Overexpression of Tbx3 Predicts Poor Prognosis of Patients with Resectable Pancreatic Carcinoma

  • Background: To determine the expressions of Tbx3, a member of subgroup belonging to T-box family, and its prognostic value in pancreatic carcinoma. Materials and Methods: We determined the expression levels of Tbx3 on both mRNA and protein levels in 30 pairs of fresh tumor tissues and paratumor tissues by quantitative real-time polymerase chain reaction (qRT-PCR) and Western blotting, respectively. In addition, protein level of Tbx3 were identified using immunochemistry in 80 pairs of paraffin-embedded specimen. The correlations between Tbx3 expression and various clinicopathological parameters as well as overall survival were evaluated. Results: Tbx3 mRNA and protein levels in tumor tissues were significantly higher than in the paratumor tissues by qRT-PCR ($0.05{\pm}0.007$ vs. $0.087{\pm}0.001$, p<0.001) and western blotting ($1.134{\pm}0.043$ vs. $0.287{\pm}0.017$, p<0.001). The statistical analysis based on immunohistochemical evaluation suggested that Tbx3 aberrant expression was significantly associated with several conventional clinicopathological variables, such as gender, age, tumor position, preoperative CA19-9 level, pathological T staging and N staging. Univariate and multivariate analyses revealed that Tbx3 expression was an independent prognostic factor for overall survival (<0.001). Conclusions: Our results suggest that overexpression of Tbx3 is associated with poor prognosis of pancreatic cancer patients. However, additional clinical trials are needed to accurately validate this observation.

Application of KOMPSAT-5 SAR Interferometry by using SNAP Software (SNAP 소프트웨어를 이용한 KOMPSAT-5 SAR 간섭기법 구현)

  • SeNtinel's Application Platform (SNAP) is an open source software developed by the European Space Agency and consists of several toolboxes that process data from Sentinel satellite series, including SAR (Synthetic Aperture Radar) and optical satellites. Among them, S1TBX (Sentinel-1 ToolBoX)is mainly used to process Sentinel-1A/BSAR images and interferometric techniques. It provides flowchart processing method such as Graph Builder, and has convenient functions including automatic downloading of DEM (Digital Elevation Model) and image mosaicking. Therefore, if computer memory is sufficient, InSAR (Interferometric SAR) and DInSAR (Differential InSAR) perform smoothly and are widely used recently in the world through rapid upgrades. S1TBX also includes existing SAR data processing functions, and since version 5, the processing capability of KOMPSAT-5 has been added. This paper shows an example of processing the interference technique of KOMPSAT-5 SAR image using S1TBX of SNAP. In the open mine of Tavan Tolgoi in Mongolia, the difference between DEM obtained in KOMPSAT-5 in 2015 and SRTM 1sec DEM obtained in 2000 was analyzed. It was found that the maximum depth of 130 meters was excavated and the height of the accumulated ore is over 70 meters during 15 years. Tidal and topographic InSAR signals were observed in the glacier area near Jangbogo Antarctic Research Station, but SNAP was not able to treat it due to orbit error and DEM error. In addition, several DInSAR images were made in the Iraqi desert region, but many lines appearing in systematic errors were found on coherence images. Stacking for StaMPS application was not possible due to orbit error or program bug. It is expected that SNAP can resolve the problem owing to a surge in users and a very fast upgrade of the software.

A Numerical Investigation on the Isentropic Efficiency of Steam Turbine Nozzle Stage with Different Nozzle Vane Thickness and Mass Flow Rate (증기 터빈 노즐 베인의 두께 변화와 유량별 등엔트로피 효율 변화에 관한 수치해석)

  • In this study, the influence of mass flow rate on the isentropic efficiency of the steam turbine nozzle stage is investigated. A realistic three-dimensional numerical model, which is based on the compressible Navier-Stokes equations, is developed for the steam phase. The comprehensive conservation laws and a kinetic model for steam are investigated. With two different models for the three-dimensional geometry of the nozzle stage, the pressure and temperature distributions, velocity, Mach number. and Markov energy loss coefficient are calculated. A maximum efficiency of 96.66% is found at a mass flow rate of 0.9 kg/s in model A. In model B, a maximum efficiency of 97.32% is found at a rate of 1.6 kg/s. It is determined that the isentropic nozzle efficiency increases as the Markov energy loss coefficient decreases through a nearly linear relationship.

Effect of Linoleic Acid Content and $omega6/$omega3$ Ratio on the Induction of Gastric Ulcer in the Experimental Rat Model (Linoleic acid 함량과 $omega6/$omega3$비율이 침수속박 스트레스에 의한 위궤양의 발병에 미 치는 영향)

  • This study was designed to determine the effects of linoleic acid contents and $\omega$ 6/$\omega$3 ratios on the induction of gastric ulcer by water immersion and restraint stress. Sprague-Dawley rats were fed 5diets containing 7% fat(w/w) for 6weeks. These diet groups were Lh, Mh, Hh, Mm, Ml, : 3 different linoleic acid levels(0.3% of energy(L). 3.5(M), 10(H) and 3 different $\omega$6/$\omega$3 ratios (11(1), 33(m), 100(h) with beef tallow, sunflower or fish oil. The Lh group showed a significantly higher ulcer index (UI) than the Mh and Hh groups(p<0.05). At the same linoleic levels, the UI had no significant difference within the $\omega$6/$\omega$3 ratios. The Mh group showed significantly higher (p<0.05) PGE2 and TBX2 content than any other group. Pearson's correlation coeffcients between UI and PGE2 and TBX2 had a negatively significant correlation(p<0.05). Linoleic acid of gastric mucosal phospholipids was reflected by the diet, but was not significantly different. The most significant finding of this study is that not only the absolute amount of linoleic acid, but also the $\omega$6/$\omega$3 ratios are important factors for the prevention of gastric ulcer.

  • In this thesis, we evaluate the performance of the TDX-IB/ISDN Switching System ISS (ISDN Subscriber Subsystem) which is the ISDN user-network interface module. For this evaluation, performance indices are established and major performance parameters which influence message processing are extracted by studying the ISS structure and mechanism. To reflect these parameters, simulation model is developed and simulated. From the result of maximum throughput, message delay time, etc. , ISS message processing capability is evaluated and several method to enhance the system performance is proposed, by analyzing the system bottleneck element.

Genome-wide association study on immune-response for improving healthiness in Holstein dairy cattle (Holstein 젖소의 호흡기 질병 백신에 대한 면역반응성과 전장 유전체 연관 분석 연구)

  • To detect Single nucleotide polymorphisms (SNP) markers associated with Bovine viral diarrhea virus (BVDV) and Bovine respiratory syncytial virus (BRSV) S/P ratio in Korean Holstein dairy cattle, Genome-wide association study (GWAS) was performed using Illumina BovineSNP50 Beadchip. The number of phenotype data and genotype data were 107, and 294. respectively. Phenotype data were collected for four periods (0 week, 1 week, 4 week, 24 week) after having vaccinated (0 week no vaccinated period). A total of 36,257 SNPs was remained after quality control had been done by PLINK. The result of GWAS showed 6 SNP markers (BTB-01704243, BTB-01594395, ARS-BFGL-NGS-118070, ARS-BFGL-NGS-111365, BTA-65410-no-rs, Hapmap38331-BTA-61256) under BVDV and 4 SNP markers (ARS-BFGL-NGS-109861, Hapmap53701-rs29017064, ARS-BFGL-NGS-71055, BTA-11232-no-rs) under BRSV. And also, 10 candidate genes found through 10 SNP markers (TBX18, CEP162, PAFAH1B1, METTL16, BRCA1, RND2, POLK, ENSBTAG00000051724, ADAM18, NRG3).

Effect of Different Amount of Dietary n-3 PUFA on Colon Carcinogenesis in DMH-treated Rats (쥐에서 식이에 첨가한 n-3 PUFA함량에 따라 대장 암화과정에 미치는 영향)

  • The objective of the study was to observe the effect of n-3 PUFA on cell proliferation and apoptosis by determining mRNA and protein of COX-2 and eicosanoid product and the mRNA and protein of Bu and Bcl-2 related to apoptosis in colon carcinogenesis of 1,2- dimethylhydrazine (DMH)-treated rats. Ninety male Sprague Dawley rats weighing about 170g were divided into 3 groups, control and n-3 PUFA supplemented groups (FO group: 6.2 mmoles n-3 PUFA; 2FO group: 12.4 mmoles n-3 PUFA) and fed experimental diet for 14 weeks. All rats were intramuscularly injected with DMH 15 mg/kg twice a week for 6 weeks to deliver total dose of 180 mg/kg body weight. Compared with the control group, 6.2 mmoles n-3 PUFA significantly reduced the levels of mRNA and protein expression of COX-2 and 2-series eicosanoids ($TXB_{2}$ and $PGE_{2}$) and decreased cell proliferation in colonic mucosa. However, high levels of n-3 PUFA supplementation significantly increased the levels of mRNA and protein expression of COX-2, TXB2 and PGE2. and increased cell proliferation which was similar level to that of control group. Compared with the control group, n-3 PUFA, regardless of the amount, significantly increased apoptotic index in colonic mucosa. Western blot and RT-PCR analyses showed that the levels of mRNA and protein expression of Bax were significantly increased by 6.2 mmoles n-3 PUFA, but decreased by 12.4 mmoles n-3 PUFA. The analyses also showed the levels of mRNA and protein expression of Bcl-2 were significantly reduced by 6.2 mmoles n-3 PUFA, but increased by 12.4 mmoles n-3 PUFA. The ratio of Bcl-2/Bax in mRNA and protein was significantly reduced by 6.2 mmoles n-3 PUFA but increased by 12.4 mmoles n-3 PUFA. Overall, these results indicate that n-3 PUFA could be effective in preventing colon carcinogenesis by reducing cell proliferation with lower level of COX-2 and 2-series eicosanoid, and increasing apoptosis by inducing pro-apoptotic gene, Bax and inhibiting anti-apoptotic gene, Bcl-2 in the colonic mucosa of DMH-treated rats. However, high level of n-3 PUFA supplementation could stimulate colon carcinogenesis by increasing cell proliferation and inhibiting apoptosis. (Korean J Nutrition 38(10): 807$\sim$816,2005)
