A medium energy ion scattering spectroscopy(ME1S) system has been developed and tested.In the MEIS system a toroidal electrostatic energy analyzer(TEA) and a two dimensional position sensitivedetector(PSD) were used. The energy resolution of MEIS system was estimated to be less than $4\times 10^{-3}$ and the overall angular resolution was less than 0.3". From the MEIS spectrum of $Ta_2O_5$(300 $\AA$)/ onSi analyzedousing 60 keV $H^+$, the energy loss factor(S.1 and depth resolution were estimated to he 42 eV/$\AA$ and 9.7 $\AA$, respectively. Also Si(100) surface was analyzed using the MEIS system. A random MElSspectrum was obtained from thc Si(100) covered with native oxide layers. At the double alignment condition, MElS spectrum showed ;i Si surface peak, a oxygen peak and a carbon peak.nd a carbon peak.
This study was carried out to investigate the effect of intra-uterine infusion of lipopolysaccharides (LPS) on uterine involution in dairy cows with retained placenta. The LPS isolated from Bacteroides helcogenes and Fusobaoterium varium was infused at the rate of 100 ug (n=11) and 200 ug (n=11) with 30 ml of phosphate buffer saline in each cow at 20 days post-partum. The cows in conrol group (n=11) with retained placenta were infused with 30 ml phosphate buffer saline in each cow at 20 days postpartum. Cross-sectional ultrasonography was performed at days 40 postpartum to examine uterine involution and exudate in the all dairy cows in study. The cows in control group have shown 45.5, 27.3 and 27.3% gravid horn with less than 30 mm (normal), $31{\sim}50mm$ (medium) and more than 51 mm (large) in diameter. Respective average values of gravid horn diameter in cows treated with LPS 200 ug were noticed 72.7, 18.2 and 9.1%. However, the gravid horn diameter in all the cows treated with 100 ug of LPS was less than 30 mm. The 18.2, 63.6 and 54.6% cows in the control, LPS 100 ug and LPS 200 ug, respectively, have shown no exudate in uterine cavity. However, the respective rates of small amount of exudate retention in uterine cavity were 45.5, 0.0, and 9.1%. Approximately 36.4% cows in all 3 treated groups have shown minute quantity of exudate. In conclusion, the LPS intra-uterine infusion promoted postpartum uterine involution in dairy cows with retained placenta.
There are a lot of efforts to develop new compounds having skin whitening effect from natural products. Sparassis crispa is a medicinal mushroom containing more than 40% β-glucan, which exhibits anticancer and immunostimulating effects. The aim of this study was to assess the availability of β-glucan extracted from cauliflower mushroom S. crispa as a skin whitener through the evaluation of inhibitory effects of melanin synthesis and tyrosinase activity and their mechanisms. B16F1 cells were treated with S. crispa β-glucan (10, 100, and 1,000 ㎍/ml, respectively) and α-melanocyte stimulating hormone (α-MSH), simultaneously. Content of melanin synthesis and tyrosinase activity were determined. The expressions levels of tyrosinase, tyrosinase related protein-1 (TRP-1), TRP-2 and microphthalmia-associated transcription factor (MITF) were also measured by western blotting. Treatment with 10, 100 and 1,000 ㎍/ml S. crispa β-glucan and 200 nM α-MSH significantly decreased melanin synthesis by 13.9%, 18.7% and 39.5%, respectively, and tyrosinase activity by 15.6%, 26.9% and 43.2%, respectively, compared to the α-MSH alone group. In addition, S. crispa β-glucan inhibited expressions of tyrosinase, TRP-1, TRP-2 and MITF induced by α-MSH. These results indicated that S. crispa β-glucan inhibited MITF expression, thereby reducing tyrosinase expression and inhibiting melanin production in B16F1 melanoma cells. Therefore, S. crispa β-glucan might be available as a skin whitener.
Immunohistochemical studies on S-100 protein and lactoferrin were carried out to evaluate the existence and distribution pattern of S-100 protein and lactoferrin positive cells in salivary gland tumors. The specimens used were 25 cases of pleomorphic adenoma, 2 cases of monomorphic adenoma, 2 cases of mucoepidermoid tumor, 2 cases of acinic cell tumor, 3 cases of adenoid cystic carcinoma and 2 cases of adenocarcinoma occured in parotid and submandibular salivary gland. ABC kits(Dako corp. Copenhagen. Denmark) for S-100 protein and lactoferrin were used. The results obtained were summarized as follows: In the normal salivary gland. positive immunoreaction for S-100 protein was observed in myoepithelial cells of acini and intercalated ducts. Positive immunoreaction for lactoferrin was observed in serous acinic cells, epithelial cells of intercalated ducts, and excretory material in the ductal lumina. In the pleomorphic and monomorphic adenomas. most of tumor cells were positive for S-100 protein, while luminal tumor cells in gland-like or duct-like structures were rarely positive for lactoferrin. In mucoepidermoid tumor, most of squamous cells and a few of intermediate cells were positive for S-100 protein, but all of tumor cells were negative for lactoferrin. In acinic cell tumor, most of tumor cells were positive for lactoferrin, but all of tumor cells were negative for S-100 protein. In adenoid cystic carcinoma, basaloid tumor cells in trabecular structure were focally positive for S-100 protein. and in adenocarcinoma, many of tumor cells were posivive for both S-100 protein and lactoferrin. Thus, according to the embryonic stage of the development of the tumor cell origin, it was possible to classify the salivary gland tumor as followings: mucoepidermoid carcinoma which originated from the earliest stage, acinic cell tumor which originated from the end stage. Between these two extremes, there were pleomorphic adenoma, adenoid cystic carcinoma and adenocarcinoma which originated in the middle stage of the development of .the salivary glands. Based on the above results, it can be stated that S-100 protein is demonstrated in tumor cells orginated from myoepithelial cells and lactoferrin in glandular differentiated tumor cells.
The mutagenicities and antimutagenicities of butanol (PL I) and water (PL II) extracts from the filtrate of the cultured broth of Phellinus linteus were examined using the Ames/Salmonella test. No mutagenic activity of PL I and PL II was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PL I and PL II showed inhibitory effects on the mutagenic activities induced by the directly-acting mutagens, 4-nitro-o-phenylenediamine (NPD) using the tester strain TA98 and sodium azide (NaN3) using the tester strain TA 100 in the absence of S9 mix. PLI and PL II also showed inhibitory effects on the mutagenicities of the indirectly-acting mutagens, 2-aminofluorene (2-AF) using the tester strain TA98 and benzo[a]pyrene (B[a]P) using the tester strain TA 100 in the presence of S9. These results suggest that P. linteus has an antimutagenic activity and may play a role in the prevention of cancer.
5-fluorouracil (5-FU), a pyrimidine analog, is a widely used anticancer drug, which works through irreversible inhibition of thymidylate synthase. In the present study, it was investigated the anti-proliferative effects and molecular mechanisms of 5-FU using Ewing's Sarcoma CHP-100 Cells. The present data indicated that treatment of 5-FU to CHP-100 cells induced a G1 phase arrest of the cell cycle in a time-dependent manner. 5-FU-induced G1 arrest was correlated with the accumulation of the hypophosphorylated form of the retinoblastoma protein (pRB) and association of pRB with the transcription factors E2F-1 and E2F-4. Although 5-FU treatment did affect the levels of cyclin-dependent kinases, the levels of cyclin A and B were markedly down-regulated as compared with the untreated control group. In addition, 5-FU-induced G1 arrest of CHP-100 cells was also associated with the induction of apoptosis, as determined by apoptotic cell morphologies, degradation of poly(ADP-ribose) polymerase and Annexin V staining. Furthermore, 5-FU induced the loss of mitochondrial membrane potential with up-regulated pro-apoptotic Bax expression, down-regulated anti-apoptotic Bcl-2 expression and cytochrome c release from mitochondria to cytosol. Collectively, the data suggest that 5-FU is effective in inducing cell growth reduction and apoptosis, in part, by reducing phosphorylation of pRB and activating mitochondrial dysfunction in CHP-100 cells.

In this study, we analyzed the flooded area around Samcheok Middle School caused by typhoons MAYSAK·HAISHEN in September 2020. To analyze the confluence of Samcheok Osipcheon, local stream Deungbongcheon, we utilized Iber, a two-dimensional hydraulic model. We simulated the water depth and flood extent based on the peak flows on September 3 and September 7, 2020, and the 80 year and 100 year frequency floods. The simulation results showed that the 80-year frequency flood and the 100-year frequency flood on September 7 were insignificantly different, but the maximum flow rate from September 3 to September 7 was significantly different at 401 m3/s, resulting in a difference of 0.8 m in water depth and 7.1 m2 in flood area. In addition, the analysis that considered only the contour lines using contour lines predicted inundation of not only the Samcheok Middle School playground but also the building, confirming the need to apply DSM.
Two growth performance experiments were undertaken in order to compare the nutritional benefit of micronized dehulled barley (MDB) with wheat and maize. In the first experiment, ninety-six 24 d old Cotswold pigs with an average body weight of 9.5 kg were randomly allotted into one of four dietary treatments. The wheat and MDB ratio in the four complex diets were 100% wheat, 75% wheat+25% MDB, 50% wheat+50% MDB and 100% MDB; cereal was about 47% of the whole diet and varied by using different ratios of wheat and MDB. Starter phase was started at a bodyweight 9.9 kg; grower phase continued at liveweight 37.1 kg and finished at 74.0 kg. The ADFI, ADG and feed efficiency (F/G) were not affected (p>0.05) by the level of MDB in the diet from 9.9 to 37.1 kg live weight. However, ADFI was (p<0.05) less (14%) for the pigs fed the 75 or 100% of MDB based diets without affecting F/G during the second phase (37.1 to 74.0 kg live weight). Pigs fed the MDB based diet had a lower (p<0.05) plasma urea nitrogen (PUN) concentration than the pigs fed the wheat-based diet. In the second experiment, the trial started at 9.9 kg body weight and ended at market weight (107.6 kg), with three different phases in the trial. One hundred and twenty Cotswold pigs were randomly assigned to one of three dietary treatments. The maize and MDB ratio in the three diets were 100% maize, 50% maize+50% MDB and 100% MDB. Diet treatments did not affect (p>0.05) growth performance, PUN level, carcass characteristics and organ size, except for an improvement (p<0.05) in ADG for the pigs in the starter phase fed 50% and 100% MDB based diets.
A study on the performance and exhaust emissions of diesel engine with reducing exhaust gas temperature is performed experimentally. In this paper, experiments are performed at engine speed 2200rpm, 2600rpm and load 0%, 25%, 50%, 75% and 100% by test engine with F.W. cooler passing through exhaust gas. Main measured & analyzed parameters are exhaust gas temperature, specific fuel consumption, NOx and soot emissions etc. The obtained conclusions are as follows. (1) Specific fuel consumption is the least value at load 75% and it is decreased 1.5% after remodeling F.W. cooler. (2) NOx emission is the most value at load 100% and it is increased 30.1% after remodeling F.W. cooler. (3) Soot emission is the most value at load 100% and it is decreased 20.0% after remodeling F.W. cooler.
Stanozolol (STZ, 17$\alpha$-methyl-17$\beta$-hydroxy-5$\alpha$-androstano-(3,2-c) pyrazole), an anabolic steroid, is an abused drug by body-builders or atheletes, as well as medicine for treatment of aplastic anemia and vascular thrombosis. In human volunteers, the major urinary metabolite of STZ was reported to be 3'-hydroxystanozolol that was identified by gas chromatography-mass selective detector (GC/MSD). The objective of this experiment is to investigate the in vitro metabolism of STZ in liver S-9 faction of polychlorinated biphenyl-induced rats. Reaction mixture including STZ as substrate and the S-9 faction was extracted with diethyl ether and quantified by the selected ion monitoring mode of GC/MSD. The selected concentration of substrate STZ is 100 nmole and the selected time for incubation in the reaction mixture was determined to 60 min. The amount of 3'-hydroxystanozolol produced was increased by about 6-fold in the reaction medium including the liver S-9 fraction of polychlorinated biphenyl-induced rats, compared to that of untreated rats. Inhibitors of cytochrome P450, SKF-525A and 7,8-benzoflavone, decreased the production of 3'-hydroxystanozolol by about 89~100% and 65~75%, respectively; In conclusion, hydroxylation of STZ into 3'-hydroxystanozolol is confirmed by GC/MSD and is catalyzed by cytochrome P450.
