P6 decreased significantly at 16 week after birth comparing to the control. Total T4 levels were siginificantly increased although cortisol and total T3 concentrations were the same in SAM R1 groups. Especially, the after birth of creatinine, alkaline phosphatase, T4 of SAM P6 at 16 week were significantly different form those of SAM R1. At 12 week after birth, pilose antler extract was given 5g/kg/day p.o. for 0, 7, 14, 21, and 30 days each in both SAM R1 and SAM P6. The RBC, HGB, and HCT levels started to increase significantly from 7 days after the dose at SAM P6 only. Total T4 conectrations were elevated gradually during the study although the antler extract administration did not prevent or inhibit the increase in total T4 concentration during the study. Therefore, the elevation of erythrocytes after administration of the extract needs to be studied in future.
Physiological effects in SAM P6 and Rl by administration of Cervus cornu, Astragali Radix, Rehmanniae Radix, and Angelicae Radix extracts were screened to know in vivo activities of each extracts. We measured complete blood cells (CBC) such as aBC, HGB, and HCT using coulter's method. Plasma concentrations of albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea and total iron ere also analyzed using biochemical clinical autoanalyzer. Plasma concentrations of cortisol, total T$_3$, and total T$_4$ were measured by chemiluminescent immunoassay methods. At 12 weeks after birth, Cervus conu or Astragali Radix or Rehmanniae Radix extracts were given 5 g/Kg.day p.o. for 0,7,14,21, and 30 days each in both SAM Rl and SAM P6. Angelicae Radix study was done the same as the others except the mice were 16 weeks after birth. The RBC, HGB, and HCT levels after administration of Astragali and Rehmanniae were elevated in SAM Rl, but those in Cervi study were increased in SAM P6 the most. Decreases in alkaline phosphatase concentration of SAM Rl and P6 after Cervi administration were detected. Total plasma iron concentration was decreased by Angelicae administration in SAM P6. In general, Angelicae and Rehmanniae stimulate increases in cortisol, but total T$_3$ and T$_4$ levels were also elevated by all these extracts. In conclusion, these herval medicine extracts help hematopoiesis in SAMs through probably different mechanisms.
모든 김치군 중 I김치가 가장 높은 SAM 함량을 나타냈으며, 김치별 SAM 함량이 가장 높은 발효시점은 A, K김치는 pH $4.6{\sim}4.9$, 산도 $0.38{\sim}0.57$0.57, D, H김치는 pH 6.3, 산도 $0.24{\sim}0.27$, E, G김치는 pH $6.1{\sim}6.6$, 산도 $0.05{\sim}0.14$0.14, B, C, F, I, J김치는 pH $4.3{\sim}5.3$, 산도 $0.45{\sim}1.02$에서 나타났다.
In previous studies we reported that the levels of RBC, hemoglobulin (HGB) and hematocrit (HCT) in SAM P6 were increased significantly from 7 day after oral administration of the pilose antler extract, 5 g/kg/day, and were lasted during the study. Thus, this study was performed to elucidate mechanism of erythropoietic action by the extract administration. SAM R1 and SAM P6 were chosen as experimental animals. At age of 12 weeks, pilose antler extract were given 0.3 and 5 g/kg/day (p.o.) each for 0, 7 and 14 days in both animals. Complete blood cells such as RBC, HGB, and HCT were counted. And plasma concentration of erythropoietin (EPO) which is the major regulator of erythropoiesis was measured using $^{125}I-antierythropoietin$ IgG. Total iron concentration in plasma was also analyzed. The levels of RBC and HCT were increased significantly after administration at both doses of 0.3 and 5 g/kg/day in SAM P6, however, these were increased only at dose of 5 g/kg/day in SAM R1. The plasma EPO concentration was increased significantly after administration in SAM P6. The plasma concentrations of total iron were significantly decreased after administration of the extract in SAM P6. These results suggest that the changes in erythropoietic effects after the administration of pilose antler extract may be mediated, at least in part, through the change in the plasma EPO.

The effect of herbal medicine on osteoporosis was studied using ovariectomized rats as an animal model of Type I osteoporosis and SAM P6 mice as that of Type II. Each traditiona l boiling water extract of Achyranthis Radix, Psoraleae Radix, Rehmanniae Radix Preparat, Corni Fructus and Mycelia of Ganoderma, and systemic water fraction of Astragali Radix was given 5g(dried herbal weight)/kg/day p.o. for 30 days in each group of ovariectomized rats, SAM R1 and SAM P6. The extract of Cervi parvum Cornu was given for 14 days only. One ml of blood was taken by tail vein at day 0, 7, 14, 21 and 30 days after administration of the extract. Plasma levels of alkaline phosphatase, calcium, creatinine, inorganic phosphate, blood urea nitrogen, cortisol, total $T_3\;and\;total\;T_4$ were measured. In ovariectomized rats, administration of Achyranthis Radix or Corni Fructus decreased in alkaline phosphatase and that of Achyranthis Radix or Psoraleae Radix decreased in calcium comparing to the control (p<0.05). The administration of Psoralese Radix decreased in calcium and increased in urea comparing to day o(P<0.05)(Table I). There were not much changes in plasma calcium, inorganic phosphate, and alkaline phosphatase concentrations after uptake of these herbal medicine used in SAM P6(Table III). However, administration of Astragali Radix altered plasma inorganic phosphate and creatinine levels in SAM R1(p<0.01)(Table UU). The administration of Corni Fructus or Psoralease Radix induced the changes in plasma concentrations of cortisol, total $T_3$ and total $T_4$ in Type I(p<0.05) (Table IV). The uptake of Cervi parvum Cornu increased in total $T_3$ concentration and that of Mycelia of Ganodtragali Radix in SAM P6. However, the uptake of Mycelia of Ganoderma induced changes in cortisol and $T_4$ concentrations in SAM R1(p<0.05). Thus, there were significant differences in responses of herbal medicine in different types of osteoporosis.

Objectives : The purpose of this study was to examine the effects of the Hominis placenta extracts on improvement of senile osteoporosis (Type 2) in SAM P6. In traditional Oriental medicine, Placenta (Jahageo) is indicated for insufficiency of blood and tidal fever due to steaming bone disorder. Methods : At 10 weeks after birth, Placenta ex. was given 100 mL, 200 ml/kg/day, i.p. 3 times a week for 4 weeks in SAM P6 mice. And then we measured complete blood cells, blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : As a result, in SAM P6, injection of Placenta ex. (200 mL) increased in RBC, HB and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased significantly in the injection groups of the Placenta ex. from that of the control group, but blood urea nitrogen was no significant. Moreover, Placenta ex. were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales and tibia in P6 mice. Conclusions : These findings suggest that Placenta ex. are effective in preventing bone loss in SAMP6.

The effects of herbal medicine on travecular bone area were studied using ovariectomized rat as an animal model of Type I osteoporosis and SAM P6 as that of Type II. We counted red blood cells(RBC), hemoglobin(Hb), and hematocrit(Hct) using Couter`sR method. Each traditional boiling water extract of Achyrathis Radix, Psoraleae Fructus, Rehmanniae Radix Preparat, and Cornii Fructus and a systemic water extract of Astragali Ractiex was given 5g/kg/day, p.o., for 30 days in a group of 4-5 ovariectormized rats. One ml of blood was taken by tail vein at day 0, 7, 14, 21, and 30 days after administration of the extract. The traditional hot water extract of Cervi parvumn Corni (Cervi) was given the same dose as described above for 14 days in a group of 10 SAM P6 mice and systemic water extract of Astragali Radix was administered as the same dose as above for 30 days in 10 SAM P6 mice. Travecular bone area was measured 5 mcm decalcified and stained thin bone slice by image analysis using a digitalizer. In Type I, ovariectomized rats, administration of Astragali Radix, Rhemanniae Radix Preparat, and Corni Fructus decreased in RBC, Hb and Hct. In Type II, administration of Cervi increased in RBC and Hct and that of Astragali Radix was also elevated RBC. In Type I, any administration of herbal medicine used in this study did not elevate travecular bone area significantly except Corni Fructus showed a trend of increase in travecular bone area. However, Type II, Cervi and Astragali Radix increased in both mean and total travecular bone area. Thus, there are significant difference in response of herbal medicine in different types of osteoporosis.
Folate and vitamin $B_{12}$ are essential cofactors for homocysteine (Hcy) metabolism. Homocysteinemia has been related with cardiovascular and neurodegenerative disease. We examined the effect of folate and/or vitamin $B_{12}$ deficiency on biomarkers of one carbon metabolism in blood, liver and brain, and analyzed the correlation between vitamin biomarkers in mild and moderate homocysteinemia. In this study, Sprague-Dawley male rats (5 groups, n = 10) were fed folatesufficient diet (FS), folate-deficient diet (FD) with 0 or 3 g homocystine (FSH and FDH), and folate-/vitamin $B_{12}$-deficient diet with 3 g homocystine (FDHCD) for 8 weeks. The FDH diet induced mild homocysteinemia (plasma Hcy 17.41 ${\pm}$ 1.94 nmol/mL) and the FDHCD diet induced moderate homocysteinemia (plasma Hcy 44.13 ${\pm}$ 2.65 nmol/mL), respectively. Although liver and brain folate levels were significantly lower compared with those values of rats fed FS or FSH (p < 0.001, p < 0.01 respectively), there were no significant differences in folate levels in liver and brain among the rats fed FD, FDH and FDHCD diet. However, rats fed FDHCD showed higher plasma folate levels (126.5 ${\pm}$ 9.6 nmol/L) compared with rats fed FD and FDH (21.1 ${\pm}$ 1.4 nmol/L, 22.0 ${\pm}$ 2.2 nmol/L)(p < 0.001), which is the feature of "ethyl-folate trap"by vitamin $B_{12}$ deficiency. Plasma Hcy was correlated with hepatic folate (r = -0.641, p < 0.01) but not with plasma folate or brain folate in this experimental condition. However, as we eliminated FDHCD group during correlation test, plasma Hcy was correlated with plasma folate (r = -0.581, p < 0.01), hepatic folate (r = -0.684, p < 0.01) and brain folate (r = -0.321, p < 0.05). Hepatic S-adenosylmethionine (SAM) level was lower in rats fed FD, FDH and FDHCD than in rats fed FS and FSH (p < 0.001, p < 0.001 respectively) and hepatic S-adenosylhomocysteine (SAH) level was significantly higher in those groups. The SAH level in brain was also significantly increased in rats fed FDHCD (p < 0.05). However, brain SAM level was not affected by folate and/or vitamin $B_{12}$ deficiency. This result suggests that dietary folate- and vitamin B12-deficiency may inhibit methylation in brain by increasing SAH rather than decreasing SAM level, which may be closely associated with impaired cognitive function in nutritional homocysteinemia.
