Bacillus species have been involved in metal association as biosorbents, but there is not a clear understanding of this chelating property. In order to evaluate this metal chelating capacity, cultures and spores from Grampositive bacteria of species either able or unable to produce surface layer proteins (S-layers) were analyzed for their capacity of copper biosorption. Only those endowed of S-layers, like Bacillus sphaericus and B. thuringiensis, showed a significant biosorption capacity. This capacity (nearly 50%) was retained after heating of cultures, thus supporting that structural elements of the envelopes are responsible for such activity. Purified S-layers from two Bacillus sphaericus strains had the ability to biosorb copper. Copper biosorption parameters were determined for strain B. sphaericus 2362, and after analyses by means of the Langmuir model, the affinity and capacity were shown to be comparable to other bacterial biosorbents. A competitive effect of $Ca^{2+}$ and $Zn^{2+}$, but not of $Cd^{2+}$, was also observed, thus indicating that other cations may be biosorbed by this protein. Spores that have been shown to be proficient for copper biosorption were further analyzed for the presence of S-layer content. The retention of S-layers by these spores was clearly observed, and after extensive treatment to eliminate the S-layers, the biosorption capacity of these spores was significantly reduced. For the first time, a direct correlation between S-layer protein content and metal biosorption capacity is shown. This capacity is linked to the retention of S-layer proteins attached to Bacillus spores and cells.
Two hundred and sixteen single comb white egg layers of the White Leghorn hens of 24 weeks of age were randomly allocated to 12 groups with three replications of six hens in each. Hens were fed in a factorial arrangement 2${\times}3{\times}$2, on diets containing either 16 or 18% crude protein with 0, 3 or 6% fish meal, replacing groundnut meal with soybean meal. Soybean meal incorporation improved (p<0.05) egg production, feed intake, feed conversion efficiency and egg weights. Egg quality traits of specific gravity, shape index, albumen index, yolk index and shell thickness remained unchanged. Laying performance was significantly (p<0.05) better at 18% than on 16% dietary protein level. Use of fish meal linearly improved egg production and feed conversion efficiency on diets supplemented with groundnut meal and fish meal incorporation showed quadratic improvement on feed conversion efficiency with SBM diets at 16% dietary protein level. Therefore, use of soybean meal as substitute of groundnut meal is recommended in layer diets, at 16% dietary protein level and fish meal incorporation could be beneficial for layers.

A Lactobacillus sp. has been isolated from infant feces and characterized according to its physiological properties and identified as Lactobacillus acidophilus KLA1012. A gene coding surface layer protein (SLP) has been cloned and the sequence has been determined. The nucleotide sequence of slpA was 1,338 bp in size and was identical to that of L. acidophilus ATCC 4356 (100%). Amino acid sequence of SLP-A was deduced from the nucleotide sequence and it had signal sequence at N-terminal, consisting of positively charged amino acid mainly lysine. slpA was cloned and heterologously expressed in E. coli M15 and the 45.2 kDa surface-layer protein band was examined by SDS-PAGE and confirmed by Western blotting using polyclonal antibody against L. acidophilus KLA 1012 SLP-A protein.

This study was carried out to enhance the nutritional value of whole crop corn silage and the utilization of animal waste as a ruminant feedstuffs. Whole crop corn were ensiled with cage layer excreta and concentrate. Treatments included whole crop corn silage (control), 10% cage layer excreta and concentrate-corn silage (CES lo), 20% cage layer excreta and 10% concentrate-corn silage (CES 20) and 30% cage layer excreta and 10% concentrate-corn silage (CES 30). The characteristics of silage fermentation was evaluated. Digestibility and palatability of the silage were observed through the feeding trial with four male Corridale sheep. The results obtained are summarized as follows: 1. Cage layer excreta-corn silage showed higher contents of crude protein and crude ash than control silage, while crude fibre content was significantlv lower than that of control silage (P(.01). 2. Total nitrogen and ammonia nitrogen content of the cage layer excreta-corn silage was significantly (P< .01) higher than that of control, but CES 10 silage was good in quality. 3. Lactic acid content of CES 10 silage was the highest, but that of CES 20 and CES 30 silage was lower than that of control. CES 20 and CES 30 silage was lower lactic acid content and higher pH than control. Therefore Flieg's score was wrose than that of control silage. 4. The numbers of total bacteria and lactic fermentation bacteria were estimated lo7-10' and 10"1OH, respectively, and there were no significant differences among treatments. Coliform numbers were not detected with CES 10 silage while were detected lo3 per gram in CES 30 silage. 5. Digestibility of crude protein, crude fibre and ADF was improved significantly in the animals fed the cage layer excreta-corn silage compared with animals fed the control silage. DCP and TDN were the highest in the CES 10 silage. 6. The intake of daily nitrogen and dry matter per metabolic body weight was the highest in animals fed the CES 10 silage.lage.
The study on the nutritive value and the potential use of two kinds (local vs. import) of sesame meal (SSM) in layer diets as substitute for soybean meal (SBM) at 25, 50, and 75% was carried out. Three hundred and thirty six layers were kept individually on a battery cage and fed isonitrogenous diets, 16% crude protein (CP), for 252 days. The local and the imported SSM contained on air dry basis 35.7 and 36.3% CP, 24.7 and 9.1% ether extract (EE), respectively. Their amino acid contents are all lower than those reported by NRC (1984). The local material contained remarkably lower lysine (0.44 vs. 0.94), methionine + cystine (1.06 vs. 1.75) and threonine (0.52 vs. 1.22) than the imported meal. Imported SSM could be substituted for SBM as high as 50% in layer ration without statistically adverse effect on egg production, feed intake, body weight gain, average egg weight and egg size, while local SSM was inferior to the control in all aspects. However, when production performance of the groups fed either kind of SSM was compared, no significant difference was found at the two lower substitution levels.
Using recombinant Vaccinia virus(vSC8) that express ${\beta}$-galactosidase, a model heterologous protein, conditions for virus and protein production were investigated in tissue culture flask. As host animal cells HeLa and HeLa S3 were used. It was demonstrated that cells infected during the exponential growth phase gave higher protein yield than those infected during the stationary growth phase and calf serum concentration after virus infection did not significantly alter protein yield. Pretreatment of cell layer with hypotonic solution enhanced the virus infectivity. Optimum cell growth and recombinant protein production was achieved at $37^{\circ}C$. But, during 2 hours of virus infection period incubation temperature must be lowered to 20∼$30^{\circ}C$ for maximum recombinant protein yield. To enhance virus replication, the effects of adrenal glucocorticoid hormone (Dexamethasone) and silkworm hemolymph were evaluated. Only dexamethasone increased about 20% of ${\beta}$-galactosidase yield in HeLa S3 cells when added with 10-7∼10-5M concentration 24 hours before infection.
To study the effects of utilizing extruded full fat soybean (FFS) in layer diets, 300 White Leghorn pullets were put in trial divided into 5 treatments; control (commercial soybean meal as protein source), FFS 50% (replacing 50% of soybean meal with FFS), FFS 100% (replacing all amount of soybean meal with FFS), soybean oil and tallow added treatment. Each treatment consisted of 5 replicates of 12 birds. Egg production and feed efficiency from hens fed control diet were significantly lower than from hens fed the other diets. Significant differences in egg production and feed efficiency were found between the control and the other treatments. Cholesterol concentration of serum and egg yolk were not affected by any dietary treatment. Feeding extruded full fat soybean did not cause pancreatic hypertrophy nor change in mortality. There was an indication that linoleic acid (C18:2), linolenic acid (C18:3) and iodine contents increased in the thigh and egg yolk lipid of the groups fed FFS or soybean soybean can serve as effective protein source for layer diets if economically justified.
Uremia is associated with defective protein binding of weakly acidic drugs, whereas the protein binding of basic drugs tends to be normal. The exact chemical nature of compound(s) and mechanism for these changes as yet is unknown, and has not been defined. Organic solvent extraction of pooled normal human urine following hydrolysis by hydrochloric acid produced an extract, which when added to normal human serum, was capable of inducing binding defects similar to those in uremia. Binding defects were observed with the weakly acidic drugs such as nafcillin, salicylate, sulfamethoxazole and phenytoin while the binding of the basic drugs such as trimethoprim and quinidine were unaffected. The binding defects induced by the hydrolyzed urine extract could readily be corrected by same organic solvent extraction of acidified serum and the defects could be transferred to the normal human serum using the organic solvent layer at the physiologic pH (7.4). Followed by reacidification ind extraction of the binding defects induced serum with the same solvent, separated several fractions were obtained on thin-layer chromatography. One of these fractions could reinduce the binding defects and this factor(s) is apparently weakly acidic compounds and tightly bound to serum at physiologic pH, but extractable at acidic pH, and its molecular weight range is approximately 500 or less similar to those seen in uremia. These findings strongly support the hypothesis that the drug binding defect in uremia is due to the accumulation of endogenous metabolic products which arc normally excreted by the kidneys but accumulate in renal failure.
Corn distiller's dried grains with solubles (DDGS) is a completely new feed ingredient in the Korean feed market. There is an ever increasing need for the Korean feed industry to import and make the best of it as a high protein and high energy feed ingredient. A layer feeding trial was conducted for 10 weeks to investigate the effects of addition of light-colored DDGS to layer diets on laying performance, egg qualities and yolk fatty acid composition. Also, the economics of using DDGS in the Korean situation was analyzed. Nine hundred Hy-line Brown layers, 24 weeks of age, were employed in a feeding trial consisting of four dietary treatments (0, 10, 15, and 20% DDGS), and five replicates per treatment. All experimental diets were prepared as iso-protein (17%) and iso-calorie (TMEn 2,780 kcal/kg). The use of DDGS up to 20% in layer diets did not exert any influence on feed intake, laying rate, total egg mass, mean egg weight and feed conversion ratio (p>0.05). The color and breaking strength of eggshell, as well as the albumin height and Haugh unit were not affected by the addition of DDGS up to 20% in the diet. The yolk color was significantly increased by DDGS supplementation (p<0.05). As the DDGS level increased, the oleic acid content decreased, and the linoleic acid increased (p<0.05) in egg yolk. The degree of saturation of yolk fatty acids was not affected by DDGS supplementation. The inclusion of light-colored DDGS up to 20% in layer diets resulted in a decrease of feed cost per kg without any undesirable effect on laying performance. In conclusion, the light-colored DDGS (L* 56.65) could be used up to 20% in layer diets without any harmful effect on laying performance, and possibly provide economic benefits to the Korean poultry industry.

Background: The objective of this study was to evaluate the changes in gene expression after incubation of cells with proteins released from different silk mat layers. Methods: A silk cocoon from Bombyx mori was separated into four layers of equal thickness. The layers were numbered from 1 to 4 (from the inner to the outer layer). The proteins were released by sonication of a silk mat layer in normal saline. The concentration of proteins was determined by spectrophotometry. They were incubated with RAW264.7 cells, and changes in the expression of genes were evaluated by cDNA microarray analysis and quantitative reverse transcriptase-polymerase chain reaction (qRT-PCR). Results: Layer 1 and 4 groups had higher protein concentrations compared to those in layer 2 and 3 groups. The genes associated with inflammation and angiogenesis showed significantly higher expression in layer 1 and 4 groups. The results of qRT-PCR were in agreement with those of the cDNA microarray analysis. Conclusions: The silk mat from the middle portion of the silkworm cocoon yielded a lower protein release and caused an insignificant change in the expression of genes that are associated with inflammation and angiogenesis.
