To investigate the pharmacological activity of chaga mushroom (Inonotus obliquus) on extraction conditions, chaga was extracted using water (reflux at $50^{\circ}C$, decoction over $90^{\circ}C$, pressure at $121^{\circ}C$) or ethanol (reflux at 50, 70, or $90^{\circ}C$). When water extract was further fractionated into crude polysaccharide (IO-CP), yields of IO-CP (4.8~16.8%) were higher than those of ethanolic extracts (IO-E, 1.9~2.7%) at increased temperature. For antioxidant activity, crude polysaccharide (IO-CP-121) obtained by pressurized extraction showed the highest polyphenolic and flavonoid contents (35.10 mg TAE/g and 18.48 mg QE/g, respectively) as well as DPPH and ABTS free radical scavenging activities (26.08 and 27.99 mg AEAC/100 mg, respectively). Meanwhile, IO-CP-D (decoction) and IO-CP-50 (reflux) had more potent mitogenic effects (2.10- and 1.95-fold of saline control at 100 ${\mu}g/mL$) as well as intestinal immune system modulating activities (6.30- and 5.74-fold) compared to IO-CP-121, whereas ethanolic extracts showed no activity. Although no IO-CP showed cytotoxicity against RAW 264.7 cells at 0.1 mg/mL, IO-CP-121 significantly inhibited TNF-${\alpha}$ and NO production as pro-inflammatory factors in LPS-stimulated RAW 264.7 cells (29.2 and 63.5%, respectively). Ethanolic extracts also showed no cytotoxicity at 0.1 mg/mL, whereas inhibition of TNF-${\alpha}$ and NO production was significantly low compared to that of IO-CP-121. In addition, active IO-CP-D was further fractionated into an unadsorbed (IO-CP-I) and seven adsorbed fractions (IO-CP-II~VIII) by DEAE-Sepharose CL-6B column chromatography in order to isolate immunostimulating polysaccharide. IO-CP-II showed the most potent mitogenic effect and macrophage stimulating activity (4.51- and 1.64-fold, respectively). IO-CP-II mainly contained neutral sugars (61.86%) in addition to a small amount of uronic acid (2.96%), and component sugar analysis showed that IO-CP-II consisted mainly of Glc, Gal, and Man (molar ratio of 1.00:0.55:0.31). Therefore, extraction conditions affect the physiological activity of chaga, and immunostimulating polysaccharide fractionated from chaga by decoction is composed mainly of neutral sugars.
An optimum seeding date for carrots growing in Jeju was determined. In early growth stage, the number of leaves was more in Hyang-yang #2 cultivar (HY2) than that in the others, meanwhile that of Hukjun 5 chon (H5) and Shinhukjun 5 chon (S5) was higher in the latter half of the growing season. The weight of leaves was greater in HY2 until 2 months after seeding, but thereafter drastically increased and resulted significant difference at harvest in S5. The length of roots was greater in H5 and S5 than in HY2 when seeded on July 17 and measured after September 13, but it was similar in all cultivars when seeded on August 1 and later. The diameter of the roots was greatest in HY2, regardless of seeding dates, meanwhile no difference was found among other cultivars. The weight of roots was greatest in HY2 when seeded on July 17, August 1 or later, meanwhile it was greater in H5 after November, when seeded on August 15. The percentage of roots cracked was 20, 15, and 10% respectively for HY2, S5 and H5. The percentage of roots branched was 0.3% in HY2. The percentage of roots cracked or branched was much higher when seeded on August 1 than on July 17, and was high (40%) in HY2 and S5, compared to H5 when seeded on August 15. The content of sucrose, glucose and fructose increased until 110 days after seeding in all cultivars, but fructose and glucose contents decreased in about 135 days after seeding, but sucrose contents increased continuously even after 135 days. Sucrose content increased and fructose and glucose contents decreased in S5, but this trend was reversed after late November. Sugar contents was different among the cultivars seeded on August 15, while sucrose content decreased and glucose and fructose contents increased with time. The content of ${\beta}$-carotene was more than 12,000 IU in all cultivars, but decreased in 170 days after seeded on July 17. The content was highest in HY2 and followed by S5 and H5 in order. It was not different among the cultivars in 170 days after seeding on August 1, but was higher in S5 than the other cultivars in 170 days after seeded on late and decreased with seeding date.

Direct conversion of starchy materials to single cell protein of Sporobolomyces holsaticus FRI Y-5 was investigated. Effect of yeast extract concentration on its cell growth showed that it could utilize more of starch in the medium containing 2.5 g/l of yeast extract. In case of jar fermentor culture, the specific growth rate and cell yield of Sp. holsaticus on soluble starch were calculated to be $0.14\;hr^{-1}$ and 0.425, respectively and its maximum cell concentration was 13.4 g/l. After 80 hr of incubation time, 45.96% of starch was consumed and 45.1% of relative blue value was decreased. Reducing sugars in the starch medium seemed to increase from 4.06 g/l to 6.08 g/l and then to decrease. During fermentor culture, pH of medium was almost not changed in the range of $pH\;7.0{\pm}0.5$. The optimal temperature and pH of Sp. holsaticus amylase activity were $40^{\circ}C$ and pH 7.5, respectively. It was shown from the effect of Tapioca starch concentration on the cell growth that the optimal concentration of Tapioca starch for Sp. holsaticus was lower than that of soluble starch. FRI Y-5 cells settled much slower than Sp. holsaticus IFO 1032 cells and the viscosity vs cell concentration relationship was related to be linear.

Whole soybean cereal was fermented with four Aspergillus strains in pilot meju fermentation system. The pH range of the product was 7.40~7.98, the contents of reducing sugar and amino-nitrogen were 0.04~2.78%, 178~309 mg%, respectively and that of free fatty acid ranged 2.67~5.05%. The components of the amino acid, organic acid, free sugars and fatty acid showed distinctive patterns among four groups of fermented soybean cereals. Amylase activity and carbohydrate degradation rate of A. usami was higher than other strains. But protease and protein degradation rate, lipase and lipid degradation rate were similar in four strains. The odor concentrates of soybean cereals fermented with Aspergillus strains were different from Bacillus strains. Especially, pyrazine components, the main and common flavor chemicals in Bacillus strains, were not determined in this study and Aspergillus specific components were 9-methyl-acridine, dl-limonene and 2,3-butanediol. Soybean paste, made from A. oryzae fermented soybean cereal, showed excellent sensory evaluation.
When a low-temperature treatment was given to a small sweet pepper variety Zairaisisi, the seed browning effect appeared soon. This change attracted the studies to determine and discuss the browning metabolites, polyphenolic compounds, and changes in their between-step components. (1) Chlorogenic acids were found as a polyphenolic compound in seed, whereas no flavanol-type polyphenol was observed. (2) There was sharp increase in total polyphenol content and chlorogenic acid with a low-temperature treatment. The contents of these substrates dropped below that of room-temperature treatment after the browning effect took place. (3) A marked increase in between-step metabolites phenylalanine, tyrosine, shikimic acid contents, and thus assumed activated shikimate pathway in this process. (4) It was suggested by determining the effect of specific metabolic inhibition and respiratory inhibitor administrations on enzymes that active biosynthesis of polyphenolic compounds takes place in shikimate pathway with combination of phosphoenolpyruvate and erythrose-4-phosphate connected to TCA cycle jaming after an active EMP pathway was gone through with sugars in pepper seeds at a low-temperature. (5) It was also suggested from the observation of increased K ion flow-out in pepper seeds with a low-temperature treatment that there is an abnormality in the plasma membrance.
To determine contents of general constituents of Ganoderma lucidum in Korea, the dried carpophores were analyzed. The contents of water, ash, crude lipid, crude protein and crude cellulose were 14.6, 2.0, 3.3, 23.6, and 59.0%, respectively. Among reducing sugars, maltose was the most abundant. Seventeen free amino acids were detected, showing alanine the highest value. The pH of hot water extract was 4.1-4.2. The spores of Ganoderma lucidum was flat and ovoidal long. Their size was $6.3-7.1{\times}3.5-4.3{\times}2.0-2.5\;{\mu}m$ long. To examine effects of life span against sarcoma-180 cells, Fractions A, B and C, were obtained from the extract of Ganoderma lucidum. The survival rates of Fractions A, B and C were 131.7, 162.5, and 141.7 %, respectively. In addition, to examine effects of Fraction B on cell-mediated immunity, delayed type hypersensitivity (=DTH) test was conducted. It restored the suppressed DTH in the sarcoma-180 bearing group up to 66.7%.
In the previous study, we have reported the effect of lowering water activity of sodium chloride, sugars and polyols. In this study, water activity($a_w$)of various model Preparations of Kamaboko was measured and ability of lowering $a_w$ of starch, glycine and sodium lactate was discussed. In addition, the aw measurements were also compared to the predicted values of au which were derived from the equation of Raoult's law and the linear slope method by Sloan ana Labuza. The effect of starch on lowering $a_w$ was very low, and glycine was more effective than starch. Sodium lactate was shown to be the most effective among humectants examined. When it was added by $7.5\%$, the $a_w$ of model Kamaboko A was reduced to 0.93-0.94. Generally, prediction by the equation of Raoult's law turned out to be useful for determining $a_w$ lowering effect of humectants, excepting sodium chloride and sodium lactate. The equation of regression derived from the least squares could be applied to Predicting au in the model Kamaboko.
The nutritional compositions of different parts (roots, leaves, and seeds) of two lotus species (Nelumbo nucifera Gaertner), Muan's white-flowered and Naju's pink-flowered, are as follows; crude protein content in the seed was four times higher and the carbohydrate content was three times higher than those in the root and leaf. Comparing between the species, the crude protein content of the white lotus was the higher than that of the pink lotus, but the carbohydrate content was comparatively lower. The potassium content of the minerals in all samples was much higher than those in others. Additionally, the iron content of the root was much higher than those in the leaf and seed. In both species, the major free sugar in the root was sucrose; the main sugars in the leaf were fructose and alucose, and those in the seed were stachyose, raffinose, and sucrose. Among amino acids, glutamic acid showed the highest level in the leaf and seed, while aspartic acid was the highest in the root.
Fifty endophytic bacteria, which produced slime around the colonies, were isolated from Pueraria roots. In particular, HDN-14, TDG-3, and TNB-3 strains, which appeared to be high viscosity producers, were selected. These strains produced high levels of polysaccharides in Puerara root medium extract. The purified polysaccharide was digested with 1N HCI and analyzed by HPLC, with glucose ($45.6{\sim}63.1%$), maltose ($14.6{\sim}23.7%$), and fructose ($17.4{\sim}23.7%$) detected as constitutive sugars. When determined by the homology relationship of the 16S rDNA sequence with the relative taxa, the HDN-14 and TNB-3 strains were closely ($99.06{\sim}99.32%$) related to the Pseudomonas $koreensis^T$ and Pseudomonas $jessenii^T$, while TDG-3 were closely ($99.48{\sim}99.74%$) related to Pseudomonas $plecoglossicida^T$, Pseudomonas $mosselii^T$, and Pseudomonas $monteilii^T$. The major cellular Pseudomonas acids are $3OH-C_{10:0}$, $2OH-C_{12:0}$, $3OH-C_{12:0}$, and $3OH-C_{12:1}$, with these strains being further differentiated in species belonging to the genus Pseudomonas.
This study was attempted to analyze amino acids, fatty acids, vitamins and minerals, reducing sugars and dietary fiber of Artemisia mongolica Fischer to establish the value as an useful vegetable. Total free amino acids of mugworts plucked in spring and autumn were 1048.1 mg% and 2187.1 mg% respectively. The contents of linoleic and linolenic acids were 76.7% in spring mugworts and 69.5% in autumn ones. Mugworts gathered in spring and autumn showed 0.16 mg% and 0.15 mg% for thiamin, 1.81 mg% and 1.74 mg% for riboflavin, 5.34 mg% and 5.36 mg% for niacin and 26.12 mg% and 25.21 mg% for ascorbic acid. Mineral contents of spring and autumn mugworts were 17.8 mg% and 20.5 mg% for iron, 0.3 ppm and 0.4 ppm for selenium. The contents of reducing sugar were fructose 9.75 mg% for spring, 9.78 mg% for autumn and the contents of dietary fiber were 37.57% for spring and 38.41% for autumn.
