UPLC를 이용한 지역별 헛개나무(Hovenia dulcis) 플라보노이드의 정량분석 (Quantitative Analys is of Flavonoids in Hovenia dulcis by Region Using UPLC)

  • Hovenia dulcis is a herbal plant, which belongs to the Rhamnaceae family and is a native of Japan, China and Korea. Its fruit stalk is called 'Jiguja' in Korea. It has been traditionally used as a medicinal plant in East Asia. It was reported to have detoxification effects on alcohol poisoning, and antioxidant, antidiabetic etc. Sample of 5 g was extracted with 50 mL of 70% EtOH. The supernatant was filtered by 0.45 ㎛ membrane filter before analysis. The UPLC system was performed on Waters alliance UPLC HSS T3 column (2.1 × 100 mm, 1.7 ㎛) with a UV detector. The gradient system was a binary eluent of 0.1% formic acid in water(A) and 0.1% formic acid in acetonitrile(B) with gradient conditions as follows: Initial, 10% B; 1 min, 10% B; 4 min, 20% B; 10 min, 25% B; 12 min, 30% B; 14 min, 90% B; 17 min, 90% B; flow rate of 0.2 mL/min. The samples were injected by 2 µL and were detected at UV 355 nm. As a result of analysis, chromatographic patterns appeared in two cases: samples analyzed for ampelopsin and myricetin, and samples analyzed for taxifolin and quercetin. Among the four compounds, the largest regional difference was found to be taxifolin.

한국인 상용 식품 중 망간 함량 분석 (Analysis of Manganese Content in Frequently Consumed Foods by Koreans)

  • Using ICP-AES, we analyzed manganese content in 366 foods consumed frequently by Koreans. For the analysis, it was included a total of 366 foods such as 51 kinds of grains, 7 kinds of potatoes and starches, 7 kinds of sugars and sweeteners, 12 kinds of legumes, 11 kinds of nuts and seeds, 68 kinds of vegetables, 7 kinds of mushrooms, 33 kinds of fruits, 13 kinds of meats, 4 kinds of eggs, 48 kinds of fishes and shellfishes, 7 kinds of seaweeds, 16 kinds of milks, 8 kinds of oils and fats, 27 kinds of beverages, 34 kinds of seasonings, 13 kinds of processed foods and others. Among the grains, starches and sugars, manganese content of rice was 0.745 mg/100g. As for legumes, the content of manganese in soybean milk was 0.033 mg/100g and in black beans was 4.075 mg/100g. In nuts and seeds, the content of manganese in gingko nuts was 0.268 mg/100g while that in pine nuts was 8.872 mg/100g. Among the vegetables, manganese contents were 0.061 mg/100g in cherry tomato and 14.017 mg/100g in ginger. In mushrooms, the highest manganese content was displayed in ear mushroom at 10.382 mg/100g. Dried jujube and shrimp were found to be the fruits and fishes with high manganese contents at 2.985 mg/100g and 3.512 mg/100g, respectively. Among dairy foods, oils and beverages, manganese content was the highest in instant coffee powder at 2.577 mg/100g. Seasonings and processed foods posted 0.010 mg/100g in Sagolgomtang, instant soup and 23.846 mg/100g in pepper. In a furture, more various food for manganese content needs to be analyzed and a reliable food database should be compiled from the findings of researches in order to estimate manganese consumption accurately.

Twelve previously unrecorded bacterial species, isolated from the Nakdong River, South Korea

  • During a survey of indigenous prokaryotic species diversity of the upstream Nakdong River, South Korea, 12 bacterial strains were isolated for further analysis. These bacterial strains were identified showing at least 98.7% 16S rRNA gene sequence similarity with known bacterial species that were previously unreported in South Korea. The 12 bacterial strains were phylogenetically diverse and assigned to four classes, eight orders, nine families, and ten different genera. The isolates were identified as Leucobacter holotrichiae (99.1%), Leucobacter tardus (99.9%), Rhodococcus rhodochrous (99.9%), Tessaracoccus oleiagri (100%), and Paeniglutamicibacter cryotolerans (99.3%), of the class Actinobacteria; Bacillus coagulans (99.7%) and Bacillus wudalianchiensis (99.1%) of the class Bacilli; Ochrobactrum pseudogrignonense (99.2%) and Paracoccus thiocyanatus (100%) of the class Alphaproteobacteria; and Ideonella azotifigens (99.0%), Polaromonas glacialis(99.3%), and Herbaspirillum seropedicae (99.5%) of the class Betaproteobacteria. The cellular and colonial morphology, biochemical properties, and phylogenetic position of these isolates were examined, and species descriptions are provided.

Multilevel Analysis on Spatial Distribution and Socio-Environmental Factors of Dental Caries in Korean Children

  • This study aims to identify the regional distribution in the prevalence of dental caries and related multidimensional factors among 12-year-old children in Korea. Data from the 2018 Child Oral Health Survey were used to calculate the average DMFT index of 12-year-old children in metropolitan cities, and a multi-level regression model was applied to explain the regional distribution of dental caries prevalence and related factors. Factors were divided into two levels by administrative structure. This study finds a significant regional difference in the prevalence of dental caries in 12-year-old Korean children across metropolitan cities. Multilevel analysis showed that district-level factors (average number of pit and fissure-sealed permanent teeth, dental treatment demand rate, preventive treatment rate, sex ratio, and number of dentists per 100,000 people) and metropolitan-level factors (intakes of cariogenic beverages and number of pediatric dental hospitals and clinics per 100,000 people) had a significant effect on dental caries prevalence (p < 0.05). Individual characteristics and local socio-environmental factors influence the prevalence of dental caries. Especially considering the strong dependence on preventive treatment and accessibility to dental care services, it is necessary to provide adequate preventive treatment and expand health care resources in high-risk areas of dental caries.

유색미 색소체 및 페놀성화합물 함량과 항산화.항변이원성 간의 상관관계 (Correlation of Antioxidant and Antimutagenic Activity with Content of Pigments and Phenolic Compounds of Colored Rice Seeds)

  • 국내 외에서 수집 재배한 23품종 유색미의 색소 및 페놀성 화합물의 양을 측정하고 항산화성 및 항변이원성과의 상관성을 각각 비교하였다. 색소의 함량은 LK 1-3-6-12-1-1>LK ID 2-7-12-1-1>Elwee>Mutumanikam>IR 1544-38-2-2-1-2-2>wx 124-163-45-7-1-1-1, LK 1A-2-12-1-1 등의 순서로 나타났다. 폴리페놀화합물의 양은 IR 17491-5-4-3-3 품종이 현미 100g 당 0.244g으로서 가장 높은 함량을 보였으며, 그 다음은 LK 1-3-6-12-1-1>LK ID2-12-1>Elwee>Mutumanikam>IR 1544-38-2-2-1-2-2, LK IA-2-12-1-1 등의 순이었다. 유색미 품종별 색소의 함량은, 유색미 함유 생리활성 성분 중 총 폴리페놀 화합물의 함량과 높은 정의 상관성이 있었으며, 색도상의 lightness는 hydroxyl radical 소거활성과 정의 상관성이 있었으나, redness는 지질과산화 억제활성과 부의 상관성을 보였다.

가공식품 중 돈육 검출을 위한 샌드위치 ELISA 개발 (Development of Sandwich ELISA for the Detection of Pork in Processed Foods)

  • 가공식품 중 돈육의 검출을 위한 샌드위치 ELISA (sELISA)의 조건을 확립하기 위해 항돼지IgG 항체(goat anti-pig IgG antibody)를 이용하였다. 이때, 돈육의 검출범위는 $3-1,000ppm({\mu}g/mL)$이며, 검출한계는 2 ppm이었다. 특이항체의 교차반응 결과, 돼지IgG에 대한 반응성을 100%로 하였을 때, 돈육에 대한 반응성은 0.18%로 나타났으나, 여타의 식품원료에 대하여 반응하지 않았다. 열처리한 돈육에 대한 항체의 반응성은 $70^{\circ}C$까지는 32% 이상으로 양호하였으나 $80^{\circ}C$ 이상에서는 0.11% 이하로 반응성 급격히 감소하였다. 크림스프, 이유식, 어묵, 소스에 대한 spike test에서 돈육의 분석회수율은 각각 8.8, 45, 36, 39%로 나타났다. sELISA에 의하여 12점의 시판 가공식품 시료 중 돈육의 함유 유무를 조사한 결과, 정성적으로 원료의 표시사항과 100% 일치하였다.

애기장대의 AP2/ERF 전사인자인 AtERF73/HRE1의 프로모터에 있어서 저산소 반응 cis-조절 요소의 분석 (AtERF73/HRE1, an Arabidopsis AP2/ERF Transcription Factor Gene, Contains Hypoxia-responsive Cis-acting Elements in Its Promote)

  • 환경 스트레스 신호 인지부터 스트레스 반응 유전자의 발현에 이르는 신호전달 네트워크에 있어서, 스트레스 반응 프로모터의 cis-조절 요소와 거기에 결합하는 다양한 전사인자는 환경 스트레스에 대한 식물의 적응을 조절한다. 애기장대 AP2/ERF 전사인자 패밀리 중 그룹 VII ERF는 RAP2.12, RAP2.2, RAP2.3, AtERF73/HRE1, AtERF71/HRE2 유전자를 포함하며, 저산소 스트레스 반응에서 중요한 역할을 하는 것으로 알려져 있다. 본 연구에서는 HRE1 프로모터의 저산소 반응 부위를 동정하였다. 이를 위해 1,000 bp, 800 bp, 600 bp, 400 bp, 200 bp, 100 bp, 그리고 50 bp HRE1 프로모터 부위를 포함하는 벡터를 제작하여 프로모 터 활성을 분석한 결과, -200에서 -100 프로모터 부위가 HRE1의 저산소 반응에서 중요함을 확인하였다. HRE1의 -200에서 -100 프로모터 부위에는 저산소 반응 cis-조절 요소로 알려진 ERF-결합 부위와 DOF-결합 부위가 존재하는데, 이는 HRE1의 발현이 ERF 전사인자와 DOF 전사인자에 의해 조절될 수 있음을 시사한다. 전체적으로, 본 연구를 통해 HRE1 의 저산소 스트레스 반응에는 -200에서 -100 프로모터 부위에 존재하는 cis-조절 요소가 중요한 역할을 한다는 것을 확인하였다.

  • QPSK 통신 방식의 고속 통신 단말기에 필요한 저 전력 3V 6-bit 100MSPS CMOS ADC를 설계하였다. 제안된 ADC는 폴딩 블록, 래치 블록과 디지털 블록으로 구성하였다. 인터폴레이션 블록에서 pMOS를 전류원과 캐스코드형태로 합성하여 기존의 블록보다 선형적인 폴딩신호를 얻었으며 Kickback를 감소시키는 새로운 래치구조로 고속 ADC를 구현하였다. 설계된 칩의 Post-layout 시뮬레이션을 통하여 각 부분의 성능을 평가하였으며, 0.65um 2-poly 2-metal CMOS 공정으로 칩을 제작하였다. 제작된 칩은 대략 $1500{\mu}m{\times}1000{\mu}m$의 유효 칩 면적을 가지며, 실험결과 100MSPS의 속도로 3V 전원에서 40mW의 전력을 소모하며 INL은 ${\pm}0.6LSB$ 이내, DNL은 ${\pm}0.5LSB$ 이내, SNDR은 10MHz 입력 주파수에서 약 33dB의 실험결과를 얻었다.

녹조 납작파래 (Enteromorpha compressa)의 질산염 흡수, 배아 생장 및 지방산 조성에 대한 광 및 온도의 영향 (Effects of Light and Temperature on Nitrate Uptake, Germling Growth and Fatty Acid Composition of Enteromorpha compressa (Chlorophyta))

  • Effects of light and temperature on the nitrate uptake and germling growth of Enteromorpha compressa (L.) Greville (Chlorophyta) were studied based on samples from Cheongsapo near Busan, Korea. In addition, their effects on fatty acids composition in thallus were examined. Nitrate uptake showed saturation kinetics. $V_{max}$ (maximal uptake rate) and its $K_s$ (half-saturation constant) at $20^{\circ}C,\;80\;{\mu}mol\;m^{-2}s^{-1},$ white light were $1.571\;{\mu}mol{\cdot}g\;fr\;wt^{-1}{\cdot}h^{-1}$ and 3.56 ${\mu}M$, respectively. In nitrate uptake with irradiance, wavelength and temperature, its rate represented respectively the highest value as $1.405\pm0.020,\;0.623\pm0.040,\;1.422\pm0.022\;{\mu}mol{\cdot}g\;fr\;wt^{-1}{\cdot}h^{-1}\;at\;100\;{\mu}mol\;m^{-2}s^{-1},$ red light, $20^{\circ}C$ and exhibited significant difference among the examined conditions (p<0.001). Germling growth of E. compressa also showed saturation kinetics, and $V_{max}$ and its $K_s$ value at $20^{\circ}C,\;100\;{\mu}mol\;m^{-2}s^{-1},$ 12:12 h were $56.18\%\;day^{-1}$ and 0.33 ${\mu}M$, respectively. SGR (specific growth rate) recorded a maximal value as 49.33-54.80, 39.07-50.72, $47.20-54.53\%\;dat^{-1}$ at $120\;{\mu}mol\;m^{-2}s^{-1},$ blue light and $18^{\circ}C$ respectively, and showed significant difference (p<0.001). Red light made the effective nitrate uptake, but germling growth was largely limited by the light. In fatty acids analysis, PUFAs (polyunsaturated fatty acids) were high at blue light, $18^{\circ}C,\;100\;{\mu}M\;NO_3^-.$ However, irradiance did not affect the production of PUFAs. In conclusion, nitrate uptake and germling growth of E. compressa showed saturation kinetics to external nitrate concentration, and were significantly affected by irradiance, wave length and temperature. Fatty acid composition was also influenced by the factors except for irradiance. Their maximal values, together with the highest production of PUFAs, were found at blue light band, $20^{\circ}C,\;100\;{\mu}mol\;m^{-2}s^{-1},\;and\;100\;{\mu}M\;NO_3^-.$

윤충란 정량검사를 위한 Stoll 희석충란계산법과 Kato 후층도말변법의 비교 (Comparative Study on the Effectiveness of Modified Kato's Cellophane Thick Smear and Stoll's Dilution Egg Counting Technique for Quantitative Fecal Examination of Helminth Eggs)

  • Kato후층도말변법 (M.C.T.S.)과 Stoll 회석충란계산법(D.E.C.T)으로 회충, 괸충, 간흡 충란 양성자 197명에 대하여 충란 정량검사를 실시하고 그 결과를 비교하여 상관함수식을 도출하였다. M.C.T.S.법에 소요된 검경시간은 표본 1매당 평균 12.6분으로 D.E.C.T.법의 14.6분보다 짧았으며, 층계부하가 낮은 감염자에 있어서도 위음성이 적었다. 각 윤충류 충란에 있어서 M.C.T.S.으로 얻은 결과를 대변 1 g당 충란수(E.P.G.)로 바꾸어 주는 전환함수식은 회충의 경우 E.P.G.=47.86×100.87 logM.C.T.S., 편충은 E.P.G.=41.69×100.83 logM.C.T.S., 간흡충은 E.P.G=63.10×100.55 logM.C.T.S.이었다. 장내 윤충류 감염에 대한 정량적 대변검사법으로 Kato후층도말변법이 Stoll희석충란계산 법보다 더 유용하다고 생각되었다.
