Antitumor Activity and Nephrotoxicity of the Novel Platinum(II) Coordination Complex (새로운 Platinum (II) Complex [Pt (II)(trans-d-dach)(DPPE)] $(NO_3)_2$의 항암효과 및 신독성에 관한 연구)

  • Platinum coordination complexes are currently one of the most compounds used in the treatment of solid tumors. However, its use is limited by severe side effects such as renal toxicity. Our platinum-based drug discovery program is aimed at developing drugs capable of diminishing toxicity and improving antitumor activity. We synthesized new Pt (Ⅱ) complex analogue containing 1,2-diaminocyclohexane (dach) as carrier ligand and 1,2-bis(diphenylphosphino) ethane (DPPE) as a leaving group. Furthermore, nitrate was added to improve the solubility. A new series of [Pt(trans-ddach)(DPPE).$2NO_3(PC)$ was synthesized and characterized by their elemental analysis and by various spectroscopic techniques [infrared (IR), $^{13}carbon$ nuclear magnetic resonance (NMR)]. PC demonstrated acceptable antitumor activity aganist P388, L-1210 lymphocytic leukemia cells and SK=OV3 human ovarian adenocarcinoma cells, and significant. activity as compared with that. cisplatin. The toxicity of PC was found quite less than thar of cisplatin using MTT, $[^3H]$ thymidine uptake and glucose consumption tests in rabbit proximal tubule cells, human kidney cortical cells and human renal cortical tissues. Based on these results, this novel platinum compound represent a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low toxicity.

Effect of Long Term Buchu (Chinese chives) Diet on Antioxidative System of ICR Mice (장기간의 부추식이가 ICR 마우스의 항산화시스템에 미치는 영향)

  • To evaluate the antioxidative and antiaging effects of buchu in vivo system, 2% or 5% buchu diets were fed to ICR mice for 13 months and lipid peroxidation, protein oxidation, activities of antioxidative enzymes and total glutathione content on liver were measured. Hepatic TBARS contents did not show differences among diet groups, while buchu diet suppressed the protein oxidation significantly. SOD activities of control diet group decreased slowly after 7 month but buchu diet increased its activities steeply for first 3 month and continued to increase twice or three times higher than control diet during 13 month. While GSH-Px activities of control diet group were increased slightly with age, buchu diet increased its activities twice or three times higher than control. While catalase activities of control diet group were almost not changed with age, buchu diet increased its activities in both 2% and 5% diet groups. Total hepatic glutathione contents were gradually increased with age, while buchu diets In-creased its contents remarkably. According to this study, many antioxidative materials and sulfides compounds containing buchu seems to protect antioxidative systems on ICR mice.

Gene Expression Profiling in Diethylnitrosamine Treated Mouse Liver: From Pathological Data to Microarray Analysis (Diethylnitrosamine 처리 후 병리학적 결과를 기초로 한 마우스 간에서의 유전자 발현 분석)

  • Diethylnitrosamine (DEN) is a nitrosamine compound that can induce a variety of liver lesions including hepatic carcinoma, forming DNA-carcinogen adducts. In the present study, microarray analyses were performed with Affymetrix Murine Genome 430A Array in order to identify the gene-expression profiles for DEN and to provide valuable information for the evaluation of potential hepatotoxicity. C57BL/6NCrj mice were orally administered once with DEN at doses of 0, 3, 7 and 20 mg/kg. Liver from each animal was removed 2, 4, 8 and 24 hrs after the administration. The histopathological analysis and serum biochemical analysis showed no significant difference in DEN-treated groups compared to control group. Conversely, the principal component analysis (PCA) profiles demonstrated that a specific normal gene expression profile in control groups differed clearly from the expression profiles of DEN-treated groups. Within groups, a little variance was found between individuals. Student's t-test on the results obtained from triplicate hybridizations was performed to identify those genes with statistically significant changes in the expression. Statistical analysis revealed that 11 genes were significantly downregulated and 28 genes were upregulated in all three animals after 2 h treatment at 20 mg/kg. The upregulated group included genes encoding Gdf15, JunD1, and Mdm2, while the genes including Sox6, Shmt2, and SIc6a6 were largely down regulated. Hierarchical clustering of gene expression also allowed the identification of functionally related clusters that encode proteins related to metabolism, and MAPK signaling pathway. Taken together, this study suggests that match with a toxicant signature can assign a putative mechanism of action to the test compound if is established a database containing response patterns to various toxic compounds.

Comparison of Antifungal Activites of Monoterpenes and Sesquiterpenes in Essential Oil from Chamaecyparis obtusa against Dermatophytes (피부사상균에 대한 편백정유의 Mono- 및 Sesquiterpene 항진균 활성 비교)

  • This study was to compare the antifungal activitiy of monoterpenes with that of sesquiterpenes, which were major constituents of antifungal active essential oil of Chamaecyparis obtusa, to investigate which constituents had more effective against dermatophytes, and to evaluate the synergistic effect of combination of the antifungal active constituents. The antifungal activities of seven fractions (A, B, C, D, E, F, and G) from C obtusa essential oil by column chromatography were tested against Trichophyton mentagrophytes. Fraction D, E, F, and G were more active than fraction A, B, and C, and their major compounds were mono- and sesquiterpenes analyzed by GC/MS. Borneol, linalool, and ${\alpha}$-terpineol were selected as monoterpenes, and ${\alpha}$-cedrol, nerolidol, and ${\beta}$-eudesmol as sesquiterpenes, respectively. The antifungal activities of the constituents were respectively examined against Trichophyton mentagrophytes (KCTC6077), Microsporum canis (KCTC6591), and Microsporum gypseum. The constituents of sesquiterpenes were more active than those of monoterpenes. By comparing single and combined sesquiterpenes with fraction F containing the higher ratio of sesquiterpenes, combined sesquiterpenes were the most active. This result indicated that there was the synergistic antifungal effect against dermatophytes when sesquiterpenes were combined together.

Effect of a Soil Amendment for Controlling Fusarium Wilt of Cucumber caused by Fusarium oxysporum f. sp. cucumerinum (오이 덩굴쪼김병(Fusarium oxysporum f. sp. cucumerinum) 방제에 대한 토양첨가제의 효과)

  • In order to find out effect on the inorganic and organic compounds as a soil amendment to control Fusarium wilt of cucumber caused by Fusarium oxysporum f. sp. cucumerinum, this study was conducted during the last two years from 1993 to 1995. According to add 14 inorganic chemicals (1%, w/v) including $Al_2(SO_4)_3$ individually in vitro, these chemicals were confirmed their suppression, and especially Alum, CaO and $Al_2(SO_4)_3$ suppressed not only $20.9{\sim}25.0$ percent on mycelial growth of the fungus, but also inhibited $72.8{\sim}97%$ on conidial germination. $Ca(NO_3)_2$ suppressed mycelial growth only, while KCl, $K_2SO_4,\;NH_4NO_3$, and Urea suppressed conidial germination. The 7 chemicals were finally seleced. Composted pine bark (CPB) suppressed definely more than 90% on conidial germination in the different extract concentration (2,5 and 10%), although mycelial growth on extract medium of CPB and milled alfalfa leaves (MAL) were not remarkable. The antagonist Trichoderma sp. (Tr-3) mixed with an amended soil (1%, w/w) containing composted pine bark showed a good mycelial growth to compete the causal fungus. And the antagonist Pseudomonas sp. (7-1-3) was also confirmed its antagonistic ability with culture filterate. It is known that a CPB soil amendment mixed with the two antagonists (1%, w/w) controlled almost completely Fusarium wilt of cucumber in greenhouse pots and a field experiment. It is therefore expected that biocontrol on Fusarium wilt of cucumber by a soil amendment can be applied to farmmer's fields.

Identification of feeding Stimulants to Improve Efficiency of Diet for flatfish (사료 효율 개선을 위한 넙치 섭식촉진물질의 검색)

  • In order to identify the feeding stimulants for flatfish, the feeding tests were conducted using some diets added the amino acids, nucleotides, betaine and TU and acid hydrolysates to basal diet formula, respectively, The feeding stimulant activities and synergistic effects of those compounds were evaluated by a bioassay method. The KH, a kind of acid hydrolysate, possessed a remarkable feeding stimulant activity. It's stimulant activities were increased up to the concentration of $1.05\%$ (w/v), and were independent of pH. The formulation of KH and glycine had a most synergistic effect, The feeding rate of diet with feeding stimulation was 1,4 fold than that of diet without one. The costs for optimum formulation of feeding stimulants were cheaper than some additives in diets. It suggest that the results can practically used in preparation of diet containing feeding stimulant effects for flatfish.

Screening for Antimutagenic Effects of the Wild Mushrooms in Korea (한국산 야생버섯류중 항변이원성 효과의 검색)

  • To detect naturally occuring antimutagenic substances from wild mushrooms in Korea, the screening for the antimutagenic compounds containing in ethanol and water extracts of 13 wild mushrooms toward benzo(a) pyrene (B(a)P) and $N-methyl-N'-nitro-N-nitrosoguanidine\;(MNNG)$ using the Ames assay system with Salmonella typhimurium TA98 and TA100 were studied. The ethanol extracts of Polyporus dispansus, Cantharellus infundibuliformis, Agaricus subrutilescens, Daedalea dickinsii, Panaeolus papilionaceus and Hydnum repandum showed significantly antimutagenic activity toward B(a)P. The water extracts of Hydnum repandum showed the strong antimutagenic activity toward B(a)P in S. typhimurium TA100, however the water extracts of the mushrooms did not show antimutagenic activity. Whereas 5 out of 13 samples exhibited antimutagenicity toward a direct mutagen of MNNG. The water extracts from mushrooms also not showed antimutagenic activity. The antimutagenic effect increased with increasing concentraion of the ethanol extracts from Polyporus dispansus.

  • Two major flavonoid compounds having inhibition activity of xanthine oxidase from onion skin were separated, isolated and identified by ODS chromatography, Sephadex LH-20 chromatography, UV/visible absorption spectroscopy and FAB Mass. Spectral analyses indicated that $F_1$ was a flavonol having 3,5,7,3'-OH and 4'-glucoside (quercetin 4'-glucoside), and $F_2$ was a flavonol having 3(5),7,3',4'-OH (quercetin). FAB-Mass of fraction $F_1\;and\;F_2$ in positive-ion-mode produced a spectra containing main ions at m/z 465, corresponding to the $(M+H)^+$ ion of quercetin 4'-glucoside, and m/z 303, corresponding to the $(M+H)^+$ ion of quercetin. The inhibition mechanisms of $F_1\;and\;F_2$ were a mixture of the uncompetative and non-competative modes, with respect to xanthine as a substrate.

Antioxidant Properties in Microbial Fermentation Products of Lonicera japonica Thunb. Extract (금은화 추출물을 이용한 미생물 발효 생성물의 항산화 특성)

  • The purpose of this study is to investigate antioxidant properties in microbial fermentation products of Lonicera japonica Thunb extract. The bacterium Lactobacillus plantarum NHP1 was isolated from conventional fermented foods. Modern pharmacological studies show that Lonicera japonica Thunb and its active principles of wide pharmacological actions. For instance, they show a strong efficacy in antibacterial, anti-inflammatory, antiviral, anti-endotoxin, blood fat reducing, antipyretic, and antioxidant activities. The extract of Lonicera japonica Thunb was obtained by extracting dried Lonicera japonica Thunb using either hot water or 70% ethanol as a solvent. Fermentation was performed in a 2L fermentor containing 1.2 L of extractat conditions of $30^{\circ}C$ and 100 rpm for 48 hr. The amount of cholorogenic acid was $2.65{\mu}g/g$ in hot water extract. The total phenolic content (GAE, gallic acid equivalent) in hot water and 70% ethanol were $56.5{\pm}4.9$ GAE mg/g and $72.7{\pm}5.3$ GAE mg/g, respectively. After fermentation, the phenolic content increased to 30.2% in hot water and 12.9% in ethanol extract. In the same manner, flavonoid content increased to more than 75% regardless of extract solvent. ABTS (2,2-azino-bis-3-ethylbenzthiazoline-6-sulfonic acid) value noticeably increased to 50% after fermentation.

Antiaging and Whitening Activities of Ethanol Extract of Yuza (Citrus junos SIEB ex TANAKA) By-product (유자 부산물 에탄올 추출물의 항노화 및 미백효과)

  • Yuza (Citrus junos Sieb ex TANAKA) is a citrus fruit that is cultivated in northeast Asia. Citron is known for containing abundant antioxidants such as vitamin C, flavonoids, for example hesperidin and hesperetin, and terpenoids such as limononin. When mature citron is processed for tea or other beverage food products in Korea, massive amounts of seeds and pericarp are remained as waste. This study aimed to exploit the processed remnant of Citron for developing functional cosmetic applications. Ethanol extracts of Yuza seed and pericarp did not show significant radical scavenging activities measured by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. But they contained significantly high phenolic compounds. Cultured human dermal fibroblasts and HaCaT keratinocytes were irradiated with 25 mJ UVB and the citron extracts were added to the medium of each culture. Cellular damages caused by UVB irradiation were prevented by the addition of the Yuza extract. In addition, the reduction of the enhanced MMP-1 expression after irradiation of UVB in human dermal fibroblasts was observed. Also the increased level of pro-inflammtory TNF-$\alpha$ in the UVB irradiated HaCaT cells was decreased. The collagen expression was enhanced by the extract. Yuza extract markedly inhibited melanin production from $\alpha$-MSH treated B16F1 melanoma cells. Melanin assay, tyrosinase zymography results indicated that Yuza extract had strong depigmenting activity. In conclusion, Yuza ethanol extracts have good anti-photoaging and strong anti-melanogenic efficacies.
