Acute toxicity of antifouling agents(TBT, Sea-nine, Cu-pyrithione and Zn-pyrithione) to rockfish Sebastes schlegeli and amphipod Monocorophium acherusicum (방오도료(TBT, Sea-nine, Cu-pyrithione과 Zn-pyrithione)의 조피볼락 Sebastes schlegeli 과 단각류 Monocorophium acherusicum에 대한 급성독성 비교)

  • Since the usage of tributyltin(TBT) has been banned, many chemicals including Sea-Nine 211, Cu-pyrithione, and Zn-pyrithione were developed to use as antifouling agents for ships and coastal structures. However, the toxicity of these antifouling chemicals have not been systematically evaluated in ecotoxicological and biological studies. In this study, we investigated the effect of four antifouling substances on survival of estuarine rockfish, Sebastes schlegeli and amphipod, Monocorophium acherusicum. Survival of S. schlegeli and M. acherusicum during the 96-h exposure period were used to estimate the median lethal concentrations(LC50s) of test chemicals for each test species. Among antifouling agents, Cu-pyrithione($56{\mu}g{\cdot}1^{-1}$;96-h LC50) was most toxic to S. schlegeli, followed by $TBT(73{\mu}g{\cdot}1^{-1}),\;Sea-Nine(184{\mu}g{\cdot}1^{-1})\;and\;Zn-pyrithione(l707{\mu}g{\cdot}1^{-1})$, while TBT($26{\mu}g{\cdot}1^{-1}$) was most toxic to M. acherusicum followed by Sea-Nine($49{\mu}g{\cdot}1^{-1}$), Cu-pyrithione($119{\mu}g{\cdot}1^{-1}$) and Zn-pyrithione($334{\mu}g{\cdot}1^{-1}$). Effect concentrations of the antifouling chemicals estimated in this study can be used when assessing the potential risks of these substances, of which usage is increasing in the coastal environment.

Effect of Fish Number in Respiratory Chamber on Routine Oxygen Consumption of Black Porgy Acanthopagrus schlegeli Reared in Seawater or Freshwater (해수 및 담수사육 감성돔, Acanthopagrus schlegeli의 호흡측정실내 수용개체수에 따른 산소소비 비교)

  • Comparison of fish number on routine oxygen consumption (OC) of black porgy (Acanthopagrus schlegeli) reared in seawater (SW) versus freshwater (FW) was performed in closed water-recirculating system containing respiratory chamber. Fish acclimated in separate indoor tanks with SW or FW were sampled as two groups (one or three of fish used, $mean\;{\pm}\;S.D.=36.4{\pm}9.8$). OC of fish reared in both SW and FW showed clear temporal rhythms, with higher values in the daytime and lower values at night, in accordance with light (09:00-21:00 h) and dark (21:00-09:00 h) phases of the diel cycle (12L:12D). The OC of the fish increased linearly with the water temperature. The OCs of black porgy reared in SW and FW at 15, 20, $25^{\circ}C$ were 140.0, 174.8, 282.3 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ and 177.5, 307.8, 413.1 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ for the one of fish used, and 200.5, 274.7, 339.1 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ and 118.2, 188.2, 252.8 mg $O^2{\cdot}kg^{-1}{\cdot}h^{-1}$ for three of fish used, respectively. Black porgy reared in SW had higher ventilation rates than those in FW. These results indicate that black porgy reared in FW had higher OC than those in SW at the one of fish used, but the fish reared in SW had higher OC than those in FW at the three of fish used.

Regulation of Copulation and Parturition in Black Rockfish, Sebastes Schlegeli by Photoperiod and Temperature (광주기와 수온에 의한 조피볼락, Sebastes schlegeli의 교미 및 출산 조절)

  • Regulation of copulation and parturition of black rockfish, Sebastes schlegeli were induced by artificially controlled photoperiod and temperature. for the treated group, daylength of copulation and parturition period was maintained at 9L : 15D and 14L : l0D, respectively. Temperature was maintained at 12~13$^{\circ}C$ during the copulation period, and at 14~15$^{\circ}C$ during the parturition period. Gonadosomatic index (GSI) of male was low 0.12 from January to August and peak to 1.36~0.10 during November in the control group but 1.27 ~ 0.05 during October in treated group. GSI of female was about 1.82 from January to November 1996 and reached the maximum of 46.5 ~ 5.72 during April 1997 in the control group but 42.0~4.24 during february 1997 in the treated group. The pattern of ovary and testis development did not much differ between the treated and control groups. However, the copulation and parturition of black rockfish could be advance 2~3 months by controlling photoperiod and temperature.

Effects of Dietary Supplement of Probiotics on Growth and Blood Assay of Rockfish Sebastes schlegeli (생균제 첨가사료가 조피볼락 Sebastes schlegeli의 성장 및 혈액성분에 미치는 영향)

  • We determined the effects of dietary supplements for lactic acid bacteria(LAB) such as Lactobacillus brevis(Lb) and Lactobacillus plantarum(Lp) in juvenile rockfish Sebastes schlegeli cultured in flow-through system for 10 weeks. The experimental diets contained $10^4cfu/g,\;10^6cfu/g\;and\;10^8cfu/g$ level each LAB(Lb-4, Lb-6, Lb-8 or Lp-4, Lp-6, Lp-8), respectively. The effects of LAB supplementation was determined by various factor such as weight gain(WG), specific growth rate(SGR), feeding efficiency(FE) and blood assay. For rearing experiment, Lp-8 treatment had significantly high growth rate than control diet treatment. However, all Lb treatment had no significance effect with control diet treatment. In case of the blood assay, hematocrit(Ht) and hemoglobin(Hb) of fish were not affected by LAB supplemental levels. On the other hand, total cholesterol in plasma of Lb-8, Lp-6 and Lp-8 treatments were significantly low than the control diet treatment. We verified the influence of LAB which was originated from species specificity and amount in diet. Consequently, the dietary supplementation as $10^8cfu/g$ level of L. plantarum could be of help for growth enhancement to the juvenile rockfish.

Pathogenicity of Iridovirus against Marine Fish and Its Detection in Culturing Seawater (Iridovirus의 해산 양식어류에 대한 병원성과 사육수에서의 검출)

  • The susceptibility of five different marine fish to iridovirus IVS-1 infection was analyzed and found a higher the cumulative mortality in the order of rock bream (Oplegnathus fasciatus), red sea bream (Pagrus major), sea perch (Lateolabrax sp.), rockfish (Sebastes schlegeli) and black porgy (Acanthopagrus schlegeli). However, the concentrations of virus in the infected spleens of these species did not differ significantly. To determine the release of iridovirus from infected fish into culturing seawater, rock bream were challenged with iridovirus IVS-1 and the concentration of virus in the water was analyzed using PCR. Over the 10 days of the analysis, the linear relationship between the number of dead fish and viral DNA concentration found in culturing seawater should be considered direct evidence of horizontal iridovirus transmission.

Species Composition and Behavioral Characteristics of Released Black Rockfish, Sebastes inermis in the Coastal Waters off Namhae Island, Korea (남해도 주변해역에서 서식하는 어류의 종조성 및 양식산 방류 볼락(Sebastes inermis)의 행동특성)

  • Seventeen fish species were counted in an underwater visual census, and the most common species, Sebastes inermis and Halichoeres poecilopterus, were released. More than 60% of the released S.inermis occurred near a fish farm 5 days after release, but they moved to a distant rocky area, and decreasing numbers were seen near the farm 90 days after release. In pots, a total of 722 individuals belonging to 43 species occurred, and released S.inermis dominated with 174 individuals recaptured during the study period. The other common fish species were Stephanolepis cirrhifer and Acanthopagrus schlegeli. The percentage of recapture was 0.23%, but declined to 0.12% at 90 days after release. The potential predators of released S.inermis included Lateolabrax japonicus and Sebastes schlegeli.

Differences by RAPD-PCR Analysis within and between Rockfish (Sebastes schlegeli) Populations from the Yellow Sea and the Southern Sea in Korea (황해 및 남해산 조피볼락 (Sebastes schlegeli) 개체군 사이의 RAPD-PCR 분석에 의한 차이)

  • Polymerase chain reaction (PCR) amplification of DNA as 30 different arbitrary primers and random amplified polymorphic DNAs (RAPD) analysis were performed on genomic DNA extracted from the blood of the marine rockfish (Sebastes schlegeli) from the Yellow Sea and the Southern Sea. The unique properties of the genomic DNA were used to investigate the features of the population dynamics and origins of the species. Out of 30 primers, seven generated 207 highly reproducible RAPD polymorphic products, producing approximately 2.7 polymorphic bands per primer. About 67.4% of total amplified products (307) were either polymorphic (207) to rockfish. The degree of similarity varied from 0.22 to 0.63 as calculated by bandsharing analysis. Also, the average level of bandsharing was 0.39$\pm$0.02 within the rockfish strains. The electrophoretic analysis of RAPD-PCR products showed the relatively high levels if variation between different individuals in rockfish from the Yellow Sea. However, the RAPD outlines obtained with DNA of different rockfish strains from the Yellow Sea and the Southern Sea in Korea were very similar. Also, a small number of polymorphic bands were identified. Even if further analyses or more rockfish populations are required, this result implies RAPD analysis reflects genetic differences between the geographical strains of the rockfish.
