Cloning and Expression of Antifungal Protein (PR5) Genes from Hot Pepper (Capsicum annuum L.) (고추(Capsicum annuum)의 항균성 단백질(PR-5) 유전자의 클로닝과 발현 분석)

  • We have isolated and artificially expressed three cDNA clones of Capsicum annuum PR5 genes for elucidating the antifungal activity against Phytophthora capsici which contracted a hot pepper root rot in field condition. Three divergent PR5 proteins from hot pepper were designated as CAPR5-1 and CAPR5-2 from susceptible cultivar (Subicho) as well as CAPR5-3 from resistant cultivar (CM331) in response to P. capsici. The cDNA similarity was found over 80% of identity among the three CAPR5s, and deduced amino acid sequence was characterized that all of CAPR5s contained 16 cysteine residues which possibly had a significant role in the structural formation. The result of genomic DNA blot showed that CAPR5-1 and CAPR5-2 existed as single copy in the Subicho genome. Three recombinant CPARs in E. coli were identified by SDS-PACE, and each expressed protein was treated on the PDA medium which contained cultured pathogens. Although three CAPR5 proteins did not affected the hyphal growth of Glomerella glycines and Colletotrichum fagenarium, CAPR5-1, CAPR5-2, and CAPR5-3 showed a specific antifungal activities against P. capsici.

  • To evaluate the antitumor activity of Paridis Rhizoma(PR), studies were done experimentally. The results were obtained as follows: 1. In cytotoxicity against MCF-7, SK-OV-3, HCT15, concentration inhibiting cell growth up to below 50% of control was recognized at $100-200{\mu}g/m{\ell}$ of PR and also against A549, XF498 was recognized at $50-100{\mu}g/m{\ell}$. 2. In Inhibitory effect on activity of DNA topoisomerase I, the $IC_{50}$ was shown $50-100{\mu}g/m{\ell}$ of PR. 3. The concentration inhibiting adhesion of A549 and SK-OV-3 to complex extracellular matrix up to below 50% of control was recognized at $10-100{\mu}g/m{\ell}$ of PR. 4. The T/C% was 137.9 in PR-treated group in S-180 bearing ICR mice. From above results it was concluded that PR could be usefully applied for the prevention and treatment of cancer.

  • Two new formulae for the design of beam-columns at room temperature have been proposed into Eurocode 3, prEN 1993-1-1 (2002), and are the result of great efforts made by two working groups that followed different approaches, a French-Belgian team and an Austrian-German one. Under fire conditions the prEN 1993-1-2 (structural fire design) presents formulae, for the design of beam-columns based on the prENV 1993-1-1 (1992). In order to study the possibility of having, in part 1-1 and part 1-2 of the Eurocode 3, the same approach, a numerical research was made using the finite element program SAFIR, developed at the University of Liege for the study of structures subjected to fire.

  • Magnetohydrodynamics(MHD) dynamo depends on many factors such as viscosity ${\gamma}$, magnetic diffusivity ${\eta}$, magnetic Reynolds number $Re_M$, external driving source, or magnetic Prandtl number $Pr_M$. $Pr_M$, the ratio of ${\gamma}$ to ${\eta}$ (for example, galaxy ${\sim}10^{14}$), plays an important role in small scale dynamo. With the high PrM, conductivity effect becomes very important in small scale regime between the viscous scale ($k_{\gamma}{\sim}Re^{3/4}k_fk_f$:forcing scale) and resistivity scale ($k_{\eta}{\sim}PrM^{1/2}k_{\gamma}$). Since ${\eta}$ is very small, the balance of local energy transport due to the advection term and nonlocal energy transfer decides the magnetic energy spectra. Beyond the viscous scale, the stretched magnetic field (magnetic tension in Lorentz force) transfers the magnetic energy, which is originally from the kinetic energy, back to the kinetic eddies leading to the extension of the viscous scale. This repeated process eventually decides the energy spectrum of the coupled momentum and magnetic induction equation. However, the evolving profile does not follow Kolmogorov's -3/5 law. The spectra of EV (${\sim}k^{-4}$) and EM (${\sim}k^0$ or $k^{-1}$) in high $Pr_M$ have been reported, but our recent simulation results show a little different scaling law ($E_V{\sim}k^{-3}-k^{-4}$, $EM{\sim}k^{-1/2}-k^{-1}$). We show the results and explain the reason.

Selection of Primula malacoides Lines with Different Growth and Flowering Characteristics (생장이 다양한 Primula malacoides 순계 선발)

  • To obtain pure lines for breeding $F_1$ varieties of Primula malacoides, 10 lines that were considered uniform in growth and flowering characteristics were selected from self-crossings of the sixth generation ($S_6$). Plant height of the selected lines ranged from 20.8 to 33.2 cm, and number of leaves ranged from 33.6 to 54.2. The selected lines flowered at 181.4 to 207.8 days after sowing. Three lines, including 'PR-03-81', flowered in less than 190 days from sowing, while six lines, including 'PR-03-228', only flowered after more than 200 days from sowing. In terms of petal color, six lines, including 'PR-03-81', were pink, three, including 'PR-03-236', were purple, and line 'PR-03-275' was white. Flower diameter ranged from 2.4 to 2.9 cm, and number of pedicels per plant was between 15.4 and 26.4. Number of flowers per pedicel ranged from 20.2 to 30.2. Germination was more than 70%, and number of seeds per capsule (pod) ranged from 110.0 to 162.8. In summary, plant height was low and flower diameter was small and plants had many flowers and readily formed seeds. The selected lines are not only valuable as ornamental plants but could also be used as breeding materials for $F_1$ hybrids of Primula polyantha.

Effect of Eu in Partial Oxidation of Methane to Hydrogen over Ln(1)-Ni(5)/SBA-15 (Ln = Dy, Eu, Pr, and Tb) Catalysts (Ln(1)-Ni(5)/SBA-15 (Ln = Dy, Eu, Pr, Tb) 촉매상에서 수소제조를 위한 메탄의 부분 산화 반응에서 Eu의 효과)

  • The catalytic yields of partial oxidation of methane (POM) to hydrogen over Ln(1)-Ni(5)/SBA-15 (Ln = Dy, Eu, Pr, and Tb) were investigated in a fixed bed flow reactor under atmosphere. As 1 wt% of Eu was added to Ni(5)/SBA-15 catalyst, the O1s and Si2p core electron levels of Eu(1)-Ni(5)/SBA-15 showed the chemical shift by XPS. XPS analysis also demonstrated that the atomic ratio of O1s, Ni2p3/2, and Si2p increased to 1.284, 1.298, and 1.058, respectively, and exhibited O-, and O2- oxygen and metal ions such as Eu3+, Ni0, Ni2+, and Si4+ on the catalyst surface. The yield of hydrogen on the Eu(1)-Ni(5)/SBA-15 was 57.2%, which was better than that of Ln(1)-Ni(5)/SBA-15 (Ln = Dy, Pr, and Tb), the catalytic activity was kept steady even 25 h. As 1 wt% of Eu was added to Ni(5)/SBA-15, the oxygen vacancies caused by strong metal-support interaction (SMSI) effect due to the strong interaction between metals and carrier are made. They are resulted in increasing the dispersion of Ni0, and Ni2+ nano particles on the surface of catalyst, and are kept catalytic activity.

Antibacterial Activity of Bioconverted Linoleic Acid Produced by Pseudomonas aeruginosa PR3

  • Crude extract of bioconverted linoleic acid using Pseudomonas aeruginosa PR3 was evaluated for its antibacterial activity against food-borne pathogenic bacteria. Crude extract showed antibacterial activity against four Gram-positive bacteria, Staphylococcus aureus (ATCC 6538), S. aureus (KCTC 1916), Listeria monocytogenes (ATCC 19166), and Bacillus subtilis (ATCC 6633), and one Gramnegative bacterium, Pseudomonas aeruginosa (KCTC 2004), with minimum inhibitory concentrations ranging from 750 to $1,500\;{\mu}g{\cdot}ml^{-1}$. S. aureus and B. subtilis were selected for growth inhibition assays with bioconverted linoleic acid. Major antibacterial effects occurred at lag phase.

  • Das Forschungsziel dieser Untersuchung ist es, den linguistischen Begriff der Analogie zu definieren, und die auftretenden analogischen Ausgleichsprozesse vom Mittelhochdeutschen zum Neuhochdeutschen und auch im Neuhochdeutschen selbst insbesondere im System der starken Verben vorzuzeigen und zu $erkl\"{a}ren$. Und diese Forschungsergebnisse sind wie folgt. $Au{\ss}er$ dem Phonemwandel, der immer das Phonemsystem tangiert, gibt es M$Lautver\"{a}nderungen$, die auf assoziativer Ubertragung von Lautungen aus konkurrierenden sprachlichen Formen beruhen, die sog. Analogiebildungen. $N\"{a}mlich$ in der sprachlichen Entwicklung vom Mittelhochdeutschen zum Neuhochdeutschen zeigt sich eine sehr starke innere Tendenz der Sprache, diese durch $gesetzm\"{a}{\ss}igen$ Lautwandel entstandene Formenvielfalt innerhalb eines Paradigmas zu vereinheitlichen. Das $hei{\ss}t$; Analogischer Ausgleich ist als universaller sprachlicher Simplifizierungsprozess zu werten.   • Hydrogen energy, a field of low-carbon substitute energy, can be produced by fossile fuel reforming and electrolysis of water etc. We developed 1kW class flat type reformer for PEM Fuel Cells. The PEMFC is highly sensitive to carbon monoxide because CO has detrimental effects on the performance of the fuel cell. Thus, reformed gas supplied to Fuel cell system, which maintained CO concentration below 10ppm. After applying optimum drive condition, reformed gas was measured with gas chromatography and could find out about each experimental condition of $H_2$ and CO concentration. As a results, The 1kW class plate type hydrogen generation system's optimum condition is A/F ratio ${\alpha}=1.3$, STR temperature 1023K, S/C ratio 3, and $PrOx1{\cdot}2$ 30cc/min. It turns out that installation of PrOx 2 stage is more efficient for reducing CO concentration.

  • $La_{2/3}TiO_3$, $La_2Ti_2O_7$ are used in various parts by dielectric properties. The purpose of the present study is to understand the photoluminescence properties of $Al^{3+}\;and\;Pr^{3+}$ doped perovskite-type $La_{2/3}TiO_3$ and pyrochlore-type $La_2Ti_2O_7$ phosphor, which characterized by the red emission $(^1D_2{\rightarrow}^3H_4)\;of\;Pr^{3+}$ of $Pr^{3+}$ ion. The explanation for the energy transfer and the corresponding critical distance were proposed on the role of Al^3+ ions as energy transfer mediates in perovskite-type $La_{2/3}TiO_3$:Pr phosphor. In order to clarify the distinction of photoluminescence propoerties between the perovskite-type $La_{2/3}TiO_3$ and the pyrochlore $La_2Ti_2O_7$, the trap-involved process and the charge transfer band have been investigated.
